Description
L-エルゴチオネイン、及び、S-リボシルホモシステインを含有する組成物であって、
該S-リボシルホモシステインの含有量が、低減されてなる、組成物。
前記S-リボシルホモシステインの含有量が低減されることにより、前記組成物の着色が抑制されてなる、[1]に記載の組成物。
前記L-エルゴチオネイン100質量部に対して、前記S-リボシルホモシステインの含有量が、15質量部以下である、[1]又は[2]に記載の組成物。
前記L-エルゴチオネイン100質量部に対して、前記S-リボシルホモシステインの含有量が、0.001質量部以上である、[1]~[3]のいずれか1項に記載の組成物。

EGTは、ヒスチジン誘導体(N,N,N-トリメチル-L-2-チオヒスチジン)であり、その構造は次式(1)で表される。
SRHの構造は次式(2)で表される。
EGT及びSRHを含有する組成物において、SRHの含有量を低減させることにより、安定性を高めることができるため、本発明は、上記組成物の着色を抑制するために用いることが可能である。
本発明の組成物は、例えば、錠剤、カプセル剤、顆粒剤、散剤などの固形製剤;または液剤、シロップ剤、注射剤、クリーム、ローション、ペースト、軟膏、エマルジョン(水中油型エマルジョン、油中水型エマルジョン、多重エマルジョン、ミクロエマルジョン、PET-エマルジョン、ピッカリング・エマルジョン)、ゲル(ヒドロゲル、アルコールゲル)、懸濁液などの液状製剤として経口または非経口的(外用を含む)に投与することもできる。固形製剤には、賦形剤、滑沢剤、結合剤、崩壊剤;液状製剤には、溶剤、溶解補助剤、乳化剤、乳化安定剤、増粘剤、保湿剤、懸濁化剤、等張化剤、緩衝剤、無痛化剤などを用いることができる。また必要に応じて、防腐剤、抗酸化剤、着色剤、甘味剤、香料などの添加物を用いることもできる。
本発明の組成物は、飲食品組成物としても使用することができ、食品または機能性食品に含有させて提供され得る。このような食品または機能性食品としては、米飯;そば、うどん、はるさめ、中華麺、即席麺、カップ麺を含む各種の麺類;清涼飲料、炭酸飲料、栄養飲料、果実飲料、乳酸飲料、スポーツ飲料等の飲料;カレールー、シチュー、各種スープ類;アイスクリーム、アイスシャーベット、かき氷等の冷菓;飴、クッキー、キャンディー、ガム、チョコレート、錠菓、スナック菓子、ビスケット、ゼリー、ジャム、クリーム、その他の焼き菓子等の菓子類;かまぼこ、はんぺん、ハム、ソーセージ等の水産・畜産加工食品;加工乳、発酵乳等の乳製品;サラダ油、てんぷら油、マーガリン、マヨネーズ、ショートニング、ホイップクリーム、ドレッシング等の油脂及び油脂加工食品;ソース、ドレッシング、味噌、醤油、たれ等の調味料;スープ、サラダ、惣菜、ふりかけ、漬物;その他種々の形態の健康・栄養補助食品・機能性表示食品・特定保健用食品などが例示される。
本発明の組成物が適用される対象者は、EGTが本来有する生理活性を必要とする年代であれば、特に制限されないが、仕事等の生活環境からストレスを受けやすい年代である30歳頃以上であってもよく、ホルモンバランスの変化や生活環境の変化等により身体の変化を感じやすい年代である中年齢者(45歳頃以上55歳未満)であってもよく、高年齢者(55歳以上)であってもよい。
本発明の組成物のpHは、他の配合成分の種類及び含有量、製剤形態、使用方法等に応じて適宜設定され、薬学的又は生理学的に許容できる範囲であれば制限されないが、例えば、pH2~10とすることができる。本発明の効果を安定的に発揮する観点から、本発明の組成物のpHは、例えば、pH2~10、pH2~9、pH2~8、pH2~7、pH3~10、pH3~9、pH3~8、pH3~7、pH4~10、pH4~9、pH4~8、pH4~7、pH5~10、pH5~9、pH5~8、pH5~7、pH6~10、pH6~9、pH6~8、pH6~7等とすることが可能である。
本発明において、EGT及びSRHを含有し、着色が抑制された組成物の製造方法は、SRHの含有量を低減させる工程を含む。
本発明において、EGT、及び、SRHを含有する組成物であって、
