Description
Claims (18)
- 1.リガンドとの相互作用を調べることによる巨大分子の特性決定の方法であっ て、 前記方法は巨大分子を少なくとも1つのリガンドと相互作用させた後、巨大分子 を少なくとも1つの追加のリガンドと接触させ、巨大分子又は追加のリガンド( 1つ又は複数)はセンサー表面に結合しており、センサー表面の物理化学的性質 に結果として起こる変化を検出することにより相互作用を測定し、及び測定した リガンド相互作用の間の相互依存に基づいて巨大分子のエピトープを識別し及び それらの相対的位置をマッピングすることによりリガンド相互作用の相互の影響 を測定することを特徴とする巨大分子の特性決定の方法。
- 2.a)巨大分子をセンサー表面に結合させ、b)少なくとも2つのリガンドを センサー表面に結合させた巨大分子に連続的に接触させ、及びc)各リガンドを 巨大分子を持つセンサー表面と接触させた後それぞれのリガンドの巨大分子との 相互作用をセンサー表面の物理化学的性質に結果として起こる変化を検出するこ とにより測定する段階からなることを特徴とする請求の範囲第1項記載の方法。
- 3.a)リガンドをセンサー表面に結合させ、b)巨大分子をセンサー表面に結 合させる巨大分子がセンサー表面に結合させたリガンドと相互作用することを許 容し、 c)1つ又はそれより多い追加のリガンドをセンサー表面に結合させた巨大分子 と連続的に接触させ、及び d)各々の追加のリガンドを巨大分子を持つセンサー表面と接触させた後、それ ぞれのリガンドの巨大分子との相互作用をセンサー表面の物理化学的性質に結果 として起こる変化を検出することにより測定する段階からなることを特徴とする 請求の範囲第1項記載の方法。
- 4.a)リガンドをセンサー表面に結合させ、b)巨大分子が少なくとも1つの 他のリガンドと相互作用することを許容し、 c)前記少なくとも1つの他のリガンドと相互作用させた巨大分子をリガンドを 持つセンサー表面と接触させ、及び d)巨大分子のセンサー表面に結合させたリガンドとの相互作用をセンサー表面 の物理化学的性質に結果として起こる変化を検出することにより測定する段階か らなることを特徴とする請求の範囲第1項記載の方法。
- 5.変更した順序で少なくとも2つのリガンドの複数のリガンドとの相互作用を 繰り返すことからなることを特徴とする請求の範囲第1項〜第4項のいずれか一 項記載の方法。
- 6.段階b)と段階c)を変更した順序で複数のリガンドと1回又はそれより多 い回数繰り返すことを特徴とする請求の範囲第2項及び第5項のいずれか一項記 載の方法。
- 7.段階c)と段階d)、及び場合によりa)を変更した順序で複数のリガンド と1回又はそれより多い回数繰り返すことを特徴とする請求の範囲第3項及び第 5項のいずれか一項に記載の方法。
- 8.段階a)〜段階d)を変更した順序で複数のリガンドと1回又はそれより多 い回数繰り返すことを特徴とする請求の範囲第4項及び第5項のいずれか一項記 載の方法。
- 9.方法をリガンドのより小さい群で繰り返して実行することを特徴とする多数 のリガンドを使用する請求の範囲第1項〜第8項のいずれか一項記載の方法。
- 10.多数のリガンドの各々の1つをセンサー表面のそれぞれのサブ領域に結合 させ、及びそれに結合した少なくとも1つのリガンドを持つ巨大分子を得られる 多機能性センサー表面に結合させ、巨大分子の前記センサー表面のサブ領域のす べてとの相互作用を各巨大分子/リガンド組合せにつき測定することを特徴とす る請求の範囲第4項記載の方法。
- 11.センサー表面の物理化学的性質の変化はセンサー表面の屈折率の変化であ ることを特徴とする請求の範囲第1項〜第10項のいずれか一項記載の方法。
- 12.巨大分子ハタンパク質であることを特徴とする請求の範囲第1項〜第11 項のいずれか一項記載の方法。
- 13.リガンドは抗体からなることを特徴とする請求の範囲第1項〜第12項の いずれか一項記載の方法。
- 14.リガンドの数は少なくとも3つであることを特徴とする請求の範囲第1項 〜第13項のいずれか一項記載の方法。
- 15.巨大分子の構造修飾のそれへのリガンド結合性質に与える影響を調べるた めの請求の範囲第1項〜第14項のいずれか一項記載の方法の使用。
- 16.診断的又は予知的臨床目的のための請求の範囲第1項〜第14項のいずれ か一項記載の方法の使用。
- 17.巨大分子上の異なる可能性のあるエピトープを限定するリガンドのサブセ ットを用いて巨大分子又はその複合体、病理学的条件の指標である巨大分子上の 1つ又はそれより多い数の前記エピトープのそれぞれ存在又は不存在を特性決定 することからなることを特徴とする請求の範囲第16項記載の使用。
- 18.患者の試料で限定されたエピトープを持つ巨大分子を最初に接触させ、次 いで巨大分子を前記エピトープに結合させてリガンドのサブセットに接触させる ことにより試料中に存在する巨大分子による1つ又はそれより多い数のエピトー プの可能な封鎖を測定することによる免疫応答を文するための請求の範囲第16 項記載の使用。
