AAVの作製
AAVベクターは、コトランスフェクション法を使用して以前に記載されたようにして作製し、イオジキサノール勾配超遠心分離によって精製した(49)。AAVベクターストックは、SYBR Green(Thermo Fischer Scientific社)を使用して定量的PCRにより力価を測定した(50)。
本研究では、野生型C57BL6/j(Janvier Labs社)又はrd10マウス(Vision Institute社の動物施設にて繁殖及び飼育)、並びにマカクザル(Noveprim社、モーリシャス)を使用した。
瞳孔拡張後、Spectralis HRA+OCTシステム(Heidelberg Engineering社、ドイツ)を用いて、488nmの励起波長と500nmのバリアフィルターで構成される「眼底自己蛍光モード」を使用して、OCT画像とGFPの蛍光画像を取得した。
パルスフェムト秒レーザー(InSight(商標) DeepSee(商標)、Newport Corporation社)を備えた40×水浸対物レンズ(LUMPLFLN40x/W/0.80、Olympus社)を備えた2光子顕微鏡を使用して、ホールマウントの(視細胞側を上にした)網膜又は網膜スライス(垂直断面)でGFP陽性網膜細胞を画像化した。AAVで処置したマカク網膜を単離し、その後、酸素化(95%O2、5%CO2)Ames培地(Sigma-Aldrich社)中で画像化した。ライブ2光子イメージングでは、網膜を顕微鏡の記録チャンバーに配置し、波長930nmの励起レーザーを使用してzスタックを取得した。画像は、ImageJ(NIH)を使用してオフラインで処理した。全細胞パッチクランプ記録には、Axon Multiclamp 700B増幅器を使用した。電極は、ホウ珪酸ガラス(BF100-50-10、Sutter Instruments社)で製作し、6~9MΩに引き合わせた。ピペットは、115mM Kグルコン酸、10mM KCl、1mM MgCl2、0.5mM CaCl2、1.5mM EGTA、10mM HEPES、及び4mM ATP-Na2(pH 7.2)で満たした。細胞は、電圧クランプ構成で-40mVの電位でクランプするか、電流クランプ(電流ゼロ)構成で記録した。網膜は、記録に先立ち、記録チャンバー内で少なくとも30分、暗順応させた。
我々は、以前に公開されたプロトコールに基づき、ヒトiPSCから網膜オルガノイドを作製した(37)。クローンhiPSC-2は、既に記述されているように(37)、「前神経培地」中において、出生後ヒト包皮(ATCC CRL 2429)由来の線維芽細胞フィーダー上で増幅及び分化させた。非常にコンフルエントな付着性のiPS細胞培養から開始して、線維芽細胞増殖因子2(FGF2)の非存在下で、2週間後に自己形成網膜オルガノイドを確認することができる。この時点で、オルガノイドを機械的に単離し、3D条件で最長43日間培養した。オルガノイドを機械的に単離した後、FGF2を3つの条件で7日間培地に補充し、成長を促進させた。分化の28日目に、網膜オルガノイドにpR1.7プロモーターの制御下でGFP遺伝子を保持するAAV2-7m8ベクターを5×1010vg/オルガノイドの用量で感染させた。既存の細胞集団の細胞周期停止を促進するために、28日目から1週間、10μMのDAPT(Selleck社)を培地に添加した。蛍光強度は、感染の5日後に最初に観察され、43日目まで増加し続けた。
ヒト網膜外植片は、以前に記載されたプロトコル(38)を用いて調製した。簡単に言うと、眼をCO2非依存培地(Thermo Fischer Scientific社)中で解体した。前部を取り除き、網膜を分離して、細かく切った。これらの外植片を、Transwell細胞培養インサート(Corning社)に光受容体側を上にして置き、B27(Thermo Fischer Scientific社)を補充した2mLのNeurobasal培地(Thermo Fischer Scientific社)を各外植片の下の各ウェルに加えた。翌日、各外植片を、1010個のウイルス粒子を含む0.5μLのAAV-pR1.7-GFPを1滴用いることで感染させた。ベクターの感染した外植片を10~15日間インキュベートしてGFPを発現させ、落射蛍光マクロスコープを使用して確認した。
