Description
Claims (12)
- 1.サンプル中の抗凝固剤の濃度を前記サンプル中のトロンビン生成阻害の関数 として測定する方法であって、a.前記サンプルを希釈するのに充分な量の緩衝 液を添加すること、 b.前記サンプルを前記サンプル中のトロンビン生成を阻害するのに充分な量の 凝固因子試薬と混合すること、c.凝固カスケードを開始させるのに充分な量の 賦活剤試薬を添加すること、 d.トロンビンと複合体を形成するのに充分な量の緩衝化された検出試薬が添加 すること、 e.前記サンプル中のトロンビン濃度を抗凝固剤の既知量と相関させ、前記サン プル中の抗凝固剤の濃度を決定すること、よりなる前記方法。
- 2.前記凝固因子試薬は、サンプル中に存在する前記凝固因子の濃度より大きい 与えられた量の凝固因子と、前記因子を安定化するのに充分な量のカルシウムイ オンと、反応を触媒するのに充分な量のリン脂質と、前記サンプル中の血小板因 子4を中和するのに充分な量の硫酸デキストランと、前記サンプル中のトロンビ ン生成を抑制するのに充分な量の弱いトロンビン阻害剤とよりなる請求項1の方 法。
- 3.前記抗凝固剤は、ヘパリン、α−NAPAPおよび硫酸デルマタンよりなる 群から選ばれる請求項2の方法。
- 4.前記弱いトロンビン阻害剤は、抗トロンビンIII,ヘパリン助因子II、 または抗凝固剤によって増強される合成もしくは生理トロンビン阻害剤からなる 群から選ばれる請求項3の方法。
- 5.前記凝固因子試薬は、サンプル中に存在する前記凝固因子の濃度より大きい 与えられた量の凝固因子と、前記因子を安定化するのに充分な量のカルシウムイ オンと、反応を触媒するのに充分な量のリン脂質と、前記サンプル中の血小板因 子4を中和するのに充分な量の硫酸デキストランとよりなる請求項1の方法。
- 6.前記抗凝固剤は、ヒルジンおよび合成もしくは生理的トロンビン阻害剤より なる群から選ばれる請求項5の方法。
- 7.前記凝固因子は、因子X,因子VIII,因子V,因子II,因子IXまた は因子IX−因子XI複合体よりなる群から選ばれる請求項1の方法。
- 8.前記賦活剤試薬は、凝固カスケードを活性化するのに充分な量の因子IXa ,CaCl2および不活性タンパクを含んでいる請求項1の方法。
- 9.前記緩衝化基質試薬は、色素原性、発光原性、螢光原性または電磁性である 請求項1の方法。
- 10.前記緩衝化試薬はフィブリノーゲンである請求項1の方法。
- 11.a.前記凝固因子試薬を含んでいる第1の容器と、b.前記賦活剤試薬を 含んでいる第2の容器と、c.前記緩衝化試薬を含んでいる第3の容器からなる 請求項1の方法を実施するためのキット。
- 12.a.前記凝固因子試薬および緩衝化試薬を含んでいる第1の容器と、 b.ある凝固因子試薬を含んでいる第2の容器からなる請求項1の方法を実施す るためのキット。
