Claims (1)
- 【特許請求の範囲】 1.以下の工程を含む、全血試料から少数細胞を濃縮する方法: a)1.119g/mlの密度のFicollの存在下で、500g以下で全 血の密度勾配遠心を行い; b)密度勾配遠心から求める少数細胞集団を回収し、及び適用電場の存在下で 血液試料中のそれぞれの細胞成分の移動が緩衝液の向流により妨害されている、 比重ー勾配をもつ、向流安定化荷電ー流動分離装置に血液試料を通し、それによ り他の細胞性の血液成分から少数細胞集団の濃縮を引き起こし; c)荷電ー流動分離装置から溶出される濃縮された少数細胞集団が存在する、 少なくとも一画分を回収し; d)濃縮された少数細胞集団から、不要な血液細胞をネガティブに選択し; e)解析に供するため少数細胞集団を採取する。 2.荷電ー流動分離装置から得られる有核血液細胞である分離された細胞集団 を得、そして分離された細胞集団の試料を白血球を捕捉する親和性結合材に通す 工程をさらに含む、請求項1の方法。 3.荷電ー流動分離装置から得られる有核血液細胞である分離された細胞集団 を得、そして分離された細胞集団の試料を白血球を捕捉する吸着ーろ過結合材に 通すという工程をさらに含む、請求項1の方法。 4.ネガティブ選択工程が、成熟赤血球を選択的に溶解するために十分な時間 行われるアンモニウム溶解の工程をさらに含む、請求項1の方法。 5.ネガティブ選択工程が、リンパ性細胞及び骨髄性細胞を選択的に溶解する ためのアンモニウム溶解の工程の後に補体溶解の工程をさらに含む、請求項4の 方法。
