【発明の詳細な説明】
医薬調製物
本発明は、1種または数種の担体を1種または数種のレセプタ類/結合蛋白質類
との組合せにおいて含む医薬調製物に関するものである。該レセプタ類/結合蛋
白質類は、場合により、例えば成長ホルモン等の適当なリガンド類に結合されて
いてよい。
本発明による組合せ、担体+レセプタ類/結合蛋白質類十例えば活性ペプチドに
よれば徐放性調製物が得られる。裏返せば本発明による原理は、例えば腫瘍成長
因子等の成長因子の異常な増大した産生の場合においても有用であり、この場合
には担体+レセプタ類/結合蛋白質類は成長因子類の選択的吸着剤として作用す
る。
レセプタ類は、ホルモン類および成長因子類(=リガンド類)を結合し得る蛋白
質分子である。各レセプタ型は、そのリガンドに対して特異的である。レセプタ
の機能は、外部の、例えばホルモン性信号を標的細胞に伝達することである。レ
セプタ研究における新たな成功は、特定の純粋なレセプタ蛋白質を大量に入手で
きる可能性を生んだ。この可能性は、レセプタ類の新規な治療的役割を定義する
本発明について基本的なことである。数種の既知のレセプタ類は、インシュリン
様成長因子−1−レセプタ、インシュリン様成長因子−2−レセプタ、血小板誘
導成長因子−レセプタ、繊維芽成長因子−レセブタ、表皮成長因子−レセプタ、
神経成長因子−レセプタ、コロニー刺激因子−レセプタ、転換成長因子−レセプ
タ、成長ホルモン−レセプタ、上皮小体ホルモン−レセプタ、カルシトニン−レ
セプタ、エストロゲン−レセプタ、腫瘍壊死因子−レセプタ、インシュリン様成
長因子血清結合蛋白質およびコルチコステロイド結合グロブリンによって例示さ
れる。
成長因子類およびホルモン類は、動物およびヒトの両者において細胞分裂、成長
、変異、分化等に関する重要な細胞工程を刺激することか知られている。加えて
、治癒および再生工程もまた、これらの因子により制御されている。成長因子類
/ホルモン類は、例えば、インシュリン様成長因子−1および2 ; IGF−
1,IGF−2、血小板誘導成長因子: PDGF、表皮成長因子、EGF、線
維芽成長因子: FGF、神経成長因子:NGF、コロニー刺激因子; CSF
、転換(Transforming)成長因子;TGF、腫瘍壊死因子、TNF
、カルシトニン:CT、上皮小体ホルモン;PTH,成長ホルモン;GH。
エストロゲン類、ボンベシン、骨形態発生蛋白質−BMP、インシュリンおよび
コルチコステロイド類を含む。
インシュリン、エストロゲン類、コルチコステロイド類、CTおよびGHは、日
常の臨床的実務において周知の医薬である。該研究は、その他の前述した成長因
子類およびホルモン類に関して黙然である。種々の研究および成果に関して言え
ば、例えばPDGFおよびIGF−1は、傷の治癒を促し、GHは、骨折の治癒
を増大させる等がある。これらの成長因子類およびホルモン類のほとんどは、種
々の記録データによれば興味深い効果を有するものではあるが、臨床的意味はこ
れから見出されるへきものである。従来得られている結果は、現在使用されてい
る投与経路か、例えばペプチド類の短い半減期、該物質の有効性および起こりつ
る毒性のために該物質の薬剤としての使用を制限していることを示している。徐
放を提供する本発明によれば、該ペプチド類の生物活性は、担体物質との組合せ
によって延長される。ヒアルロン酸等の担体は、それ自体でも好ましい生物学的
活性を −育し、またポリラクチドのロッドは、安定化骨形成材料であること以
外に、骨折治癒促進性ポリペプチド類を分配する徐放性担体として作用する。
かくして、本発明は、(2種または)3Flの成分;担体、レセプタ類およびリ
ガンド類に関するものである。
これらの成分のそれぞれに関する知識は拡大している(教科書的情報については
、“The molecular biologyof the cell”
、Albertら、Garland publ、inc、NYおよびLondo
n、1989の12章および14章参照)。この情報に基づけば、本発明に従っ
