内容物を内包したリポソームおよび/または生物細胞の製造方法ならびにその用
途
本発明は、請求の範囲に記載した対象、つまり、製造の間に、外部から添加した
物質を収容したリポソームおよび/または生物細胞の製造方法に関する。双方の
場合に、物質の収容は穏やかな条件で行われる。負荷されたリポソームおよび/
または細胞は標的に向かう薬物担持体として適している。
リポソームは脂質、特にリン脂質から構成されている。これは水性媒体中で自然
発生的に、閉じたラメラ状の構造になっている脂質二重層を構成する。一つ以上
のラメラは内部コンパートメントを外部の溶液と隔てている。脂質の極性の親水
性の先頭基(たとえばセファリンの場合のエタノールアミン、レシチンの場合の
コリン)は水相に向い、非極性の疎水性の脂肪酸基は、脂質二重層の内側に向か
う、この二重層は2分子層であるかまたは4〜5nmの厚さである。リポソーム
はその大きさおよびラメラの数に応じて区別され、半径1100nまでの小さな
単ラメラ小胞(Sma1] unilamellar vesicles(SO
’/)) 、大きな単うメラ小M1(Jarge unilamellar v
esicles(LUV) )ならびに多重ラメラ小胞(multilamel
Jar vesicles(MLV) )がある、水相中でリン脂質は、何も
干渉せずにた重ラメラリポソームに集合し、その際タマネギ状の多数の脂質二重
層は水相により隔てられて配置されている。
ラメラ配置した脂質二重層の形成は、物理的な力および立体的効果の結果である
。この脂質二重層は、生物細胞を取り囲み、jl!2の構成要素としてタンパク
質を脂質マトリックス中に封じ込めた形質膜の構造ブロックを形成する。
リポソームの物理的および生理的特性は、脂質の組成、大きさ、温度、pH,a
m液の種票および濃度、製造条件、ヒストリー(l(istorie )および
エイジ(Alter)によって決まる。薬物を負荷したリポソームの利用は、診
断および治療において多大な関心が生じている。リポソームは薬物の標的に向か
う担持体として新規の診断形式および治療形式を開拓している(たとえば、R,

O−99(1987)参照)、製剤学的目的にとって、脂質膜の透過性およびリ
ポソームの安定性が重要であり、さらに、生理学的には溶解性および細胞収容の
速度が重要である。
広範囲の関心に基づき、物理学、化学、生物学、リポソームの製造および用途に
関する多数の刊行物が存在する。最近発行された薬理学的成果を有する概要文献

単ラメラおよびオリゴラメラならびに負荷されたリポソームの製造のために、多
様な方法が開発され(Liebtenberg & Barenholz報告)
、これらの方法は、自然発生的に形成されたリポソームを1.剪断力による物理
的破壊および2.「ヘルパー分子」による化学的分解として概略的に分票できる
。剪断力の適用は、不安定な物質を封じ込める場合に適当でない、ヘルパー分子
は、界面活性剤または有機溶剤分子の形で提供される。このヘルパー分子を用い
て、脂質分子の非二重層凝集体(たいていはミセル)が形成される。ヘルパー分
子の除去は、再度自然発生的にリポソームの二重層を形成させ、同時に添加した
物質の収容させる。界面活性剤分子の完全な除去はしばしば困難であり、さらに
、タンパク質含有の封入分子の場合に望ましくない変性が妨げとなりえる。生物
細胞による収容は、後続する等優性の調節による減膜分解(hypo−osmo
lares^ufbrechen) [分解(Lyse) ]または電気的に誘
導した極形成により達成することができる。この場合、外部に存在する物質を収
容し、膜が閉じる間に閉じ込められる。等張的な、界面活性剤および電圧の無い
条件下での穏やかな分解は、今まで公知でなかった。従って、本発明の課題は、
前記の欠点を有しない内容物含有リポソームおよび/または生物細胞の製造方法
を開発することであった。前記課題は本発明により解決された。
物質、有利に治療および予防の目的の薬物をリポソームおよび/または細胞に収
容することは、意想外にガス状のヘルパー分子を適用することによっても達成可
能であることが見出された。この方法は、前記した方法の欠点を有していない、
この新規方法において、脂質または細胞のほかに封入すべき物質を含有する水性
脂質または細胞懸濁液に、本発明により、ガスを添加する。界面活性効果を有す
るガスは、たとえば静水圧と友対に、圧力が高まると共に脂質分子の配列を減少
させ、および/または、膜の可溶化を持たらすことができるガスである。その都
度適用する圧力で、水性脂質または細胞懸濁液での熱力学的平衡の調節が期待さ
れる。使用したガスは、脂質および水相に関して化学機に不活性でなければなら
ず、二酸化炭素またはアンモニアのように水に取り込まれてはならない、100

