〔実施例1〕
活動によって誘導されるセマフォリン3A分泌は、学習を媒介する。
セマフォリンファミリーは、軸索ガイダンスおよび細胞遊走という活動非依存的な神経発達過程、ならびに免疫システムに関与する分泌型または膜結合型タンパク質として、よく特徴づけられている。発達中の軸索の化学忌避物質(chemo-repellents)としてのセマフォリンの生理学的機能は十分に確立されているが、活動依存的なシナプス可塑性およびさらに学習・記憶におけるセマフォリンの役割は、いまだに殆ど分かっていない。本研究において、我々は、恐怖学習(恐怖条件付け)(inhibitory avoidance task)(これは海馬に依存した文脈的学習パラダイムである)が海馬において分泌型セマフォリンであるセマフォリン3Aの分泌を増加させることを見出した。さらに、海馬において分泌されたセマフォリン3Aは、ニューロピリン1/プレキシンA1セマフォリン受容体複合体を介してAMPA受容体を海馬シナプス中に移行させることによって、文脈的記憶形成を媒介する。このシグナル伝達過程は、セマフォリンシグナル伝達の下流分子として特徴付けられているコラプシン反応媒介タンパク質2(collapsin-response-mediator-protein 2; CRMP2)のリン酸化状態の変更を伴う。このことは、学習の制御因子としてのセマフォリンファミリーの生理学的役割を提供する。
セマフォリンは、軸索誘導、細胞遊走および免疫システムの制御に関与する、分泌型および膜結合型分子の最も大きいファミリーの一つを形成する(Kolodkin and Tessier-Lavigne, 2011)。セマフォリンは、その構造に基づいて幾つかのグループに分類される(Jongbloets and Pasterkamp, 2014; Kolodkin and Tessier-Lavigne, 2011)。セマフォリン3A (Sema3A)は、クラス3の分泌型セマフォリンであり、発達中の軸索の反発分子としてよく特徴付けられている(Jongbloets and Pasterkamp, 2014; Luo et al., 1993)。ニューロピリン1 (NRP1)は、Sema3Aの主要な結合部位であり、そして大きな細胞内ドメインを有するプレキシンと複合体を形成して、Sema3Aシグナルを伝達する(He and Tessier-Lavigne, 1997; Kolodkin et al., 1997; Nakamura et al., 1998; Takahashi et al., 1999; Takahashi et al., 1998; Tamagnone et al., 1999)。活動非依存的な軸索ガイダンスおよび神経細胞遊走におけるセマフォリンの生理学的役割は徹底的に研究されているが、活動依存的な神経現象、すなわち学習、記憶、等におけるセマフォリンの役割は未だに不明である。
Sema3Aは、海馬初代培養物のニューロン表面へのAMPA受容体の輸送を制御する。
セマフォリンファミリーは、神経科学、免疫システムから腫瘍学に至るまで種々の生物学的分野において大いに注目されてきた(Gurrapu et al., 2018; Kolodkin and
Tessier-Lavigne, 2011; Vadasz and Toubi, 2018)。神経科学の分野では、セマフォリンは軸索ガイダンスおよび細胞遊走に関して徹底的に研究されてきた。本研究において我々は、Sema3Aはセマフォリンファミリーの神経系における新規な役割を意味する、学習の決定的なメディエーターであることを報告した。文脈的恐怖条件付けの一形態であるIA学習は、Sema3Aの分泌を誘導した。そして、Sema3A機能の中和抗体の海馬への注入は、IA記憶形成を妨げた。さらに、Sema3A受容体複合体の各コンポーネント(NRP1および PlexinA4)のノックダウンは、背側海馬におけるIA学習およびIA学習依存的なGluA1のCA3-CA1錐体シナプスへの移行を遮断した。我々の所見は、軸索ガイダンスおよびシナプス形成の両者を制御する分子および細胞メカニズムの間の関係を明らかにし、それによって軸索ガイダンス分子を神経可塑性と結び付ける証拠を提供する。

