本発明は、2-ヒドロキシベンジルアミン(2-HOBA)のようなγ-ケトアルデヒド捕捉化合物を含んでなる組成物と、肝臓の線維症を治療する、予防する、減弱させる、抑制する、その進行を遅らせる、又は改善するためにγ-ケトアルデヒド捕捉剤を投与する方法に関する。
本発明には、有害な酸化脂質(ケトアルデヒド)を捕捉して、肝臓の線維症(肝線維症)を治療する、予防する、減弱させる、抑制する、その進行を遅らせる、又は改善するための、2-HOBAが含まれるγ-ケトアルデヒド捕捉剤の使用が含まれる。
本発明のもう1つの目的は、肝線維症を治療する、予防する、減弱させる、抑制する、その進行を遅らせる、又はその症状を改善する、及び/又は肝線維症の症状の重症度を抑えるために、本発明の化合物の調製品の治療又は有効量を提供することである。
本明細書に記載される組成物は、肝線維症を治療する、予防する、減弱させる、抑制する、その進行を遅らせる、及び/又は改善するために使用される。

実施例1
DIAMOND(非アルコール性脂肪性肝疾患の食餌性動物モデル)は、高脂肪、高糖分の食餌に反応して、非アルコール性脂肪性肝疾患、非アルコール性脂肪性肝炎、線維症、及び肝細胞癌を順次発症する、専用の同系マウス系統である。DIAMONDマウスにおける疾患進行は、ヒトの疾患進行と、組織病理学を含めて、比類なく相似する。
γ-KAは、肝線維症の主要な推進因子である、肝星細胞(HSC)の活性化を誘導する。HSCの好炎症性/好線維形成性の表現型への活性化を予防すれば、肝臓における線維症の発現を阻止し得るだろう。肝線維症にはHSCの筋線維芽細胞への変換が必須であると考えられるので、固定肝切片に関する免疫組織化学によって、デズミン(HSCのマーカー)とα-平滑筋アクチン(SMA)(活性化HSCのマーカー)を使用して、HSC活性化について測定する。
エンドポイント:RNA:線維形成性の活性化、サイトカイン産生、及び接着分子に関連した選択される転写産物の発現について、RT2ProfilerTMPCR Array(キアジェン、メリーランド州フレデリック)と単一遺伝子プローブベースのqRT-PCR遺伝子発現アッセイを適宜使用して測定する。タンパク質:免疫ブロット分析を使用して、主要な細胞シグナル伝達経路(ERK1/2、JNK、NFκB、及びp38MAPK)の含量及び活性化状態について測定する。サイトカイン:15-E2-IsoLGと2-HOBAとのインキュベーション後に採取する培地における、炎症性サイトカインの濃度を決定する。ROS/RNS:5-(及び-6-)-カルボキシ-2’-7’-ジクロロジヒドロフルオレセインジアセテート(カルボキシ-H2)蛍光プローブ(ThermoFisher Scientific)を使用して、細胞内ROS/RNS形成について測定する。ヘキスト(Hoechst)33342での核染色によって、全細胞分布を可視化する。蛍光顕微鏡により画像を獲得する。

Claims (3)
- 肝線維症を有するか又はそのリスク状態にある被験者に対して肝線維症を治療する、予防する、又は改善することによって肝線維症を防止するか又は治療する方法に用いるための、化合物を含む組成物であって、その機能不全を哺乳動物において治療するのに有用な投与量及び量で該化合物を、肝線維症を治療、予防、又は改善する既知の副作用を有する薬物とともに、該被験者へ投与する、該化合物は、以下の式:
- 肝線維症を治療、予防、又は改善する既知の副作用を有する薬物は、ビタミンE、チアゾリジンジノン類、及びペントキシフェリンからなる群から選ばれる、請求項1に記載の組成物。
- 肝線維症を有するか又はそのリスク状態にある被験者に対して肝線維症を治療する、予防する、又は改善することによって肝線維症を防止するか又は治療する方法に用いるための、化合物を含む組成物であって、その機能不全を哺乳動物において治療するのに有用な投与量及び量で該化合物を、該被験者へ投与する、該化合物は、以下の式:
