抗ヒト4-1BBモノクローナル抗体ハイブリドーマ細胞をELISAによってスクリーニングした。ELISAプレートを10μg/mlのヒト4-1BB(配列番号5)で被覆し、4℃で一晩ブロックした。試験細胞培養上清を順番に添加し、37℃で1時間インキュベートし、プレートをPBSTで3回洗浄した。4000倍で希釈されたホースラディッシュ・ペルオキシダーゼで標識されたヤギ抗マウス(ThermoFisher社)を添加し、37℃で45分間インキュベートした。プレートをPBSTで3回洗浄した後、テトラメチルベンジジン(tetramethylbenzidine、TMB)(ThermoFisher社)基質で発色させた後、OD値を450nmの波長で測定した。
スクリーニングして得た、陽性で且つよく増殖している抗ヒト4-1BB抗体ハイブリドーマ細胞株を、段階的に拡大培養し、遠心分離(10000rpmで10分間)して上清を採取した。得た上清をProtein Gアフィニティクロマトグラフィーカラムにより精製した。詳細な操作としては、まず、Protein Gカラム(GE社)をPBSで平衡化し、次に、培養上清はカラムを通過させた。液体A(配合:溶媒は水であり、溶質及び濃度は、20mMリン酸ナトリウム、500mMNaCl、pH5.0である)を使用して、5つのカラムの容積分の予溶出を行った後、液体B(配合:溶媒は水であり、溶質及び濃度は、20mM酢酸ナトリウム、150mMNaCl、pH3.5である)を使用して、5つのカラムの容積分の溶出を行い、溶出ピーク分を収集した。その後、30KDa濃縮遠心管を使用して濃縮することで、抗体を得た。
4種のハイブリドーマが分泌する抗体がヒト4-1BBを認識する親和性を、ELISAにより評価した。96ウェルプレート(96ウェルマイクロプレート、Nunc社)をヒト4-1BB(配列番号5)で被覆し、濃度1μg/ml、100μl/ウェルの条件で、4℃で一晩放置した。プレートを3回洗浄し、300μlブロッキング溶液(配合は上記の通りである)をウェル毎に加え、37℃で1時間ブロッキングした。プレートを洗浄液(配合は上記の通りである)で3回洗浄し、抗体を試料希釈液(1%ウシ血清アルブミンを加えたPBS-T)で4000ng/mlに希釈し、7本のEpチューブで4倍希釈し、勾配溶液とした。濃度ごとに、2つの複製ウェルを設け、100μl/ウェルの条件で、1時間インキュベートした。プレートを洗浄液で3回洗浄し、8000倍希釈された、ホースラディッシュ・ペルオキシダーゼで標識されたヤギ抗マウス(ThermoFisher社)100μlを各ウェルに加え、0.5時間振とうした。プレートを3回洗浄し、テトラメチルベンジジン(tetramethylbenzidine,TMB)(ThermoFisher社)100μlを各ウェルに加えた。遮光下で発色させ、1M硫酸を加えて反応を停止させた。OD値をBIO-TEKELX-800マイクロプレートリーダーを用いて450nmで測定した。
ハイブリドーマが分泌する抗体がサル4-1BBを認識する親和性を、ELISA方法により評価した。具体的に、サル4-1BB(Cy4-1BB)(配列番号6)抗原をプレートコーティングバッファー(配合は上記の通りである)で1μg/mlに希釈し、各ウェルに100μlを加えてプレートコーティングし、4℃で一晩放置した。ハイブリドーマから分泌された抗体、及び陽性対照抗体Utomilumabを試料希釈液で1000ng/mlに希釈し、7本のEpチューブを用いて10倍希釈し勾配溶液とした。濃度ごとに、2つの複製ウェルを設け、100μl/ウェルでプレートに添加し、1時間振とうした。プレートを再度洗浄した後、8000倍希釈された、ホースラディッシュ・ペルオキシダーゼで標識されたヤギ抗マウス(ThermoFisher社)100μlを各ウェルに加え、0.5時間振とうした。洗浄後、テトラメチルベンジジン(tetramethylbenzidine,TMB)(ThermoFisher社)100μlを添加し、発色させた。
