Description
Claims (1)
- 【特許請求の範囲】 1. 遠心分離機内で濃縮装置を回転させることによって溶液から巨大分子を 濃縮するための方法であって、前記装置は、濃縮あるいは精製する試料を受ける ための試料貯蔵器(1)、試料を濾過するための濾過手段、濾液を回収するため の遠心分離管(5)、及び濃縮物を保持するための濃縮物ポケット(4)を有し 、液体の試料が試料貯蔵器内に入れられる方法において、さらに、試料貯蔵器( 1)及び濾過手段(2)を含む装置が少なくとも部分的に濾液を回収するための 遠心分離管内に入れられる工程、このようにして構成された組み合わせ装置が遠 心分離機にかけられて巨大分子に作用しまたフィルター表面を掃引する力のベク トルが生成される工程、巨大分子がフィルター表面から離れた濃縮物ポケット( 4)内に回収されそれによって乾燥状態に達するまで濃縮が行なわれる危険が除 去される工程を含むことを特徴とする方法。 2. 前記力のベクトルが少なくともフィルターの表面の一部に平行なまたは そこから離れる角度を示す方向にフィルターの表面を掃引することを特徴とする 請求の範囲1に記載の方法。 3. 前記力のベクトルが少なくともフィルターの表面の一部に平行なまたは そこへ向かう角度を示す方向にフィルターの表面を掃引することを特徴とする請 求の範囲1に記載の方法。 4. 前記ベクトルとフィルターの表面の間の角度が0−75゜の間に含まれ ることを特徴とする請求の範囲2に記載の方法。 5. 前記ベクトルとフィルターの表面の間の角度が0−55゜の間に含まれ ることを特徴とする請求の範囲3に記載の方法。 6. 濃縮物ポケット(4)内の停滞物がピペットまたは他の回収装置によっ てポケットから引き出されることを特徴とする請求の範囲1〜5の いずれか一項に記載の方法。 7. 濃縮あるいは精製する試料を受けるための試料貯蔵器(1)、試料を濾 過するための濾過手段(2)、濾液を回収するための遠心分離管(5)、及び濃 縮物を保持するための濃縮物ポケット(4)を有する装置において、前記貯蔵器 (1)には容量が試料貯蔵器の全容量の小さな一部分にすぎず側壁と底壁を有す る濾過チャンバー(3)を形成する部分が配設されること、前記濾過手段は前記 濾過チャンバーの側壁に配置されること、及び前記濃縮物ポケット(4)は前記 濾過チャンバーの底壁に配置されることを特徴とする請求の範囲1に記載の方法 を実施する微量濃縮装置。 8. 前記濾過手段が前記濾過チャンバーの側壁に液漏れしないように取り付 けられる膜支持手段が配設された平坦な膜であることを特徴とする請求の範囲第 7項に記載の微量濃縮装置。 9. 濾過チャンバー(3)の外形が半円筒形を示し、濾過手段が前記半円筒 形の平坦な側壁に配置され、前記濾過チャンバー(3)の周囲には対応する半円 筒形のスリーブ(25)が配設されて濾過手段を前記チャンバー(3)の平坦な 側壁にしっかり固定することを特徴とする請求の範囲7または8に記載の微量濃 縮装置。 10. 濾過チャンバー(3)が濃縮物ポケット(4)の方向にテーパの付いた 外形を有し、濾過手段は前記チャンバー(3)の平坦な側壁に配置され、前記濾 過チャンバー(3)の周囲には対応する内形を有するスリーブ(25)が配設さ れて濾過手段を平坦な側壁にしっかり固定することを特徴とする請求の範囲7ま たは8に記載の微量濃縮装置。 11. 前記濾過手段が前記濾過チャンバーの側壁に液漏れしないように取り付 けられる膜支持手段が配設された外側に凸面の膜であることを特徴とする請求の 範囲7に記載の微量濃縮装置。 12. 前記濾過手段が前記濾過チャンバーの側壁に液漏れしないように取り付 けられる膜支持手段が配設された外側に凹面の膜であることを特徴とする請求の 範囲7に記載の微量濃縮装置。 13. 前記膜が1マイクロメートルの最大の孔の大きさを有することを特徴と する請求の範囲7に記載の微量濃縮装置。 14. 前記濾過手段が濾過チャンバーの側壁を構成する円筒状多孔質の膜支え を有することを特徴とする請求の範囲7に記載の微量濃縮装置。 15. 前記円筒状の膜支えが試料貯蔵器の下方部分に液漏れしないように前記 貯蔵器に対して同軸状に取り付けられることを特徴とする請求の範囲14に記載の 微量濃縮装置。 16. 前記膜支えがセラミック材料でつくられることを特徴とする請求の範囲 14または15に記載の微量濃縮装置。 17. 前記膜支えが炭素でつくられることを特徴とする請求の範囲14または15 に記載の微量濃縮装置。 18. 前記膜が前記膜支えの表面上に一体化されていることを特徴とする請求 の範囲14に記載の微量濃縮装置。 19. 前記膜が前記膜支えの中に挿入される別個の円筒状装置を形成すること を特徴とする請求の範囲14に記載の微量濃縮装置。 20. 試料貯蔵器(1)の底部が濾過溝の入口の方向に縮小する漏斗状に形成 され、前記壁が試料貯蔵器の軸に垂直の方向に対して5゜と46°の間の角度で 配置されることを特徴とする請求の範囲7〜19のいずれか一項に記載の微量濃縮 装置。 21. 溝の側壁または周囲のスリーブの内側または外側に残留液の量を正確に 示すことができる目盛が配設されることを特徴とする請求の範囲7に記載の微量 濃縮装置。 22. 停滞物貯蔵器(4)を濾過手段の表面から分離する仕切り壁(28)に よって確実停止機能がえられること、及び、この壁の上方部分の断面が斜面また は台形の形状を有することを特徴とする請求の範囲7〜21のいずれか一項に記載 の微量濃縮装置。