全ての実験は、「実験動物の管理と使用に関する指針」(日本神経科学学会)および横浜市立大学の指針に従って実施された。全ての動物実験は、横浜市立大学の「動物の管理および使用委員会」によって承認された(承認番号:F-A-14-026)。全ての外科的処置は麻酔下で実施され、そして苦痛を最小限にするためのあらゆる努力がなされた。
雄および雌ICR E16(チャールス・リバー・ラボラトリーズ)または雄および雌ウィスターラットE17-18(チャールス・リバー・ラボラトリーズ)由来の海馬ニューロンを、0.01%ポリ-L-リシン(和光純薬)でコーティングした培養皿で、2% B27サプリメント(Gibco)および0.5 mM L-グルタミン(Nacalai)を添加したneurobasal medium (Gibco)中で培養した。ニューロンは、プラスチック製カバーグラスの上で、21~28日間 in vitro(DIV)で培養した。
PlexA4 IPTは、以前に記述されているように(Yamashita et al., 2014)作製した。

免疫細胞化学は、0.1% Triton X-100 (ref)を用いた透過処理のもとで、標準プロトコールにより実施した。培養ニューロンの染色に用いた抗体は以下の通りであった:抗N末端GluA1マウスモノクローナル抗体(1:500希釈)、抗MAP2ウサギポリクローナル抗体(PRB-547C、1:1000希釈、Covance)、抗MAP2マウスモノクローナル抗体(M1406、1:1000希釈、Sigma)、抗GFPニワトリ抗体(1:1000希釈、Aves)、抗PlexA4ハムスターモノクローナル抗体(1:500希釈、Sutoにより提供される)、および抗NRP1ウサギポリクローナル抗体(1:1000希釈、T. Kawasakiにより提供される)。ニューロンは、抗MAP2ウサギポリクローナル抗体を用いて対比染色した。免疫染色したニューロンを、40倍水浸対物レンズ (C-Apochromat/1.2W corr)を有し、Axioplan 2イメージング顕微鏡(Carl Zeiss)を備えたレーザー走査顕微鏡(LSM510)を用いて分析した。GluA1の表面発現レベルを可視化するため、我々は、非透過処理条件下で抗GluA1マウスモノクローナル抗体(1:500希釈)を用いて免疫細胞化学を実施した。次に、0.1% TritonX-100を用いて培養ニューロンを透過処理し、抗MAP2ウサギポリクローナル抗体を用いて対比染色した。免疫染色したニューロンを、40倍水浸対物レンズ (C-Apochromat/1.2W Korr M27)を有し、AxioImager Z1イメージング顕微鏡(Carl Zeiss)を備えたレーザー走査顕微鏡(LSM700)を用いて分析した。
HEK293T細胞を6 cmの培養皿に4.0 x 105細胞/皿の密度で播種した。1日後、FuGENE6トランスフェクション試薬(Promega)を用いて、EGFP-GluA1、全長PlexA4、 Δect、IPTドメイン(Yamashita et al., 2014)およびPlexA4-Myc発現ベクターにより細胞をトランスフェクションした。48時間後、免疫沈降緩衝液(20mM Tris-HCl (pH 8.0), 100mM NaCl, 10mM NaF, 1mM Na3VO4, 1mM EDTA, 1% Nonidet P-40, cOmplete Mini プロテアーゼ・インヒビター・カクテル (Roche))を用いて細胞を溶解し、次に2 mgの抗Mycマウスモノクローナル抗体(M5546, Sigma)を用いて免疫沈降させた。得られたサンプルは、抗Mycマウスモノクローナル抗体(1:5000希釈)および抗GFPマウスモノクローナル抗体を用いたイムノブロット分析に付した。
