抗体薬物の開発は現在、主にその他物種由来の抗体をヒト化させること、又は遺伝子改変動物及び体外スクリーニング技術で完全ヒト抗体をスクリーニングすることにより実現し、これは抗体が人体における免疫原性をより低下させ、対応する副作用を軽減し、同時に成薬性を向上させているので、抗体薬物の発展重要方向の一つである。
本実施例において、細胞によるファージディスプレイスクリーニングに使用するために、CLDN18.2を安定発現するCHO細胞株(CHO-CLDN18.2)を製作した。その具体的な実験過程は次のとおりである:ヒトCLDN18.2のcDNA配列をpCDHレンチウイルスベクター中にクローンし、その後レンチウイルスパッケージングベクターと共に293t細胞にトランスフェクションする。48又は72時間培養後、レンチウイルス粒子を多く含む細胞上清を収集し、そのままCHO細胞、CT26細胞又はSNU601細胞を感染する。レンチウイルス感染後の細胞をピューロマイシンでスクリーニング培養をし、2-3周後にCLDN18.2特異性抗体(IMAB362、Ganymed)によるフローサイトメトリーで細胞のCLDN18.2発現を検出し、その結果を図1に示す。
CHO-CLDN18.2安定発現細胞株を利用し、ファージディスプレイライブラリーの選択を三回行った。実験手順は主に以下の文献を参考した:Targeting membrane proteins for antibody discovery using phage display Jones ML et al.Scientific Reports 2016。
モノクローナルファージの重鎖と軽鎖可変領域のcDNA配列をすでに抗体定常領域を含有するpcDNA3.4ベクター(Invitrogen)中にそれぞれクローンし、合計6種類のモノクローナル抗体の重鎖と軽鎖発現プラスミドを獲得した。PEI法でプラスミドをEXPI-293細胞(Invitrogen)中にトランスフェクションし、7-10日一過性トランスフェクションし、遠心して上清液を回収した。上清液をprotein Aで精製し、精製した抗体を得た。前記6種類のモノクローナル抗体をそれぞれ18.2-A1(A1)、18.2-A2(A2)、18.2-A3(A3)、18.2-A4(A4)、18.2-A5(A5)と18.2-A6(A6)に命名した。これらの抗体の重鎖と軽鎖可変領域のアミノ酸配列並びにコードする配列は以下の表1と表2に示す。Kabat CDRシステムで確定した抗体のCDR配列は表3に示す。組換え抗体の重鎖と軽鎖定常領域配列は表4に示す。
組換えモノクローナル抗体のCLDN18.2に対する結合活性を検出した。具体的には、成長が良好なCT26-CLDN18.2安定発現細胞株を取る。細胞をPBSで洗浄した後、倍数で希釈した抗体と混合し、4℃で1時間インキュベートした。細胞をフローサイトメトリーバッファーで一回洗浄し、PE標識抗ヒトIgG抗体を入れ、4℃で30分インキュベートした。遠心して上清を取り除き、フローサイトメトリーバッファーで洗浄した後、フローサイトメーターで細胞表面の蛍光強度を検出した。平均蛍光強度で各種抗体の相対結合活性を計算した。
ヒトClaudin18タンパク質には二つアイソフォーム、即ちCLDN18.1とCLDN18.2があり、この二つアイソフォームはエクソン1以外の他のエクソンを共有し、且つCLDN18.1とCLDN18.2のエクソン1配列は極めて似ていて、そのなか、第一の細胞外ドメインにおいて、異なるアミノ酸が僅か8つしか存在しない。CLDN18.1とCLDN18.2エクソン1は、それぞれ異なるプロモーターの制御下で、特異的に異なる組織に発現する。そのなか、CLDN18.1は肺上皮細胞に特異的に発現し、CLDN18.2 は胃上皮細胞に特異的に発現する。スクリーニングされたCLDN18.2抗体の特異性を鑑定するために、抗体とCLDN18.1及びCLDN18.2の結合を検出した。
すでに報告されたように、一部の抗体は細胞表面の抗原に結合した後、抗原と抗体の復合物のエンドサイトーシスを引き起こすことができ、これにより細胞表面抗原の発現レベルを低下させ、抗体が体内における代謝を加速させる(Schrama D et al.2006、Nat Rev Drug Discov)。本実施例では、CLDN18.2安定発現SNU601細胞において、抗体18.2-A1、18.2-A3、18.2-A4及び18.2-A6の細胞にエンドサイトーシスされる能力を検出した。具体的に、濃度が10μg/mlであるCLDN18.2抗体18.2-A1、18.2-A3、18.2-A4、18.2-A6又はIMAB362抗体(例えば、実施例2に記載)、空白コントロール(PBS)及びアイソタイプコントロール(ISO)をそれぞれ細胞と4℃で又は37℃で4時間インキュベートした。その後、4℃で洗浄を行い、PE標識抗ヒトIgG抗体で染色をした。4%ホルマリンによる固定後、フローサイトメーターで細胞表面の抗体結合を検出した。