可能で、工業的に提供可能なガスの使用が有利である。液体状態でのp、’r−
ダイアダラムにおける沸騰曲線を上回るのが有利である、適当なガスは、笑気(
N:O)、 ハoタン(Halothan) 、アルゴン、キセノン、六フッ化
硫黄(SF@)アルカン、たとえばメタン、エタン、プロパン、ブタン、ヘキサ
ン、ヘプタン、アルケン、たとえばエデン、プロペン、ブテン、ヘキセン、ヘプ
テンである。笑気の場合、圧力は55barまでに調節され、工業的耐圧フラス
コが提供される。温度については、その都度の凝固点と沸点との範囲内、有利に
0〜37℃である。
前記のガス分子は界面活性剤に似た効果を有し、脂質二重層および/または膜の
開放および/または可溶化を持たらす、放圧用の間に、自然発生的に新たなリポ
ソームが形成されるかもしくは膜が再び閉じ、その結果添加した物質が収容され
る。この工程は、周期的に繰り返される。リポソームの場合に、リポソームサイ
ズの限定は、大気圧への放圧において、大きさを選択する分離フィルタにより可
能である。リポソームの脂質の組成は、治療および/または予防的有用性に応じ
てまたは目的器官に応じて最適にされる。生物細胞の場合に、血液細胞、たとえ
ば赤血球、白血球、リンパ球、血小板の他に、有利に培養中に保持されている細
胞が挙げられる。
「ガスヘルパー法」でのリポソーム製造および細胞負荷の利点は、次のようなも
のである:この方法は工業的に容易に実現化でき低コストである。
ガス分子は従来の界面活性剤分子とは反対に容易に除去できる。
不活性の性質および痕跡量で残留する量のため、使用した分子の型性は、あった
としてもまったく問題にならない。
生理学的温度以下での作業が有利である。
前記圧力範囲内での不活性ガスの適用はたいていの物質または細胞にとって害に
ならない。
この方法は、親水性および脂肪親和性物質をリポソームおよび細胞に付加、加入
および/または補充することに適している。
負荷したリポソームは表面的に変性され目標または器官特異的な薬物担持体また
は薬物カプセルとして使用することができる。
[実施例]
例1
シミリストイル−レシチン20mgをクロロホルム1ml中に溶かした。この溶
液を耐圧容器にいれた。
クロロホルムを窒素で後洗浄し、および引き続き真空で習去した。カルボキシフ
ルオロレシチン(洗浄し、エタノールから再結晶)1mMおよびpH8に調節し
たトリスHCl20mMを含有した水20m1を満たした。CO2および0.を
笑気で洗浄することにより追い出した。この耐圧容器を閉じ、室温で50bar
の笑気圧に調節した。この溶液を0.5時間振盪し、その後ゆっくりと等張に大
気圧に放圧した。この工程を5回繰り返した。この溶液を耐圧容器から取り出し
、外部に存在するカルボキシフルオロレシチンの除去のためにアニオン交換器を
通した。このリポソームフラクションは封入された色素を含有し、蛍光試験のた
めに使用することができる。
例2
例1と同様に行うが、シミリスドールレシチンの代わりに卵黄レシチンおよびコ
レステリンからなる4:1混合物200mgを溶かした。カルボキシフルオロレ
シチンを、MR造影剤としてガドリニウムDTPA20mMに換えた。得られた
リポソームはMR診断に使用することができる。
例3
カルボキシフルオロレシチンを負荷された赤血球または赤血球ゴーストの血液等
張条件下での製造。
新鮮なウシ血清を、pH7,4のPBS!1m液中で3回洗浄し、赤血球に関し
てその他の成分を除去した。
暗赤色の、濁った赤血球!!!濁液を、カルボキシフルオロレシチン含有のPB
SII!濁液で希釈し、その結果、カルボキシフルオロレシチン濃度は10’M
であった。
この溶液を圧力測定セル中にいれた。N、Oで洗浄した後に37℃でNユ0を5
0barにし、撹拌し、光学密度を675nmで追跡した。400〜600分間
に細胞の分解を生じさせ、これは光学密度の典型的なS字形の減少により明らか
に確認できた。大気圧または50barの静水圧での参照測定は18時間まで赤
血球の分解を示さなかフた。この溶液をゆっくりと大気圧に放圧し、透明および
蛍光において顕微鏡で試験した。この赤血球は分解の前および後で典型的な円板
状の外観を示した0分解していない細胞(対照)は中間時で分解したものとは反
対にカルボキシフルオロレシチンを含有していなかった。細胞の蛍光は広い範囲
での成長の後でも変化せず確認可能であった。従って、赤血球は分解状態で等張
条件下で色素を収容し、放圧の間に再度封入した。
例4
例3と同様であるが、ウシ赤血球の代わりにKH。
Po、2mM、Na、HPo、9mM、KCI 3mM。
NaC1137mM中でpH7,5で羊赤血球を使用した6分解は55〜70分
で行った。結果は例3からのものと比較可能であった。

Claims (9)
- 1.内容物を含有するリポソームおよび/または生物細胞を製造する方法におい て、リポソームまたは生物細胞を、水性媒体中で、圧力が高まると共に脂質配列 の切断を増加させる不活性ガスを用いて、ガス状態または液体状態で、所望の内 容物を添加しながら加圧下で処理し、引き続き大気圧に放圧を行うことを特徴と する内容物を含有するリポソームおよび/または生物細胞を製造する方法。
- 2.リポソームまたは生物細胞により収容される内容物が薬物である請求項1記 載の方法。
- 3.不活性ガスとして、笑気(N2O)、ハロタン、アルゴン、キセノン、六フ ッ化硫黄(SF6)、アルカンたとえばメタン、エタン、プロパン、ブタン、ヘ キサン、ヘブタン、アルケンたとえばエテン、プロベン、ブテン、ヘキセン、ヘ プテン、および/またはそれらの混合物を使用する請求項1記載の方法。
- 4.1000bar、有利に200barの圧力を使用する請求項1記載の方法 。
- 5.放圧のために、リポソーム懸濁液を大きさのを選択するフィルターにより圧 搾する請求項1記載の方法。
- 6.内容物を加入、付加および/または置換する請求項1記載の方法。
- 7.生物細胞が血液細胞、たとえば赤血球、白血球、リンパ球および血小板であ る請求項1記載の方法。
- 8.生物細胞が、培養を保持しかつ植物的および/または動物的および/または 微生物的起源である細胞である請求項1記載の方法。
- 9.請求項1により製造したリポソームお上び/または生物細胞の、予防的およ び/または治療的目的にたいする薬物の輸送および/または貯蔵のための付形剤 としての用途。
