免疫チェックポイント分子は、核酸配列、アミノ酸配列、小分子、またはそれらの組み合わせであり得る。核酸配列は、DNA、RNA、cDNA、それらのバリアント、それらの断片、またはそれらの組み合わせであり得る。核酸は、ペプチド結合により免疫チェックポイント阻害剤に連結されるリンカーまたはタグ配列をコードする追加の配列も含み得る。小分子は、低分子量、例えば800ダルトン未満の、酵素基質として機能し得る有機もしくは無機化合物、タンパク質もしくは核酸により結合されるリガンド(またはその類似体)、または生物学的プロセスの調節因子であり得る。アミノ酸配列は、タンパク質、ペプチド、それらのバリアント、それらの断片、またはそれらの組み合わせであり得る。

本明細書で考察されるMUC16抗原をコードする核酸分子を含むベクターの調製方法が本明細書に提供される。哺乳類発現プラスミドへの最終的なサブクローニングステップ後、当該技術分野において既知の方法を使用して、ベクターを大規模な発酵タンクにおいて細胞培養物に播種するために使用することができる。

上述の医薬製剤を使用して、限定されないが、卵巣がんなどのMUC16発現がんを治療および/または予防するための方法が本明細書に提供される。対象における、限定されないが、卵巣がんなどのMUC16発現がんの治療および/または予防に上述の医薬製剤を使用する方法も本明細書に記載される。対象にワクチン接種する方法も本明細書に記載される。本明細書に記載の医薬製剤を、それを必要する対象に投与する方法も本明細書に記載される。本明細書に記載の医薬製剤を使用する治療方法と集合的に称される本明細書に記載の方法は、治療的および/または予防的免疫応答を誘導するために、本明細書に記載される1つ以上のワクチンを、それを必要とする対象に投与することを含み得る。ワクチンは、対象の免疫系の活性を調節し、免疫応答を増強するために対象に投与され得る。ワクチンの投与は、細胞において発現され、細胞の表面に送達されると、免疫系が認識して、細胞性、液性、または細胞性および液性応答を誘導する核酸分子としての本明細書に開示されるがん抗原のトランスフェクションであり得る。ワクチンの投与は、本明細書で考察されるワクチンを対象に投与することにより、対象において、MUC16に対して免疫応答を誘導または誘発するために使用され得る。
ワクチンを使用して、治療的または予防的免疫応答を含む、哺乳動物または非哺乳動物対象において免疫応答を生成することができる。免疫応答は、本明細書に開示される1つ以上のがん抗原に指向される抗体および/またはキラーT細胞を生成することができる。そのような抗体およびT細胞が単離され得る。
ワクチンを使用して、哺乳動物において、限定されないが、卵巣がん、より具体的には上皮卵巣がん、最も具体的には漿液性卵巣がんなどのMUC16発現がんに対して反応性であるかまたはそれに指向される、免疫応答を生成または誘発することができる。誘発された免疫応答は、卵巣がんまたは腫瘍成長を阻止することができる。
ワクチンまたは医薬組成物は、経口、非経口、舌下、経皮的、直腸に、経粘膜的、局所的、吸入により、頬側投与により、胸膜内、静脈内、動脈内、腹腔内、皮下、筋肉内、鼻腔内 くも膜下腔内、および/もしくは関節内、またはそれらの組み合わせにより投与され得る。獣医学的使用において、本組成物は、通常の獣医学的実践に従い、好適に許容可能な製剤として投与され得る。獣医は、特定の動物に最も適切な投与レジメンおよび投与経路を容易に決定することができる。ワクチンは、従来の注射器、無針注射装置、「微粒子衝撃遺伝子銃(microprojectile bombardment gene guns)」、または電気穿孔法(「EP」)、「流体力学法」、もしくは超音波などの他の物理的方法により投与され得る。