IA条件付けの10分後に、マウスに深い麻酔をかけ、経心的にPBSを灌流し、次に4%パラホルムアルデヒド(PFA)を灌流した。脳を摘出し、4% PFA中で室温で2時間インキュベートした。脳をPBS中に移し、ビブラトームを用いて50 μmの冠状スライスを調製した。免疫染色のために、各スライスを3%正常ヤギ血清を含むPBS+0.2% TritonX-100 (PBS-T)中に2時間置き、次に一次抗体と共に4℃で24時間インキュベートした。PBS-Tを用いて各スライスを1回につき10分かけて3回洗浄し、二次抗体と共に1時間インキュベートした。PBS-Tを用いて1回につき10分かけてさらに3回洗浄した後、各スライスを顕微鏡スライド上にマウントした。染色に用いた抗体は以下の通りであった:抗N末端GluA1マウスモノクローナル抗体(1:500希釈、EMD Millipore)、抗MAP2ウサギポリクローナル抗体(1:1000希釈、Covance)、抗MAP2マウスモノクローナル抗体(1:1000希釈、Sigma)、抗PSD 95(1:500希釈、Alomone labs)、抗Synapsin1(1:1000希釈、Millipore)、および抗GFPニワトリ抗体(1:1000希釈、Aves)。
培養ニューロンをSema3Aで30分処理した後、染色のため固定した。抗GluA1抗体および抗PlexA4抗体によって染色された平均免疫染色強度をImageJソフトウエアを用いて測定した。ニューロンあたりの樹状突起の平均強度を求めるため、
各MAP2またはEGFP陽性樹状突起の免疫染色強度を、樹状突起の容積によって正規化した。これら3つの領域の各領域の相対免疫染色強度を計算した。
flag-SEPノックインマウスを作製するため、我々は、flagでタグ付けしたsuper ecliptic pHluorin cDNA(flag-SEP)をSema3Aの分泌シグナルコード配列(25 aa)のすぐ後ろに配置したターゲティング構築物を設計した(図3B、中及び図5-1、5-2)。ターゲティングベクターの構築には、Quick and Easy BAC modification Kit (Gene Bridges, Dresden, Germany)を用いた。すなわち、我々は、C57/BL6 BACライブラリー (Advance GenoTechs、つくば、日本)のゲノムクローン(RP23-181I7)から 11.92 kb のホモロジーアームを回収し、これをCAGプロモーターによって駆動されるジフテリア毒素遺伝子を含有するpDT-MC#3中にサブクローン化した。次に、我々は、ノックイン断片〔flag-SEP、エクソン2の12個のアミノ酸をコードする配列およびそれに続くイントロン2-3の608bp、およびFRT配列によって挟まれたネオマイシン耐性カセット(Neo)〕をpDT-MC#3のホモロジーアームの正確な位置に挿入した。ES細胞の培養、組換えES細胞の同定、およびキメラマウスの作製は、以前に記述されているように(Nishina & Sakimura, 2007)実施した。FLP発現マウスと交配してSema3A遺伝子座からNeoカセットを除去した後、我々はSema3Aflag-SEPマウスを確立した。マウス尾DNAのPCR遺伝子型判定は、以下のプライマーを用いて実施した。

動物は、一定の14時間/10時間の明暗サイクルの元で、水および餌を自由に摂取できる状態で管理した。全ての動物管理および外科的処置は、横浜市立大学医学部大学院、動物実験センター、動物の管理および使用委員会のガイドラインに従って実施した。
イソフルラン-酸素混合物を用いてラットに深い麻酔をかけた。頭蓋を覆う皮膚を切り、そっと両側に広げた。座標は、ブレグマから3 mm前側、正中線の外側2 mm、および脳表面から2.5 mm下であった。組換えレンチウイルスを、ガラス製マイクロキャピラリー(ナリシゲ)から海馬CA1領域へ圧力注入した。ホールセル記録のためには、海馬右側への1回の片側注入を実施した。行動分析のためには、ウイルス溶液を両側に注入した(片側につき1回から約2回注入)。
