Description
「生体高分子」は、少なくとも500の分子量を有する細胞の構成物または産物、例えばタンパク質、炭水化物、脂質または核酸を意味し;
「濾過層」は、シングルシートを提供するために組み合わせることができる一層以上の個々の層を含むシート様織物または不織物多孔性材料を意味し;

「フィルターカートリッジ」は固定相微粒子が充填され得る濾過デバイスを意味し;
「フィルターカートリッジハウジング」は、フィルターカートリッジのための支持構造を意味し;
「マクロ多孔性」は、乾燥状態においても永久多孔性構造を有する粒子を指す。溶媒と接触時に樹脂は膨張し得るが、多孔性構造を通して粒子の内部への接近を可能にするために膨張は必要でない。
「ゲル型樹脂」または「ゲル」は乾燥状態において永久多孔性構造を有さないが、適切な溶媒によって膨張して粒子の内部への接近を可能にする。マクロ多孔性およびゲル粒子は、Sherrington、Chem.Commun.、2275−2286(1998)にさらに記載される。マクロ多孔性イオン交換樹脂は、典型的に20〜3000オングストロームの径の細孔を有する(すなわち細孔径は、窒素吸着を使用して様々な相対圧力で低温条件下で、または水銀圧入ポロシメトリーによって特徴づけられる)。
「分離フィルターアセンブリ」は、その上流面上に固定相微粒子が位置する複合フィルター媒体を含むフィルターカートリッジ、好ましくはデッドエンドフィルターカートリッジを含有するハウジングを意味し;
「分離システム」は、貯蔵器中に含有された少なくとも一種の生体高分子溶質を含む溶液混合物、分離フィルターアセンブリまたはクロマトグラフィカラム、ポンプおよびフィルターカートリッジと貯蔵器とポンプと関連するチューブを意味し;

「流動速度」は、フィルタリングエレメント中を通過する液体流の速度を意味し、そして濾過層の断面表面積によって分割される流速と等しい。このように記載されて、液体流の流れを特徴づけることができ、そしてこれは濾過層のサイズに依存しない。流動速度はフィルターを横切る圧力低下にも関与し、すなわち、流動速度の増加は一般的に系圧の増加を意味する。市販フィルターカートリッジの適用において、液体流の最大量を処理する最小サイズのフィルターを提供することは非常に所望である。従って、流速を増加させることによって流動速度を増加させることは望ましい;

2)(任意に再循環を伴って)固定相微粒子への選択された生体高分子の結合を達成するためにフィルターカートリッジアセンブリを通して溶液混合物をポンピングする工程とであって、フィルターエレメントを通してのポンピングが、最高0.34MPa、好ましくは最高0.25MPa)のフィルターカートリッジ圧力において少なくとも0.01cm/分、好ましくは少なくとも0.10cm/分、そしてより好ましくは少なくとも0.30cm/分の流動速度で実行される工程と;
3)任意に、開ループまたはワンパス手順において、選択された吸着、イオン交換、疎水性または親和性結合相互作用によって、固定相微粒子に結合されない望ましくない生体高分子および他の溶質を除去するために適切な液体によって生体高分子:固定相微粒子生成物を洗浄する工程と、
4)任意に、分離および精製された生体高分子を遊離するために生体高分子:固定相微粒子結合相互作用を逆転する脱結合溶液の好ましくは(最初の溶液混合物の容積と比較して)減少された容積をポンピングする工程と、
を含む生体高分子の分離または精製の方法を提供する。
i)表面濾過−この手順は、微粒子を含有する液体流が、大きさを設定された穴または孔の分布を有するフィルターエレメントに直面し、そして微粒子に濾過層を通して湾曲した経路を提供するものである。従来技術においては、微粒子は濾過層内での吸着および/またはエントラップメントによって大部分が除去された。表面濾過は、しばしば系に適用される粗いかまたは第1の濾過手順であり、数百マイクロメートル(直径で最大の寸法)から約1マイクロメートルまでの粒度を有する微粒子を除去するように設計されたものであって、不明確な細孔径のため微粒子の不完全な除去の問題があり、そしてフィルターが充填されるとフィルターカートリッジ圧は一定に迅速に増加する。