ワクチンまたは医薬組成物は、電気穿孔法により投与され得る。電気穿孔法によるワクチンの投与は、可逆性細孔を細胞膜において形成させるのに有効なエネルギーのパルスを哺乳動物の所望の組織に送達するように構成され得る電気穿孔装置を使用して達成することができ、好ましくは、エネルギーのパルスは、ユーザによって入力される予め調整された電流と同等の定電流である。電気穿孔装置は、電気穿孔構成要素および電極アセンブリまたはハンドルアセンブリを備え得る。電気穿孔構成要素は、コントローラ、電流波形発生装置、インピーダンステスター、波形ロガー、入力素子、ステータス報告要素、通信ポート、メモリー構成要素、電源、および電源スイッチを含む、電気穿孔装置の様々な要素のうちの1つ以上を含み、かつそれらを組み込むことができる。電気穿孔は、プラスミドによる細胞のトランスフェクションを容易にするために、インビボ電気穿孔装置、例えば、CELLECTRA(登録商標)EPシステム(Inovio Pharmaceuticals,Inc.,Blue Bell,PA)またはElgenエレクトロポレーター(electroporator)(Inovio Pharmaceuticals,Inc.)を使用して達成され得る。
本明細書で考察されるDNAプラスミドの調製方法が本明細書において提供される。哺乳類発現プラスミドへの最終的なサブクローニングステップ後、当該技術分野において既知の方法を使用して、DNAプラスミドを大規模な発酵タンクにおいて細胞培養物に播種するために使用することができる。
天然ヒトMUC16は、長さが22,152個のアミノ酸の大きな強くグリコシル化された(heavily glycosylated)タンパク質である。これは、N末端ドメイン、中央タンデム反復ドメイン、およびカルボキシ末端ドメインの3つのドメインで構成されている。天然ヒトMUC16のN末端ドメインは非常に大きく(12,068個のアミノ酸)、反復配列を欠くため、中央タンデム反復およびカルボキシ末端ドメインのみが標的とされる(図1)。異なる設計戦略をタンデム反復およびカルボキシ末端ドメインに適用した。
天然ヒトMUC16のカルボキシ末端ドメインは、長さが284個のアミノ酸であり、細胞外外部ドメイン(extracellular ectodomain)、膜貫通ドメイン、および短い細胞質側末端からなる。中央タンデム反復ドメインと違い、カルボキシ末端ドメインは多くの種の間で十分に保存されている。しかしながら、MUC16の細胞質側末端は、複数の潜在的なS/T/Yリン酸化部位を含有し、シグナル伝達経路(複数可)におけるMUC16の改善を示唆する(図4)。図4において、ボックスは潜在的なS/T/Yリン酸化部位を表す。N-およびO-グリコシル化部位は、それぞれ、XおよびOで示される。O’Brien,T.J.et al.Tumour biology:Journal of the International Society for Oncodevelopmental Biology and Medicine 22,348-366(2001)。望ましくない下流シグナル伝達を引き起こすのを阻止するために、細胞質側末端(天然のカルボキシ末端ドメインのアミノ酸254~284)は、MUC16免疫原のいずれにも含まれない。

コンセンサスMUC16外部ドメインおよび膜貫通ドメインを生成した後、3つのアスパラギン(N)のアラニン(A)への変異(N->A)を導入して、得られたコンセンサスMUC16外部ドメインおよび膜貫通ドメインタンパク質の生物学的機能を無効化した。これら3つの変異は、MUC16のカルボキシ末端外部ドメインが、腫瘍発生、腫瘍浸潤、および転移の促進に関係していたため導入された。Rao,T.D.et al.PloS one 10(2015)。Raoらは、Nグリコシル化を無効化するためにMUC16の外部ドメインにこれら3つのN->A変異を導入することにより、ERK1/2およびAKTのリン酸化、ならびにトランスフェクトされた3T3線維芽細胞を移植された無胸腺ヌードマウスにおけるインビボ腫瘍成長が劇的に低減されたことを示した。合成コンセンサスMUC16構築物に3つのN->A変異を導入した後の生物学的機能の無効化は、インビトロまたはインビボで評価されなかった。
コンセンサスMUC16外部ドメインおよび膜貫通ドメインを生成し、3つの変異(N->A)を導入して、Nグリコシル化を無効化した。(図5)。修飾されたコンセンサスMUC16外部ドメインおよび膜貫通ドメインは、ヒト天然MUC16の外部ドメインおよび膜貫通ドメインと95.3%の同一性を共有する。表2は、生成されたMUC16外部ドメインおよび膜貫通ドメインのヒト天然MUC16外部ドメインおよび膜貫通ドメインとの配列相同性パーセントを示す。
