Claims (1)
- 【特許請求の範囲】 1.(a)第1図の推定アミノ酸配列を有するポリペプチド、または該ポリペ プチドのフラグメント、アナログもしくは誘導体をコードするポリヌクレオチド ; (b)ATCC寄託番号第75927号に含まれるcDNAによりコードされる アミノ酸配列を有するポリペプチド、または該ポリペプチドのフラグメント、ア ナログもしくは誘導体をコードするポリヌクレオチド; よりなる群から選択される、単離されたポリヌクレオチド。 2.ポリヌクレオチドがDNAである、請求項1に記載のポリヌクレオチド。 3.ポリヌクレオチドがRNAである、請求項1に記載のポリヌクレオチド。 4.ポリヌクレオチドがゲノムDNAである、請求項1に記載のポリヌクレオ チド。 5.該ポリヌクレオチドが、第1図の推定アミノ酸配列を有するポリペプチド をコードする、請求項2に記載のポリヌクレオチド。 6.該ポリヌクレオチドが、ATCC寄託番号第75927号のcDNAによ りコードされるポリペプチドをコードする、請求項2に記載のポリヌクレオチド 。 7.第1図に示すポリペプチドのコード配列を有する、請求項1に記載のポリ ヌクレオチド。 8.ATCC寄託番号第75927号として寄託されたポリペプチドのコード 配列を有する、請求項2に記載のポリヌクレオチド。 9.請求項2に記載のDNAを含むベクター。 10.請求項9に記載のベクターで遺伝的に操作された宿主細胞。 11.ポリペプチドを製造する方法であって、該DNAによりコードされるポ リペプチドを請求項10に記載の宿主細胞から発現させることを含んでなる方法 。 12.ポリペプチドを発現させることができる細胞を製造する方法であって、 細胞を請求項9に記載のベクターで遺伝的に操作することを含んでなる方法。 13.請求項2に記載のDNAにハイブリダイズすることが可能であって、T NF−γ活性を有するポリペプチドをコードする、単離されたDNA。 14.(i)第1図の推定アミノ酸配列を有するポリペプチド、並びにそのフ ラグメント、アナログおよび誘導体;および (ii)ATCC寄託番号第75927号のcDNAによりコードされるポリペプ チド、並びに該ポリペプチドのフラグメント、アナログおよび誘導体; よりなる群から選択されるポリペプチド。 15.ポリペプチドが第1図の推定アミノ酸配列を有する、請求項14に記載 のポリペプチド。 16.請求項14に記載のポリペプチドに対する抗体。 17.請求項14に記載のポリペプチドのアゴニスト。 18.請求項14に記載のポリペプチドに対するアンタゴニスト。 19.TNF−γを必要とする患者の治療方法であって、治療上有効な量の、 請求項14に記載のポリペプチドを患者に投与することを含んでなる方法。 20.TNF−γを必要とする患者の治療方法であって、治療上有効な量の、 請求項17に記載のアゴニストを患者に投与することを含んでなる方法。 21.TNF−γを阻害する必要がある患者の治療方法であって、治療上有効 な量の、請求項18に記載のアンタゴニストを患者に投与することを含んでなる 方法。 22.該ポリペプチドをコードするDNAを患者に与えて、該ポリペプチドを インビボにおいて発現させることにより、治療上有効な量の該ポリペプチドを投 与する、請求項19に記載の方法。 23.請求項14に記載のポリペプチドに対するアゴニストおよびアンタゴニ ストを同定する方法であって、 TNF−γの存在下または不存在下、内皮細胞、コンカビリン(Concavilin) −A、スクリーニングすべき化合物、[3H]チミジンを選択に混合し; 内皮細胞による[3H]チミジンの取り込みを測定し;および 該化合物が[3H]チミジンの取り込みを高めたか、またはブロックしたかどう かを測定する; ことを含んでなる方法。 24.患者における腫瘍細胞増殖を阻害する方法であって、場合により、薬学 上許容され得る担体の存在下、治療上有効な量のTNF−γタンパク質を患者に 投与することを含んでなる方法。 25.腫瘍または腫瘍に対する感受率を診断する方法であって、宿主から得ら れた試料中、変異型の、TNF−γをコードする核酸配列を検出することを含ん でなる方法。