ii)表面(ケーキ)濾過−この手順は本発明において好ましく、液体流の処理において表面濾過に続いてしばしば行なわれる。従来技術において、これは明確に画定された細孔径を有するガラスまたはポリマーマイクロファイバーの多層を使用して一般的に実行され、そして微粒子は一般的に濾過層内に浸透しないが、層の上流面上に閉じ込められたままである。約0.1マイクロメートルまでの粒度の微粒子が効率よく充填され得る。高い流動速度は容易に達成可能であり、そしてフィルターがほぼ充満されるまで、比較的多量の微粒子が比較的低い系圧において充填される。本発明において、液体流の多くの逆転によって濾過層の表面上で濾過された粒子を充填または再調整することは都合がよい;このような機会は深層フィルターで存在しない。
iii)膜(スクリーンまたは篩分け)濾過−この濾過メカニズムは、精密に画定された非常に小さい孔が存在することを除いて表面濾過と非常に類似しており、0.05マイクロメートル程度の小さい径の微粒子の充填が可能である。
i)ハウジングに含有される本発明の複合フィルター媒体を含むフィルターカートリッジと、少なくとも0.01cm/分の流動速度を送達可能であるポンプおよびフィルターカートリッジと貯蔵器とポンプと関連するチューブとを含むアセンブリを提供する工程と、
ii)分離フィルターアセンブリ中で循環を受けて生体高分子の分離が実行されるように、貯蔵器中の少なくとも一種の生体高分子溶質を含む生物学的溶液混合物を導入する工程と
を含む。
i)ハウジングに含有される本発明の複合フィルター媒体を含むフィルターカートリッジと、少なくとも0.01cm/分の流動速度を送達可能であるポンプおよびフィルターカートリッジと貯蔵器とポンプと関連するチューブとを含むアセンブリを提供する工程と、
ii)適切な溶媒中の微粒子のスラリーを提供する工程と、
iii)所望の量の固定相微粒子が充填されるまで、再生利用の様式でフィルターカートリッジを通してスラリーをポンピングする工程と
を含む「湿式」充填技術によって充填され得る。好ましくは、フィルターカートリッジ圧力は約0.15MPa未満、より好ましくは0.10MPa未満、そして最も好ましくは0.05MPa未満である。
リゾチームに対するカチオン交換容量
0.8×4センチメートルのポリプロピレン使い捨てクロマトグラフィカラム(カリフォルニア州、ヘラクレスのポリ−プレップ カラム、バイオ−ラド ラボラトリーズ(Poly−Prep Column,Bio−Rad Laboratories,Hercules,CA))を1mlのイオン交換樹脂で充填した。10mLの充填緩衝液、pH7.5の10mM MOPS(4−モルホリノプロパンスルホン酸)の溶液で洗浄することによって、カラム床を平衡化した。次いで、MOPS緩衝液中12mg/mLの濃度を有する30mLのタンパク質溶液(鶏卵白リゾチーム、純度約95%、シグマ ケミカル カンパニー(Sigma Chemical Co.))でカラム床を充填した。全ての緩衝液およびタンパク質溶液を脱イオン水中で調製した。いずれの非結合性リゾチームも30mLのMOPS緩衝液(10mLのフラクションで3回)によって洗浄除去した。最終的に、結合タンパク質をMOPS緩衝液中1M NaClの15mLで溶出した。