さらにCLDN18.2抗体の脱フコシル化形態を製作した。具体的に、抗体の重鎖と軽鎖をコードする配列を安定発現できる哺乳動物GS発現ベクター中にクローンした。重鎖と軽鎖をコードする配列はすべてCMVプロモーターに制御される。発現ベクターを無血清及び懸濁培養で家畜化されたCHO-K1細胞とFut8遺伝子がノックアウトされた家畜化CHO-K1細胞株にトランスフェクションした。MSXスクリーニングで安定発現細胞株を選択した。安定発現細胞株を得た後、振とうフラスコで12-14日培養し、期間内に必要に応じてサプリメント培地を補充した。
その後、Fut8遺伝子ノックアウトCHO-K1細胞株に発現した脱フコシル化抗体とCHO-K1細胞に生成した通常抗体の糖鎖化修飾を分析した。100μgの抗体をトリプシンとグリコシルペプチダーゼで分解した後に精製して抗体の糖鎖を獲得し、その後蛍光タンデム質量分析を行った。各種糖鎖の鑑定はその質量電荷比m/zによるものであり、蛍光により検出した面積百分比で糖鎖百分比を計算した。
フローサイトメトリーにより、18.2-A1、18.2-A3及び18.2-A4とCLDN18.2との結合活性と比べての、脱フコシル化抗体18.2-A1F、18.2-A3F及び18.2-A4FとCLDN18.2との結合活性を検出し、その検出方法は実施例5に記載したものと同じであり、実験結果は図7に示す。
その後、Biacore方法でCLDN18.2抗体及びその脱フコシル化形態とFcγRIIIaとの結合活性を検出した。具体的に、FcγRIIIa(Sino Biological Inc、10389-H08C1)をHBS-EPバッファーで0.1μg/mlに希釈し、配体とする。そして18.2-A1Fと18.2-A1の抗体サンプルをそれぞれ360μg/ml、120μg/ml、40μg/ml、13.3μg/ml及び4.4μg/mlに希釈して、分析物とする。間接的な捕獲の方法で配体FcγRIIIaを固定し、まず50μg/mlのAnti-His IgGを使い、アミノカップリングでCM5チップの表面に共有結合し、その後に配体及び分析物に結合する。Biacore Wizardモードで複数循環方式を使い、FcγRIIIaを配体とし、18.2-A1Fと18.2-A1の抗体サンプルを分析物とし、親和力分析実験を行った。
次に、我々はさらにCLDN18.2抗体及びその脱フコシル化形態と細胞表面で発現したFcγRIIIaとの結合活性を検出した。具体的に、ヒトFcγRIIIa(V158、高FC結合サブタイプ)遺伝子をピューロマイシンスクリーニング標識を持つ哺乳動物細胞発現ベクターにクローンし、Jurkat細胞株中にトランスフェクションした。ピューロマイシンでFcγRIIIaを安定発現する細胞株(FcγRIIIa-Jurkat)をスクリーニングした。また、NK92MIはヒトNK細胞株であり、FcγRIIIa受容体(F158、低FC結合サブタイプ)を天然的に発現する。フローサイトメトリーでCLDN18.2抗体及びその脱フコシル化形態とFcγRIIIa-jurkat細胞及びNK92MI細胞表面で発現したFcγRIIIaとの結合能力を検出した。具体的なフローサイトメトリー検出方法は実施例6に記載したものを参照する。
FcγRIIIa受容体は抗体FC領域と結合した後、エフェクター細胞内のNF-AT転写因子通路を活性化させる。したがって、NF-ATが媒介するレポーター遺伝子強度の検出はFcγRIIIa受容体の活性化強度を反映することができる。本実施例において、NF-AT結合サイトを含有するプロモーターをルシフェラーゼレポーター遺伝子発現ベクターに挿入し、これをJurkat細胞株に安定的にトランスフェクションした。同時に、高FC結合のFcγRIIIa(V158)もJurkat細胞に安定的にトランスフェクションし、FcγRIIIa受容体の活性化強度を感知できる機能性細胞株(FcγRIIIa-Jurkat)を構築した。