イソフルラン-酸素混合物を用いてラットに深い麻酔をかけた。誘導用カニューレ(AG-4; EICOM、京都、日本)を海馬CA1領域内へ定位的に植え込んだ。座標は、ブレグマから3.5 mm前側、正中線の外側2.5 mm、および脳表面から2.15 mm下であった。手術の7日後、抗体溶液4-2 CMchおよび対照IgG(Yamashita et al., 2015)を、テフロンチューブを用いて頭蓋ウインドウより圧力注入した。
訓練の日、IA訓練装置(長さ:25 cm、幅:62 cm、高さ:45 cm)を備えた、電磁気および音を遮断した部屋(長さ:1.2 m、幅:2.2 m、高さ:2.3 m)にラットを移した。上記装置は、トラップドアによって分けられた明るい安全な側と暗いショック側から成る、2チェンバーのPerspexボックスである。訓練中、ラットはドアと反対側のコーナーを向いて、ボックスの安全な側に置かれた。トラップドアを開けた後、ラットは随意に暗いボックスに入った。新規な暗いボックスへ入るまでの反応時間を行動パラメーターとして測定した(すなわち、IA訓練前の反応時間)。動物が暗いボックスに入って4秒後に、我々はドアを閉じて、ボックスの床に配置した帯電されたスチールロッドを介して足にスクランブル電気ショック(2秒、1.6 mA)をかけた。ホームケージに戻される前に、ラットは暗いコンパートメントに10秒間置かれた。学習の30分後に、ラットを再びボックスの明るい側に置いた。体験した暗い側に入るまでの反応時間を、学習成績として測定した(すなわち、IA学習後の反応時間)。次に、ペントバルビタールの過剰投与によってラットを屠殺した。未訓練対照(non-IA)については、IA訓練をせずにホームケージでラットに同用量のペントバルビタールを注射した。ウォークスルー対照(walk-through)については、ラットをIA訓練装置に入れ、1分間探検することを許した。
レンチウイルス注入の5から7日後に、ラットをイソフルレンガスで麻酔し、脳を摘出した。脳は、5% CO2/95% O2でガス処理した氷冷解剖緩衝液(25.0 mM NaHCO3, 1.25mM NaH2PO4, 2.5mM KCl, 0.5mM CaCl2, 7.0 mM MgCl2, 25.0 mM グルコース, 110.0 mM 塩化コリン, 11.6 mM アスコルビン酸, 3.1 mM ピルビン酸)中に速やかに移した。冠状脳スライスは解剖緩衝液中で切断し(350 μm、Leicaビブラトーム)、生理学的溶液(5% CO2/95% O2でガス処理した、22℃-25℃, 118 mM NaCl, 2.5 mM KCl, 26 mM NaHCO3, 1 mM NaH2PO4, 10 mM グルコース, 4 mM MgCl2, 4 mM CaCl2 [pH 7.4])中に移した。記録チェンバーを、0.1 mMピクロトキシンおよび4 mM 2-クロロアデノシンを含む生理学的溶液22℃-25℃で灌流した。パッチ記録ピペット(3-7 MΩ)を細胞内溶液(115 mM メタンスルホン酸セシウム, 20 mM CsCl, 10 mM HEPES, 2.5 mM MgCl2, 4 mM Na2ATP, 0.4 mM Na3GTP, 10 mMクレアチニンリン酸 二ナトリウム, 0.6 mM EGTA [pH 7.25])で満たした。ホールセル記録は、Multiclamp 700B(Axon Instruments)を用いて、ラット海馬の感染した、または未感染のCA1錐体ニューロンから得た。二極タングステン刺激電極をCA1領域に、記録した細胞から約200から300 μm外側に設置した。シナプス応答の振幅が>約10 pAを記録するまで刺激強度を増大させた。AMPA/NMDA 比を、- 60 mVにおけるピーク電流の、刺激開始50ミリセカンド後の+40 mV におけるピーク電流に対する比として計算した(各保持電位について30-50トレースを平均した)。
