Description
本出願は、2018年9月24日に出願された米国仮出願62/735,531号の出願日の利益を主張し、その教示を参照により本願に援用する。
細胞増殖のための表面領域を有する複数のボイド(前記複数のボイドは直径Dを有する)、前記ボイド間の複数の細孔開口部(細孔開口部は、D>dとなる直径dを有する)を含む、細胞増殖のための3Dバイオリアクターであって、
ここで、(a)前記ボイドの90%以上が、+/-10.0%を超えては変わらない、選択されたボイド体積(V)を有し;かつ、(b)前記ボイド間の前記細孔開口部の90%以上が、+/-10.0%を超えて変わらないdの値を有し、
そして、前記バイオリアクターは置換又は非置換ポリ(p-キシリレン)でコーティングされている。

ここでの開示は、灌流に基づくスケーラブルなバイオリアクターの設計、及び免疫療法目的のT細胞増殖又は再生医療のための幹細胞増殖を達成するための対応する操作性能に関する。患者からの、例えば遺伝子改変後の、T細胞又は幹細胞の活性化及び増殖は、治療用のT細胞又は幹細胞の産生物をもたらし、それは患者に注入して戻すことができ、患者の腫瘍細胞を選択的に標的にし且つ殺すやり方で患者自身の免疫システムを利用するか、あるいは老化関連疾患を治療するために、患者自身の再生細胞を使用する。

ここでの3Dバイオリアクターは、好ましくは、追加の印刷後の手順にかけられる。このような手順は、3Dバイオリアクターの印刷に使用された可能性のある残留モノマー及び/又は光開始剤の浸出を低減又は防止する能力があるように設計される。そのような場合は、特に、積層造形手順が、液体樹脂の硬化(重合)を含むステレオリソグラフィー(SLA)又はデジタルライトプロセッシング(DLP)を伴う場合である。したがって、図18に注意が向けられ、これは、好ましい印刷後の手順をまとめたものである。図示しているように、最初に光重合(光誘起重合)による積層造形によって製造されたここでの3Dバイオリアクターは、好ましくは後硬化、好ましくはUV光への追加の曝露によって後硬化される。UV光へのそのような追加の曝露は、印刷中の基礎となる重合を増加させ、及び/又は比較的低分子量の材料、例えば重合していないモノマー材料の量を低減させることが考えられる。さらに、そのような後硬化に続いて、そのような比較的低分子量の材料をさらに低減し又は除去するために熱及び/又は真空での処理を行うことができる。たとえば、周囲温度から125℃までの温度において10-3Torr以下の真空を適用することができる。したがって、後硬化を単独で、又は真空及び/又は加熱と組み合わせて使用することによって、比較的低いMWの材料(500以下のMW)の量を2.0質量%以下に、より好ましくは0.01質量%~1.50重量%、なおさらに好ましくは0.01質量%~0.75質量%の量まで低下させることができると考えられる。
Miltenyi MACSiBeadシステムに似せて、アビジン(ストレプトアビジン)及びビオチン結合メカニズムを使用して、ポリドーパミンでコーティングされたバイオリアクター表面に抗-CD2、CD3、及びCD28抗体を固定した。最初に、様々な濃度の蛍光標識したアビジン及びストレプトアビジンを試験して(図13)、100μg/mLのアビジン又は30μg/mLのストレプトアビジンが、バイオリアクター表面上の比較的高いアビジン又はストレプトアビジンのコーティング密度を達成することが発見された。図13は、様々な濃度でのアビジン及びストレプトアビジンのコーティングの蛍光強度を示している。蛍光標識されたアビジン/ストレプトアビジンを使用して、ポリドーパミンプライムコーティング上に結合することができるアビジン又はストレプトアビジンの濃度を試験した。アビジン/ストレプトアビジンのコーティングの好ましい手順は以下のとおりである。アビジン又はストレプトアビジンのコーティング濃度は、さらに最適化することができる。
