Description
Claims (20)
- 下記の構成を含む、補体依存性細胞傷害(Complement Dependent
Cytotoxicity、CDC)が抗CD20対称抗体と比較して向上した抗CD20非対称抗体:
(a)配列番号16のLCDR1、配列番号18のLCDR2、配列番号20のLCDR3、配列番号24のHCDR1、配列番号26のHCDR2及び配列番号28のHCDR3配列を含むscFv(Single-chain variable fragment);及び
(b)配列番号33のLCDR1、配列番号35のLCDR2、配列番号37のLCDR3、配列番号42のHCDR1、配列番号44のHCDR2及び配列番号46のHCDR3配列を含むFab(Fragment antigen-binding)。 - 前記scFvは、配列番号51の軽鎖可変領域(VL)配列を含むことを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記scFvは、配列番号52の重鎖可変領域(VH)配列を含むことを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記scFvは、配列番号15のLFR1、配列番号17のLFR2、配列番号19のLFR3、配列番号21のLFR4、配列番号23のHFR1、配列番号25のHFR2、配列番号27のHFR3及び配列番号29のHFR4で構成された群から選ばれる配列を含むことを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記scFvは、VL-リンカー-VHの形態であることを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記リンカーは、Gly及びSerの組み合わせであることを特徴とする、請求項5に記載の抗CD20非対称抗体。
- 前記Fabは、配列番号53の軽鎖可変領域(VL)配列を含むことを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記Fabは、配列番号54の重鎖可変領域(VH)配列を含むことを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記Fabは、配列番号32のLFR1、配列番号34のLFR2、配列番号36のLFR3、配列番号38のLFR4、配列番号41のHFR1、配列番号43のHFR2、配列番号45のHFR3及び配列番号47のHFR4で構成された群から選ばれる配列を含むことを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記Fabは、配列番号39のCκ(C kappa)配列を含むことを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記Fabは、配列番号48のCH1配列を含むことを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記Fabは、VL-Cκ-リンカー-VH-CH1の形態であることを特徴とする、請求項1に記載の抗CD20非対称抗体。
- 前記リンカーは、Gly及びSerの組み合わせであることを特徴とする、請求項12に記載の抗CD20非対称抗体。
- 請求項1の抗CD20非対称抗体をコーディングする核酸分子。
- 請求項14の核酸分子を含むベクター。
- 請求項15のベクターで形質転換された単離された宿主細胞。
- 請求項1の抗CD20非対称抗体、前記抗体をコーディングする核酸分子、又は前記核酸分子を含むベクターを有効成分として含む癌の予防又は治療用薬剤学的組成物。
- 請求項1の抗CD20非対称抗体、前記抗体をコーディングする核酸分子、又は前記核酸分子を含むベクターを有効成分として含む癌の診断用組成物。
- 下記の段階を含む、補体依存性細胞傷害(Complement Dependent
Cytotoxicity、CDC)が抗CD20対称抗体と比較して向上した抗CD20非対称抗体の製造方法:
(a)配列番号16のLCDR1、配列番号18のLCDR2、配列番号20のLCDR3、配列番号24のHCDR1、配列番号26のHCDR2及び配列番号28のHCDR3配列を含むscFv(Single-chain variable fragment)を製造する段階;及び
(b)配列番号33のLCDR1、配列番号35のLCDR2、配列番号37のLCDR3、配列番号42のHCDR1、配列番号44のHCDR2及び配列番号46のHCDR3配列を含むFab(Fragment antigen-binding)を製造する段階。 - 下記の段階を含む、補体依存性細胞傷害(Complement Dependent
Cytotoxicity、CDC)が抗CD20対称抗体と比較して向上した抗CD20非対称抗体をコーディングする核酸分子の製造方法:
(a)配列番号16のLCDR1、配列番号18のLCDR2、配列番号20のLCDR3、配列番号24のHCDR1、配列番号26のHCDR2及び配列番号28のHCDR3配列を含むscFv(Single-chain variable fragment)をコーディングする核酸分子を製造する段階;及び
(b)配列番号33のLCDR1、配列番号35のLCDR2、配列番号37のLCDR3、配列番号42のHCDR1、配列番号44のHCDR2及び配列番号46のHCDR3配列を含むFab(Fragment antigen-binding)をコーディングする核酸分子を製造する段階。
