Claims (1)
- 【特許請求の範囲】 1、酵素グルコースオキシダーゼの存在下、酸素によるグルコースの酵素的酸化 を使用することを含み、測定セルによって酸素消費量もしくは過酸化水素生成量 を決定し、溶解されたまたは測定セルに到達する前に溶解されたグルコースオキ シダーゼを含む灌流液は、一定速度で供給管を介し、皮下組織に備えられたグル コースを透過する中空糸を通過し、中空糸から気密排出管を介して体外に備えら れた測定セルを通過し、透析は、皮下でグルコースが局地的に拡がつたグルコー ス濃度に比例して皮下組織から中空糸の壁を介して灌流液に移動して行われ、灌 流液に受け取られたグルコースは測定セルに到達する前に完全に酸化されること を特徴とする連続的もしくは断続的に皮下組織におけるグルコース濃度を決定す る方法。 2、酸素消費量を測定することを特徴とする請求項1の記載の方法。 3、使用される測定セルが作用電極、電解物で満たされた電解物空間および参照 電極を含んで成り、灌流液はフロー要素に備えられたフロー室を介して測定セル を通過し、前記フロー室は中空糸から排出された灌流液の入口と灌流液の出口と を備え、かつ測定セルと酸素ガスを透過する膜によつて隔離されていることを特 徴とする請求項2記載の方法。 4、貴金属好ましくはプラチナの作用電極および銀の参照電極を使用し、使用さ れる電解物がリン酸カリウム緩衝液、好ましくは0.5MK2HP04、使用す る酸素ガスを透過する膜がテフロン膜であり、参照電極に関して約0.6Vの陰 圧が作用電極にかけられることを特徴とする請求項3記載の方法。 5、灌流液が体外に備えられた貯蔵器から密封供給管好ましくはポリエチレン製 の密封供給管または大気透過供給管好ましくはテフロンもしくはシリコンゴムの 大気透過供給管を介して供給され、測定セルを通過した後、体外に備えられた排 出器に排出されることを特徴とする請求項1−4記載の方法。 6、貯蔵器から供給される使用される灌流液は中空糸を通過した後、測定セルを 通過する前に体外でグルコースオキシダーゼと接触する生理的食塩水であること を特徴とする請求項5記載の方法。 7、貯蔵器から供給され使用される灌流液が生理的食塩水中のグルコースオキシ ダーゼ溶液であることを特徴とする請求項5記載の方法。 8、灌流液が生理的食塩水1mlにつき少なくとも0.05mg、好ましくは少 なくとも0.10mgのグルコースオキシダーゼを含むことを特徴とする請求項 7記載の方法。 9、灌流液が1mlにつき0.20−0.40mgのグルコースオキシダーゼを 含んでいることを特徴とする請求項7記載の方法。 10、使用される灌流液が溶解状態の酵素グルコースオキシダーゼおよびカタラ ーゼを含む生理的食塩水であり、灌流液は測定セルを通過した後、少なくとも1 つの大気透過部分を含む装置を介して中空糸に戻り、測定セルを通適する前もし くは後に灌流液は体外に備えられている酵素計量装置を通過し、この装置では灌 流液に新しいグルコースオキシダーゼとカタラーゼを溶解させることを特徴とす る請求項2−4記載の方法。 11、灌流液は測定セルを通過した後、大気透過供給管好ましくはテフロンもし くはシリコンゴム製の大気透過供給管を介して中空糸に戻ることを特徴とする請 求項10記載の方法。 12、灌流液は1mlの生理的食塩水につき少なくとも0.05mg好ましくは 少なくとも0.10mgのグルコースオキシダーゼおよび少なくとも0.05m g、好ましくは少なくとも0.10mgのカタラーゼを含むことを特徴とする請 求項10もしくは11記載の方法。 13、灌流液は1mlにつき0.20−0.40mgのグルコースオキシダーゼ および0.20−0.40mgのカタラーゼを含むことを特徴とする請求項10 もしくは11記載の方法。 14、約10kDの分子量カットオフ値を持つセルロースエステルの中空糸を使 用することを特徴とする請求項1−13記載の方法。 15、内径100−500μm、好ましくは120−200μm、外径130− 550μm、好ましくは150−250μm、長さ0.1−3cm、好ましくは 0.5−2.5cmの中空糸を使用することを特徴とする請求項1−14記載の 方法。 16、ポリスルホンもしくはアクリル共重合体の中空糸を使用することを特徴と する請求項1−13記載の方法。 17、内径500−1200μm、好ましくは900−1100μmの中空糸を 使用することを特徴とする請求項16記載の方法。 18、使用される気密排出管がポリエチレン管であることを特徴とする請求項1 −17記載の方法。 19、供給および排出管の内径が0.2−0.6mm、好ましくは0.25−0 .35mm、外径が0.4〜1.0mm、好ましくは0.6−0.8mmである ことを特徴とする請求項1−18記載の方法。 20、中空糸とフロー室との間の気密排出管の長さが1−10cm、好ましくは 1−5cmであることを特徴とする請求項1−19記載の方法。 21、フロー室の灌流液の入口が作用電極のさらされた表面の向かい側もしくは 蓬横ぎつた側に備えられ、互いに酸素透過膜によって隔離されている入口端面と 作用電極のさらされた表面との距離が5mmよりも少なく、好ましくは1mmよ りも小さいことを特徴とする請求項1−20記載の方法。 22、使用されるフロー室が実質的に使用しない空間を生じないような大きさ、 形、灌流液入口および出口の位置を持つことを特徴とする請求項1−20記載の 方法。 23、灌流液がフロー速度0.1−1.0ml/時間、好ましくは0.2−0. 