Claims (1)
- 【特許請求の範囲】 1.非ヒト哺乳動物の生殖系列中の遺伝子または遺伝子断片を他の哺乳動物の 相同遺伝子または相同遺伝子断片で置換する方法であって、その際、 (i)選択可能なようにマーキングした組換えビヒクルで胚性幹細胞株をトラン スフェクションし; (ii)安定にトランスフェクションされた細胞クローンをマーカー遺伝子の存在 について選択し; (iii)これら細胞クローンをPCRおよび/またはサザーンブロットによるタ ーゲティング選択に供し; (iv)該細胞クローンを非ヒト哺乳動物の胞胚中に注入し; (v)該胞胚を代理母に移行させ、 選択可能なようにマーキングした組換えビヒクルにより、組換え事象において内 生遺伝子または内生遺伝子断片が相同遺伝子または相同遺伝子断片によって1工 程で機能的に置換されることを特徴とする方法。 2.導入した遺伝子または導入した遺伝子断片がヒト由来であり、非ヒト哺乳 動物が齧歯類、とりわけマウスである、請求項1に記載の方法。 3.相同遺伝子または相同遺伝子断片が、免疫系;神経系、とりわけシグナル 媒体分子および接着性分子;ウイルスレセプター;造血系;および支持組織、と りわけ筋肉、腱および骨をコードする、請求項1または2に記載の方法。 4.置換された遺伝子断片が、免疫系をコードするものであり、とりわけ、抗 体遺伝子、T細胞レセプター遺伝子、サイトカイン、サイトカインレセプター遺 伝子、MHC遺伝子、接着性分子遺伝子およびシグナル媒体分子である(抗体遺 伝子、サイトカインおよびMHC遺伝子が好ましい)請求項1〜3のいずれか1 または2以上に記載の方法。 5.マウスの抗体遺伝子または抗体遺伝子断片がヒトの抗体遺伝子または抗体 遺伝子断片で置換される請求項1〜4のいずれか1または2以上に記載の方法。 6.選択可能なようにマーキングした組換えビヒクルが置換ベクターであり、 導入しようとする遺伝子断片または導入しようとする遺伝子;置換されるべき内 生DNA断片の両側に位置する配列に相補的な配列;ネオマイシンおよびハイグ ロマイシンから選択されたマーカー遺伝子;並びに、別の機能性の遺伝子配列、 とりわけウイルス認識配列、遺伝子発現を増幅させるための配列、および原核お よび真核組換え系のための標的配列、とりわけCre認識配列LoxPおよびf lip認識配列Frtを含有する、請求項1〜5のいずれか1または2以上に記 載の方法。 7.マーカー遺伝子がネオマイシンである請求項6に記載の方法。 8.選択可能なようにマーキングした組換えビヒクルがpTZ2−CkN(P A−)(DSM7211)である請求項1〜7のいずれか1または2以上に記載 の方法。 9.マーカー遺伝子および依然として存在する標的遺伝子断片を標的配列によ り第二の反応工程で除去する、請求項6に記載の方法。 10.請求項1〜9のいずれか1または2以上に記載の、相同的組換えのため の選択可能なようにマーキングした組換えビヒクル。 11.請求項1〜9のいずれか1または2以上に記載の安定にトランスフェク ションした細胞クローン。 12.請求項11に記載の安定にトランスフェクションした細胞クローンを胞 胚中に注入し、これら胞胚を代理母に移し、生まれたキメラ非ヒト哺乳動物を交 配させ、ついでその子孫を変異の存在について選択することを特徴とする、トラ ンスジェニック非ヒト哺乳動物の製造方法。 13.キメラ非ヒト哺乳動物が齧歯類、とりわけマウスである請求項12に記 載の方法。 14.請求項12または13に記載の方法によって得られるトランスジェニッ ク非ヒト哺乳動物。 15.もともとの遺伝子断片によってコードされた産物の代わりに他の種の遺 伝子産物を発現させるための請求項14に記載の選択的に変異された非ヒト哺乳 動物の使用。 16.該遺伝子産物をヒト化モノクローナル抗体を産生させるのに使用する、 請求項15に記載の使用。 17.該遺伝子産物をウイルスを産生させるのに使用する、請求項15に記載 の使用。 18.薬物および治療モデルを試験するための請求項14に記載の選択的に変 異された非ヒト哺乳動物の使用。
