Claims (1)
- 【特許請求の範囲】 1.アルファ9ニコチン性アセチルコリン受容体(nAChR)サブユニット をコードする単離された核酸。 2.核酸はDNAを含む、請求の範囲第1項に記載の単離された核酸。 3.DNAはcDNAである、請求の範囲第2項に記載のDNA。 4.DNAは配列番号2に記載のアミノ酸配列をコードする、請求の範囲第2 項に記載のDNA。 5.DNAは、高厳密性条件下で配列番号1に記載のヌクレオチド配列とハイ ブリダイズする、請求の範囲第2項に記載のDNA。 6.DNAのヌクレオチド配列は、配列番号1に記載のヌクレオチド配列と実 質的に同じである、請求の範囲第2項に記載のDNA。 7.請求の範囲第2項に記載のDNAを含むベクター。 8.請求の範囲第7項に記載のベクターを含有する宿主細胞。 9.細胞は、少なくとも1つのアルファ9nAChRサブユニットを含む機能 性nACh受容体を発現する、請求の範囲第8項に記載の宿主細胞。 10.受容体はホモマーである、請求の範囲第9項に記載の宿主細胞。 11.受容体はヘテロマーである、請求の範囲第9項に記載の宿主細胞。 12.請求の範囲第2項に記載の少なくとも14個の連続ヌクレオチドを含む 核酸プローブ。 13.請求の範囲第2項に記載のDNAに相補的な単離されたmRNA。 14.請求の範囲第13項に記載のmRNAに特異的に結合しその翻訳を調節 する、アンチセンスオリゴヌクレオチド。 15.単離されたアルファ9ニコチン性アセチルコリン受容体サブユニット、 およびその生物活性を有する断片。 16.サブユニットは、配列番号2に記載のアミノ酸配列と同じアミノ酸配列 を有する、請求の範囲第15項に記載の単離されたアルファ9ニコチン性アセチ ルコリン受容体サブユニット。 17.サブユニットは、配列番号2に記載のアミノ酸配列と実質的に同じアミ ノ酸配列を有する、請求の範囲第15項に記載の単離されたアルファ9ニコチン 性アセチルコリン受容体サブユニット。 18.宿主細胞中で組換え発現される機能性ニコチン性アセチルコリン受容体 であって、 (i)下垂体、嗅上皮、蝸牛および舌で発現され、 (ii)アセチルコリンにより活性化されるが、ニコチンとムスカリンにより阻 止され、そして (iii)少なくとも1つのアルファ9アセチルコリン受容体サブユニットを含 む、上記受容体。 19.アルファ9ニコチン性アセチルコリン受容体サブユニットは、配列番号 2に記載のアミノ酸配列と同じアミノ酸配列を有する、請求の範囲第18項に記 載の受容体。 20.アルファ9ニコチン性アセチルコリン受容体サブユニットは、配列番号 2に記載のアミノ酸配列と実質的に同じアミノ酸配列を有する、請求の範囲第1 8項に記載の受容体。 21.アルファ9ニコチン性アセチルコリン受容体サブユニットに特異的に結 合する抗体。 22.抗体はモノクローナル抗体である、請求の範囲第21項に記載の抗体。 23.抗体はポリクローナル抗体である、請求の範囲第21項に記載の抗体。 24.少なくとも1つのアルファ9サブユニットを含むニコチン性アセチルコ リン受容体の発現を調節するのに有効な量の請求の範囲第14項に記載のオリゴ ヌクレオチド、および 細胞膜を通過できる許容される疎水性担体、を含む組成物。 25.少なくとも1つのアルファ9サブユニットを含むニコチン性アセチルコ リン受容体への、天然に存在するリガンドの結合を阻止するのに有効な量の請求 の範囲第21項に記載の抗体、および許容される担体、を含む組成物。 26.アルファ9ニコチン性アセチルコリン受容体サブユニットをコードする DNAを発現するトランスジェニック非ヒト哺乳動物。 27.発現される受容体が野生型のアルファ9ニコチン性アセチルコリン受容 体サブユニットを含有しないように、受容体をコードするDNAが突然変異され ている、請求の範囲第26項に記載のトランスジェニック非ヒト哺乳動物。 28.トランスジェニック非ヒト哺乳動物はマウスである、請求の範囲第26 項に記載のトランスジェニック非ヒト哺乳動物。 29.アルファ9ニコチン性アセチルコリン受容体サブユニットをコードする 核酸の同定方法であって、 核酸の集団を含有する試料に請求の範囲第12項に記載のプローブを接触させ( ここで、接触は高厳密性ハイブリダイゼーション条件下で行われる)、そして該 プローブにハイブリダイズする核酸を同定することよりなる、上記方法。 30.請求の範囲第9項に記載の細胞に試験化合物を接触させ、そこに結合す る化合物を同定することよりなる、アルファ9ニコチン性アセチルコリン受容体 サブユニットに結合する化合物の同定方法。 31.少なくとも1つのアルファ9サブユニットを含むニコチン性アセチルコ リン受容体の活性を調節する化合物の同定のためのバイオアッセイであって、請 求の範囲第9項に記載の細胞に、少なくとも1つの化合物(この化合物が受容体 のイオンチャネルを調節する能力は未知である)に接触させ、その後にイオンチ ャネル活性の変化について細胞をモニタリングすることよりなる、上記バイオア ッセイ。 32.細胞が少なくとも1つの可能性のあるアゴニストと接触する、請求の範 囲第31項に記載の方法。 33.細胞が、一定量のアゴニストと増加する量の少なくとも1つの可能性の あるアンタゴニストと接触する、請求の範囲第31項に記載の方法。 34.アルファ9ニコチン性アセチルコリン受容体サブユニット遺伝子の制御 領域を調節する化合物の同定方法であって、 アルファ9遺伝子の制御領域の少なくとも一部(制御遺伝子の該部分は受容体遺 伝子に機能的に結合している)を含む核酸構築体で哺乳動物を形質転換し、形質 転換した細胞に、少なくとも1つの化合物(ここで、この化合物がアルファ9受 容体サブユニット遺伝子の制御領域を調節する能力は未知である)を接触させ、 そして レポーター遺伝子の発現について細胞をモニタリングすることよりなる、上記バ イオアッセイ。