1)バイオリアクター表面を、最初に、ここではポリドーパミンによってコーティングする。
2)アビジン又はストレプトアビジンをTRISバッファー(pH8.5)に溶かして、100μg/mL又は30μg/mLのアビジン又はストレプトアビジンの濃度をそれぞれ有するコーティング溶液を調製する。暗所で穏やかに振とうしながら、バイオリアクターをコーティング溶液中に12時間浸す。
3)リン酸緩衝生理食塩水(PBS)でその足場(scaffold)を完全に洗浄する。次に、その足場をPBS中に残しておき、これで、ビオチン化抗体でコーティングする準備ができる。
1)BPSからバイオリアクターの足場を取り出す。
2)足場からできるだけ多くのPBDを取り除くが、足場は乾燥させない。
3)100μg/mLのCD2-ビオチン(200μL)、100μg/mLのCD3-ビオチン(200μL)、及び100μg/mLのCD28-ビオチン(200μL)を15mLのチューブ中で混合し、1.4mLの抗体標識バッファー(Ca2+及びMg2+なしのPBS、pH=7.2に加えて、0.5%の熱不活化ウシ胎児血清及び2mMのEDTA)を添加し、良く混合して、各抗体が10μg/mlの濃度をもつ合計2mLのビオチン化抗体を作る。
4)上記の混合した抗体2mLを添加して、12ウェルのプレート中のバイオリアクターマトリクスを覆い、ピペットで液を上下させて混合する。
5)2℃~8℃の温度において、穏やかに振とうしながら、冷蔵庫中暗所で2時間、マトリクス中でインキュベートする。
6)PBS洗浄により、足場から、未結合の抗体を完全に除去する。
抗体でコーティングされたマトリックス(入口及び出口なし)を、Miltenyi MACSiBeadTMシステムと比較した。磁気ビーズ(すでにストレプトアビジンでコーティングされている)を、上述したコーティング手順と同様に、製造元のプロトコルに従ってビオチン化抗-CD2、CD3、及びCD28抗体でコーティングした。表1に挙げた、直径2.1cm×高さ0.7cmの比較的小さいバイオリアクター(図7e)を使用した。このバイオリアクターマトリックスは12ウェルプレートのウェルに収まるので、ビーズとマトリックスの性能を簡単に比較できる。3つのバイオリアクターマトリックスを最初にアビジン又はストレプトアビジンで、次に上述した好ましい手順を使用してビオチン化CD2、CD3、CD28抗体でコーティングした。同じ濃度の抗体を使用して、バイオリアクターマトリックスとMACSiBeadsをコーティングした。ストレプトアビジンコーティングを施した1つのバイオリアクターマトリックスは、抗体でさらにコーティングされていない(ネガティブコントロール)。
上と同様の研究を、単離されたT細胞の代わりにPBMCを用いて行った。通常、T細胞及び他の単核細胞、例えば、B細胞、NK細胞、単球を含むPBMCを患者から採取し、T細胞を分離することなく細胞増殖に直接用いた。これは、非T細胞はCD2、CD3、及びCD28によって活性化されることはなく、それらは活性化せずに数日後に自然に排除されるからである。この実験と図15の実験との別の違いは、抗体でコーティングされたマトリックスを使用するときにサイトカイン活性化ステップを組み込んでいることである。手短に言えば、2.5×106個の細胞/mLの密度でPBMCを含む培地2mLを12ウェルプレートの3つのウェルに添加した。その3つのウェルには、それぞれ、1)7.5×106個の、抗体でコーティングされた磁気ビーズ、2)抗体でコーティングされたストレプトアビジンマトリックス、及び3)抗体でコーティングされていないストレプトアビジンマトリックスが含まれている。ストレプトアビジンは最後の2つの実験でアビジンよりも優れていることが示されているので、この実験ではアビジンでコーティングされたマトリックスは使用しなかった。PBMCを、活性化のために各ウェル中で2日間インキュベートした。次に、IL2サイトカインを含む追加の培地をそれらのウェルに添加し、培地に20IU/mLのヒトIL2が含まれるようにして増殖期を開始した。5日間の培養中の総細胞数の変化を図16に示している。これは、抗体でコーティングされたマトリックスで増殖したT細胞が、抗体でコーティングされた磁気ビーズと比較した場合、より多くのT細胞をもたらしたことを確認するものである。これはまた、抗体でコーティングされたマトリックスが、同等以上のT細胞の活性化及び増殖をもたらすことができることを確認するものである。
