【発明の詳細な説明】
発明の背景
A 発明の分野
この発明は、ヒトを含む動物体における新生物
細胞の増殖を阻害するため、及び腫瘍状態又は癌
状態に感受性の個体の遺伝的又は環境的素質に由
来する免疫系不調の治療のための、インターフエ
ロン(IFN)と組合わせた又はインターフエロン
を伴わない二重鎖リボース核酸(ds RNA)の使
用に関する。この発明はさらに、腫瘍状態又は癌
状態を治療するための、他の化学的薬剤及び生物
学的薬剤と併用するds RNAの使用に関する。 B 発明の背景 IFNの機能及びIFN誘発物質としてのds
RNA IFNは、動物細胞中でウイルスの増殖を阻害
することができる蛋白質である。これらは細胞
に由来し、試験管内又は生体内で生産され、そ
して非常に広範囲の物質により刺激される
〔Encyclopedia of Biochemistry,ロジヤー・
ウイリアムス(Roger Williams)及びエドウ
イン・ランスフオード(Edwin Lansford)
編、レインフオールド・パブリツシング
(Reinhold Pablishing)社、1967年〕。IFNが
抗ウイルス作用のほかに抗細胞増殖作用を有す
ることが知られている(Nature、262、300、
1976年;Exptl.Cell Res.,121、111、1979
年)。さらに、インターフエロンがプラスミノ
ーゲン活性化物質の分泌を低下せしめ
(Nature、276、828、1978年)、接触阻害を確
立し(J.Cell Biol.,56、846、1973年)、そし
て半固形寒天中での新生物細胞の増殖を阻害す
る(Nature、231、20、1971年)ことが知られ
ている。 ds RNAは種々の分子形(白血球性及び線維
芽細胞性を含む)及び免疫型のヒト−IFNのイ
ンデユーサーであつて(J.Reticuloendothelial
Society、23、299、1978)、天然ds RNA及び
合成ds RNAのいずれもIFN生産を刺激するこ
とができる。インターフエロン誘発物質として
のポリリボイノシン酸とポリリボシチジン酸の
複合体の形の合成ds RNA(rIo・rCo)の使用
が、例えばランプソン(Lampson)等の米国
特許第3666646号において知られている。ds
RNAはIFN誘発物質であるほか、IFNに誘導
される2種類の酵素、すなわちプロテインキナ
ーゼ及び2′−5′オリゴアデニレートシンセター
ゼの活性化物質でもある(Pro.natl.Acad.Sci.,
75,5893,1978年)。しかしがら、インターフ
エロンの抗細胞増殖作用及び抗新生物作用の分
子的基礎は未知であり、そしてこの発明に先立
つてインターフエロン誘発物質としての機能以
外のds RNAの抗細胞増殖効果もなんら知られ
ていなかつた。 ds RNAの合成不均整類似体 さらに最近、Ts′o及びカーター(Carter)
の米国特許第4130641号、及び第4024222号に開
示されているごとく、IFNはrIo・rCoの不均整
類似体によつても誘発され得ることが見出され
た。これらの特許の記載を引用によりこの明細
に組み入れる。これらの類似体は、ポリリポシ
チジレート(rCo)鎖にそつて非対塩基(ウラ
シル又はグアニン)を導入することにより
rIo・rCoを変形することにより、又はポリリポ
イノシン酸(rIo)のリポース主鎖を変形する
ことにより形成される。不均整類似体、特に
rCo鎖において変形された類似体は、第1図に
示すごとき一定の構造的要求に合致する限りイ
ンターフエロン誘発能力を保有する。第1図に
おいて、完全な塩基対からなるds RNAの長い
領域はインターフエロン誘発のために必要でな
い。実際に、完全な塩基対ds RNAの1/2〜1
回転の螺旋が保持されていればインターフエロ
ン合成を開始する(引金を引く)ための構造上
の先行条件が満たされる。 発明者等は、不均整重合体がヌクレアーゼに
暴露された場合加水分解速度は上昇する(J.
