Description

本発明の一態様において、コンセンサス抗原は、以下のうちの1つ以上を有することを含む、改善された転写および翻訳を提供することが望ましい:転写を増加するための低いGC含有量のリーダー配列、mRNA安定およびコドン最適化、シス作用配列モチーフ(すなわち、内部TATA-ボックス)を可能な限り排除すること。
本明細書において使用される用語法は、特定の実施形態を記載するだけの目的であり、限定的であることを意図しない。明細書および添付の特許請求の範囲で使用されるとき、単数形「a」、「an」、および「the」は、特に文脈から明白に示されない限り、複数の対象を含む。
「アジュバント」は、本明細書で使用されるとき、本明細書に記載されるDNAプラスミドワクチンに添加されて、DNAプラスミドおよび以下に記載されるコード核酸配列によりコードされる1つ以上のコンセンサスフィロウイルス免疫原の抗原性を増強する任意の分子を意味し得る。
「抗体」は、IgG、IgM、IgA、IgD、もしくはIgEの部類の抗体、またはFab、F(ab′)2、Fdが含まれるそのフラグメント、フラグメント、もしくは誘導体、ならびに一本鎖抗体、二特異性抗体、二重特異性抗体、二官能性抗体、およびその誘導体を意味し得る。抗体は、哺乳動物の血清試料から単離された抗体、ポリクローナル抗体、親和精製抗体、または所望のエピトープもしくはそれから誘導される配列に対して十分な結合特異性を示すそれらの混合物であり得る。
「コード配列」または「コードする核酸」は、本明細書で使用されるとき、タンパク質をコードするヌクレオチド配列を含む核酸(RNAまたはDNA分子)を意味し得る。コード配列は、核酸が投与される個体または哺乳動物の細胞において発現を指示することができるプロモーターおよびポリアデニル化シグナルを含む、調節エレメントに機能的に結合している開始および終結シグナルを更に含み得る。幾つかの実施形態では、コード配列は所望により、N末端メチオニンまたはIgEもしくはIgGシグナルペプチドなどのシグナルペプチドをコードする開始コドンを更に含んでもよい。
「補体」または「相補」は、本明細書で使用されるとき、核酸が、核酸分子のヌクレオチドまたはヌクレオチド類縁体の間のワトソン・クリック(例えば、A-T/UおよびC-G)またはフーグスティーン塩基対を意味し得ることを意味する。

「フラグメント」は、特定のフィロウイルス抗原を認識することによってフィロウイルスに対する哺乳動物における免疫応答を誘発することが可能な、フィロウイルス免疫原のポリペプチドフラグメントを意味し得る。フィロウイルスエンベロープ糖タンパク質免疫原は所望により、位置1のシグナルペプチドおよび/またはメチオニン、本明細書に記載されるコンセンサス配列に98%以上相同なタンパク質、本明細書に記載されるコンセンサス配列に99%以上相同なタンパク質、ならびに本明細書に記載されるコンセンサス配列に100%同一なタンパク質を含んでもよく、いずれの場合も位置1にシグナルペプチドおよび/またはメチオニンを伴っても伴わなくてもよい。フラグメントは、例えば、シグナルペプチド、例えば、IgEシグナルペプチドまたはIgGシグナルペプチドなどの免疫グロブリンシグナルペプチドなどに連結される、フィロウイルス免疫原のフラグメントを含んでも含まなくてもよい。
「同一」または「同一性」は、2つ以上の核酸またはポリペプチド配列の文脈において本明細書で使用されるとき、配列が、特定の領域にわたって同じである残基の特定の率(%)を有することを意味し得る。率(%)は、2つの配列を最適に整列すること、2つの配列を特定の領域にわたって比較すること、同一残基が両方の配列に発生してマッチした位置の数を生じる、位置の数を決定すること、マッチした位置の数を、特定の領域内の位置の総数で割ること、および結果に100を掛けて、配列同一性の率(%)を生じることによって、計算することができる。2つの配列が異なる長さである、または整列が1つ以上の付着末端を生じ、特定の比較領域が、単一配列のみを含む場合、単一配列の残基は、計算の分母に含まれるが、分子には含まれない。DNAおよびRNAを比較すると、チミン(T)およびウラシル(U)は、同等であると考慮することができる。同一性は、手作業により、またはBLASTもしくはBLAST2.0のようなコンピューター配列アルゴリズムを使用して実施することができる。

