Claims (9)
- 透明な素材からなる基板と、
前記基板上に一定の大きさと深さに形成された一つまたはそれ以上の反応チャンバーと、
前記反応チャンバーの内部に一定量の液体状態のプローブを収容することができる大きさと深さに形成された多数の粒子形成溝と、を含み、
空間的に分離された互いに異なる粒子形成溝に注入される前記プローブは、互いに異なる核酸分子の配列と特異的に混成化される互いに異なる種類のプライマー、前記プライマーと結合する蛍光色素、及び前記プライマーが固着される支持体が含まれ、
前記プローブに含まれている支持体は、物理的刺激(光、温度、超音波)によって3次元の網構造体を形成して、前記粒子形成溝に対応する形状のプローブ粒子を形成し、
前記粒子形成溝の内部には、前記プローブ粒子が離脱することを防止するための粒子固定体が形成されることを特徴とする、マルチプレックスPCRチップ。 - 透明な素材からなる基板と、
前記基板上に一定の大きさと深さに形成された1つ又はそれ以上の反応チャンバーと、
前記反応チャンバーの内部に一定量の液体状態のプローブを収容することができる大きさと深さに形成された多数の粒子形成溝と、を含み、
前記粒子形成溝は、様々な平面形状をし、互いに異なる平面形状をする粒子形成溝に注入される前記プローブは、互いに異なる核酸分子の配列と特異的に混成化される互いに異なる種類のプライマー、前記プライマーと結合する蛍光色素、及び前記プライマーが固着される支持体が含まれ、
前記プローブに含まれている支持体は、物理的刺激(光、温度、超音波)によって3次元の網構造体を形成して、前記粒子形成溝に対応する形状のプローブ粒子を形成し、
前記粒子形成溝の内部には、前記プローブ粒子が離脱することを防止するための粒子固定体が形成されることを特徴とする、マルチプレックスPCRチップ。 - 前記粒子固定体は、前記粒子形成溝の底部または内側面に一定の長さ突出するように設けられる多数の突起で構成され、前記プローブ粒子と一体に結合することを特徴とする、請求項1又は2に記載のマルチプレックスPCRチップ。
- 前記粒子固定体は、互いに異なる形状又はパターンからなり、空間的に分離された前記粒子形成溝に形成された粒子固定体は、互いに異なる形状又はパターンを有し、前記粒子形成溝に注入される前記プローブは、互いに異なる核酸分子の配列と特異的に混成化される互いに異なる種類のプライマーが含まれることを特徴とする、請求項1又は2に記載のマルチプレックスPCRチップ。
- 前記基板上に形成された反応チャンバーは、多数で構成され、前記多数の反応チャンバーは、一つの連結チャンネルを介して直列に連結され、前記連結チャンネルの一端には、試料を注入するための注入口が設けられ、前記連結チャンネルの他端には、試料を排出する排出口が設けられることにより、前記注入口を介して注入される試料がすべての反応チャンバーに供給されて充填されることを特徴とする、請求項1又は2に記載のマルチプレックスPCRチップ。
- マルチプレックスPCR方法であって、
多数の反応チャンバーが一つの連結チャンネルを介して直列に連結され、各反応チャンバーの内部には、空間的に分離された多数の粒子形成溝が形成されているPCRチップを準備するPCRチップ準備ステップ(S110)と、
前記多数の粒子形成溝に互いに異なる核酸分子の配列と特異的に混成化されるプライマー、前記プライマーと結合する蛍光色素、及び硬化されるときに3次元網状構造を形成する支持体が備えられた溶液状態のプローブを注入するプローブ注入ステップ(S120)と、
前記粒子形成溝に注入されたプローブに物理的刺激を与えて前記支持体を硬化させるとともにプライマーを前記支持体の内部に固着させ、前記粒子形成溝に対応する形状のプローブ粒子を形成するプローブ粒子固定ステップ(S130)と、
前記プローブ粒子が固定された多数の反応チャンバーに洗浄液を注入して前記支持体に多孔を形成するプローブ粒子洗浄ステップ(S140)と、を含み、
前記多数の粒子形成溝の内側面には、前記粒子形成溝の内部に形成される前記プローブ粒子を固定することができるように一定の長さ突出するように設けられる多数の突起からなる粒子固定体が備えられることを特徴とする、マルチプレックスPCR方法。 - 前記多数の反応チャンバーと連結チャンネルの開放部を密封するために前記PCRチップの一面に密封フィルムを付着させ、前記連結チャンネルの一端に設けられる注入口の開口部と前記連結チャンネルの他端に設けられる排出口の開口部を密封するためにステッカーを付着させる密封ステップ(S150)と、
前記注入口を介して多数の反応チャンバーに試料を注入して充填する試料注入ステップ(S160)と、
前記PCRチップを繰り返し加熱及び冷却して、試料に含まれている特定の塩基配列とプライマーが混成化されて連鎖的にターゲット核酸を複製し、増幅させた後、前記多数の反応チャンバーを貫通するように励起光を照射し、前記プライマーと結合した蛍光色素から発光する蛍光色と前記プローブ粒子の位置及び形状に基づいて一度に多数のターゲット遺伝子を検出するPCRステップ(S170)と、をさらに含むことを特徴とする、請求項6に記載のマルチプレックスPCR方法。 - 前記プローブ粒子洗浄ステップ(S140)で洗浄されたプローブ粒子を乾燥させるプローブ粒子乾燥ステップをさらに含むことを特徴とする、請求項6に記載のマルチプレックスPCR方法。
- 前記粒子固定体は、互いに異なる形状又はパターンで構成され、空間的に分離されている粒子形成溝に形成された粒子固定体は、互いに異なる形状又はパターンを有し、前記粒子形成溝に注入される前記プローブは、互いに異なる核酸分子の配列と特異的に混成化される互いに異なる種類のプライマーが含まれることを特徴とする、請求項6に記載のマルチプレックスPCR方法。