脱イオン水中のカチオン交換ビーズの50%v/vスラリーは、ビーズを水と混合し、300相対遠心力(rcf)で20分間遠心分離処理し、次いで総容積が充填ビーズ床の二倍であるように水の量を調節することによって調製された。スラリーを十分に混合し、ビーズ懸濁させ、次いでスラリーの400マイクロリットルの試料をピペットで5ミリリットルの0.45マイクロメートル酢酸セルロース セントレックス(Centrex)MF 遠心ミクロフィルター(ミネソタ州、エーガン(Eagan,MN)のVWRを通して入手可能であるシュライヒャー&シュル(Schleicher & Schuell))に加えた。水を5分間、3000rcfで遠心分離によって除去し、80mM塩化ナトリウムを含有する4mLの50mM酢酸ナトリウム、pH4.5と混合し、そして10分間3000rcfで再び遠心分離した。濾液をデカンテーションした。次いで同一酢酸塩緩衝液中のヒトIgGの4.5mL試料(約7mg/ml)を、ビーズを含有するフィルターに添加した。混合物を一晩回転させることによって混合し、次いで20分間3000rcfで遠心分離によって上澄をビーズから除去した。
ホリバ(Horiba)LA−910機器(カリフォルニア州、アービンのホリバ ラボラトリー インストルメンツ(Horiba Laboratory Instruments,Irvine,CA))を使用して、光散乱によって粒度を測定した。
コンピュータ制御された試験装置は、1cm×10cmガラスカラム、カラム末端固定具、ポンプ、圧力計およびリン酸緩衝塩水(PBS)を含有する貯蔵器に連結された適切なチューブからなった。カラムに粒子を充填して測定した。ポンプの電源を入れてカラム中の流動を開始し、そして典型的に2ml/分(約150cm/時)で開始した。圧力低下の安定を確実とするために、流動をこの速度で維持し(典型的に5〜15分)、次いで流動速度を2または4ml/分増加によって増加させ、再びカラムを横切って圧力低下を監視した。カラムが不具合になるまで、この手順を続けた。不具合とは、コンピュータが自動的に系をシャットダウンする時点である170psiを超える圧力として定義した。
ヒトIgG容量に関して試験される粒子をタンパク質Aで充填し、そしてフィルターを通してビーズスラリーをポンピングし、続いて緩衝液交換のため、PBS緩衝液をポンピングすることによってカートリッジフィルター(ポール フィリング マシン カプセル(Pall Filling Machine Capsule)カートリッジ、5マイクロメートル細孔径、300cm2の濾過層面積またはCUNO ベタピュア カプセル(CUNO Betapure Capsule)フィルターカートリッジ、2マイクロメートル細孔径、900cm2の濾過層面積のいずれか)上に充填した。緩衝液をカートリッジフィルターハウジングから排出した後、ヒトIgG溶液(10mM PBS pH7.2中1.0mg/mL、1,500mL総容積)を貯蔵器に充填し、フィルターカートリッジハウジング中にポンピングし、そして再循環のために貯蔵器に戻した。35psi(0.24MPa)未満の圧力低下で、流速は400mL/分であった。280nm波長におけるUV吸光度によるIgG濃度のオンライン監視によって決定されるIgG捕捉速度がゼロ付近に達するまで、溶液を4〜30分間再循環した。インレットチューブを通してフィルターハウジングから溶液を排出し、そしてフィルターカートリッジ中のビーズの洗浄、捕捉されたIgGの溶出および緩衝液交換によって再生した。次いで、カートリッジフィルターの中の再生ビーズは、溶液に残るIgGを捕捉するために使用された。このサイクルを5回繰り返し、そして溶液中の大部分のIgG(93〜99%)を捕捉した。各サイクルにおける捕捉性能および全捕捉速度を記録した。
米国特許第5,403,902号明細書に記載されるように、重量で35:65のAMPS/MBAコポリマーを逆相懸濁液重合によって調製した。メカニカルスターラー(撹拌速度450rpm)、窒素インレット、温度計、温度制御装置を有する加熱マントルおよび凝縮器を備えたフラスコに、ポリマー安定剤(0.28グラム)、トルエン(132mL)およびヘプタン(243mL)を添加した。ポリマー安定剤は重量で91.8:8.2のイソオクチルアクリレートおよび2−アクリルアミドイソブチラミドのコポリマーであった(ラスムッセン(Rasmussen)ら、Makromol.Chem.、Macromol.Symp.、54/55、535−550(1992)に記載の通りに調製した)。フラスコ中の非水溶液を撹拌しながら35℃まで加熱し、そして15分間窒素スパージングした。