「核酸」または「オリゴヌクレオチド」または「ポリヌクレオチド」は、本明細書で使用されるとき、一緒に共有結合している少なくとも2つのヌクレオチドを意味し得る。一本鎖の描写は、相補鎖の配列も定義する。したがって、核酸は、描写される一本鎖の相補鎖も包含する。核酸の多くの変種を、所定の核酸と同じ目的で使用することができる。したがって、核酸は、実質的に同一の核酸およびその相補体も包含する。一本鎖は、厳密なハイブリッド形成条件下で標的配列とハイブリッド形成することができるプローブを提供する。したがって、核酸は、厳密ハイブリッド形成条件下でハイブリッド形成するプローブも包含する。
「機能的に連結する」は、本明細書で使用されるとき、遺伝子の発現が、空間的に連結しているプロモーターの制御下にあることを意味し得る。プロモーターは、その制御下の遺伝子の5′(上流)または3′(下流)に位置することができる。プロモーターと遺伝子との間隔は、プロモーターが誘導される遺伝子における、プロモーターと、それが制御する遺伝子との間隔とほぼ同じであり得る。当該技術において知られているように、この間隔の変動は、プロモーター機能を失うことなく適応され得る。

「厳密なハイブリッド形成条件」は、本明細書で使用されるとき、核酸の複合混合物におけるように、第1の核酸配列(例えば、プローブ)が第2の核酸配列(例えば、標的)とハイブリッド形成する条件を意味し得る。厳密な条件は、配列依存性であり、異なる状況において異なる。厳密な条件は、確定されたイオン強度のpHで特定の配列の熱融点(Tm)よりも約5~10℃低くなるように選択することができる。Tmは、標的に相補的なプローブの50%が標的配列と平衡(標的配列がTmで過剰に存在するので、50%のプローブが平衡を占める)にハイブリッド形成する(確定されたイオン強度、pH、および核濃度下での)温度であり得る。厳密な条件は、塩濃度が、pH7.0~8.3で約0.01~1.0Mのナトリウムイオン濃度のような約1.0M未満のナトリウムイオン(または他の塩)であり、温度が、短プローブ(例えば、約10~50個のヌクレオチド)では少なくとも約30℃、長プローブ(例えば、約50個を超えるヌクレオチド)では少なくとも約60℃であるものであり得る。厳密な条件は、ホルムアミドのような不安定化剤の添加により達成することもできる。選択的または特定的なハイブリッド形成では、陽性シグナルは、バックグラウンドハイブリッド形成の少なくとも2~10倍であり得る。例示的な厳密なハイブリッド形成条件には、以下が含まれる:50%ホルムアミド、5×SSC、および1%SDS、42℃でのインキュベーション、または5×SSC、1%SDS、65℃でのインキュベーションと、0.2×SSCでの洗浄および65℃での0.1%SDS。
「実質的に相補的」は、本明細書で使用されるとき、第1の配列が第2の配列の補体と、8個、9個、10個、11個、12個、13個、14個、15個、16個、17個、18個、19個、20個、21個、22個、23個、24個、25個、30個、35個、40個、45個、50個、55個、60個、65個、70個、75個、80個、85個、90個、95個、100個、またはそれ以上のヌクレオチドもしくはアミノ酸の領域にわたって、少なくとも60%、65%、70%、75%、80%、85%、90%、95%、97%、98%、または99%同一であること、あるいは2つの配列が厳密なハイブリッド形成条件下でハイブリッド形成することを意味し得る。
「実質的に同一」は、本明細書で使用されるとき、第1の配列および第2の配列が、8個、9個、10個、11個、12個、13個、14個、15個、16個、17個、18個、19個、20個、21個、22個、23個、24個、25個、30個、35個、40個、45個、50個、55個、60個、65個、70個、75個、80個、85個、90個、95個、100個、またはそれ以上のヌクレオチドもしくはアミノ酸の領域にわたって、あるいは第1の配列が第2の配列の補体と実質的に相補的である場合、核酸に関して、少なくとも60%、65%、70%、75%、80%、85%、90%、95%、97%、98%、または99%同一であることを意味し得る。