該EGT100質量部に対して、該SRHの含有量が、15質量部以下とすることによる、前記組成物に対して着色抑制効果を付与する方法を提供することも可能である。EGT及びSRHの好ましい含有比率については、上記に準じる。
上述した実施の形態に関し、限定はされないが、本発明は以下の実施形態を開示する。
該S-リボシルホモシステインの含有量が、低減されてなる、組成物。
該L-エルゴチオネイン100質量部に対して、該S-リボシルホモシステインの含有量が、15質量部以下である、
前記組成物の着色を抑制するための組成物。
該S-リボシルホモシステインの含有量を低減させる工程を含む方法。
該L-エルゴチオネイン100質量部に対して、該S-リボシルホモシステインの含有量が、15質量部以下である、
前記組成物に対して着色抑制効果を付与する方法。
L-エルゴチオネイン(EGT)及びS-リボシルホモシステイン(SRH)を含む組成物について、30℃における保存安定性を下記の通り調べた。なお、SRHの含有量による影響を確認する試験モデルとして、EGTを含有する組成物に対してSRHを添加する方法を採用しているが、抽出法における夾雑物、合成法、発酵法等における副生成物等により内在的にEGTとSRHが含有されていてもよい。
(1)EGT 500mg(テトラエドロン社製)を50mLに溶解した。これを5mL×8本サンプリングしそれぞれにSRH 0.108w/w%水溶液(9.3mL、6.98mL、4.63mL、2.33mL、0.463mL、0.232mL、46.3μL、4.63μL)を含有させ、EGT100質量部に対しSRHを20質量部、15質量部、10質量部、5質量部、1質量部、0.5質量部、0.1質量部、0.01質量部で含有するサンプルを調製した。
(2)それぞれのサンプルを5等分したのち、凍結乾燥した。
(3)凍結乾燥直後に各含有量について1本ずつ1mLの水で溶解し、200μLずつ96wellマイクロプレートに採取し、MULTISKAN GO(Thermo SCIENTIFIC社製)を用いて230-600nmの吸光度測定を行った。
(4)30℃にて保管し、3日目、24日目、6か月後に各含有量について1本ずつ1mLの水で溶解し、230-600nmの吸光度測定を行った。表1は上記(3)、(4)における400nmの吸光度のデータを用いてEGTに対するのSRH含有量と吸光度変化をまとめた。
EGT50mg(テトラエドロン社製)に0.108w/w%SRH水溶液2.31mL(2.5mg、EGT100質量部に対するSRH含有量 5質量部)を含有させ、濃縮後、60℃で水200μLに溶解後、エタノールを2mL加え、析出した結晶を室温にてろ過した。得られたEGTの収量は34mg、HPLC定量値は98.5%であった(定量値はテトラエドロン社製のEGT標準品とした絶対定量)。同様に、EGT100質量部に対して、SRH 10質量部を含有させて結晶化を行った結果、及び、20質量部を含有させて結晶化を行った結果を表2に示す。
EGTの定量は以下のHPLC条件で行った。
カラム:YMC-Pack ODS-AQ S-5μM,12nm、250x4.6mm I.D.
流速:0.5mL/min、温度:30℃
検出器:PDA(257nm)
溶離液:0.1%ギ酸水溶液
保持時間:8.9分付近
EGT 7.1mg(テトラエドロン社製)をSRH 0.108w/w%水溶液(1mL)に溶解し、EGT100質量部に対しSRHが15質量部のサンプルを調製した。
このうち100μLを水で10倍希釈してHPLC分析を行った。残りの900μLについては、30℃でエバポレータにて減圧濃縮した。室温で4日放置後に水 0.9mLに溶解し、このうち100μLを水で10倍希釈してHPLC分析を行い濃縮前とHPLC面積を比較した結果を表3に示す。
HPLC分析は以下の条件で行った。

検出器:PDA(210nm)
溶離液:0.1%ギ酸水溶液
注入量:5μL
保持時間:SRH 6.6分付近 SRH脱水二量体 7.5分付近
EGT 8.7分付近
以下にSRH脱水二量体についての高分解能質量分析の結果を示す。

該L-エルゴチオネイン100質量部に対して、該S-リボシルホモシステインの含有量を、0.01~15質量部とする工程を含む方法。 - L-エルゴチオネイン、及び、S-リボシルホモシステインを含有する組成物において、
