Claims (1)
- 【特許請求の範囲】 1.分析物を含有すると見なされる試料内の分析物の存在を決定する確認する方 法であって、前記分析物は酵素又は触媒と反応し、その際検出可能な信号の量が 検定媒体中の分析物の量の関数であり、前記検出可能な信号を電極で測定する方 法において、 a)電極が、該電極を電解液から分離する電気絶縁ポリマーの薄膜で被覆さ れており、 b)前記分析物との酵素又は触媒反応が直接又は間接的に前記電気絶縁ポリ マー層との反応を引き起こし、該反応の際にポリマー層は電解液に対して多孔質 又は一層多孔質になり、電極表面での電気的特性の変化を惹起する ことを特徴とする、試料中の分析物の存在を確認する方法。 2.電気絶縁材料がpH感応性ポリマーであり、かつ反応性又は触媒種が電極被 覆層の透過性を増大するために十分なpH変化を惹起する、請求項1記載の方法 。 3.反応性又は触媒種が、pH変化を惹起する反応を行う酵素である、請求項1 記載の方法。 4.反応性触媒成分がウレアーゼ又はグルコースオキシダーゼである、請求項2 記載の方法。 5.酵素−抗体又は酵素−抗原抱合体が特殊な免疫化 学反応を介してポリマー層に又はその極近くに結合され、該ポリマー層が電極を 被覆しておりかつ抱合体の酵素活性が電解液に対するポリマー層の透過性を増大 させ、該ポリマー層の下にある電極によりキャパシタンス又は通過電流の変化に 基づき測定する、免疫検定法。 6.電極を、電極が電解液に曝されるようになって初めてポリマーの分解が促進 されるように、電解液の成分とレドックス反応を起こすことができる電位で分極 する、請求項1記載の方法。 7.触媒種がヒドロキシル基の生成を惹起し、該ヒドロキシル基がポリマー層と 反応可能であり、それにより前記層の電解液に対する透過性を増大せしめる、請 求項1記載の方法。 8.触媒種が過酸化水素産生オキシダーゼであり、かつ電解液がフェントン反応 を促進することのできる種を含有する、請求項7記載の方法。 9.触媒種が、直接ポリマー層に作用して層の透過性を増大させるポリマー減成 酵素である、請求項1記載の方法。 10.触媒種がアミラーゼ又はアミログルコシダーゼでありかつ誘電体がポリサッ カリドからなるか又はポリサッカリドを含有する、請求項9記載の方法。 11.触媒種がエンドヌクレアーゼでありかつポリマーが核酸からなるか又は核酸 を含有する、請求項9記 載の方法。 12.触媒種がリパーゼでありかつポリマーが脂質からなるか又は脂質を含有する 、請求項9記載の方法。 13.酵素又は触媒がポリマー層内に存在する、請求項1記載の方法。 14.酵素又は触媒がポリマー層に結合されている、請求項1記載の方法。 15.酵素又は触媒が、ニトロセルロース膜のようなポリマー層又はその他の膜支 持体に密接した別の支持体に結合されている、請求項1記載の方法。 16.試料が水性の全血液、血清、血漿、尿又は唾液である、請求項1記載の方法 。