ELISAにより、ヒト4-1BB分子及びサル4-1BB分子に対するscFv分子の親和性を同定した(具体的な方法については、実施例1に記載の当該手順のとおり)。具体的に表4に示すように、12種のscFv-Fc分子は、いずれもヒト4-1BB及びサル4-1BBの両方を認識できたという結果が示された。ただし、scFvB60103の認識能力が最も良かった。ELISAの結果は図5及び図6に示す。
PBSへBSAとアジ化ナトリウムを添加して得たFACSバッファー(BSAの質量%濃度が2%であり、アジ化ナトリウムの質量%濃度が0.02%である)、ヤギ抗ヒトFITC二次抗体(Sigma社、遮光保存)。
この実施例で同定された例示的な抗体Hanke10F4は、ハイブリドーマ分泌型抗体37G10F4をヒト化して得たもの、すなわち、表4のscFvB60103から変換されたものである。この実施例では、Hanke10F4の発現及び精製、その後の、ELISA、FACS、ルシフェラーゼのレポーター遺伝子の検出、T細胞活性化システムにおける同定について、詳細に説明する。
全長遺伝子配列を、配列決定された遺伝子配列に基づいて合成した。Hanke10F4抗体の重鎖をコードする遺伝子は配列番号3で示され、Hanke10F4抗体の軽鎖をコードする遺伝子は配列番号4で示される。
測定方法は、実施例1の手順1.4と同様にして、96ウェルプレートをそれぞれヒト4-1BB又はサル4-1BBで直接に被覆した。なお、抗体サンプル(Hanke10F4及びUtomilumab)を一次抗体とし、ホースラディッシュ・ペルオキシダーゼで標識されたヤギ抗マウスを二次抗体とした。また、テトラメチルベンジジン(tetramethylbenzidine、TMB)を発色剤とし、硫酸で発色を停止させた。BIO-TEKELX-800マイクロプレートリーダーを使用して、450nmでOD値を測定した。
PBSへBSAとアジ化ナトリウムを添加して得たFACSバッファー(BSAの質量%濃度が2%であり、アジ化ナトリウムの質量%濃度が0.02%である)、ヤギ抗ヒトFITC二次抗体(Sigma社、遮光保存)。
h4-1BB-His(Acro社、製品番号:Q07011-1)抗原を使用し、Fortebio octec装置によりバイオレイヤー干渉法(BLI)技術を用いてUtomilumab、Hanke10F4、又はUrelumabの親和性を測定した。
Hanke10F4抗体とヒト4-1BBLとの間の競合結合活性の比較に、CHO-K1-hu4-1BBを使用した。二次抗体は、SA-FITC(Biolegend社)であり、その他の操作(下記は除く)及び希釈液は、手順3.2に記載のFACS測定と同じである。ヒト4-1BBLアミノ酸配列は、配列番号9で示される。
Hanke10F4抗体と対照抗体であるUtomilumab、Urelumabとの間のエピトープ競合関係の比較に、CHO-K1-hu4-1BBを用いた。二次抗体はSA-FITC(Biolegend社)であり、その他の操作(下記は除く)及び希釈液は、手順3.2に記載のFACS測定と同じである。
ヒト4-1BB抗原の細胞外ドメインを、段階的にマウス4-1BBの対応する細胞外ドメインに置き換え、それぞれの目的遺伝子を合成した。それぞれpCDNA3.4ベクターに挿入して、それぞれの4-1BB抗原のDNAプラスミドを得た。一過性トランスフェクションにより、それぞれの4-1BB抗原を細胞表面において発現させる細胞株を得た。具体的には表12のとおりである。FACSを用いて、抗体と、それぞれの4-1BB抗原との結合活性を分析し、抗体がヒト4-1BBの細胞外ドメインに結合する具体的なエピトープを更に分析した。