Molec.Biol.,70、567、1972年)が、これらの
ds RNAは、インターフエロンの誘発に関して
変形されていないrIo・rCoとほとんど異らない
活性を有することを見出した。障害を受けてい
ない二重螺旋構造が長時間保持されることによ
り、ds RNAに対する薬剤毒性として一般に記
載されている他の多くの生物学的反応が生ずる
ことが報告された。種々の動物系における不均
整類似体rIo・r(C12、U)o〔アンプリゲン
(Ampligen)、HEMリザーチ社の商標〕の研
究により、 rIo・r(C12、U)oはrIo・rCoに比べて毒性が
少ないことが示された〔Poly IC with
mismatchedbases,prospects for cancer
therapy,Augmenting Agents in Cancer
Therapy,E.M.ハーシユ(Hersh)等編、レ
ーブン・プレス(Raven Press)、ニユーヨー
ク、177、1981年)。特に、rIo・r(C12、U)o
をマウスに反復投与する場合、骨髄要素、肝細
胞、胸線細胞及び脾細胞を含む非常に感受性の
器官に対する毒性が非常に低く(J.Moles.
Biol.,70、567、1972年)、他方種々の致命的
なウイルスの攻撃に対する保護において有効で
ある(Moles.Pharm.,12、299、1976年)。 さらに、rIo・r(C12、U)oはrIo・rCoに比べ
て、家兎において低下した発熱性(1/100に低
下)、マウス及びヒトの細胞を用いる試験管内
試験における低い有糸分裂毒性(1/5〜1/10に
低下)、及び家兎及びマウスにおける低下した
ds RNA抗体生産性(1/5〜1/10に低下)を示
す。この低下した細胞毒性の背景に対して、
rIo・r(C12、U)oはインターフエロンを誘発
し、天然キラー(NK)細胞機能を刺激し(J.
Immuno、124、1852、1980)、そして前記の細胞
内媒介体である2′−5′−オリゴ−アデニレート
シンセターゼ及び特異的プロテインキナーゼを
活性化する(共同発明者の未発表データ)。従
つて、種々の試験系(家兎、マウス及びヒト)
において、そして、種々の毒性及び治療効果を
測定するための種々の投与量において、不均整
類似体rIo・r(C12、U)n(アンプイゲン)は
強化された治療率を有することが明らかであ
る。この不均整誘発物質/活性化物質の調製及
び製剤はすでに記載されている(J.Molec.Bio.
70、567、1972)。 動物細胞内でインターフエロン生産を誘発し
てウイルスによる攻撃に対抗することのみを目
的とする変形rIo・rCo類似体の使用について
は、例えば前記の米国特許第4130641号及び第
4024222号に記載されている。しかしながら、
インターフエロンの抗細胞増殖剤としての使用
の観点からは、多くの個体においてIFNは周辺
的効果のみ生じさせるというIFN療法に対する
障害が存在する。従つて、比較的多量のインタ
ーフエロンを投与する必要があり、このために
通常のウイルス感染に対するヒトの通常の反応
の過程で生ずることがない程不自然に高い体内
IFNレベルが生ずる。このような高いレベルに
おいては、体はIFNを外来物質として扱うこと
ができ、そして処理された個体はIFNに対する
抗体を形成する能力を有する。 さらに、特に新生物細胞は、IFNの治療的性
質に対する耐性を獲得する能力を有する。後記
のごとく、IFNに対する耐性を発生せしめる新
生物のこの能力は非−無作為的表現型(non−
randomphenotype)として獲得され、そして
それ故にIFNのみの使用に基く細胞増殖の阻害
を意図する治療方法に対して深刻な欠点をもた
らす。この発明は、細胞がIFN耐性を獲得する
か否かにかかわらず癌細胞の増加を停止せしめ
又は劇的に妨害する新規な抗細胞増殖剤及び組