4ml/時間で通過することを特徴とする請求項1−22記載の方法。 24、グルコース透過中空糸、灌流液のための供給管、灌流液のための気密排出 管および灌流液中の酸素もしくは過酸化水素の量を測定するための測定セルを特 徴とする皮下組織における連続的もしくは断続的なグルコース測定のための装置 25、貯蔵器に含まれる灌流液を一定速度で貯蔵器に接続する供給管を介して中 空糸を通過させるための装置を備えている灌流液のための貯蔵器、および使用す る灌流液のための貯蔵器によって特徴付けられる請求項24記載の装置。 26、貯蔵器に含まれる灌流液は生理的食塩水中のグルコースオキシダーゼ溶液 から成ることを特徴とする請求項25記載の装置。 27、灌流液のための大気透過供給管、酵素計量装置、および一定速度で灌流液 を循環させるためのポンプによって特徴付けられる請求項24記載の装置。 28、固体状態での酵素グルコースオキシダーゼおよびカタラーゼの酵素計量手 段に含まれる供給手段によつて特徴付けられる請求項27記載の装置。 29、灌流流は生理的食塩水中の酵素グルコースオキシダーゼおよびカタラーゼ 溶液から成ることを特徴とする請求項28記載の装置。 30、測定セルは作用電極、電解室空間および参照電極を含んで成り、酸素ガス 透過膜によつて測定セルから隔離することができるフロー室に通じている灌流液 の入口と出口とを含んで成る適合するフロー要素を備えることによつて特徴付け られる請求項24−29記載の装置。 31、作用電極は貴金属、好ましくはプラチナ製および参照電極は銀製であるこ とを特徴とする請求項30記載の装置。 32、電解室空間がりン酸カリウム緩衝液好ましくは0.5MK2HPO4で満 たされていることを特徴とする請求項30または31記載の装置。 33、酸素ガス透過膜が嫌水性膜好ましくはテフロン製の膜であることを特徴と する請求項30−32記載の装置。 34、フロー室の灌流液の入口が作用電極のさらされた表面の向かい側もしくは 横ぎつた側に備えられ、互いに酸素透過膜によつて隔離されている入口端面と作 用電極のさらされた表面との距離が5mmよりも少なく、好ましくは1mmより も小さいことを特徴とする請求項30−33記載の装置。 35、使用されるフロー室が実質的に使用しない空間を生じないような大きさ、 形、灌流液入口および出口の位置を持つことを特徴とする請求項30−34記載 の装置。 36、約10kDの分子量カットオフ値を持つセルロースエステルの中空糸を使 用することを特徴とする請求項24−35記載の装置。 37、内径100−500μm、好ましくは120−200μm、外径130− 550μm、好ましくは150−250μm、長さ0.1−3cm、好ましくは 0.5−2.5cmの中空糸を使用することを特徴とする請求項24−36記載 の装置。 38、ポリスルホンもしくはアクリル共重合体の中空糸を使用することを特徴と する請求項24−35記載の装置。 39、内径500−1200μm、好ましくは900−110μmの中空糸を使 用することを特徴とする請求項38記載の装置。 40、使用される気密排出管がポリエチレン管であることを特徴とする請求項2 4−30記載の装置。 41、供給および排出管の内径が0.2−0.6mm、好ましくは0.25−0 .35mm、外径が0.4〜1.0mm、好ましくは0.6−0.8mmである ことを特徴とする請求項24−40記載の装置。 42、中空糸とフロー室との間め気密排出管の長さが1−10cm、好ましくは 1−5cmであることを特徴とする請求項24−41記載の装置。 43、測定セルは作用電極、電解室空間および参照電極を含んで成り、酸素ガス 透過膜によつて測定セルから隔離することができるフロー室に通じている灌流液 の入口と出口とを含んで成る適合するフロー要素を備えることによつて特徴付け られる請求項24−42記載の装置に使用されるために適合した測定セル組立品 。 44、作用電極は貴金属、好ましくはプラチナ製および参照電極は銀製であるこ とを特徴とする請求項43記載の測定セル組立品。 45、電解室空間がリン酸カリウム緩衝液好ましくは0.5MK2HP0.4で 満たされていることを特徴とする請求項43または44記載の測定セル組立品。 46、酸素ガス透過膜が嫌水性膜ましくはテフロン製の膜であることを特徴とす る請求項43−45記載の測定セル組立品。 47、フロー室の灌流液の入口が作用電極のさらされた表面の向かい側もしくは 横ぎつた側に備えられ、互いに酸素透過膜によつて隔離されている入口端面と作 用電極のさらされた表面との距離が5mmよりも少なく、好ましくは1mmより も小さいことを特徴とする請求項43−46記載の測定セル組立品。 48、使用されるフロー室が実質的に使用しない空間を生じないような大きさ、 形、灌流液入口および出口の位置を持つことを特徴とする請求項43−47記載 の測定セル組立品。 49、請求項24−42記載の皮下組織における連続的もしくは断続的グルコー ス濃度決定装置、インシュリンのような薬剤を血液中に投与する調節できる注入 装置、測定セルの測定値および適合する検量線に基づいて、特徴的な重要な因子 が数字的モデルの中に含まれている、アルゴリズムの手段によって皮下組織にお けるグルコース濃度を計算し、供給すべき薬剤の量を決定し、グルコース濃度は 組織および/または血液中において予め定められる値内にとどまるように注入装 置を制御する計算および調整装置を特徴とする連続的または断続的に皮下のグル コース温度を調整するための測定セル組立品。