マウスの眼を摘出し、直ちに凍結切片用に10%ホルマリン-4%ホルムアルデヒドで2時間固定した。解剖後、4%ホルムアルデヒドで3時間、マカクの網膜を固定した。網膜オルガノイド及びヒト網膜外植片を、培養期間の終わりにPBSでリンスし、4%パラホルムアルデヒドで10分間固定した。凍結切片の場合、マウス及びマカクの網膜、網膜オルガノイド及びヒト網膜外植片をPBS-30%スクロース中に4℃で一晩浸漬させた。マウスの眼杯、ヒトの網膜外植片、マカクの網膜をOCT(最適切断温度)培地中に埋め込み、液体窒素中で凍結させ、その一方、網膜のオルガノイドは、7.5%ゼラチン及び10%スクロース(PBS)中に埋め込み、ドライアイスで冷やしたイソペンタン中で凍結させた。Micromクライオスタットを使用して、厚さ10μmの垂直断面を切断した。ブロッキングバッファー中でインキュベートした後、切片を4℃で一次抗体と共に一晩インキュベートした:ヒトCone Arrestin(hCAR)抗体(米国ロサンゼルス、南カリフォルニア大学のCheryl Craftから贈与);M/Lオプシン抗体(Millipore社、AB5405)、及びマウスCone Arrestin抗体(Millipore社、AB15282)。切片を複数回洗浄した後、二次抗体(Alexa Fluor 488、594又は647、ThermoFisher社)とDAPIを追加し、その後、数回洗浄した。網膜のフラットマウント又は凍結切片を蛍光顕微鏡用のVectashieldマウンティング培地(Vector Laboratories社)にマウントして、網膜切片をOlympus Upright共焦点顕微鏡を使用して可視化し、Fiji Softwareで分析した。中心窩の三次元投影は、Imarisソフトウェア(Bitplane社)で作成した。
赤オプシン遺伝子プロモーター配列(pR2.1及びpR1.7配列)と錐体アレスチン3ゲノム領域について、TF結合部位分析を行った。TRANSFACデータベース8.3(http://alggen.lsi.upc.es/)をTF結合部位の予測に使用した。予測されたリスト中の各TFを感覚系ナレッジベース(KBASS、http://kbass.institut-vision.org/KBaSS/transcriptomics/index.php)を使用して分析し、トランスクリプトーム実験RNG209(51)を使用して、ヒト網膜で発現されるものを選択した。以前に記載されたように、ロバストマルチアレイ平均(RMA)により正規化した実験RNG209から調製したサンプルにおいて、フィルターを使用して、40ユニットを超える信号強度値のTFを保持した(52)。この実験において対照として使用したヒト網膜標本は、眼疾患又は糖尿病の過去の病歴のない患者の死後12時間以内に収集された死後標本であった。17人の患者に相当する19個の眼から19個のサンプルを収集した。性比は男性12人/女性7人、平均年齢は61歳(25~78歳の範囲)であった。
Graphpad Prismにおいて、一元配置Anovaテストを使用して、データを分析した(多重比較、Tukey補正)。グラフのエラーバーは、平均の標準誤差(SEM)を示している。p値は次の*p<0.033のように表される、ns:非有意。
動物に関しては、国立衛生研究所の実験動物の管理と使用に関するガイドに従って実験を行った。プロトコールは、地域の動物倫理委員会によって承認されたものを欧州議会の指令2010/63/EUに従って実施した。ドナーからの死後ヒト眼球は、外科手術部(Ecole de Chirugie、Assitance Publique Hopitauxde Paris)から入手した。プロトコールは、外科手術部及びクインゼヴィンツ国立眼科病院の組織審査委員会により承認された。死後ヒト網膜外植片に関する全ての実験は、地域の規制とヘルシンキ宣言のガイドラインに従って実施した。
齧歯類モデルにおける強力且つ特異的な錐体細胞特異的プロモーターの選択
