Claims (5)
- 1.分析している試料を含む懸濁液の容積を測定しない装置で細胞試料の単位容 積あたり細胞総数を探知する方法において、a.第1光散乱信号を有する既知の 量の位子を、第1光散乱信号と異なる第2光散乱信号を有する既知容積の細胞試 料と混合して、試料容積あたり一定の位子濃度を有する懸濁液を得; b.上記細胞懸濁液の未知の容積部分の中で上記位子及び細胞それぞれを順次に 光線を通過させて、上記粒子及び細胞のそれぞれが少なくとも一つの前進光散乱 パターンを生じさせ; c.上記懸濁液の上記未知容積部分中の細胞数及び位子数をカウントし;d.上 記細胞試料に加えられた既知量の粒子に対して、上記懸濁液分中の粒子及び細胞 の数を相関させて、試料の単位容積あたりの細胞の総数を決定することを特徴と する細胞試料の単位容積あたり細胞総数の測定方法。
- 2.上記の細胞数及び位子数のカウンティングを光散乱トリガーを使用するフロ ーサイトメータによって行うことを特徴とする請求の範囲1記載の方法。
- 3.上記の粒子と細胞試料中細胞との密度差が20%より少ないことを特徴とす る請求の範囲1又は2記載の方法。
- 4.さらに(e)位子の蛍光信号及び上記光散乱パターンを装置性能の量小限界 を表す所定の比に相関させ;(f)所定の比が得られなかった場合には、位子を 入射光線の中に置きながら、測定比が上記所定の比に達するまで、上記の装置の 操作を調節することを特徴とする、細胞試料の単位容積あたり細胞の総数を測定 する中、カウンティング装置の操作パラメーターを規定範囲内に維持することを 更に含み、かつ、上記粒子の蛍光が既知の波長を有する請求の範囲1又は2記載 の方法。
- 5.上記位子を細胞試料中の細胞に付着しないように処理してあることを特徴と する請求の範囲1記載の方法。
