Abstract
Description
Claims (1)
- 【特許請求の範囲】 1.下記工程を含む、神経幹細胞から分化細胞を調製する方法: (a)ドナーの組織から神経幹細胞を単離する工程、 (b)その単離した神経幹細胞を第1増殖因子を含む第1培養基中で増殖して 前駆細胞を生産する工程、及び (c)その前駆細胞を少なくとも第2増殖因子を含む第2培養基中で培養する ことにより、前記前駆細胞を分化して分化細胞を生産する工程、ここで前記第2 培養基は実質的に前記第1増殖因子を含まない。 2.該第1増殖因子が表皮増殖因子である請求項1記載の方法。 3.該第2培養基が血清を含む請求項1記載の方法。 4.該第2増殖因子が酸性線維芽細胞増殖因子、塩基性線維芽細胞増殖因子、繊 毛神経栄養因子、神経増殖因子、脳由来神経栄養因子、ニューロトロフィン3、 ニューロトロフィン4、インターロイキン、白血球阻止因子、サイクリックアデ ノシンーリン酸、ホルスコリン、破傷風毒素、高レベルのカリウム、アンフィレ グリン、悪性化増殖因子α、悪性化増殖因子β、インスリン様増殖因子、デキサ メタゾン、イソブチル3−メチルキサンチン、ソマトスタチン、成長ホルモン、 レチノイン酸及び血小板由来増殖因子からなる群より選ばれる請求項1記載の方 法。 5.前記分化細胞がニューロンであり、前記第2増殖因子が繊毛神経栄養因子で ある請求項1記載の方法。 6.前記分化細胞が成熟希突起膠細胞であり、前記第2増殖因子が繊毛神経栄養 因子である請求項1記載の方法。 7.前記分化細胞がアストログリア細胞であり、前記第2増殖因子が繊毛神経栄 養因子であり、前記第2培養基が実質的に血清を含まない請求項1記載の方法。 8.前記分化細胞がニューロンであり、前記第2増殖因子が脳由来神経栄養因子 である請求項1記載の方法。 9.前記分化細胞がニューロンであり、前記第2増殖因子がレチノイン酸である 請求項1記載の方法。 10.前記分化細胞がニューロンであり、前記第2増殖因子が塩基性線維芽細胞増 殖因子である請求項1記載の方法。 11.下記工程を含む、前駆細胞を調製する方法: (a)ドナーの組織から神経幹細胞を単離する工程、 (b)その単離した神経幹細胞を塩基性線維芽細胞増殖因子を含む第1培養基 中で維持する工程、及び (c)その単離した神経幹細胞を表皮増殖因子と塩基性線維芽細胞増殖因子を 含む第2培養基中で増殖して前駆細胞を生産する工程。 12.下記工程を含む、神経幹細胞から分化細胞を調製する方法: (a)ドナーの組織から神経幹細胞を単離する工程、 (b)その単離した神経幹細胞を増殖因子を含む第1培養基中で増殖して前駆 細胞を生産する工程、及び (c)その前駆細胞を前記第1増殖因子を実質的に含まない第2培養基中で基 質と接触させる工程。 13.基質がポリ−L−オルニチン、コラーゲン、フィブロネクチン、ラミニン及 びマトリゲルからなる群より選ばれる請求項12記載の方法。