成物を開示することにより従来技術が有する前
記の欠点を除去する。さらにこの発明は、従来
技術のより達成されるレベルに比べてインター
フエロンの体内レベルが20〜50の係数をもつて
低いが、非常に大きな治療効果をもたらすイン
ターフエロン含有組成物を提供する。ds RNA
を含有するこの発明の組成物は相剰的に作用す
るため投与に必要なds RNAレベルも同時に従
来技術により開示されているそれよりも低い。 腫瘍細胞に対する免疫監視機構としての天然
キラー(NK)細胞 過去2年間に、新生物細胞に対する免疫監視
機構におけるNK細胞集団の中心的役割を支持
する多くの報告が発表された。高いNK−細胞
活性を有するマウスは低いNK−細胞活性を有
するマウスに比べてNK−感受性腫瘍細胞の増
殖に対し高い耐性を有する(Immunol.Rev.,
44、165、1979)。さらに最近、選択的NK細胞
欠損を有するベージユマウスが、正常なNK細
胞活性を有する同質遺伝子的マウスに比べて腫
瘍増殖に対して感受性であることが報告された
〔Nature(ロンドン)、284、622、1980;
Nature、284、624、1980〕。チエデイアク−ヒ
ガシ(Chediak−Higashi)症候群を有するヒ
トにおいても同様の状態が存在し、そしてこの
疾患におけるNK細胞欠損は原因として、これ
らの患者におけるリンパ腫の発達と関連するこ
とが示唆された〔J.Exp.Med.,151、1939、
1980;J.Exp.Med.,151、1049、1980;
Nature(ロンドン)、284、553、1980〕。慢性リ
ンパ球性白血病(CLL)患者は二次悪性腫瘍
の高い発生率を有することが知られている。最
近、チグラー(Zigler)及びツアーリング
(Zarling)(Int.J.Cancer、27、321、1981)は、
白血病細胞を取り出した後、慢性白血病患者か
らのリンパ球中のNK細胞活性を評価した。
CLL患者の多くは、NK−感受性標的に結合し
たリンパ球の存在にもかかわらず、最小の又は
検知できないNK細胞活性を有する。さらに、
多量の免疫抑制剤により処理された賢臓移植受
容体は、約100倍高い二次悪性腫瘍発生の危険
性を有し、そして又非常に抑制されたNK細胞
活性を有し又はこの活性を喪失している
(Transplantatin、29、214、1980)。これに対
して、これらの受容体において、抗体依存性細
胞性細胞毒性 〔antibody−dependent cell−mediated cylotoxicity(ADCC)〕は抑制されない。この
ことは、NK細胞活性の欠損(又は損傷)は非
常に特異的であり、そして現在のところ、高い
二次悪性腫瘍発生傾向と関連しているように思
われる。 従つて、幾つかの別個の研究室によりなされ
た最近の証明により、ヒトにおける新生物に対
する免疫監視機構としてのNK細胞系の重要性
が確認される。発明者等はここに、ヒトにおけ
るNK活性の欠損が実際にヒト−癌を誘導し、
そしてこの欠損はインターフエロン単独ではな
くある種の不均整ds RNAを併用することによ
り完全に修正され得ることを発見した。 NK細胞機能の内的制御物質としてのIFN インターフエロンのごとき天然制御物質の主
要な有用性は、腫瘍クローン(これが発生する
場合には)の生理的コントロールを強化するこ
とにより、そしてNK細胞系のごとき天然免疫
経路を強化することによつて明白な二次腫瘍発
生過程を防止する点にある。発明者等は、内生
インターフエロンが存在しない場合これらの経
路の機能は限界レベルより低くなつており、又
は全く休止していると推論する。さらに、幾つ
かの別個の研究室による最近の証明により、
NK細胞系の内性活性化物質としてのインター
フエロンの役割が確認されつつある。ハンナ
(Hanna)及びフイルダー(Fildler)(J.Nat.
