Claims (1)
- 【特許請求の範囲】 1.アラビドプシスクリアナ(Arabidopsisthaliana)の1 1.6若しくは12.9kDaのタペータムのタンパク質またはアブラナ科(f amilyBrassicaceae)の他のものの同様のタンパク質をエンコ ードする遺伝子の発現を自然に誘導するプロモーターからなる組換えまたは単離 DNA。 2.アラビドプシスタリアナ(A.thaliana)の11.6kDaのタペ ータムのタンパク質と等価である、プラッシカナプス(Brassicanap us)の10.3kDaのタペータムのタンパク質の発現を誘導するプロモータ ーからなる請求の範囲1に記載のDNA。 3.図4に示される配列のコーディング領域(codingregion)の5 ′方向にプロモーターを有する、請求の範囲1に記載のDNA。 4.図7に示される配列のコーディング領域(codingregion)の5 ′方向にプロモーターを有する、請求の範囲1に記載のDNA。 5.該プロモーターを、発現すると、植物において雄性不稔が生じるDNAに操 作して連結した、請求の範囲1から4のいずれかに記載のDNA。 6.該雄性不稔DNAが分解酵素をエンコードする、請求の範囲5に記載のDN A。 7.該分解酵素によって核酸、タンパク質、炭水化物または脂質が分解する、請 求の範囲6に記載のDNA。 8.該分解酵素がリボヌクレアーゼまたはデオキシリボヌクレアーゼである、請 求の範囲7に記載のDNA。 9.該分解酵素によって炭水化物が分解する、請求の範囲6に記載のDNA。 10.該分解酵素がグルカナーゼである請求の範囲9に記載のDNA。 11.グルカナーゼのコード配列との転写融合物(transcriPtion alfusion)においてシグナル配列を有する請求の範囲10に記載のDN A。 12.該分解酵素によってタンパク質が分解する、請求の範囲6に記載のDNA 。 13.該タンパク質分解酵素がアクチニジンまたはパパインである、請求の範囲 12に記載のDNA。 14.該雄性不稔DNAが、植物のタペータム細胞において一般的に発見される RNAに対してアンチセンスであるRNAをコードする、請求の範囲5に記載の DNA。 15.該雄性不稔DNAが、アラビドプシスタリアナ(Arabidopsis thaliana)の11.6若しくは12.9kDaのタペータムのタンパク 質またはアブラナ科(familyBrassicaceae)の他のものの同 様のタンパク質をエンコードするRNAに対してアンチセンスであるRNAをコ ードする、請求の範囲14に記載のDNA。 16.アラビドプシスタリアナ(Arabidopsisthaliana)の 11.6若しくは12.9kDaのタペータムのタンパク質またはアブラナ科( familyBrassicaceae)の他のものの同様のタンパク質をエン コードする遺伝子において自然に転写されるDNA鎖の少なくとも一部に対して 相補的である転写可能なDNA鎖を有するアンチセンス核酸。 17.転写が構成プロモーター(constitutivepromoter) の制御下にある請求の範囲16に記載のアンチセンス核酸。 18.転写がタペータムに特異的なプロモーターの制御下にある請求の範囲16 に記載のアンチセンス核酸。 19.該雄性不稔DNAが植物のタペータム細胞において一般的に発見されるR NAを切断できるRNAをコードする、請求の範囲5に記載のDNA。 20.該雄性不稔DNAが、アラビドプシスタリアナ(Arabidopsis thaliana)の11.6若しくは12.9kDaのタペータムのタンパク 質またはアブラナ科(familyBrassicaceae)の他のものの同 様のタンパク質をエンコードするRNAを切断できるRNAをコードする、請求 の範囲19に記載のDNA。 21.アラビドプシスタリアナ(Arabidopsisthaliana)の 11.6若しくは12.9kDaのタペータムのタンパク質またはアブラナ科( familyBrassicaceae)の他のものの同様のタンパク質をエン コードする遺伝子によってエンコードされるRNAを特異的に切断できるリボザ イムをエンコードするDNA。このようなリボザイムをエンコードするDNAは 、通常、アブラナ科(familyBrassicaceae)のものを雄性不 稔化するのに有用である。 22.転写が構成プロモーター(constitutivepromoter) の制御下にある請求の範囲21に記載のリボザイムをエンコードするDNA。 23.転写がタペータムに特異的なプロモーターの制御下にある請求の範囲21 に記載のリボザイムをエンコードするDNA。 24.アラビドプシスタリアナ(Arabjdopsisthaliana)の 11.6若しくは12.9kDaのタペータムのタンパク質またはアブラナ科( familyBrassicaceae)の他のものの同様のタンパク質をエン コードする遺伝子の適当な発現を特異的に破壊することができるDNA。 25.3′転写調節配列からなる請求の範囲1から24のいずれかに記載のDN A。 26.該3′転写調節シグナルがカリフラワーモザイクウィルス(Caulif lowerMosaicVirus)35S遺伝子由来である、請求の範囲25 に記載のDNA。 27.組換体であり、ベクターの形態を有する、請求の範囲1から26のいずれ かに記載のDNA。 28.該ベクターがクローニングベクターであり、1つまたはそれ以上の選択マ ーカーを有する、請求の範囲27に記載のDNA。 29.請求の範囲27または28に記載のベクターを感染または形質転換した微 生物の宿種細胞。 30.DNAを形質転換された植物が形質転換されない植物と区別できるマーカ ー配列を有する、請求の範囲1から28のいずれかに記載のDNA。 31.該マーカー配列によって抗生物質若しくは除草剤耐性が得られるまたは該 マーカー配列がグルクロニダーゼをコードする、請求の範囲30に記載のDNA 。 32.該マーカー配列がタペータムに特異的でない第二のプロモーターの制御下 にある、請求の範囲31に記載のDNA。 33.該第二のプロモーターがカリフラワーモザイクウィルス(Caulifl owerMosaicVirus)(CaMV)35S遺伝子由来である、請求 の範囲32に記載のDNA。 34.請求の範囲1から28および30から33のいずれかに記載のDNAを有 する植物細胞。 35.少なくとも幾つかの細胞が請求の範囲34に記載されるものである植物ま たは植物の一部。 36.請求の範囲35に記載の植物からの増殖材料。