注入部位から離れた位置にある錐体の強力且つ特異的な標的化に適したベクターとプロモーターの組み合わせを見つけるために、我々は、マウス網膜における硝子体内及び網膜下の送達後に、いくつかのAAVを比較した。効率的な錐体視細胞の標的化を可能とするために、我々は、両方の投与経路を介して視細胞を効率的に標的化することが示されているAAV2-7m8と呼ばれる改変AAVバリアントを使用した(16、17)。錐体細胞の特異的な標的化が、硝子体に投与されたAAVを使用して試みられたことはこれまで無かった。臨床で適用可能な錐体における限定的な遺伝子発現に適したプロモーター配列を見つけるために、我々は、非ヒト霊長類(NHP)(18)又はヒト組織(4)のいずれかで以前に検証されているプロモーターに焦点を当てた。我々は、mCAR(マウスCone Arrestin)、pR2.1及びpR1.7プロモーター(ヒト赤オプシン遺伝子エンハンサー及びプロモーター配列に基づく合成プロモーター)の制御下にあるGFPをコードするAAV2-7m8ベクターを生成し、それらを等しい力価で6週齢の野生型マウスの眼に注入した。網膜下注入の3週間後、網膜横断面を錐体アレスチンで染色し、GFPの発現を調べた(データは示さず)。mCARプロモーターでは、桿体と錐体の両光受容体において、高いGFP発現を我々は見出したが、pR2.1とpR1.7は主に錐体で強力な発現を導いた。同じベクターを使用して、我々は、硝子体内送達後には著しく異なる発現パターンを得た(データは示さず)。mCARプロモーターは、一部の錐体においてGFPの発現を引き起こしたが、桿体並びに内核層(INL)及び神経節細胞層(GCL)の細胞に対して漏出性であった(データは示さず)。pR2.1及びpR1.7の両プロモーターは、mCARプロモーターよりも多くの錐体の標識をもたらす(データは示さず)。pR2.1は、pR1.7よりも多くの錐体に形質導入したが、INL及びGCLにおいても非特異的なGFP発現を生じさせた。pR1.7プロモーターのみが、錐体においてGFP発現を示し、桿体における発現は最小限であり、網膜の内側に対して漏出しなかった(データは示さず)。最後に、網膜疾患の状態は、AAVを介した遺伝子送達と導入遺伝子発現パターンに影響を与えうるため(20、21)、我々は、網膜変性のマウスモデルにおいてAAV2-7m8-pR1.7のベクターとプロモーターの組み合わせを検証した。我々は、網膜色素変性症のrd10マウスモデルにおいて硝子体内にAAV2-7m8-pR1.7-GFPを注入した。注入の2か月後、GFPの発現は錐体に限定されており(データは示さず)、硝子体内注入と網膜下注入の両方を介した錐体指向性の遺伝子治療に対するこのベクターの適合性が裏付けられた。
他の種での更なる研究に移る前に、我々は、3つのプロモーターで得られた異なる発現パターンの背後にある理由をより良く理解することを目指した。そのために、我々は、バイオインフォマティクスを使用して、各プロモーター配列内の転写因子(TF)結合部位を分析した(データは示さず)。本分析は、以下の疑問に答えることを目的としている:(i)なぜpR1.7は錐体においてpR2.1よりも効率的なのか、(ii)pR2.1及びmCARプロモーターが硝子体内投与後にオフターゲット発現を引き起こすのはなぜか。我々は、pR1.7とpR2.1の間で観察された異なる発現パターンは、pR2.1プロモーターの5'領域にある337bpの配列中にあるが、pR1.7プロモーター中には無い付加的なTF結合部位によるものであると仮定した(データ示さず)。興味深いことに、我々は、この337bpの配列内に、ニワトリ卵白アルブミン上流プロモーター転写因子I(COUP-TFI)結合部位を見出した(データは示さず)。COUP-TFIは、マウスの網膜における緑オプシン遺伝子(Opn1mw)の発現を抑制することが示されており(22)、よって、AAVが網膜下に送達される際、マカクの錐体におけるpR2.1プロモーターの発現低下の原因となる可能性がある。同じ特異的な337bpの領域内において、我々は、CCAAT/エンハンサー結合タンパク質ベータ(CEBPB)や基本転写因子II-I(GTF2I)等、一般的な偏在性のアクチベーターTFの結合部位も複数見出した(データは示さず)。これらの結合及び基礎的な転写機構の集合を高め、錐体において特異的に発現されるのではないTFの付加的な結合部位が、pR1.7と比較して、pR2.1で観察された一部のオフターゲット発現の原因である可能性がある(データは示さず)。また我々は、AAV構築物において使用されるこのプロモーターの短いバージョンを使用した際の特異性の欠如を説明するために、ゲノムのmCARプロモーター配列(マウスArr3しても知られる)中のTF結合部位も分析した。この短い配列は、TATAボックス、TATA様ボックス、並びにCRX(錐体-桿体ホメオボックスタンパク質)及びSP(特異性タンパク質)TFの結合部位を提示する(23)(データは示さず)ゲノムの近位CARプロモーターの521bpの部分から成る(データは示さず)。