Cancer Inst.,64(4)、80、1980)は、低NK細
胞活性を有する3週令マウスをインターフエロ
ン誘発物質で処理することによりNK細胞活性
が増強され、そして転移形成(腫瘍の核酸)が
阻害させることを示した。ライド(Reid)等
(Proc.Natl.Acad.Sci.,USA、78(2)、1171、
1981)は、ヌードマウスを抗−マウスインター
フエロン抗血清で処理することによりインター
フエロン生産及びNK細胞活性の両者を低下せ
しめた。さらに、抗−インターフエロン処理に
よりヒト−腫瘍移植成功率、及びヌードマウス
中での増殖が非常に増加した。この結果は、非
常なレベルの内生インターフエロンが、NK細
胞のごとき免疫系成分を介して間接的に作用す
ることにより、腫瘍細胞の増殖、侵入及び転移
において重要な役割を演することと一致する。
最後に、エドワード(Edwards)及びボーデ
ン(Borden)(Proceedings AACR、1153、
1981年)は、NK細胞を抗−インターフエロン
抗血清とプレインキユベートすることにより正
常なヒトのNK細胞活性が減少し又は消滅する
ことを示した。すなわち、ヒトにおいてNK細
胞が正常に機能するためには細胞表面に結合し
たインターフエロンが常に存在することが必須
のようである。すなわち、多くの研究室による
最近の証明により、天然キラー細胞系の内生制
御物質としてのインターフエロンの詳細な役割
が確認されつつある。IFNの内生誘発の概略を
第1表に示す。 【表】 ーフエロンを生産す
る。
細胞の増殖を阻害するため、及び腫瘍状態又は癌
状態に感受性の個体の遺伝的又は環境的素質に由
来する免疫系不調の治療のための、インターフエ
ロン(IFN)と組合わせた又はインターフエロン
を伴わない二重鎖リボース核酸(ds RNA)の使
用に関する。この発明はさらに、腫瘍状態又は癌
状態を治療するための、他の化学的薬剤及び生物
学的薬剤と併用するds RNAの使用に関する。 B 発明の背景 IFNの機能及びIFN誘発物質としてのds
RNA IFNは、動物細胞中でウイルスの増殖を阻害
することができる蛋白質である。これらは細胞
に由来し、試験管内又は生体内で生産され、そ
して非常に広範囲の物質により刺激される
〔Encyclopedia of Biochemistry,ロジヤー・
ウイリアムス(Roger Williams)及びエドウ
イン・ランスフオード(Edwin Lansford)
編、レインフオールド・パブリツシング
(Reinhold Pablishing)社、1967年〕。IFNが
抗ウイルス作用のほかに抗細胞増殖作用を有す
ることが知られている(Nature、262、300、
1976年;Exptl.Cell Res.,121、111、1979
年)。さらに、インターフエロンがプラスミノ
ーゲン活性化物質の分泌を低下せしめ
(Nature、276、828、1978年)、接触阻害を確
立し(J.Cell Biol.,56、846、1973年)、そし
て半固形寒天中での新生物細胞の増殖を阻害す
る(Nature、231、20、1971年)ことが知られ
ている。 ds RNAは種々の分子形(白血球性及び線維
芽細胞性を含む)及び免疫型のヒト−IFNのイ
ンデユーサーであつて(J.Reticuloendothelial
Society、23、299、1978)、天然ds RNA及び
合成ds RNAのいずれもIFN生産を刺激するこ
とができる。インターフエロン誘発物質として
のポリリボイノシン酸とポリリボシチジン酸の
複合体の形の合成ds RNA(rIo・rCo)の使用
が、例えばランプソン(Lampson)等の米国
特許第3666646号において知られている。ds
RNAはIFN誘発物質であるほか、IFNに誘導
される2種類の酵素、すなわちプロテインキナ
ーゼ及び2′−5′オリゴアデニレートシンセター
ゼの活性化物質でもある(Pro.natl.Acad.Sci.,
75,5893,1978年)。しかしがら、インターフ
エロンの抗細胞増殖作用及び抗新生物作用の分
子的基礎は未知であり、そしてこの発明に先立
つてインターフエロン誘発物質としての機能以
外のds RNAの抗細胞増殖効果もなんら知られ
ていなかつた。 ds RNAの合成不均整類似体 さらに最近、Ts′o及びカーター(Carter)
の米国特許第4130641号、及び第4024222号に開
示されているごとく、IFNはrIo・rCoの不均整
類似体によつても誘発され得ることが見出され
た。これらの特許の記載を引用によりこの明細
に組み入れる。これらの類似体は、ポリリポシ
チジレート(rCo)鎖にそつて非対塩基(ウラ
シル又はグアニン)を導入することにより
rIo・rCoを変形することにより、又はポリリポ
イノシン酸(rIo)のリポース主鎖を変形する
ことにより形成される。不均整類似体、特に
rCo鎖において変形された類似体は、第1図に
示すごとき一定の構造的要求に合致する限りイ
ンターフエロン誘発能力を保有する。第1図に
おいて、完全な塩基対からなるds RNAの長い
領域はインターフエロン誘発のために必要でな
い。実際に、完全な塩基対ds RNAの1/2〜1
回転の螺旋が保持されていればインターフエロ
ン合成を開始する(引金を引く)ための構造上
の先行条件が満たされる。 発明者等は、不均整重合体がヌクレアーゼに
暴露された場合加水分解速度は上昇する(J.
