Claims (7)
- 分析目的のために生体試料内の生細胞を固定化する方法であって、
- 分析する所定数の細胞を含有する生体試料を提供するステップと、
- 分析装置で使用するために特に適切な平面支持体を提供するステップと、
- 前記平面支持体がナノ構造材料を含む表面コーティングで官能化された表面を備えるステップと、
- 該平面支持体の該官能化表面に該生体試料の層状層を沈着させるステップと、
- 該生体試料の該層状層上に固定剤を配置するステップと、
- 該生体試料に含有される該細胞を該平面支持体の該官能化表面に接着させるステップと、を含み、
前記生体試料を提供するステップ、前記平面支持体を提供するステップ、前記備えるステップ、前記沈着させるステップ、前記配置するステップおよび前記接着させるステップを、25℃を下回る温度で実施し、
前記ナノ構造材料が、酸化亜鉛(ZnO)、二酸化ジルコニウム(ZrO2)、二酸化チタン(TiO2)のいずれか1種から選択される、方法。 - 前記生体試料の前記層状層を前記平面支持体上に配置するステップと前記層状層上に前記固定剤を配置するステップとの間の時間範囲が4分未満である、請求項1に記載の方法。
- 前記平面支持体の全表面積と生体試料の前記層状層によって占有される前記平面支持体の表面積との間の比が1.5と9との間である、請求項1または請求項2に記載の方法。
- 前記生体試料の前記層状層の体積が1マイクロリットルと2ミリリットルとの間である、請求項3に記載の方法。
- 閉じ込め疎水性物質を前記平面支持体に結びつけるステップをさらに含む、請求項1から4のいずれか一項に記載の方法。
- 前記平面支持体の前記表面コーティングに付着したままの前記生細胞が希少細胞である、請求項1から5のいずれか一項に記載の方法。
- 請求項1から6のいずれかに記載の方法を実施するステップと、
循環腫瘍細胞(CTC)を同定するために前記生体試料を分析するステップと、
画像分析によってCTCを計数するステップと、を含む、循環腫瘍細胞(CTC)の存在を同定する方法。