重量で65:35のAMPS/MBAコポリマー(調製例2)および重量で40:60のAMPS/MBAコポリマー(調製例3)が得られる試薬レベルで、予備実施例1に記載の同一の逆相重合手順に従った。
調製例2に記載されるように、重量で65:35のAMPS/MBAコポリマーを逆相懸濁液重合によって調製した。リゾチームの平衡カチオン交換能力を測定し、そして160mg/mlであることがわかった。顕微鏡試験は、直径で約10〜200マイクロメートルの範囲の球形粒子を示した。圧力/流動特性を測定する試みによって、最低流速における加圧以上のカラムが得られた。これらの粒子の試料を分類し、約45〜110マイクロメートルの粒度範囲が提供された。この分類された試料の圧力/流動特性は2ml/分(150cm/時間)において20psi(約0.14MPa)の圧力低下を生じるが、3ml/分(約230cm/時間)において不具合を生じる(>170psi=1.17MPa)。

ビーズ:AMPS/MBA(65/35);水和床容積5ml
リゾチーム充填溶液:10mM MOPS pH=7.5中2mg/ml、1000ml
緩衝液:10mM MOPS pH=7.5
(ハウジングおよびチューブの)系容積:450ml
流動:圧力低下が<5psiで1120ml/分。
充填、排出および容積測定を3回行い、そして結果を平均することによって系容積を決定した。具体的なポンプ設定に関して、ストップウォッチおよび目盛付きシリンダーを使用して流速を測定した(ここでも3回実行の平均)。次いで、50mlの緩衝液でビーズのスラリーを製造することによって、ビーズをフィルター上で充填した。
調製例3に記載されるように、重量で40:60のAMPS/MBAコポリマーを逆相懸濁液重合によって調製した。形成されたビーズを分類して、約40〜110マイクロメートルの粒度範囲を提供した。リゾチームに対する平衡能力を測定すると、113mg/mlであった。この分類された試料の圧力/流動特性は10ml/分(760cm/時間)において50psi(0.34MPa)の安定した圧力低下を生じたが、次いでより高い流速において圧力低下のわずかな増加が生じ、最終的に>1000cm/時間で不具合を生じた。またこのビーズを実施例1に記載される系において評価し、そして結果を表1に示す。
調製例2に記載されるように、AMPS(36.4gの50%水溶液)、MBA(9.8g)、DI水(31.8ml)およびイソプロパノール(100ml)から、重量で65:35のAMPS/MBAコポリマーを逆相懸濁液重合によって調製した。リゾチームに対する平衡カチオン交換能力を測定し、そして25mg/mlであることがわかり、またIgGに対する平衡カチオン交換能力は7mg/mlであることがわかった。
比較例2の水相と同一配合物を有するモノリシック媒体を調製した。MBA(0.993g)、AMPSの50重量%水溶液(3.649g)、脱イオン水(2.88mL)およびイソプロパノール(10mL)を混合し、そしてガラス容器中で撹拌しながら穏やかに加熱した。混合物を完全に溶解した後、それをポリエチレン製ポーチ(約10cm×7cm×壁厚0.15mm)へ移し、そして水(0.3mL)中の過硫酸ナトリウム(0.0512g)の溶液をTMEDA(0.05mL)と一緒に添加した。ポーチを直ちにヒートシールし、次いで一晩室温でオービタルシェーカーで穏やかに振盪した。ポーチを切断して開き、そしてポリマーの塊をフィルターロートに移した。ここでこれを水、次いでアセトンで完全に洗浄し、そして一晩真空下で乾燥させた。乾燥試料を乳鉢および乳棒で簡単に粉砕した。粒度測定値によって非常に広範囲の分布が示され、粒子は1マイクロメートル〜700マイクロメートルの粒度範囲であった。
