Claims (1)
- 【特許請求の範囲】 1.α1A-1、α1A-2、α1E-1、α1C-2及びα1E-3から成る群から選択されるα1 サブユニットをコードするヌクレオチド配列を含む単離されたDNAフラグメン ト。 2.α1サブユニットがα1A-1またはα1A-2である請求項1に記載のDNAフラ グメント。 3.α1サブユニットがα1E-1またはα1E-3である請求項1に記載のDNAフラ グメント。 4.α1サブユニットがα1C-2である請求項1に記載のDNAフラグメント。 5.β2、β3及びβ4から成る群から選択されるβサブユニットをコードするヌ クレオチド配列を含む単離されたDNAフラグメント。 6.サブユニットがβ2C、β2Dまたはβ2Eサブユニットである請求項5に記載の DNAフラグメント。 7.サブユニットがβ3サブユニットである請求項5に記載のDNAフラグメン ト。 8.サブユニットがβ3-1サブユニットである請求項7に記載のDNAフラグメ ント。 9.サブユニットがβ4サブユニットである請求項5に記 載のDNAフラグメント。 10.サブユニットが配列番号28に示すアミノ酸配列を有する請求項9に記載 のDNAフラグメント。 11.α1A-1、α1A-2、α1C-2、α1E-1及びα1E-3から成る群から選択されるα1 サブユニットをコードする異種DNAを含む真核細胞。 12.α1サブユニットをコードする異種DNAとβサブユニットをコードする 異種DNAとを含み、少なくとも1つのサブユニットが、α1A-1、α1A-2、α1C -2 、α1E-1、α1E-3、β2C、β2D、β2E、β3-1及びβ4サブユニットから成るサ ブユニットの群から選択される真核細胞。 13.βサブユニットがβ2サブユニットである請求項12に記載の真核細胞。 14.βサブユニットがβ4サブユニットである請求項12に記載の真核細胞。 15.HEK293細胞、チャイニーズハムスター卵巣細胞、アフリカミドリザ ル細胞及びマウスL細胞から成る群から選択される請求項11に記載の真核細胞 。 16.HEK293細胞、チャイニーズハムスター卵巣細胞、アフリカミドリザ ル細胞及びマウスL細胞から成る群 から選択される請求項12に記載の真核細胞。 17.ヒトカルシウムチャンネルのα1サブユニットをコードする異種核酸を細 胞に導入することを含む方法によって産生される機能性異種カルシウムチャンネ ルを有する真核細胞であって、 α1サブユニットがα1A-1、α1A-2、α1C-2、α1E-1及びα1E-3から成る群か ら選択され、 異種カルシウムチャンネルが異種核酸によってコードされる少なくとも1つの サブユニットを含み、 異種イオンチャンネルがカルシウムチャンネルだけであることを特徴とする真 核細胞。 18.ヒトカルシウムチャンネルのα1サブユニットをコードする核酸を細胞に 導入し、ヒトカルシウムチャンネルのβサブユニットをコードする核酸を細胞に 導入することを含む方法によって産生される機能性異種カルシウムチャンネルを 有する真核細胞であって、 サブユニットの少なくとも1つがα1A-1、α1A-2、α1E-1、α1E-3、β2C、β2D 、β2E、β3及びβ4サブユニットから成る群から選択され、 異種カルシウムチャンネルが異種核酸によってコードさ れる少なくとも1つのサブユニットを含み、 異種イオンチャンネルがカルシウムチャンネルだけであることを特徴とする真 核細胞。 19.HEK293細胞、チャイニーズハムスター卵巣細胞、アフリカミドリザ ル細胞、マウスL細胞及び両生類卵母細胞から成る群から選択される請求項17 に記載の真核細胞。 20.HEK293細胞、チャイニーズハムスター卵巣細胞、アフリカミドリザ ル細胞、マウスL細胞及び両生類卵母細胞から成る群から選択される請求項18 に記載の真核細胞。 21.βサブユニットがヒトカルシウムチャンネルのβ2、β3及びβ4サブユニ ットである請求項18に記載の真核細胞。 22.