しかしながら、521bpの領域のすぐ上流に位置するCARプロモーター活性を調節する「Reg」配列(23)は、短いmCAR配列からは除かれている(データは示さず)。STRINGデータベース(24)から取得したmCARプロモーターに結合するTFのインタラクトームに基づくと、CRX及びSP TFは互いに相互作用し、また、RARA(レチノイン酸受容体アルファ)、RXRA(レチノイドX受容体アルファ)及びTHRB(甲状腺ホルモン受容体ベータ)TFと相互作用する(データは示さず)。これらの3つの核内受容体(NR)は、細胞特異的転写コアクチベーター複合体(26、27)を形成することにより、MED1(25)(メディエーター複合体サブユニット1)を介して、遺伝子発現の細胞型特異的な調節に関与する(データは示さず)。CRX及びSPの結合部位は521bp領域上にあり、RXRA、RARA及びTHRBの結合部位はReg領域上に位置している(データは示さず)。更に、Reg領域は、錐体で発現される重要な核内受容体であるTHRβ2の5つの結合部位を含んでいる(28)(データは示さず)。これら全ての理由から、Reg領域の削除が、短いmCARプロモーターで観察されるオフターゲット発現の原因である可能性が高い。
他の研究者らと我々は、ユビキタスプロモーターを備えたAAVバリアントを使用したマカク錐体の形質導入を示しているが(16、29~31)、硝子体に投与したAAVによる錐体の形質導入の達成は、炎症を引き起こす高用量でのみ可能であった(16、29)。我々は、強力な錐体特異的プロモーターを使用すれば、安全性と両立できる、より低い硝子体内に注入されるAAVの用量で、中心窩錐体の標的化を達成できると考えた(29、32)。そのような「用量節約」が可能かどうかをテストするために、我々は、2つのマカクの眼にAAV2-7m8-pR1.7-GFPを、他の2つのマカクの眼にAAV2-7m8-CMV-GFPを1011ウイルスゲノム(vg)の用量で注入した(表1)。我々は、in vivo眼底イメージングを使用してCMVの注入から早くも2週間後にはGFP発現を観察し、それは注入後2か月まで増加した(データは示さず)。GFPの蛍光は主に、周辺と傍中心窩領域に見られた。pR1.7によるGFPの発現は、投与後6~8週間で検出可能となり、中心窩に限定されていた(データは示さず)。光干渉断層法(OCT)で評価したところ、中心窩に検出可能な損傷は無かった(データは示さず)。注入から3か月後に動物を屠殺した。我々はそれから、黄斑のフラットマウント(データは示さず)と中心窩のレベルの網膜凍結切片(データは示さず)を調製し、眼毎に等しい取得設定を用いて、共焦点顕微鏡を使用してGFP蛍光を画像化した。これらの画像はin vivoの所見を裏付け、そして、AAV2-7m8-pR1.7を使用した1011粒子の用量での、硝子体からの特異的且つ強力な錐体の形質導入を示している。hCAR+細胞の約58%が、中心窩において検出可能なレベルのGFPを発現していることが見出された。同様な用量を使用して、CMVにより錐体に形質導入することは不可能である(検出可能な導入遺伝子の発現無し)。
我々は次に、このプロモーターを治療用遺伝子送達に使用する可能性を評価することを目指した。遺伝子(すなわち、色覚異常の治療のためのCNGB3)補充後の機能的転帰を試験するための網膜変性の既存の盲目マカク又は霊長類モデルは存在していない。しかしながら、我々は内因性の錐体オプシンを介した応答と光遺伝学を介した光応答を区別できるため、光遺伝学的な戦略を使用して、野生型マカクにおける視力回復を評価することは可能である(4)。我々は、中心窩錐体における過分極性微生物オプシン(15)であるJawsの発現により、光遺伝学的視力回復の可能性を評価した。1匹のマカクの硝子体に1011vgのAAV2-7m8-pR1.7-Jaws-GFPを注入して、マウスにおいて以前に記述されたようにして、中期網膜色素変性症における休眠錐体の再活性化について、その治療ポテンシャルを評価した(4、15)。我々は、GFP発現のみと同様に(データは示さず)、注入を行った眼の中心窩錐体に限定された高レベルのJaws-GFP発現を見出した(データは示さず)。それから、注入の2か月後に動物を屠殺し、網膜の半分を組織学的検査のために処理した。