「変種」は、核酸に関して本明細書で使用されるとき、(i)基準ヌクレオチド配列の一部もしくはフラグメント、(ii)基準ヌクレオチド配列の補体もしくはその一部、(iii)基準核酸もしくはその補体と実質的に同一である核酸、または(iv)厳密な条件下、基準核酸、その補体もしくはそれと実質的に同一の配列とハブリッド形成する核酸を意味する。
本明細書で使用される「ベクター」は、複製の起点を含有する核酸配列を意味し得る。ベクターは、プラスミド、バクテリオファージ、細菌人工染色体、または酵母人工染色体であり得る。ベクターは、DNAまたはRNAベクターであり得る。ベクターは、自己複製染色体外ベクター、または宿主ゲノムに組み込まれるベクターのいずれかであり得る。
特定のフィロウイルス抗原の複数の亜型または血清型に対する広範な免疫を誘導するために使用することができるフィロウイルス免疫原が、本明細書に提供される。コンセンサスフィロウイルス抗原はそれぞれ、マールブルグウイルスフィロウイルス糖タンパク質MARV RAV免疫原のコンセンサスアミノ酸配列、マールブルグウイルスフィロウイルス糖タンパク質MARV OZO免疫原のコンセンサスアミノ酸配列、マールブルグウイルスフィロウイルス糖タンパク質MARV MUS免疫原のコンセンサスアミノ酸配列、マールブルグウイルスフィロウイルス糖タンパク質MARV ANG免疫原の単離アミノ酸配列、ザイールエボラウイルス糖タンパク質ZEBOV免疫原のコンセンサスアミノ酸配列、およびスーダンエボラウイルス糖タンパク質SUDV免疫原のコンセンサスアミノ酸配列を含んでもよい。幾つかの実施形態では、免疫原は、例えば、IgEシグナルペプチドまたはIgGシグナルペプチドなどの免疫グロブリンタンパク質などの異なるタンパク質に由来するシグナルペプチドを含んでもよい。
本明細書に記載されるタンパク質をコードするコード配列は、定められた方法を用いて生成することができる。コンセンサスザイールエボラウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列、コンセンサススーダンエボラウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列、マールブルグマールブルグウイルスアンゴラ2005エンベロープ糖タンパク質免疫原をコードする核酸配列を含む組成物が提供され、第1のコンセンサスマールブルグマールブルグウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列、第2のコンセンサスマールブルグマールブルグウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列、および第3のコンセンサスマールブルグマールブルグウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列を、開示されるアミノ酸配列に基づいて生成することができる。
プラスミドは、合成の、フィロタンパク質に対する免疫応答を誘発することができるコンセンサス抗原をコードするコード配列を含む、上述に開示される様々な免疫原のうちの1つ以上をコードする核酸配列を含んでもよい。
核酸分子を含む組成物が提供される。組成物は、単一のプラスミドのような単一の核酸分子の複数のコピー、2つ以上の異なるプラスミドのような2つ以上の異なる核酸分子の複数のコピーを含むことができる。例えば、組成物は、複数の2、3、4、5、6、7、8、9、もしくは10個、またはそれ以上の異なる核酸分子を含むことができる。そのような組成物は、複数の、2、3、4、5、6個、またはそれ以上の異なるプラスミドを含むことができる。