文献の分析によると、ヒト4-1BB抗原における101番目、132番目のアミノ酸はUtomilulabにおける重要な結合部位であり、42番目はUrelumabにおける重要な結合部位である。上記部位のアミノ酸を一つずつ突然変異させ、それぞれの目的遺伝子を合成した。これにより、それぞれの4-1BB抗原を有するDNAプラスミドを得た。一過性トランスフェクションにより、それぞれの4-1BB抗原を細胞表面において発現させる細胞株を得た((1)方法1の欄に記載の関連手順に従って得た)。具体的には表13に示す。
Hanke10F4抗体とHKB6との間の、抗原に結合するエピトープ競合関係を、CHO-K1-hu4-1BBを用いて比較した。二次抗体はSA-FITC(Biolegend社)であり、他の手順(下記は除く)及び希釈液は、手順3.2に記載のFACS測定と同じである。
HEK-293/NFκB-Luci/41-BB細胞は、China Hefei HankeMab Companyにより構築された、ヒト4-1BB膜タンパク質及びNFκB-Luciferaseレポーター遺伝子を安定に発現させるHEK293細胞株である。具体的には以下のように操作した。ヒト4-1BB配列(配列番号7)を目的遺伝子としてpCDNA3.4ベクターに挿入してプラスミドAを得た。同時にNFκBエレメント配列(配列番号12)とルシフェラーゼ遺伝子(配列番号13)を目的遺伝子とし、該分野の一般的手法により、pGL4.10ベクター(Youbio company, China)におけるそれぞれの部位に挿入し、プラスミドBを得た。次いでLipofectamine3000トランスフェクション試薬(Invitrogen社)を用いてAプラスミド及びBプラスミドの両方をともにHEK293細胞(Shanghai Cell Bank, Chinese Academy of Sciences)に導入した。これに、Puromycin(Gibco)とHygromycinB(Sangon Biotech (Shanghai) Co.,Ltd.)を添加し、加圧スクリーニングを行い、最終的にはHEK-293/NFκB-Luci/41-BBを得た。
抗CD3抗体(Biolegend、品番317325)をPBSで1μg/mlになるまで希釈して抗CD3抗体溶液を得た。Hanke10F4抗体をPBSで希釈して高、中、低の3つの濃度としたことで、Hanke10F4抗体の濃度が4μg/ml、0.4μg/ml、0.04μg/mlである溶液をそれぞれ得られた。抗CD3抗体溶液を、3つの濃度のHanke10F4抗体溶液と1:1の体積比でそれぞれ均一に混合した後、50μl/ウェルで96ウェルプレートに添加した。なお、3つの複製ウェルを設けた。96ウェルプレートを37℃で静置して1時間インキュベートした。翌日の使用の前にPBSで3回洗浄した。また、Utomilumabを陽性対照抗体とし、IgG(Biolegend、品番403601)を陰性対照抗体とした。
健康な被験者から全血を採取し、リンパ球分離液(Sigma社)を用いて、その説明書に従って操作してPBMC細胞を単離した。また、ヒトCD4+T細胞磁気ビーズ(BD社、品番557767)を用いて、説明書に従ってヒトCD4+T細胞を分離・精製し、予備とした。
抗CD3抗体をPBSで0.5μg/mlに希釈し、96ウェルプレート(Corning)に60μl/ウェルで添加し、96ウェルプレートを37℃で静置して1時間インキュベートした。PBSで洗浄し、次いでCHO-K1安定発現細胞株(CHO-K1-CD32AまたはCHO-K1-CD32BまたはCHO-K1)を100μl/ウェル、1×104個/ウェルで96ウェルプレートに加えた。次いで96ウェルプレートを細胞インキュベーターに一晩置いた。