重合時間が2.5時間から5時間まで延長されたこと以外は、実施例1に記載されるように重量で65:35のAMPS/MBAコポリマーを調製した。この試料をエラトリエーションによって3つの粒度範囲に分類した。すなわち、小留分、中間留分および大留分であった。中間留分を廃棄し、そして小留分および大留分の平均粒度を決定したところ、37.6マイクロメートル(小ビース)および160.0マイクロメートル(大ビーズ)であった。次いで、実施例1に記載のカートリッジ系において、これらの2つの試料を評価した。リゾチーム捕捉の結果を表2に記載する。
CM−セファデックス(Sephadex)C50は、小規模タンパク質精製のために有用なイオン交換樹脂である。製造業者は45cm/時間の最大の流速を推奨しており、この樹脂が大規模なカラム使用のためには軟質であることを示す。40℃で一晩混合することによって、試料を10mMリン酸ナトリウム緩衝液中で水和した。沈殿した樹脂10mlを実施例1に記載の分離システムに充填し、そして1134ml/分の流速で、充填されたフィルターカプセルを通してウサギIgGの溶液(10mMリン酸塩中0.17mg/ml、pH7.2)を再循環することによってIgG吸着に関して評価した。時間経過に従って採取された試料のUV分析はIgGの急速な吸着を示し、6分で3.9mg/mlの樹脂が吸着し、そして25分以内で4.5mg/mlの平衡吸着が達成された。製造業者によると、有効容量(飽和容量)は7mg/mlであると規定されている。実験の間、流速の変化または圧力増加は観測されなかった。
調製例1に記載の一般手順に従って、重量で95:5のMBA/VDMコポリマービーズを調製した。有機相はヘプタン(348ml)、安定剤(0.13g)およびVDM(0.72g)からなった。水相は、イソプロパノール(90ml)、水(55ml)、MBA(13.33g)、過硫酸ナトリウム(0.55g)およびTMEDA(0.55ml)からなった。P.R.ジョンソン(P.R.Johnson)ら、J.Chromatogr.A、1994、667、1−9に記載の通り、脱イオン水中の水和容積に関して、そしてミオグロビン結合容積に関して、このビーズ(5A)を評価した。結果を表3に示す。イソプロパノールの容積を125mlまで増加し、そして水を75mlまで増加したこと以外は、同一成分および量を使用して第2のMBA/VDMコポリマーを調製した。このビーズ(5B)も水和容積およびミオグロビン結合容積に関して評価した。結果を表3に記載する。
トルエン(188ml)を有機相に添加し、そして水の総量を60mlまで増加したこと以外は、実施例5Aに記載の手順に従って、重量で95:5のMBA/VDMのコポリマービーズを調製した。実施例6に関して、使用されるポリマー安定剤は、重量で92.5:7.5のイソオクチルアクリレートおよびアクリル酸のコポリマー(0.27g)であり、そして撹拌速度は600rpmまで増加され;実施例7に関して、使用されるポリマー安定剤は、重量で90:10のイソオクチルアクリレートおよびアクリル酸のコポリマーであり、そして水酸化ナトリウム(3.7mlの0.1M溶液)を水相に添加し、アクリル酸を中和し;そして実施例8に関して、ポリマー安定剤は、重量で95:5のイソオクチルアクリレートおよびアクリル酸のコポリマー(1.06g)であり、水酸化ナトリウム(0.74mlの1M溶液)を水相に添加し、アクリル酸を中和し、ドデシル硫酸ナトリウム(3mlの10重量%水溶液)を水相に添加し、そして撹拌速度を750rpmに増加した。乾燥後、ビーズを乾燥分類し、32マイクロメートル篩を通過する留分を得た。