哺乳動物においてフィロウイルス、具体的には、マールブルグウイルス、エボラウイルス・スーダン、および/またはエボラウイルス・ザイールに対して免役応答を生成することができるワクチンが、本明細書に提供される。ワクチンは、上で考察されたそれぞれのプラスミドを含むことができる。ワクチンは、複数のプラスミドまたはそれらの組み合わせを含むことができる。ワクチンは、治療または予防免疫応答を誘導するために提供され得る。
それらに対する免疫応答が誘導され得るフィロウイルス、具体的には、マールブルグウイルス、エボラウイルス・スーダン、および/またはエボラウイルス・ザイールの特に有効な免疫原となるエピトープを含む、コンセンサス抗原の遺伝子構築物およびタンパク質を提供するためのワクチンを送達するため方法が、本明細書において提供される。ワクチンを送達する、またはワクチン接種の方法は、治療的および予防的な免疫応答を誘導するために提供することができる。ワクチン接種プロセスは、フィロウイルス、具体的には、マールブルグウイルス、エボラウイルス・スーダン、および/またはエボラウイルス・ザイールに対する免疫応答を哺乳動物において生成することができる。ワクチンを個体に送達して、哺乳動物の免疫系の活性を調節し、免疫応答を増強することができる。ワクチンの送達は、細胞内で発現し、細胞の表面に送達され、その時点で免疫系が認識し、細胞性、液性、または細胞性および液性の応答を誘導する核酸分子としてのコンセンサス抗原の形質移入であり得る。ワクチンの送達は、上記に考察されたワクチンを哺乳動物に投与することによって、フィロウイルス、具体的には、マールブルグウイルス、エボラウイルス・スーダン、および/またはエボラウイルス・ザイールに対する免疫応答を哺乳動物に誘導または誘発するために使用することができる。

ワクチンのプラスミドの電気穿孔を介したワクチンの投与は、細胞膜に可逆的細孔を形成させるのに有効なエネルギーパルスを、哺乳動物の所望の組織に送達すように構成され得る電気穿孔装置を使用して達成することができ、好ましくは、エネルギーパルスは、使用者により予め設定された電流入力に類似した定電流である。電気穿孔装置は、電気穿孔構成要素および電極アセンブリーまたはハンドルアセンブリーを含むことができる。電気穿孔構成要素は、制御装置、電流波形発生器、インピーダンス試験器、波形自動記録器、入力要素、状況報告要素、通信ポート、記憶装置構成要素、電源、および電源スイッチが含まれる、電気穿孔装置の様々な要素の1つ以上を含み、組み込むことができる。電気穿孔は、インビボ電気穿孔装置、例えば、CELLECTRA EPシステム(VGX Pharmaceuticals,Blue Bell,PA)またはElgen電気穿孔機(Genetronics,San Diego,CA)を使用することにより達成され、プラスミドによる細胞の形質移入を促進することができる。
本明細書で考察されているDNAワクチンを含むDNAプラスミドを調製する方法が、本明細書において提供される。DNAプラスミドは、哺乳類発現プラスミドへの最終サブクローニングステップの後、当該技術において既知の方法を使用して、大規模発酵タンク中の細胞培養物への接種に使用することができる。
方法
プラスミドワクチン構築
pMARV、pEBOS、およびpEBOZプラスミドDNA構築物は、全長GPタンパク質をコードする。アミノ酸コンセンサス戦略を、pEBOSおよびpEBOZのために使用し、一方2005Angola発生株に由来する型マッチ配列(GenBank #VGP_MABVR)を、pMARVのために使用した(Towner JS,et al.(2006).Marburgvirus genomics and association with a large hemorrhagic fever outbreak in Angola.J Virol 80:6497-6516)。コンセンサス配列を、主流のZEBOVおよびSUDV GPアミノ酸配列の整列、およびそれぞれに関するコンセンサスの生成によって決定した。各ワクチンGP遺伝子を、ヒトにおける発現において遺伝的に最適化し(タンパク質発現を増強するためのコドン最適化およびRNA最適化を含む(GenScript,Piscataway,NJ))、商業的に合成し、次にサイトメガロウイルス初期(CMV)プロモーターの制御下で改変されたpVAX1哺乳動物の発現ベクター(Invitrogen,Carlsbad,CA)へとサブクローニングした(GenScript,Piscataway,NJ)。