健康な被験者から全血を採取し、リンパ球分離液(Sigma社)を用いて、その説明書に従って操作してPBMC細胞を単離した。また、ヒトCD8+T細胞磁気ビーズ(BD社、品番557766)を用いて、説明書に従ってヒトCD8+T細胞を分離・精製し、予備とした。
実験では、B-hCD137(4-1BB)マウスを選択して、Hanke10F4のインビボでの抗腫瘍薬効を測定した。B-hCD137(4-1BB)マウスモデルは、具体的には遺伝的背景C57BL/6マウスのゲノムにヒト由来のh4-1BB遺伝子を嵌合してなる遺伝子改変マウスであり、Biocytogen Pharmaceuticals (Beijing) Co., Ltdから入手した。
実験では、B-hCD137(4-1BB)マウスを選択して、Hanke10F4のインビボでの抗腫瘍薬効を測定した。B-hCD137(4-1BB)マウスモデルは、具体的には遺伝的背景C57BL/6マウスのゲノムにヒト由来のh4-1BB遺伝子を嵌合してなる遺伝子改変マウスであり、Biocytogen Pharmaceuticals (Beijing) Co., Ltdから入手した。
高(30mg/kg)、中(10mg/kg)、低(3mg/kg)の3つの投与ドーズを設定して、40匹のSDラット(メス半分、オス半分)へHanke10F4を単回の静脈注射で投与し、それぞれの時点における機能観察総合評価(FOB)試験に基づいて、SDラットの中枢神経系に対する、Hanke10F4の影響を評価した。
4匹のカニクイザルを、性別に基づいて分け、体重に基づいてドーズクライミンググループ、固定ドーズグループの合計2つのグループにランダムに割り当てた。なお、1グループにつき2匹、且つメス半分、オス半分であった。それらに静脈注射のルートで投薬した。詳細は表16を示す。
調製したF01(pH5.2)とF02(pH5.5)の2種類の緩衝液を用い、Hanke10F4を20mg/mlとし、4℃の条件で3ヶ月間(3M)放置して、抗体濃度、凝集分解等の各物理化学的特性を測定した。具体的には、光路長可変型分光光度計によりHanke10F4タンパク質の濃度を測定し、高速液体クロマトグラフィーによりHanke10F4の分子サイズ、ヘテロ純度(SEC-HPLC)及び電荷ヘテロ純度(CEX-HPLC)を測定し、キャピラリー電気泳動分析システムによりHanke10F4非還元キャピラリーゲル電気泳動タンパク質純度(NR-CE)を測定した。結果は表19に示す。Hanke10F4は、それぞれのpHの緩衝液中に4℃で3ヶ月間放置した後、初期時のサンプル(Hanke10F4/T0)と比較して、明らかな変化がなく、Hanke10F4の安定性が良好であった。
示差走査蛍光(differential scanning fluorimetry、DSF)により、両波長(350nmと330nm)における、抗体Hanke10F4の蛍光強度を測定し、蛍光強度比を算出した。また、タンパク質のアンフォールディング期間中の蛍光強度及び蛍光ピーク値の変化を測定し、Hanke10F4のタンパク質が半分アンフォールディングされた時点の温度、すなわち、Tm値を得た。結果として、Hanke10F4のTm1値は64.4℃であり、Tm2値は71.5℃(Tm1とTm2は、タンパク質のアンフォールディング期間中に蛍光ピークが変化したときの2つの温度である)であった。つまり、Hanke10F4抗体の安定性は良好であった(図19)。

- 抗ヒト4-1BB抗体またはその抗原結合フラグメントであって、前記抗体または前記抗原結合フラグメントは各々、軽鎖可変領域および重鎖可変領域を含み、