網膜フラットマウントは、我々の高い視力を担う密集して詰まった錐体を含む中心窩の領域である中心窩における錐体の典型的な解剖学的形態を示した。錐体アレスチンの免疫染色を使用して、形質導入された錐体を定量化した(データは示さず)。錐体アレスチン陽性細胞(hCAR+)の約50%が、この網膜で検出可能なレベルのJaws-GFPを発現していることが見出された。網膜の残りの半分は、過分極性ポンプJawsから生じる光遺伝学的光応答の化学分析のための外植片(33)として保存された(データは示さず)。Jawsを発現する形質導入された錐体及びGFP発現なしの対照錐体において、電気生理学的記録を行った(データは示さず)。GFP陽性Jaws錐体における全細胞パッチクランプ記録は、オレンジ色の光フラッシュ(n=4)に対して強い光応答を示した(データは示さず)。記録された細胞の活性スペクトルは、Jawsについて以前に示されたように(14)、575~600nmのオレンジ色の光を使用して最大の光応答が得られることを示した(データは示さず)。電流クランプ構成で記録されたJawsを発現する錐体は、光誘発過分極に続いて短い脱分極を示したが(n=4個の細胞)、対照の錐体は同じ光刺激に対して反応しなかった(n=3個の細胞)(データは示さず)。
強力な錐体プロモーターを伴うAAV2-7m8の硝子体内注入による中心窩錐体の形質導入は、主に中心窩に存在する休眠錐体を有する網膜色素変性症患者の脆弱な網膜の錐体を治療するための理想的なアプローチとなる可能性がある。しかしながら、色覚異常症の患者と、AAV2に対する強い中和抗体力価を有する網膜色素変性症の一部の患者(32)に対しては、網膜下アプローチが有利となりうる。以前の研究は、AAV9の網膜下注入が、低用量の中心及び周辺の両送達で錐体における効率的な導入遺伝子発現を導くことを示しているが、これは、AAV9の主要な受容体であるガラクトシル化グリカンが錐体光受容体上に豊富であるためであると考えられる(30、34)。これに基づき、強化されたAAV9バリアントは、遠位小疱からの中心窩錐体の効率的な形質導入をもたらす可能性があると我々は考えた。遠位の網膜下注入部位を介した中心窩錐体遺伝子の送達を促進するために、我々は、野生型AAV9よりも30倍の感染力の増加をもたらすAAV9-7m8バリアントを使用した。我々は、Jaws-GFPをコードする5×1010粒子のこのベクターを1匹の動物の周辺網膜(図1、A~B)に、中心窩を剥離させることなく、網膜下注入した。注入後早くも2週間で、小疱(破線で区切られている)において、そして中心窩においても、強いJaws-GFP蛍光を我々は観察した(図1C)。蛍光強度は、硝子体内処置した網膜よりも、中心窩において高かった。我々は、同じベクターを1010vgの低用量で注入した第2の眼で同じデータを観察した(図2A)。動物が直立した状態において上位周辺小疱が中心窩に向かって下降しないことを更に確認するため、また、更なる用量の低減が可能かどうかを調べるために、我々は、他の2つの眼に510vgの用量で注入を行ったが、今回は中心窩よりも下位とした(Table 1(表1))。OCTを使用して、我々は、中心窩が手術後に剥離しないことを観察した(図3)。中心窩錐体にまで及ぶ同じ発現パターンがこれらの網膜において得られた(図2B)。これらの結果はあわせて、このアプローチの再現性と低ウイルス用量との適合性を示している。
全体的にみて、非ヒト霊長類における我々のデータは、視覚回復のための光遺伝学的適用に適合性のある非侵襲的で特異的且つ高レベルな霊長類の中心窩錐体形質導入を初めて示したものである。しかしながら、プロモーター活性は種間で重要な変動を示すため(29、35、36)、ヒトの細胞及び組織でpR1.7を検証する必要があると我々は考えた。錐体プロモーター活性の効率をテストするために使用できる優れたヒト視細胞細胞株又は他のモデルが無いため、我々は、ヒト人工多能性幹(iPS)細胞由来の3D網膜オルガノイドを使用した(37)。我々は、光受容体の豊富な網膜オルガノイドを生成し、pR1.7プロモーターの制御下にあるGFPをコードするAAV2-7m8ベクターで感染させた(データは示さず)。GFPの発現は、感染から早くも5日後に観察され、分析のため43日目に実験を終了させるまで増加し続けた。これらのオルガノイドにおけるGFPの発現は、ヒトCone Arrestin(hCAR)の免疫染色と共局在していた(データは示さず)。最後に、ヒト網膜オルガノイドは成熟したヒト網膜の全ての機能を表すわけではないため、我々は、死後ヒト網膜外植片においてpR1.7プロモーターの有効性と特異性を検証した。ヒト網膜外植片を以前に記述されたようにして培養し(38)、1滴のAAV2-7m8-pR1.7-GFPに感染させた(データは示さず)。感染から15日後、凍結切片上でGFPの発現を分析した。発現はONLに限定され(データは示さず)、錐体細胞マーカーであるM/L-オプシンと共局在していた(データは示さず)。これらのデータはあわせて、ヒトの錐体における限定的な遺伝子発現の誘導におけるpR1.7の高い効率と特異性を示している。