改変は、2A>C、3C>T、4T>G、241C>G、1,942C>T、2,876A>-、3,277C>T、および3,753G>Cを含む。系統発生分析を、MEGAバージョン5ソフトウエアを用いてClustalWによる多重整列によって実施した。別法として、MARV間のGP多様性は、比較において遥かに高かったため(約70%の同一性)、コンセンサス戦略は適用されなかった。MARVの対応範囲のため、我々は、アンゴラにおける2005発生に由来するMGP配列(GenBank #VGP_MABVR)が、今日までで最大規模かつ最も致命的なMARV発生の唯一の原因であったため、その利用を選択した。この配列は、Musoke、Popp、およびLeiden(10.6%分岐)、またはUganda(01Uga07)、Durba(05DRC99および07DRC99)、およびOzolin(10.3%分岐)を含む、関連株の最も近いクラスターのいずれかからも10%超分岐した。全体として、3つのプラスミド戦略は、我々の新規の三価多価フィロウイルスワクチン戦略の基盤を形成した。
ヒト胎児性腎臓(HEK)293T細胞を培養し、形質移入し、採取した。手短に述べると、細胞は、10%FBS、1%Pen-strep、ピルビン酸ナトリウム、およびL-グルタミンと共にDMEM中で成長させた。細胞を、150mmコーニング皿内で培養し、5%CO2を伴う37°インキュベーター内で一晩、70%集密まで成長させた。皿を、製造者プロトコールに従って、Calphos(商標)Mammalian Transfection Kitプロトコル(Clonetech)かまたはLipofectamine(商標)2000試薬(Invitrogen)かを用いて10~25μgのフィロウイルス科pDNAで形質移入し、次に24~48時間インキュベートした。細胞を、冷たいPBSで採取し、遠心分離および洗浄した後、ウエスタン免疫ブロットまたはFACS解析のためにペレット化した。標準ウエスタンブロットを使用し、GP1検出のためにGP特異性MAbを生成した。ウエスタン免疫ブロット法実験からのデータが、図1Bに示される。FACS解析からのデータが、図1Cに示される。
成体メスC57BL/6(H-2b)、BALB/cJ(H-2d)、およびBl0.Br(H-2k)マウスを、The Jackson Laboratory(Bar Harbor,ME)から購入し、一方ハートレイ系モルモットをCharles River(Wilmington,MA)から購入した。全ての動物実験は、UPenn IACUC and School of Medicine Animal Facility、またはNML Institutional Animal Care Committee of the PHAC and the Canadian Council on Animal Care guidelines for housing and care of laboratory animalsに従って行い、上述の機関による関連チェックおよび認可後に、Guide for the Care and Use of Laboratory Animals of NIHにおける推奨事項に従って実施した。UPennおよびNMLは、動物の福祉に関するNIHポリシー、Animal Welfare Act、および他の全ての該当の連邦法、州法、および現地法に適合する。