MUC16設計に含まれる要素には、4つのRMC、3つの天然反復、ならびに合成コンセンサスMUC16外部ドメインおよび膜貫通ドメイン(CTD)が含まれた。最初の設計戦略は、図6A~6Bに示される要素を組み込むことにより、MUC16免疫原(予備合成MUC16免疫原)を生成することであった。しかしながら、この免疫原におけるRMC1-RMC2、RMC2-RMC3、RMC3-RMC4、RMC4-R61、およびR63合成コンセンサスMUC16外部ドメインおよび膜貫通ドメイン接合部は天然MUC16配列では生じないため、これらの接合部が少なくとも1つの天然MUC16反復接合部と高い配列同一性を共有しない場合、無関係なオフターゲットエピトープが潜在的に導入され得る可能がある。合成免疫原接合部のMUC16の天然反復接合部のすべてとの配列同一性を決定するために、すべての隣接する天然反復の最後の11個のアミノ酸および最初の11個のアミノ酸を、すべての隣接する合成免疫原接合部(RMC1-RMC2、RMC2-RMC3、RMC3-RMC4、RMC4-R61、およびR63合成コンセンサスCTD)の最後の11個のアミノ酸および最初の11個のアミノ酸とアライメントさせた。分析結果は、RMC4-R61合成免疫接合部を除いて(すべての天然反復接合部と最大76.2%の配列同一性を共有したのみであった)、すべての合成免疫原接合部が少なくとも1つの天然反復接合部と>95.2%の配列同一性を共有したことを示した(図6A)。
pGX0001

pGX1439は、SalIおよびMluI部位において合成コンセンサスMUC16 RMC DNA配列を、そしてPmeI およびXhoI部位において合成コンセンサスMUC16 NRC DNA配列をpGX0003にクローニングすることにより作製された。
pGX1435、pGX1436、pGX1437、pGX1438、およびpGX1439による抗原タンパク質の発現は、ウエスタンブロッティングにより確認された。10%のFBS(ThermoFisher)を含むDMEM培地中に維持されたヒト横紋筋肉腫(RD)細胞(ATCC、CCL-136)を、Turbofectin 8(Origene)を使用して、pGX1435、pGX1436、pGX1437、pGX1438、pGX1439、またはpGX0001(6μg/10cm2皿)でトランスフェクトした。トランスフェクションの48時間後、RIPA細胞溶解緩衝液(ThermoFisher)を使用して細胞を溶解し、細胞溶解物を収集した。総タンパク質濃度を決定するためのBCAアッセイ(ThermoFisher)後、15μgの細胞溶解物を、4~12%のSDS-PAGEゲル(ThermoFisher)で電気穿孔し、抗MUC16モノクローナル抗体(Abcam、クローンEPSISR23-96、ab168360)を使用して検出を行い、次いでECLウエスタンブロット分析システム(GE Amersham)を使用して、西洋ワサビペルオキシダーゼ(HRP)コンジュゲート抗ウサギIgG(Santa Cruz Biotech #sc-2004)で可視化した。ローディング対照として、抗β-アクチンモノクローナル抗体(Santa Cruz Biotech、クローン、C4)を使用して、アクチン発現についてブロットを再検出した。
動物および免疫化
雌の8週齢のCB6F1マウスをJackson Laboratoriesから購入した。すべての動物を、BTS Research(San Diego,CA)の温度制御された光サイクル施設に収容した。動物のケアは、National Institutes of Health and the Animal Care and Use Proposal(ACUP)(BTS ACUP #15-091)のガイドラインに従って行われた。表14に詳述されるように、マウスを16の群に分けた。