前記重鎖可変領域におけるHCDR1のアミノ酸配列が配列番号1のN末端から31~35番目の位置で示され、前記重鎖可変領域におけるHCDR2のアミノ酸配列が配列番号1のN末端から50~64番目の位置で示され、前記重鎖可変領域におけるHCDR3のアミノ酸配列が配列番号1のN末端から98~106番目の位置で示され、
前記軽鎖可変領域におけるLCDR1のアミノ酸配列が配列番号2のN末端から24~34番目の位置で示され、前記軽鎖可変領域におけるLCDR2のアミノ酸配列が配列番号2のN末端から50~56番目の位置で示され、前記軽鎖可変領域におけるLCDR3のアミノ酸配列が配列番号2のN末端から89~97番目の位置で示されることを特徴とする、抗ヒト4-1BB抗体またはその抗原結合フラグメント。 - 前記重鎖可変領域のアミノ酸配列が、配列番号1のN末端から1~117番目の位置であるか、または
前記軽鎖可変領域のアミノ酸配列が、配列番号2のN末端から1~107番目の位置である
ことを特徴とする、請求項1に記載の抗ヒト4-1BB抗体またはその抗原結合フラグメント。 - 前記抗体は、IgG、IgM、IgE、IgA、またはIgDであり、前記IgGは、IgG1サブタイプ、IgG2サブタイプ、IgG3サブタイプ、またはIgG4サブタイプであり、および/または
前記抗体の軽鎖タイプは、κ鎖である
ことを特徴とする、請求項1に記載の抗ヒト4-1BB抗体またはその抗原結合フラグメント。 - 前記抗体の重鎖におけるアミノ酸配列は、配列番号1であるか、または
前記抗体の軽鎖におけるアミノ酸配列は、配列番号2である
ことを特徴とする、請求項1に記載の抗ヒト4-1BB抗体またはその抗原結合フラグメント。 - 前記抗原結合フラグメントは一本鎖抗体であり、前記一本鎖抗体のアミノ酸全配列は、配列番号16の1~239番目の位置であることを特徴とする、請求項1に記載の抗ヒト4-1BB抗体またはその抗原結合フラグメント。
- 下記の(i)~(iii)のいずれか一つである抗体誘導体:
(i)請求項1に記載の前記抗体またはその抗原結合フラグメントと、他のタンパク質とから形成される抗体融合タンパク質;
(ii)請求項1に記載の前記抗体またはその抗原結合フラグメントと、他のターゲット抗体とから形成される二重抗体;
(iii)請求項1に記載の前記抗体若しくはその抗原結合フラグメント、または前記抗体融合タンパク質、または前記二重抗体と、生物活性を有する小分子薬物とのカップリングにより形成されるADC抗体。 - 前記抗体融合タンパク質における前記他のタンパク質は、タグタンパク質であることを特徴とする、請求項6に記載の抗体誘導体。
- 前記タグタンパク質は、Fcであることを特徴とする、請求項7に記載の抗体誘導体。

- 前記Fcのアミノ酸配列は、配列番号16のN末端から240~472番目の位置で示されることを特徴とする、請求項8に記載の抗体誘導体。
- 前記タグタンパク質を有する一本鎖抗体のアミノ酸配列は、配列番号16で示される通りであることを特徴とする、請求項11に記載の抗体誘導体。
- 以下のいずれか一つである組成物:
(C1)請求項1に記載の抗体若しくはその抗原結合フラグメント、又は請求項6に記載の抗体誘導体を含む、4-1BBを検出するための組成物;
(C2)請求項1に記載の抗体若しくはその抗原結合フラグメント、又は請求項6に記載の抗体誘導体を含む、4-1BB/4-1BBLシグナル伝達経路を遮断するための組成物;
(C3)請求項1に記載の抗体若しくはその抗原結合フラグメント、又は請求項6に記載の抗体誘導体を含む、T細胞活性化を刺激するための組成物;

- 前記癌は4-1BB発現の調節不全を示す癌であり、前記自己免疫疾患は4-1BB発現の調節不全を示す自己免疫疾患であり、前記炎症性疾患は4-1BB発現の調節不全を示す炎症性疾患であり、前記感染性疾患は4-1BB発現の調節不全を示す感染性疾患であることを特徴とする、請求項14に記載の組成物。