抗体(Ab)価を、スクロース精製したMARV Ozolin GPもしくはZGPか、または負の対照スクロース精製したNipah Gタンパク質かで1:2,000の濃度でコーティングした96ウエルELISAプレートを用いて決定した。手短に述べると、プレートを次に、4℃で18時間インキュベートし、PBSおよび0.1%Tween-20で洗浄し、100μl/標本の血清を三連で試験した(5%スキムミルクおよび0.5%Tween-20を伴うPBS中で1:100、1:400、1:1,600、および1:6,400の希釈)。湿潤容器中で37℃にて1時間インキュベーションした後、プレートを洗浄し、次に100μlのヤギ抗マウスIgG-共役HRP抗体(Cedarlane)を添加し(1:2,000希釈)、湿潤容器中で更に37℃で1時間インキュベートした。洗浄後、100μlのABST(2,2’-アジノ-ビス(3-エチルベンチアゾリン-6-スルホン酸)およびペルオキシダーゼ基質(Cedarlane)を添加して、Ab結合を可視化した。再び湿潤容器中で、プレートを37℃で30分間インキュベートし、次にその後405nmで読み取った。正の結合結果を、負の対照血清から正の対照を減じ、3標準偏差超とすることによって特徴付けした。
マウスを、最終免疫化の8~11日後に屠殺し、脾臓を採取した。手短に述べると、脾臓を、10%FBS、1XAnti-anti(Invitrogen)、および1X β-ME(Invitrogen)を補充したRPMI 1640培地(Mediatech Inc.,Manassas,VA)中に定置した。脾細胞を、Stomacher machine(Seward Laboratory Systems Inc.,Bohemia,NY)を用いた脾臓の機械的分裂によって単離し、得られた産生物を、40μm細胞濾過器(BD Falcon)を用いて濾過した。細胞を次に、RBCの溶解のためにACK溶解緩衝液(Lonza,Switzerland)で5分間処理し、PBS中で洗浄し、次にELISPOTまたはFACS分析での使用のためにRPMI培地中に再懸濁した。
標準IFNγ ELISPOT分析を実施した。手短に述べると、96ウエルプレート(Millipore,Billerica,MA)を、抗マウスIFN-γ捕捉抗体でコーティングし、4℃で24時間インキュベートした(R&D Systems,Minneapolis,MN)。翌日、プレートをPBSで洗浄し、次にブロッキング緩衝液(PBS中の1%BSAおよび5%スクロース)で2時間ブロッキングした。脾細胞(1~2x105細胞/ウエル)を、三連で定置し、RPMI 1640(負の対照)か、Con A(正の対照)か、またはGPペプチドかのいずれかの存在下で、個別に(9個のアミノ酸による15mer重複、およびそれらのそれぞれのGPの長さにわたる)、または全てプールして(最終で2.5μg/ml)、5%CO2内で37℃にて一晩刺激した。刺激の18~24時間後、プレートをPBS中で洗浄し、次にビオチン化抗マウスIFN-γ mAb(R&D Systems,Minneapolis,MN)で4℃で24時間インキュベートした。次に、プレートを再びPBS中で洗浄し、ストレプトアビジンアルカリホスファターゼ(MabTech,Sweden)を各ウエルに添加し、室温で2時間インキュベートした。最後に、プレートを再びPBSで洗浄し、次にBCIP/NBT Plus基質(MabTech)を、スポット現像のために各ウエルに5~30分間添加した。目視上で現像プロセスが完了すると直ちに、プレートを蒸留水で濯ぎ、次に室温にて一晩乾燥した。スポットを、自動ELISPOTリーダー(Cellular Technology Ltd.,Shaker Heights,OH)を用いて計数した。
脾細胞を、96ウエルプレート(lxl06細胞/ウエル)に添加し、個々のペプチドか、または「最小ペプチドプール」(最終2.5μg/ml)かのいずれかで5~6時間刺激した。個々のペプチド刺激を、改変されたELISPOTによって確認された全ペプチド(表1~6)の機能的確証および表現型特徴付けのために使用した。まず、脾細胞および形質移入293Tを、LIVE/DEAD(登録商標)Fixable Violet Dead Cell Stain Kit(Invitrogen)で予備染色した。脾細胞に関して、細胞を、CD19(V450;クローン1D3)、CD4(PE-Cy7;クローンRM4-5)、CD8α(APC-Cy7;クローン53-6.7)、およびCD44(PE-Cy5;クローンIM7)(BD Biosciences,San Jose,CA)に関して表面染色し、PBS+1%FBS中で3回洗浄し、BD Cytofix/Cytoperm(商標)キットで透過化し、次にIFNγ(APC;クローンXMG1.2)、TNF(FITC;クローンMP6-XT22)、CD3(PE-cy5.5;クローン145-2C11)、およびT-bet(PE;クローン4B10)(eBioscience,San Diego,CA)で細胞内を染色した。