我々は次に、AAV2-7m8-PR1.7-Jaws-GFPの硝子体内注入、及びAAV9-7m8-PR1.7-Jaws-GFPの網膜下注入後の免疫反応を評価した。
炎症(免疫反応)は、注入の2か月後に網膜凍結切片のH&E染色及び炎症細胞免疫化学(IHC)によって評価した。H&E染色と網膜切片の顕微鏡観察に基づくと、AAV2-7m8-PR1.7-Jaws-GFPの硝子体内投与(図4A)とAVV9-7m8-PR1.7-Jaws-GFPの網膜下注入(図4B)のいずれも、細胞浸潤又は網膜の組織崩壊とは関連しておらず、これは、以前の部分のKiller Red及びGFPの眼とは対照的であった。
局所的な適応免疫応答を分析するために、我々はAAV2-7m8-PR1.7-Jaws-GFP及びAVV9-7m8-PR1.7-Jaws-GFP構築物の両方を注入したサル網膜のIHC分析を行った。網膜下で処置した動物及び硝子体内で処置した動物の凍結切片を抗ヒトHLA ABC(MHC I)、抗HLA-DR(主要組織適合性複合体II(MHC II))、抗ヒトCD8、抗F4/80で染色した。上記のマーカーについて陽性の切片は無かった。ミクログリアとマクロファージの造血マーカーであるグリア線維性酸性タンパク質(GFAP)とイオン化カルシウム結合アダプター分子1(IBA1)の染色は陰性であった(AVV9-7m8-PR1.7-Jaws-GFPに関しては図5;AAV2-7m8-PR1.7-Jaws-GFPについてのデータは示さず)。
中心窩は、霊長類の網膜表面積の1%未満しか占めていないが、一次視覚野の細胞の約50%に入力を提供する(1)。網膜の最も薄くて繊細な部分である中心窩における錐体の高い集中は、高い視力を可能にし、そして、治療的介入の際に、この領域における錐体の独特な機能(39)と構造(40)を維持することは最も重要である。中心窩の錐体は、網膜下又は硝子体内注入のいずれかを使用して、異なる投与経路を介して標的化できるが(35、41、42)、中心窩の剥離は、特に変性網膜において、機械的な損傷を導くおそれがある(43)。これら全ての理由により、この領域を剥離させることなく、治療薬を中心窩に送達する方法が必要とされる。硝子体内注入は、網膜剥離を伴わずに治療薬を送達する外科的にシンプルな方法である。遺伝子治療ベクターは、げっ歯類において、光受容体に損傷を与えることなく、硝子体内注入により網膜の外側を標的化することができる(17、35)。しかしながら、硝子体内注入による霊長類の錐体への安全で効率的な遺伝子送達は、おそらく硝子体内でのベクターの実質的な希釈とその結果としての効力の喪失が原因で、これまで達成されていなかった。この課題を克服するためには、より低い局所濃度で高レベルの遺伝子発現を提供する細胞型特異的なプロモーターの使用が重要となる(29、44)。

- 組換えAAV9由来ベクターを含む、錐体視細胞に影響を及ぼす網膜疾患の治療における使用のための組成物であって、前記ベクターが、

前記網膜疾患が、加齢性黄斑変性症、バッセン-コルツヴァイク症候群、脈絡膜血症、旋回性萎縮、レフサム症候群、アッシャー症候群、色盲、青色錐体単色症、色覚異常、不完全色覚異常、乏錐体三色性、網膜色素変性症(RP)、黄斑変性症、スターガルト病、バルデット-ビードル症候群、ボーンホルム眼病、ベスト病、及びレーバー先天性黒内障から成る群から選択される、組成物。 - 目的のポリヌクレオチドが、遺伝子補充療法において使用されうる遺伝子であり、好ましくは、網膜色素変性症GTPaseレギュレーター(RPGRORF15)、CNGB3(錐体環状ヌクレオチド感受性陽イオンチャネルのベータサブユニット)、CNGA3(錐体環状ヌクレオチド感受性陽イオンチャネルのアルファサブユニット)又はGNAT2をコードする遺伝子から選択される、請求項1に記載の組成物。