て医薬調製物を調製した場合に、上述したペプチド類について徐放が起こるであ
ろうことが明らかである。
ll例えば、ヒアルロン酸等の担体ゲルとレセプタ蛋白質との間の交差結合は、
種々の手段、例えばイミドカルボネート類、カルボネート類、オキソラン類、ア
ジリジン類ならびに活性化二重結合およびハロゲン類等を介して達成されつる。
このような方策は、以前にポリサッカライドゲル上に酵素類を固定化するために
使用されている。
2、 定義により、ホルモンまたは成長因子のレセプタは、そのリガンドを高い
親和性により、かつ可逆的方法で特異的に結合し、また会合速度は解離速度より
速い。
これらのレセプタの特徴である高い親和性、高い特異性および可逆性は、レセプ
タという用語を使用するために−必須である。解離速度は、通常はレセプタを放
射標識リガンド類と平衡に達するまで熟成することにより測定される。過剰量の
非標識リガンドを添加し、非−レセプタ結合放射能の放出を時間の関数として測
定することにより解離速度か明らかになる。上述したことから、担体とレセプタ
、リガンドとの組合せは、一般的に許容される原理として該リガンドの徐放が明
らかであろう。本発明は、レセプタ類を治療的環境に導入するものであり、加え
てこれは徐放を許容する担体を定義するものである。
以下は、模式的な応用の概要を記述するもので、本発明による医薬調製物の製造
のための原理を明らかにするものである。
例: 徐放性GH−複合体の調製
a、OHレセプタの細胞外領域の単離を、MTプロモータにより制御される発現
ベクター中に挿入されたGHレセプタcDNAの部分を使用することにより行な
った。
感染細胞培地からの切断レセプタの精製を、アフイニティクロマトグラフィ(h
GHセファロース■使用)、次いて大きさ分離ゲルクロマトグラフィにより行な
った。
b、ヒアルロン酸は、商業的に入手可能であり、精製レセプタを交差結合試薬(
上記参照)を使用して結合した。該高分子量の複合体を、残留するOHレセプタ
から、遠心分離を反復して精製した。約75%の交差結合レセプタか機能的に完
全であった。
C1該交差結合ヒアルロン酸−OHレセプタ調製物を、過剰量のGHと共に37
°Cにて2時間熟成し、非結合ホルモンを遠心分離により除去した。
d、 該調製物は、例えば皮下的に注射された場合に、投与量に依存して、GH
の量のみならずゲルに結合したGH−レセプタの数にも従ってGHを緩慢に放出
するであろう。与えられた調製物の解離速度は、各バッチについてインビトロに
おいて免疫反応性GHの放出を試験することによって(あるいは動物モデルにお
いて放射性GHの放出を試験することによって)決定される。
A、 下垂体切除(HX)ラットの群において異なった型のGH−処置に続く体
重増加測定の実験を行った。3匹のHXラットに、約1.2■のレセプタ結合G
Hを含む本発明によるヒアルロン酸−〇Hレセプター〇H調製物を、それぞれ皮
下的に注射投与した。また3匹のHXラットに、同量のGHを水溶液の形態で皮
下的に注射投与し、対照とした。徐放処置の群は、対照に比べて4日間長く継続
する官憲な体重増加を示した。
B、ラットの群に、6マイクログラムのI GF−1+IGF−1結合蛋白質+
ヒアルロン酸または6マイクログラムのIGF−1の水溶液のいずれかを、尾部
に注射した。使用したIGF−1は、痕跡量(200000cpm)の放射標識
IGF−1を含有していた。各群について3匹のラッI・を使用した。異なった
時点後に、尾の端部を切断し、放射能分析を行なった。ラットの第1群は、対照
に比べて官憲に緩慢(2日より長い)にICF−1を放出することが見出された
。

これらのすべては、体内で加水分解により分解される。
異なるポリマー類は、それらの構造的および化学的面に5 おいて異なり、この
ことはそれぞれに異なった強度、作用、分解時間、および有用性を与える。例え
ば、PDSは、吸収性の縫合材料として使用されている。PGAは、例えばBi
ofixO等のロッド、板体、ネジとして骨接合材料に使用されている。PLL
