Claims (1)
- 【特許請求の範囲】 1.α(1,3)ガラクトシルトランスフェラーゼの正常な発現が少なくとも組 織の型の一つの器官において妨げられる一つの遺伝子導入ブタ。 2.請求の範囲第1項記載の遺伝子導入ブタであり、前記ブタα(1,3)ガ ラクトシルトランスフェラーゼをコード化するmRNAと結合しその翻訳を妨げ る一つの核酸配列を生産することを特徴とする遺伝子導入ブタ。 3.請求の範囲第2項記載の遺伝子導入ブタであり、そのためにプロモーター に操作可能に結合される機能的酵素を妨げあるいは発現するの必要なα(1,3 )ガラクトシルトランスフェラーゼコード化領域のその部分にアンチセンスRN Aをコード化するDNAよりなる一つの構築物を含むようにブタのゲノムが修飾 されたことを特徴とする遺伝子導入ブタ。 4.請求の範囲第3項記載の遺伝子導入ブタであって、ここでプロモーターが 強力で非組織特異的構築あるいは調節可能プロモーターであることを特徴とする 遺伝子導入ブタ。 5.請求の範囲第3項記載の遺伝子導入ブタであって、ここでプロモーターが 強力で組織特異的構築あるいは調節可能プロモーターであることを特徴とする遺 伝導入ブタ。 6.リボザイムを生産するように作られたことを特徴とする請求の範囲第1項 記載の遺伝子導入ブタ。 7.請求の範囲第6項記載の遺伝子導入ブタであり、それがプロモーターに操 作可能に結合されるα(1,3)ガラクトシルトランスフェラーゼを不活性化す るのに必要な少なくともリ ボザイムのその部分をコード化するDNAよりなる構築物を含むように修飾され たことを特徴とする遺伝子導入ブタ。 8.請求の範囲第7項記載の遺伝子導入ブタであって、ここでプロモーターが 強力で被組織特異的構成あるいは調節可能プロモーターであることを特徴とする 遺伝子導入ブタ。 9.請求の範囲第7項記載の遺伝子導入ブタであって、ここでプロモーターが 強力で組織特異的構成あるいは調節可能プロモーターであることを特徴とする遺 伝導入ブタ。 10.ブタ卵母細胞から成長するα(1,3)ガラクトシルトランスフェラー ゼフェらーゼ陰性ブタであり、その前核物質が除去されα(1,3)ガラクトシ ルトランスフェラーゼの発現に陰性である多能性ブタ胚幹細胞を導入されたα( 1,3)ガラクトシルトランスフェラーゼ陰性ブタ。 11.配列識別番号1および13から24よりなるグループから選択される配 列を持つ一つの核酸。 12.配列識別番号1で説明された配列を持つことを特徴とする請求項第11 項記載の核酸。 13.請求の範囲第11項記載の核酸であって、ここで配列識別番号が配列識 別番号13から15よりなるグループから選択されることを特徴とする核酸。 14.請求の範囲第11項記載の核酸であって、ここで配列識別番号が配列識 別番号16から21よりなるグループから選択されることを特徴とする核酸。 15.請求の範囲第11項記載の核酸であって、ここで配列識別番号が配列識 別番号22から24よりなるグループから選 択されることを特徴とする核酸。 16.請求の範囲第1項記載の遺伝子導入ブタであって、ここでβ(1,3) ガラクトシルトランスフェラーゼ遺伝子の正常な発現が同一遺伝子型DNA含有 標的化ベクターの前記ブタへの導入により妨げられることを特徴とする遺伝子導 入ブタ。 17.ブタ接合体から成長する請求の範囲第16項記載のα(1,3)ガラク トシルトランスフェラーゼ陰性ブタであり、それはα(1,3)ガラクトシルト ランスフェラーゼ染色体遺伝子座において同質遺伝子型DNA標的化ベクターを 用いて相同性組換えにより修飾されたことを特徴とするα(1,3)ガラクトシ ルトランスフェラーゼ陰性ブタ。 18.請求の範囲第17項記載のα(1,3)ガラクトシルトランスフェラー ゼ陰性ブタであって、ここで同一遺伝子型DNA標的化ベクターが少なくとも約 6キロベースの同質遺伝子型DNAおよび無プロモーター選択マーカー遺伝子よ りなることを特徴とするα(1,3)ガラクトシルトランスフェラーゼ陰性ブタ 。
