Description
Claims (1)
- 【特許請求の範囲】 1.生体高分子を分離する方法であって、 a)生体高分子と結合可能な固定相微粒子がその上流表面上に位置している複 合ろ過媒体、溶質として少くとも1つの生体高分子を含む溶液混合物を含有する 貯槽並びにポンプ及び閉鎖ループ組立体を形成するための関連する配管を含む閉 鎖型フィルターカートリッジを含有してなる分離系を供給する段階、 b)生体高分子−固定相微粒子生成物を形成させるために、前記少くとも1つ の生体高分子が固定相微粒子と結合するように、該溶液混合物をフィルターカー トリッジにポンプで繰返し循環させる段階、 c)任意に、生体高分子−固定相微粒子生成物を液体で洗浄して、固定相微粒 子に結合していない、所望しない生体高分子及び他の溶質を除去する段階、及び d)任意に、生体高分子を遊離させるために、溶離溶液を生体高分子−固定相 微粒子生成物結合相互作用を逆行可能な閉鎖ループ組立体にポンプで流す段階、 を含んでなる前記方法。 2.前記固定相微粒子が吸着、イオン交換、疎水的結合及び親和性結合による 結合可能な微粒子の群から選択される請求項1に記載の方法。 3.前記溶液混合物を少くとも0.01cm/分の流動速度、最大0.25MPa のフィル ターカートリッジ圧で、フィルターカートリッジにポンプで流す、請求項1また は2に記載の方法。 4.前記生体高分子が、タンパク質、炭水化物、脂質、及び核酸よりなる群か ら選択される請求項1〜3のいずれか1項に記載の方 法。 5.前記複合ろ過媒体が、織物または不織布の多孔性物質である請求項1〜4 のいずれか1項に記載の方法。 6.前記ろ過媒体物質が、セルロース、ガラス、ポリオレフィン、ポリエステ ル、ポリアミド及びセラミックからなる群から選択される請求項1〜5のいずれ か1項に記載の方法。 7.前記ろ過媒体物質がポリプロピレンである請求項1〜6のいずれか1項に 記載の方法。 8.前記吸着固定相微粒子が、ヒドロキシアパタイト、アルミナ、シリカゲル 及びジルコニアからなる群から選択される請求項2〜6のいずれか1項に記載の 方法。 9.前記微粒子がアフィニティクロマトグラフィー固定相微粒子である請求項 1〜8のいずれか1項に記載の方法。 10.前記生体高分子を含有する溶液混合物が、発酵培地、細胞溶解物及び体液 からなる群から選択される請求項1〜9のいずれか1項に記載の方法。 11.請求項1〜10項のいずれか1項に記載の方法により供給される、生体高分 子−微粒子生成物。
