Description
Claims (10)
- 1.以下の工程より成る、塩基性線維芽細胞増殖因子蛋白を安定化する方法: 塩基性線維芽細胞増殖因子(bFGF)のアミノ酸配列を有する影響を受けやす いペプチドを、該影響を受けやすいペプチドに含まれている少なくとも一つ以上 のシステインとS−S共有結合を形成できる、該bFGFの多量体化を防止する のに有効な量の保護試薬を用いて、pH6あるいはそれ以上であって前記bFG Fが変質しないpHの緩衝液の存在下で、該安定化操作が効果を発揮するのに十 分な時間、処理する工程。
- 2.前記保護試薬が、各Rが独立して、該保護試薬に水溶性を賦与するのに十分 な親水性をもつ有機置換基である、構造式RSSRの化合物である、請求項1の 記載の方法。
- 3.前記の構造式RSSRの化合物が、グルタチオンジスルフィド及びシスチン から成る群から選択される、請求項2に記載の方法。
- 4.前記pHが、pH6から8である、請求項1に記載の方法。
- 5.さらに、処理したbFGFをクロマトグラフィーによる精製に共する工程を 包含する、請求項1に記載の方法。
- 6.請求項1に記載の方法で調製される安定化した塩基性FGFであって、図8 あるいは図9に示した上部の番号の第24〜155番の残基で示されるアミノ酸 配列を含む場合には、前記保護試薬がグルタチオンジスルフィドではない、bF GF。
- 7.請求項5に記載の方法で調製される安定化した塩基性FGFであって、図8 あるいは図9に示した上部の番号の第24〜155番の残基で示されるアミノ酸 配列を含む場合には、前記保護試薬がグルタチオンジスルフィドではない、bF GF。
- 8.影響を受けやすい塩基性線維芽細胞増殖因子(bFGF)蛋白、および、該 影響を受けやすいbFGFに含まれている少なくとも一つ以上のシステインとS −S共有結合を形成できる、該bFGFを安定化するのに有効な量の保護試薬、 を含有する水性溶媒を含む組成物であって、該水性溶媒のpHが、少なくとも6 で該bFGFが変性しないpHを有する、組成物。
- 9.前記の保護試薬が、各Rが独立して、該保護試薬に水溶性を賦与するのに十 分な親水性をもつ有機置換基である、構造式RSSRの化合物である、請求項8 に記載の組成物。
- 10.前記の構造式RSSRの化合物が、グルタチオンジスルフィド及びシスチ ンから成る群から選択される、請求項9に記載の組成物。