Molec.Biol.,70、567、1972年)が、これらの
ds RNAは、インターフエロンの誘発に関して
変形されていないrIo・rCoとほとんど異らない
活性を有することを見出した。障害を受けてい
ない二重螺旋構造が長時間保持されることによ
り、ds RNAに対する薬剤毒性として一般に記
載されている他の多くの生物学的反応が生ずる
ことが報告された。種々の動物系における不均
整類似体rIo・r(C12、U)o〔アンプリゲン
(Ampligen)、HEMリザーチ社の商標〕の研
究により、 rIo・r(C12、U)oはrIo・rCoに比べて毒性が
少ないことが示された〔Poly IC with
mismatchedbases,prospects for cancer
therapy,Augmenting Agents in Cancer
Therapy,E.M.ハーシユ(Hersh)等編、レ
ーブン・プレス(Raven Press)、ニユーヨー
ク、177、1981年)。特に、rIo・r(C12、U)o
をマウスに反復投与する場合、骨髄要素、肝細
胞、胸線細胞及び脾細胞を含む非常に感受性の
器官に対する毒性が非常に低く(J.Moles.
Biol.,70、567、1972年)、他方種々の致命的
なウイルスの攻撃に対する保護において有効で
ある(Moles.Pharm.,12、299、1976年)。 さらに、rIo・r(C12、U)oはrIo・rCoに比べ
て、家兎において低下した発熱性(1/100に低
下)、マウス及びヒトの細胞を用いる試験管内
試験における低い有糸分裂毒性(1/5〜1/10に
低下)、及び家兎及びマウスにおける低下した
ds RNA抗体生産性(1/5〜1/10に低下)を示
す。この低下した細胞毒性の背景に対して、
rIo・r(C12、U)oはインターフエロンを誘発
し、天然キラー(NK)細胞機能を刺激し(J.
Immuno、124、1852、1980)、そして前記の細胞
内媒介体である2′−5′−オリゴ−アデニレート
シンセターゼ及び特異的プロテインキナーゼを
活性化する(共同発明者の未発表データ)。従
つて、種々の試験系(家兎、マウス及びヒト)
において、そして、種々の毒性及び治療効果を
測定するための種々の投与量において、不均整
類似体rIo・r(C12、U)n(アンプイゲン)は
強化された治療率を有することが明らかであ
る。この不均整誘発物質/活性化物質の調製及
び製剤はすでに記載されている(J.Molec.Bio.