細胞の生物学的実験については、我々は1匹の動物の実験に由来する観察を平均化した(培養物を用いた場合を除く)。したがって、我々は1匹の動物に由来する観察の平均をn=1として用いた。適切な統計試験を選択するため、サンプル分布の歪度および尖度を計算した。歪度が2未満で、かつ尖度が7未満の場合、我々はパラメトリック検定を用いてデータを分析した。歪度が2より大きい、または尖度が7より大きいとデータが示す場合、我々はノンパラメトリック検定を用いた。パラメトリック検定としては、対応のない両側t検定を用いて、2つの独立した群を比較した。また、一元配置分散分析を事後ダネット検定とともに用いて、3つ以上の群を比較した。ノンパラメトリック検定としては、マン・ホイットニーのU検定を用いて、2つの独立した群を比較した。p <0.05は、統計的に有意と考えられた。統計分析は、GraphPad Prism 7 (Graph Pad Software) または SPSS software (SPSS 22.0; IBM)を用いて実施した。箱ひげ図においては、ひげの末端は最大値および最小値によって規定された。中央の四角形は、第1四分位から第3四分位に広がっていた。四角形内のセグメントは、中央値を示した。箱ひげ図以外のグラフにおいては、エラーバーは平均値の標準誤差を示した。
ノックインベクター構築(図5-1, 5-2)
Red/ETリコンビネーションシステムを用い、マウスSema3a遺伝子のエクソン1及びエクソン2を含む11.9 kbのゲノム配列をBACクローンよりpDT-MC#3へサブクローニングした。一方、FRT配列に挟まれたNeo耐性遺伝子含むベクターに、flag-SEP, Sema3Aのエクソン2の12個のアミノ酸をコードする配列及びそれに続くイントロン2-3の608 bpを連結したベクターを作製し、それを鋳型としてPCRにより両端に5’ホモロジーアーム及び3’ホモロジーアームの50 bpを連結したノックインDNA断片を作製した。再度Red/ETリコンビネーションシステムを用い、サブクローニングしたゲノム配列へノックインDNA断片を挿入し、それを制限酵素SalIにより線状化してノックインベクターとして用いた。

エレクトロポーレーションと薬剤選別:ES細胞にはC57BL/6N系マウス由来RENKAを用いた。凍結保存されているES細胞を復元し、4日目に電気穿孔法(4mmキュベット、200 V、250 μF、R=∞)により1キュベットあたり2.5 pmolの線状化ベクターを2百万個のES細胞へ導入し、10 cmカルチャーディッシュ4枚に分割して培養した。導入2日後に175 μg/mL G418による薬剤選別を開始し、導入7日まで継続した。細胞培養のほぼ全ての過程で、ES細胞はフィーダー細胞として用いるマウス胎仔繊維芽細胞との共培養で維持された。
受精卵の準備:1回のインジェクションにつき採卵用のメスICRマウス15頭に排卵誘起処理(PMSG、hCGの投与)を行い、オスと同居させ、翌日プラグにより交配を確認したメスの卵管を潅流して受精卵を採取し、インジェクションまで培養を行った。

- セマフォリン3Aの分泌シグナルの直下にpHluorinを融合させたノックイン遺伝子が導入され、pHluorinで標識されたセマフォリン3Aを発現する非ヒトノックイン動物。
- 動物がマウスである、請求項1記載の動物。
- 請求項1又は2に記載の動物に医薬品候補物質を投与することを含む、医薬品のスクリーニング方法。
- 医薬品が、セマフォリン3Aが関与する疾患を予防及び/又は治療の対象とする請求項3記載の方法。
- セマフォリン3Aが関与する疾患が、喘息、アトピー性皮膚炎、アルツハイマー型認知症、変形性関節炎、悪性腫瘍及び循環器系疾患からなる群より選択される少なくとも一つの疾患である請求項4記載の方法。