窒素パージングされた封着ガラス小瓶中でモノマー混合物を重合させることによって、実施例2と同一組成のモノリスを調製した。小瓶を一晩33℃で水浴中に置いた。小瓶を壊し;モノリスプラグを水およびアセトンで洗浄し、乳鉢および乳棒で簡単に粉砕し、次いで真空下で乾燥させた。脱イオン水中のこれらの破砕粒子のスラリーを製造し、そしてバイオ−ラド ポリ−プレップ カラム(Bio−Rad Poly−Prep Column)中に1mlの床厚まで充填した。リゾチームの手順をIgGに適応させることによって、IgGに対する平衡カチオン交換能力を測定し、そして35mg/mlであることがわかった。
1.5mlのイソプロパノールを1.5mlのPEG400と置換したこと以外は、実施例9と同一組成のモノリス。手順および仕上げは実施例9と同一であった。IgGに対する平衡カチオン交換能力は実施例9に関して記載された通りに測定され、そして23mg/mlであることがわかった。
1.68mlのイソプロパノールを、ポロゲンとして1.68mlの1−オクタノールと置換したこと以外は、実施例9と同一組成のモノリス。手順および仕上げは実施例9と同一であった。IgGに対する平衡カチオン交換能力は実施例9に関して記載された通りに測定され、そして30mg/mlであることがわかった。
窒素パージングされたバイオ−ラド ポリ−プレップ カラム(Bio−Rad Poly−Prep Column)中、室温で一晩、1mlのモノマー混合物を重合させることによって、実施例10と同一組成のモノリスを調製した。脱イオン水(10ml)をモノリスプラグの上部に添加するが、カラムを通して流動は生じない。わずかな窒素圧をカラムの上部に適用したが、なお流動は生じなかった。フリットを含有するカラムの底部を切断したが、なお流動は生じなかった。最終的に、実施例9に記載される通りにモノリスを洗浄し、破砕および乾燥させた。IgGに対する平衡カチオン交換能力は実施例9に関して記載された通りに測定され、そして31mg/mlであることがわかった。
5重量%のAMPSモノマーを等量のn−ブチルアクリレートと置換したこと(実施例13)、10重量%のAMPSモノマーを等量のn−ブチルアクリレートと置換したこと(実施例14)および15重量%のAMPSモノマーを等量のn−ブチルアクリレートと置換したこと(実施例15)以外は、実施例10と同一組成のモノリスを調製した。手順および仕上げは実施例9と同一であった。IgGに対する平衡カチオン交換能力は実施例9に関して記載された通りに測定され、そして24mg/ml(実施例13)、16mg/ml(実施例14)および5mg/ml(実施例15)であることがわかった。
Claims (4)
- 少なくとも一層の多孔性繊維状濾過層を含むフィルターエレメントと、標的分子と結合可能な少なくとも一層の固定相微粒子とを含む複合フィルター媒体であって、前記固定相微粒子が破砕モノリシックポリマー粒子を含み、前記フィルターエレメントが上流面と下流面とを含み、微粒子層が前記上流面上に配置される、クロマトグラフィ用複合フィルター媒体。
- 請求項1に記載の複合フィルター媒体を含むフィルターカートリッジ。
- 請求項2に記載のフィルターカートリッジと、溶質として少なくとも一種の標的生体高分子を含む溶液混合物を収容する貯蔵器と、前記少なくとも一種の生体高分子を固定相微粒子に結合して標的分子と固定相微粒子との生成物を形成するためにフィルターカートリッジを通して溶液混合物をポンピングするためのポンプおよび前記フィルターカートリッジと前記貯蔵器と前記ポンプと関連するチューブとを含む生体高分子の分離システム。
- a)標的分子と結合可能な請求項1に記載の複合フィルター媒体を含むフィルターカートリッジと、溶質として少なくとも一種の標的分子を含む溶液混合物を収容する貯蔵器と、ポンプおよび前記フィルターカートリッジと前記貯蔵器と前記ポンプと関連するチューブとを含有する分離システムを提供する工程と、
b)前記少なくとも一種の生体高分子を固定相微粒子に結合して標的分子と固定相微粒子との生成物を形成するために最大344.7kPa(50psi)の圧力でフィルターカートリッジを通して溶液混合物をポンピングする工程と
を含む溶液混合物からの標的分子の分離方法。