脾細胞を無菌で単離し、5mLのR10培地(10%のウシ胎仔血清および1%の抗生物質-抗真菌薬で補充されたRosewell Park Memorial Institute medium 1640)に入れた。Stomacher機器(Seward Laboratory Systems Inc.)を使用して脾臓を機械的に破砕することにより脾細胞を単離し、40-μmセルストレーナー(BD Falcon)を使用して、得られた産物を濾過した。得られた産物を遠心分離し、RBCの溶解のためにペレットをACK溶解緩衝液(Lonza)で5分間処理した。次いで、脾細胞を遠心分離し、PBSで洗浄し、その後R10培地に再懸濁し、さらなる分析のために直ちに使用した。
MabTech(MabTech、#3321-4APW-10)からのキットを使用してマウス IFNγ ELISpotアッセイを行い、抗原特異的細胞応答を評価した。抗マウスIFNγ抗体(mAb AN18)で予めコーティングされた96ウェルプレートをPBS中で洗浄し、R10培地で室温で2時間遮断し、その後2×105細胞/ウェルの投入細胞数で(3つ組で)蒔いた。ペプチドのセットを合成し(GenScript)、それぞれ、各構築物の全合成コンセンサスMUC16タンパク質配列を表す、11個のアミノ酸が重複する15のアミノ酸残基を含有する。ペプチドのこれらのセットをDMSO(Sigma)に再懸濁し、約2μg/mlのペプチドの濃度で6つのペプチドプールにプールした。ペプチドプールは、図17に提供される略図に示される合成コンセンサスMUC16抗原タンパク質に対応するペプチドを含有した。5μg/mlのコンカバリンA(Sigma)を陽性対照として使用し、完全培養培地を陰性対照として使用した。プレートを、5%のCO2雰囲気のインキュベータ中で、37℃で18時間インキュベートした。次いで、ビオチン化抗マウスIFNγ検出抗体(MabTech、mAb R4-6A2)を添加し、プレートを室温で2時間インキュベートした。プレートを洗浄し、ストレプトアビジン-ALP抗体(MabTech)を添加し、プレートを室温で1時間インキュベートした。キット製造業者の指示(MabTech)に従い、BCIP/NBT基質を使用してスポット検出を完了した。プレート上のスポットは、自動ELISPOTリーダー(Cellular Technology)を使用して計数した。スポット形成単位(Spot Forming Units)(SFU)の平均数は、データ表示のために1×106脾細胞に調整された。
合成コンセンサスMUC16により誘導された細胞性免疫応答は、フローサイトメトリーによりさらに特徴付けされた。ワクチン接種されたおよびナイーブマウスからの2x106脾細胞を、単離後に、ブレフェルディンA(BD Biosciences)、モネンシン(BD Biosciences)、およびFITC抗マウスCD107a抗体(BD Biosciences)の存在下で6時間、合成コンセンサスMUC16ペプチドで直ちに刺激した。ペプチドでの刺激後、脾細胞を沈降させ、20μL/ウェルのマウスBD Fc Block(BD Biosciences)溶液に再懸濁した。PBS中1:40の初期希釈でFc Blockを使用し、4℃で5分間インキュベートした。
IBM SPSS Statistics 22(IBM Corporation)を使用して統計分析を完了した。多重比較を調整するためにTukeyの事後Honest Significant Difference(HSD)とともにANOVAを使用して、群間の分析を行った。等分散性は多重比較の前にF統計を使用して確認された。すべての統計分析に関して、0.050のp値が有意であるとみなされた。
全体的に、完全長MUC16構築物pGX1438(フーリン切断)およびpGX1439(デュアルプロモーター)は、pGX1435(融合)と比較して、より頑強な免疫応答を誘導した(図30A~C)。pGX1435と比較して、pGX1438は、10μgおよび30μgの投与量で、IFNγ ELISpotにより有意により頑強な応答を誘導した。また、CD4+ T細胞コンパートメントにおいて、pGX1438は、10μgの投与量でのpGX1435と比較して、有意により頑強な応答を誘発した。3つの完全長MUC16構築物により誘導された応答の規模において、有意な差は他になかったが、pGX1438およびpGX1439により誘導されたCD8+ T細胞応答がより頑強である傾向があった。