現在、EOF(エピルビシン、Epirubicin;オキサリプラチン、Oxaliplatin;5-フルオロウラシル、5-FU)を含む併用化学療法は、胃癌の主な治療手段である。EOF感受性細胞株はEOF処理後で、異なる程度の細胞分裂周期の遮断、増殖阻害及びアポトーシスが起こされる。
Ficoll-Paque Plus(GE Healthcare)で健常人(ドナー1とドナー2)の末梢血白血球(PBMC)を分離して、37℃で一晩中培養した。CLDN18.2安定発現SNU601胃癌細胞(SNU601-CLDN18.2)を取り1x105/mlに希釈し、PBMCを5x106/mlに希釈して、両者を同じ体積で混合し、エフェクター細胞と標的細胞の比率が50:1である。100μl混合細胞を取り、96ウェルプレートに入れ、そして倍比希釈した抗体をそれぞれ入れて、37℃で24時間静置培養した。その後、乳酸脱水素酵素(LDH)法(promega)で細胞生存率を検出した。そのなか、マイクロプレートリーダーによりOD490での吸光度を測定する。傷害率の計算方法は以下の通りである:

CLDN18.2安定発現HELA細胞を取り、1%不活化補体を含有する血清を含む培地で再懸濁カウントし、細胞濃度を2x105/mlに調整した。細胞生存率が90%を超えた。96ウェルプレートにウェルあたり50μlの細胞を入れ、そして倍比希釈した抗体と50μl希釈後のヒト血清をそれぞれ入れた。細胞を37℃で2時間インキュベートした。その後、CCK8法で細胞溶解率を測定した。細胞溶解率の計算方法は以下の通りである:

NPG免疫不全マウスに対して、SNU601-CLDN18.2細胞とFicollで分離したヒトPBMC細胞(SNU601-CLDN18.2細胞:PBMCが1:0.8)を皮下接種することで、SNU601細胞のマウス異種移植モデルを建築した。その後、SNU601腫瘍細胞を接種したNPG免疫不全マウスに腹腔内注射で10mg/kg又は5mg/kg用量の18.2-A1F抗体18.2-A1F、又は空白コントロール(PBS)又はIgGアイソタイプコントロール(ISO)を注射した。各組n=マウス15匹。腫瘍接種後に3日ごとに投薬して、マウス体内腫瘍の体積を測定した。
実施例14に記載した結果のように、体外でのEOF併用化学療法は、CLDN18.2抗体に誘導されたFcγRIIIa受容体活性化を向上させることができる。KATOIIIは体外IMDM培地において培養する時、レベルの低いCLDN18.2を発現し、僅か約5.2%の細胞はフローサイトメトリー検出で明らかな陽性を示した(図11)。フローサイトメトリーでソーティング後、CLDN18.2高発現のKATOIII細胞(KATOIII-18.2High)を選択して、NPG免疫不全マウスに接種し、異種移植モデル実験を行った(図16)。

b.配列番号21に示すVHにおけるCDRH1、CDRH2とCDRH3;及び配列番号26に示すVLにおけるCDRL1、CDRL2とCDRL3;
c.配列番号31に示すVHにおけるCDRH1、CDRH2とCDRH3;及び配列番号36に示すVLにおけるCDRL1、CDRL2とCDRL3;
d.配列番号41に示すVHにおけるCDRH1、CDRH2とCDRH3;及び配列番号46に示すVLにおけるCDRL1、CDRL2とCDRL3;又は
e.配列番号55に示すVHにおけるCDRH1、CDRH2とCDRH3;及び配列番号60に示すVLにおけるCDRL1、CDRL2とCDRL3。 - 前記抗体は以下のCDRHとCDRLを有する、請求項1に記載の抗体又はその抗原結合断片:
a.配列番号3-5に示すCDRH1、CDRH2とCDRH3;及び配列番号8-10に示すCDRL1、CDRL2とCDRL3;
b.配列番号23-25に示すCDRH1、CDRH2とCDRH3;及び配列番号28-30に示すCDRL1、CDRL2とCDRL3;
c.配列番号33-35に示すCDRH1、CDRH2とCDRH3;及び配列番号38-40に示すCDRL1、CDRL2とCDRL3;
d.配列番号43-45に示すCDRH1、CDRH2とCDRH3;及び配列番号48-50に示すCDRL1、CDRL2とCDRL3;又は
e.配列番号57-59に示すCDRH1、CDRH2とCDRH3;及び配列番号62-64に示すCDRL1、CDRL2とCDRL3。 - 前記抗体は以下のVHとVLを有する、請求項1又は2に記載の抗体又はその抗原結合断片:
a.配列番号1のアミノ酸配列を含むVH、及び配列番号6のアミノ酸配列を含むVL;
