Description
Claims (1)
- 【特許請求の範囲】 1.血液対照生成物が、処理された血液細胞が血液試験方法において使用する溶 菌剤による劣化に対して耐性であるように処理された処理血液細胞を有しており 、この対照生成物が、少なくとも二種の異なるヒト白血球をシミュレートする血 液対照生成物であって、それぞれがヒト白血球の少なくとも二種の物理的特性を 有しており、これらの特性が、 a.直流電流によって測定された容積 b.高周波(RF)サイズ c.不透明度および d.光散乱 からなる群より選択されていることを特徴とする、血液対照生成物。 2.前記の物理的特性の一つが光散乱であることを特徴とする、請求項1記載の 血液対照生成物。 3.ヒト白血球の一種をシミュレートするのに使用する処理血液細胞が、そのも との容積の30%以上膨張したことを特徴とする、請求項2記載の血液対照生成 物。 4.前記血液対照生成物が、少なくとも四種の異なる分析境界の中に分布してい る少なくとも四種の白血球類縁体を含有しており、この分析境界が、光散乱、容 積および不透明度に基づいて作られていることを特徴とする、請求項1、2また は3のいずれか一つの項に記載の血液対照生成物。 5.前記の処理血液細胞が、変化されたヘモグロビン内容物を含有していること を特徴とする、請求項1または2記載の血液対照生成物。 6.前記血液細胞のヘモグロビンが、処理血液細胞内で変性されたことを特徴と する、請求項5記載の血液対照生成物。 7.前記処理血液細胞のヘモグロビンが、処理血液細胞から漏出したことを特徴 とする、請求項5記載の血液対照化成物。 8.20%〜80%のヘモグロビンが、処理血液細胞から漏出したことを特徴と する、請求項7記載の血液対照生成物。 9.少なくとも二種の白血球類縁体集団を含有する血液対照生成物を使用する方 法であって: a.血液対照生成物を装置内に配置し、前記対照生成物が、血液試験方法におい て使用する溶菌剤による劣化に対して耐性であるように処理された血液細胞に由 来した少なくとも二種の異なる白血球類縁体集団を含有しており、前記の白血球 類縁体集団が、それぞれヒト白血球の少なくとも二種の物理的特性を有しており 、これらの特性が、 (1)直流電流によって測定された容積(2)高周波(RF)サイズ (3)不透明度および (4)光散乱 からなる群より選択されており; b.この対照生成物の前記の物理的特性を測定し;およびc.この測定の結果を 記録して、前記装置が仕様の範囲内で作動しているかどうかを測定することを特 徴とする、血液対照生成物の使用方法。 10.前記物理的特性の一つが光散乱であることを特徴とする、請求項9記載の 血液対照生成物の使用方法。 11.前記対照生成物が、前記装置の少なくとも四種の異なる分析境界の中に分 布している少なくとも四種の白血球類縁体を含有しており、この分析境界が、光 散乱、容積および不透明度に基づいて作られていることを特徴とする、請求項9 または10記載の血液対照生成物の使用方法。 12.前記白血球類縁体が、変化されたヘモグロビン内容物であることを特徴と する、請求項9または10記載の血液対照生成物の使用方法。 13.前記白血球類縁体が、血液細胞から漏出した20%〜80%のヘモグロビ ンを含有していることを特徴とする、請求項12記載の血液対照生成物の使用方 法。 14.前記白血球類縁体のヘモグロビンが、細胞内で変性したことを特徴とする 、請求項12記載の血液対照生成物の使用方法。 15.白血球類縁体を製造する方法であって:a.赤血球を低浸透圧溶液と混合 してこの赤血球の容積を膨張させ;b.この細胞中のヘモグロビン内容物を変化 させてヒト白血球細胞の光散乱特性をシミュレートさせ;および、 c.血液試験方法で使用する溶血剤による劣化に対して耐性であるようにこの細 胞を固定化し、この固定化された細胞が、ヒト白血球に類似している光散乱特性 および容積特性を有していることを特徴とする、白血球類縁体の製造方法。 16.前記ヘモグロビン内容物の変化を、細胞中のヘモグロビンを変性すること によって達成することを特徴とする、請求項15記載の白血球類縁体の製造方法 。 17.前記ヘモグロビン内容物の変化を、細胞からヘモグロビン内容物を漏出さ せることによって達成することを特徴とする、請求項15記載の白血球類縁体の 製造方法。 18.前記細胞のヘモグロビン内容物を、20%〜80%まで減少させることを 特徴とする、請求項17記載の白血球類縁体の製造方法。 19.前記混合が、細胞を血清物質と混合し、次いで低浸透圧溶液と混合するこ とによって特徴付けられている、請求項15記載の白血球類縁体の製造方法。 20.血清物質が、コレステロール、コレステロールエステル、リボタンパク質 コレステロール、リボタンパク質コレステロールエステル、リン脂質と結合され たコレステロールおよびこれらの混合物からなる群より選択されていることを特 徴とする、請求項19記載の白血球類縁体の製造方法。 21.前記細胞が75%以上膨張していることを特徴とする、請求項15、19 または20のいずれか一つの項に記載の白血球類縁体の製造方法。 22.前記赤血球が、同時に膨張および固定化されていることを特徴とする、請 求項15記載の白血球類縁体の製造方法。