70、567、1972)。 動物細胞内でインターフエロン生産を誘発し
てウイルスによる攻撃に対抗することのみを目
的とする変形rIo・rCo類似体の使用について
は、例えば前記の米国特許第4130641号及び第
4024222号に記載されている。しかしながら、
インターフエロンの抗細胞増殖剤としての使用
の観点からは、多くの個体においてIFNは周辺
的効果のみ生じさせるというIFN療法に対する
障害が存在する。従つて、比較的多量のインタ
ーフエロンを投与する必要があり、このために
通常のウイルス感染に対するヒトの通常の反応
の過程で生ずることがない程不自然に高い体内
IFNレベルが生ずる。このような高いレベルに
おいては、体はIFNを外来物質として扱うこと
ができ、そして処理された個体はIFNに対する
抗体を形成する能力を有する。 さらに、特に新生物細胞は、IFNの治療的性
質に対する耐性を獲得する能力を有する。後記
のごとく、IFNに対する耐性を発生せしめる新
生物のこの能力は非−無作為的表現型(non−
randomphenotype)として獲得され、そして
それ故にIFNのみの使用に基く細胞増殖の阻害
を意図する治療方法に対して深刻な欠点をもた
らす。この発明は、細胞がIFN耐性を獲得する
か否かにかかわらず癌細胞の増加を停止せしめ
又は劇的に妨害する新規な抗細胞増殖剤及び組
成物を開示することにより従来技術が有する前
記の欠点を除去する。さらにこの発明は、従来
技術のより達成されるレベルに比べてインター
フエロンの体内レベルが20〜50の係数をもつて
低いが、非常に大きな治療効果をもたらすイン
ターフエロン含有組成物を提供する。ds RNA
を含有するこの発明の組成物は相剰的に作用す
るため投与に必要なds RNAレベルも同時に従
来技術により開示されているそれよりも低い。 腫瘍細胞に対する免疫監視機構としての天然
キラー(NK)細胞 過去2年間に、新生物細胞に対する免疫監視
機構におけるNK細胞集団の中心的役割を支持
する多くの報告が発表された。高いNK−細胞
活性を有するマウスは低いNK−細胞活性を有
するマウスに比べてNK−感受性腫瘍細胞の増
殖に対し高い耐性を有する(Immunol.Rev.,
44、165、1979)。さらに最近、選択的NK細胞
欠損を有するベージユマウスが、正常なNK細
胞活性を有する同質遺伝子的マウスに比べて腫
瘍増殖に対して感受性であることが報告された
〔Nature(ロンドン)、284、622、1980;
Nature、284、624、1980〕。チエデイアク−ヒ
ガシ(Chediak−Higashi)症候群を有するヒ
トにおいても同様の状態が存在し、そしてこの
疾患におけるNK細胞欠損は原因として、これ
らの患者におけるリンパ腫の発達と関連するこ
とが示唆された〔J.Exp.Med.,151、1939、
1980;J.Exp.Med.,151、1049、1980;
Nature(ロンドン)、284、553、1980〕。慢性リ
ンパ球性白血病(CLL)患者は二次悪性腫瘍
の高い発生率を有することが知られている。最
近、チグラー(Zigler)及びツアーリング
(Zarling)(Int.J.Cancer、27、321、1981)は、
白血病細胞を取り出した後、慢性白血病患者か
らのリンパ球中のNK細胞活性を評価した。
CLL患者の多くは、NK−感受性標的に結合し
たリンパ球の存在にもかかわらず、最小の又は
検知できないNK細胞活性を有する。さらに、
多量の免疫抑制剤により処理された賢臓移植受
容体は、約100倍高い二次悪性腫瘍発生の危険
性を有し、そして又非常に抑制されたNK細胞
活性を有し又はこの活性を喪失している
(Transplantatin、29、214、1980)。これに対
して、これらの受容体において、抗体依存性細
胞性細胞毒性 〔antibody−dependent cell−mediated cylotoxicity(ADCC)〕は抑制されない。この
ことは、NK細胞活性の欠損(又は損傷)は非

- 【特許請求の範囲】 1 dsRNAと、インターフエロン又はヘミンと
の混合物を含んで成る癌治療用医薬組成物。 2 前記dsRNAがrIn・rCnである、特許請求の
範囲第1項に記載の医薬組成物。 3 前記インターフエロンが天然インターフエロ
ンであり、そしてヒト繊維芽細胞インターフエロ
ン又はヒト白血球インターフエロンである、特許
請求の範囲第1項に記載の医薬組成物。 4 前記インターフエロンがハイブリド形であ
り、そして部分的にα−インターフエロンに由来
し、そして部分的にβ又はγ−インターフエロン
に由来するものである、特許請求の範囲第1項に
記載の医薬組成物。 5 前記インターフエロンが天然形であつて組換
えDNA技法によるものであるか、又はハイブリ
ド形である、特許請求の範囲第1項に記載の医薬
組成物。 6 前記dsRNAがミスマツチ形である、特許請
求の範囲第1項に記載の組成物。 7 前記ミスマツチ形dsRNAがrIn・r(C12、
U)oである、特許請求の範囲第6項に記載の組成
物。