A帯は、研究において例えば前原十字靭帯の置換物として使用されている。
異なった原理の組合せ、すなわち安定化骨接合材料の例えば骨折治癒促進性ペプ
チド類局所的徐放との組合せは魅力的である。構造的に多孔性であり、異なった
被覆程度を有するポリマー類は、成長因子+レセプタ複合体と の吸収を促進す
る。
C9上記の原理に従った実験において、PGAロッド(1,5X8m)に放射標
識ICF−1+ IGF−1結合性蛋白質(B節参照)を真空力を用いて負荷し
、端部を溶融によ封止した。該ロッドを3匹のラットの尾の端部に移植した。3
匹の対照ラットには、水溶液中の等量の放射標識IGF−1を用いて尾の同じ部
位に注射投与した。本発明による徐放性のラットは、対照に比べて有意に高い放
射活性を尾部において有していた。

Claims (5)
- 1.1種または数種のレセプタ/結合蛋白質類(成長因子類またはホルモン類結 合性)およびヒアルロン酸(もしくはその誘導体類)または生物分解可能なポリ マー類の組合せ、場合によりそれ/それらのリガンド類(成長因子類およびホル モン類)の組合せであることを特徴とする医薬調製物。
- 2.該レセプタ類/結合蛋白質類が、ヒアルロン酸または生物分解可能なポリマ ーに混合されるか、または共有結合されている請求項1に記載の医薬調製物。
- 3.該レセプター結合蛋白質類が、 インシュリン様成長因子−1−レセプタ、インシュリン様成長因子−2−レセプ タ、インシュリンレセプタ、 血小板誘導成長因子レセプタ、 線維芽成長因子−レセプタ、 表皮成長因子−レセプタ、 神経成長因子−レセプタ、 コロニー刺激因子−レセプタ、 転換成長因子−レセプタ、 成長ホルモン−レセプタ、 上皮小体ホルモン−レセプタ、 カルシトニン−レセプタ、 エストロゲン−レセプタ、 腫瘍壊死因子−レセプタ、 インシュリン様成長因子血清結合蛋白質、コルチコステロイド結合グロブリン であることを特徴とする請求項1に記載の医薬調製物。
- 4.該生物分解可能なポリマー類が、 ポリグリコライド(PGA)、 グリコライドのコポリマー類: グリコライド/L−ラクチドコポリマー類(PGA/PLLA)、 グリコライド/トリメチレンカルボネートコポリマー類(PGA/TMC)、 ポリラクチド類(PLA)、 PLAのステレオーコポリマー類、 ポリ−L−ラクチド(PLLA)、 ポリ−DL−ラクチド(PDLLA)、L−ラクチド/DL−ラクチドコポリマ ー類、PLAのコポリマー類: ラクチド/テトラメチルグリコライドコポリマー類、ラクチド/トリメチレンカ ルボネートコポリマー類、ラクチド/δ−バレロラクトンコポリマー類、ラクチ ド/ε−カプロラクトンコポリマー類、ポリデプシペプチド類、 PLA/ポリエチレンオキシドコポリマー類、非対称3,6−置換ポリ−1,4 −ジオキサン−2,5−ジオン類、 ポリ−β−ヒドロキシブチレート(PHBA)、PHBA/β−ヒドロキシバレ レートコポリマー類(PHBA/HVA)、 ポリ−p−ジオキサノン(PDS)、 ポリ−δ−バレロラクトン、 ポリ−ε−方ブロラタトン、 メチルメタクリレート−N−ビニルピロリジンコポリマー類、 ポリエステルアミド類、 シュウ酸のポリエステル類、 ポリジヒドロピラン類、 ポリアルキル−2−シアノアクリレート類、ポリウレタン類、 ポリビニルアルコール(PVA)、 ポリペプチド類、 ポリ−β−リンゴ酸(PMLA)、 ポリ−β−アルカン酸類、 であることを特徴とする請求項1に記載の医薬調製物。
- 5.該リガンド類が、 インシュリン様成長因子−1および2:IGF−1、IGF−2、 血小板誘導成長因子;PDGF、 表皮成長因子;EGF、 線維芽成長因子;FGF、 神経成長因子;NGF、 コロニー刺激因子;CSF、 転換成長因子;TGF、 腫瘍壊死因子;TNF、 カルシトニン;CT、 上皮小体ホルモン;PTH、 成長ホルモン;GH、 エストロゲン類、 ボンベシン、 骨形態発生蛋白質;BMP、 コルチコステロイド類、 インシュリン、 であることを特徴とする請求項2に記載の医薬調製物。