b.配列番号21のアミノ酸配列を含むVH、及び配列番号26のアミノ酸配列を含むVL;
c.配列番号31のアミノ酸配列を含むVH、及び配列番号36のアミノ酸配列を含むVL;
d.配列番号41のアミノ酸配列を含むVH、及び配列番号46のアミノ酸配列を含むVL;又は
e.配列番号55のアミノ酸配列を含むVH、及び配列番号60のアミノ酸配列を含むVL。 - 前記抗体はFc領域を有する、請求項1-3のいずれか一項に記載の抗体又はその抗原結合断片。
- Euナンバリングシステムでナンバリングする場合、前記抗体はAsn297において糖鎖構造修飾を有する、請求項4に記載の抗体又はその抗原結合断片。
- 前記抗体はモノクローナル抗体である、請求項1-5のいずれか一項に記載の抗体又はその抗原結合断片。
- 前記抗体は二重特異性抗体又は多重特異性抗体である、請求項1-5のいずれか一項に記載の抗体又はその抗原結合断片。
- 前記抗体はIgG、IgA、IgM、IgEとIgDのアイソタイプから選択される、請求項1-7のいずれか一項に記載の抗体又はその抗原結合断片。
- 前記抗体はIgG1、IgG2、IgG3とIgG4のサブタイプから選択される、請求項1-7のいずれか一項に記載の抗体又はその抗原結合断片。
- 前記抗原結合断片はFab断片、Fab’断片、F(ab’)2断片、Fd断片、Fd’断片、Fv断片、scFv断片、ds-scFv断片、dAb断片、一本鎖断片、二価抗体及び線状抗体から選択される、請求項1-9のいずれか一項に記載の抗体又はその抗原結合断片。
- 請求項1-10のいずれか一項の抗体又はその抗原結合断片をコードするヌクレオチド配列を含む、核酸分子。
- 請求項11に記載の核酸分子を含む、ベクター。
- 請求項11に記載の核酸分子又は請求項12に記載のベクターを含む、宿主細胞。
- 治療剤、診断剤又は顕像剤とコンジュゲート化する請求項1-10のいずれか一項に記載の抗体又はその抗原結合断片を含む、コンジュゲート。
- 請求項1-10のいずれか一項に記載の抗体又はその抗原結合断片又は請求項14に記載のコンジュゲート、及び、一種又は多種の薬学的に許容される担体、賦形剤及び/又は希釈剤を含む、組成物。
- Euナンバリングシステムでナンバリングする場合、前記抗体のAsn297における糖鎖構造において、フコースの糖鎖構造を有する割合が50%又はそれ以下である、請求項15に記載の組成物。
- 前記抗体の前記糖鎖構造において、フコースの糖鎖構造を有する割合が30%又はそれ以下、例えば20%又はそれ以下、10%又はそれ以下、5%又はそれ以下、2%又はそれ以下、又は1%又はそれ以下である、請求項16に記載の組成物。
- 前記抗体において、フコースの糖鎖構造を有する割合が0%-1%である、請求項16に記載の組成物。
- 前記抗体はFut8遺伝子ノックアウト細胞によって生成される、請求項16-18のいずれか一項に記載の組成物。
- 前記細胞はCHO細胞とHEK293細胞から選択される、請求項19に記載の組成物。
- Fut8遺伝子ノックアウトを有しない細胞において生成した抗体と比べ、前記抗体がより高いFcγRIIIa結合活性を有する、請求項19又は20に記載の組成物。
- Fut8遺伝子ノックアウトを有しない細胞において生成した抗体と比べ、前記抗体がより高いADCC活性を有する、請求項19-21のいずれか一項に記載の組成物。
- 前記組成物は一種又は多種の他の治療剤をさらに含む、請求項15~22のいずれかに記載の組成物。
- 前記治療剤は抗体、化学療法の薬物と小分子薬物から選択される、請求項23に記載の組成物。
- 前記化学療法の薬物はエピルビシン(Epirubicin)、オキサリプラチン(Oxaliplatin)及び5-フルオロウラシル(5-FU)から選択される一種又は多種である、請求項24に記載の組成物。
- 患者のCLDN18.2の発現に関連する疾患を治療するための、請求項15-25のいずれか一項に記載の組成物。
- 前記疾患は癌である、請求項26に記載の組成物。
- 前記癌は胃癌、胆管癌、食道癌と膵臓癌から選択される、請求項27に記載の組成物。
- 一種又は多種の他の療法と併用して患者に投与することに利用される、請求項26-28のいずれか一項に記載の組成物。
- 前記他の療法は、化学療法、放射療法、免疫療法及び手術治療から選択される、請求項29に記載の組成物。
- 前記免疫療法は、免疫チェックポイント分子に対する療法、CAR-T細胞療法及びCAR-NK細胞療法から選択される、請求項30に記載の組成物。