カルシウムチャンネルがヒトカルシウムチャンネルのα2bサブユニット、 ヒトカルシウムチャンネルのα1B-1サブユニット及びヒトカルシウムチャンネル のβ3サブユニットを含む請求項18に記載の真核細胞。 23.カルシウムチャンネルが、α1B-1、α2b及びβ1-2サブユニットを含むか 、α1B-1、α2b及びβ1-3サブユニッ トを含むか、α1B-2、α2b及びβ1-3サブユニットを含むか、α1A-2、α2b及び β3-1サブユニットを含むか、または、α1B-1、α2b及びβ3-1サブユニットを含 む請求項18に記載の真核細胞。 24.カルシウムチャンネルが、ヒトカルシウムチャンネルのα2bサブユニット と、ヒトカルシウムチャンネルのα1Bまたはα1Dサブユニットと、ヒトカルシウ ムチャンネルのβ1-1、β1-2またはβ1-3サブユニットとを含む請求項18に記 載の真核細胞。 25.カルシウムチャンネルの活性を変調する化合物を同定する方法であって、 機能性異種カルシウムチャンネルを有する真核細胞を化合物とカルシウムチャ ンネル選択イオンとを含有する溶液に懸濁させ、 該細胞の細胞膜を脱分極し、 該細胞に流れる電流を検出する段階を含み、ここで、 異種カルシウムチャンネルが細胞に異種のDNAまたはRNAによってコード される少なくとも1つのヒトカルシウムチャンネルサブユニットを含み、 少なくとも1つのサブユニットがα1A-1、α1A-2、α1E -1、α1E-3、α1C-2、β2C、β2D、β2E、β3サブユニット及びβ4サブユニット から成る群から選択され、 検出される電流が、同じカルシウムチャンネル選択イオンが存在するが前記化 合物の非存在下で同じまたは実質的に等しい細胞を脱分極することによって生じ る電流とは異なることを特徴とする方法。 26.異種DNAまたはRNAがβ3サブユニットをコードすることを特徴とす る請求項25に記載の方法。 27.異種DNAまたはRNAがβ4サブユニットをコードすることを特徴とす る請求項26に記載の方法。 28.ヒトカルシウムチャンネルのαサブユニットのタイプまたはαサブユニッ トのサブタイプに結合する抗体から成る群から選択され、サブユニットがα1サ ブユニットであることを特徴とするサブユニット特異的抗体。 29.抗体がサブタイプ特異的であり、α1サブユニットがα1A、α1E、及びα1 B である請求項28に記載の抗体。 30.α1A-1、α1A-2、α1E-1、α1C-2、α1E-3、β3-1、β2C、β2D、β2E及 びβ4から成るサブユニットの群から選択されるヒトカルシウムチャンネルのサ ブユニットをコードする核酸からの少なくとも16の実質的に連続する塩 基を含む少なくとも16塩基の長さのRNAまたは一本鎖DNAプローブ。 31.α1A-1、α1A-2、α1E-1、α1C-2、α1E-3、β3-1、β2C、β2D、β2E及 びβ4サブユニットから成るサブユニットの群から選択されるヒトカルシウムチ ャンネルのサブユニットをコードする核酸からの少なくとも30の塩基を含む請 求項30に記載のプローブ。 32.少なくとも低緊縮性の条件下に請求項30に記載のプローブを核酸フラグ メントのライブラリーにハイブリダイズさせ、ハイブリダイズするフラグメント を選択する段階を含むヒトカルシウムチャンネルサブユニットをコードする核酸 の同定方法。 33.少なくとも低緊縮性の条件下に請求項30に記載のプローブを細胞もしく は組織中で発現されたmRNAまたはmRNAから産生されたcDNAにハイブ リダイズさせ、これによりサブユニットをコードするmRNAを発現する細胞ま たは組織を同定する段階を含むカルシウムチャンネルサブユニットをコードする 核酸を発現する細胞または組織の同定方法。 34.α1A-1、α1A-2、α1E-1、α1C-2、α1E-3、β3-1、 β2C、β2D、β2E及びβ4から成る群から選択される実質的に純粋なヒトカルシ ウムチャンネルサブユニット。