灌流ベースの3Dバイオリアクターを、本明細書に記載されているようにして製造した。この3Dバイオリアクターの内面は、12時間ポリドーパミンでコーティングすることによって調製した。次に、バイオリアクターの内面を12時間ストレプトアビジンでコーティングした。次に、バイオリアクターを滅菌のために70%エタノールでインキュベートした。滅菌後、上記の手順を使用して、その内面をPBSで洗浄し、CD2、CD3、及びCD28-ビオチンコンジュゲートの等量混合物(各抗体について10μg/mLの濃度)でコーティングして、バイオリアクターの内面に抗体を固定させた。
上記のように、パリレンコーティングは官能化することができる。例として、ここでのパリレンCでコーティングしたバイオリアクターの場合、それは紫外線/オゾンシステムで処理され、この処理がヒドロキシル、カルボニル、及びカルボキシレート表面基をもたらすと考えられる。より具体的には、3Dバイオリアクターを、片側にUVに透明なガラスを備えた密閉チャンバー内に配置した。UVランプ(254nm)をそのガラス上約1.75インチの距離に配置して、オゾン曝露中に同時に適用される3.2mW/cm2の曝露フラックスを生成させる。密閉チャンバーの入口はPTFEチューブを介してVMUS-4オゾン発生器(Azco Industries)に接続し、密閉チャンバーの出口は鉱油で満たされたガラス瓶に接続し、これらは全てドラフト内で行った。オゾン発生器は、2L/分の空気入口流量に設定した。出力は100%の効率に設定し、1時間あたり約2.5gのオゾンを生み出した。パリレンCでコーティングされたバイオリアクターは、それぞれ1、2、5、10、及び15分間オゾン処理した。下の表3から分かるように、パリレンC表面を10分間処理すると、表面は、ポリドーパミン/フィブロネクチン(PD/FB)で処理された表面と同じくらい親水性になった。その教示を参照により本明細書に援用する米国特許出願第15/585,812号を参照されたい。
本明細書では、パリレンフィルムタイプのコーティングを含み得るここでの3Dバイオリアクターのガンマ線滅菌の代わりに、滅菌は、非接着性T細胞増殖の場合、抗体の表面への固定化前の灌流二酸化塩素(ClO2)ガスに頼ることができることを想定している。ClO2ガスは、官能化されたパリレンフィルムコーティング(すなわち、-Cl、-CHO、-COOH、-NH2、又は-C(O)CF3)又はその下にある3Dバイオリアクターポリマー基材と反応することは予期されない。
Claims (21)
- モノマーの重合による積層造形法によって形成された、細胞増殖のための3Dバイオリアクターであって、
細胞増殖のための表面領域を有する複数のボイド(前記複数のボイドは直径Dを有する)、前記ボイド間の複数の細孔開口部(細孔開口部はD>dとなる直径dを有する)を含み、ここで、(a)前記ボイドの90%以上が、±10.0%を超えては変わらない、選択されたボイド体積(V)を有し;かつ、(b)前記ボイド間の前記細孔開口部の90%以上が、±10.0%を超えて変わらないdの値を有し;かつ
置換又は非置換ポリ(p-キシリレン)コーティングを有し、

- 3Dバイオリアクターの製造方法であって、以下のステップ:

前記3Dバイオリアクターを、置換又は非置換ポリ(p-キシリレン)でコーティングするステップ、