形質移入293T細胞におけるGP発現を、形質移入の24時間後に評価した。各々がその対応するDNAワクチンまたはpVAX1空ベクター対照で3回免疫付与されたH-2bマウスに由来する血清をプールすることによって産生された指示されたマウス由来GP特異性ポリクローナル血清試薬(1:200希釈)を含有するPBS+1%FBS中で4℃にて30分間インキュベートした後、間接染色を実施した。細胞を、次にFITC複合体化ヤギ抗マウスIgG(BioLegend,San Diego,CA)で染色し、大規模に洗浄し、次にMHCクラスIに関して染色した(HLA-ABC;PE-Cy7;クローンG46-2.6;BD)。全細胞を、1%パラホルムアルデヒド中で固定した。全データを、LSRII flow cytometer(BD)を用いて収集し、FlowJoソフトウエア(Tree Star,Ashland,OR)を用いて解析した。脾細胞を、CD3+CD44+、CD4+、またはCD8+であり、B細胞マーカーCD19および生存率判定染料に関して負である、活性化したIFNγ産生T細胞に関してゲーティングした。
無根系統樹に関する有意性を、最大公算法によって決定し、ブートストラップ解析によって検証し、有意な支持値(≧80%、1,000ブートストラップ複製)を、MEGAバージョン5ソフトウエアによって決定した。群解析を、マッチされた両側独立t検定によって完了し、生存曲線をログランク(マンテル・コックス)検定によって解析した。全ての値は、平均±標準誤差であり、統計解析はGraphPad Prism(La Jolla,CA)によって実施した。
ワクチン構築および発現
系統発生分析は、EBOV GPにおける相対的保存を示し(SUDVに関して94.4%、およびZEBOVに関して92.9%)、MARV GP(MGP)は、より分岐していた(約70%保存)。したがって、主流のZEBOVおよびSUDV GPアミノ酸配列の整列によって決定されるコンセンサス戦略が、EBOV GPに対して適用され、一方MARVに関しては、最大規模かつ最も致命的なMARV発生の唯一の原因であった2005Angola発生配列を採用する型マッチ戦略が使用された。各GPトランス遺伝子が、遺伝的に最適化され、商業的に合成され、次に改変されたpVAX1哺乳動物発現ベクターへとサブクローニングされた。全体として、3つのプラスミド戦略は、我々の新規の多価フィロウイルスワクチン戦略の基盤を形成した。
防御効果を決定するため、モルモット前臨床攻撃モデルを採用した。前臨床免疫原性および有効性試験を、モルモットおよびマウスモデルを用いて本明細書で実施した。モルモット前臨床モデルは、フィロウイルスワクチン開発のためのスクリーニングおよび「概念実証」ツールとして広く使用されている。MARVおよびEBOVの一次単離物は、モルモットに非致死性の疾病を引き起こすが、この宿主の少数継代は、フィロウイルスに感染した霊長類において見られるものに類似の病理学的特徴を伴う致死性疾患を引き起こし得る変種の選択をもたらす。同様に、マウスは、フィロウイルスワクチン開発のために広く使用されているが、しかしながら、モルモットモデルと異なり、免疫およびT細胞応答を評価するための免疫検出試薬が、広く利用可能である。マウス適合したZEBOV(mZEBOV)による感染は、標的臓器内の高レベルのウイルス、ならびにEBOV感染した霊長類において見出されるものと同種の肝臓および脾臓における病理学的変化によって特徴付けられる疾患をもたらす。
防御的DNAワクチン(プラスミドpEBOZ、pEBOS、およびpMARV、三価DNAワクチンとも称する)によって引き起こされた免疫相関物をより良く特徴付けるため、フィロウイルスワクチン開発のためのスクリーニングおよび「概念実証」ツールとして広く使用されており、広範な免疫検出試薬が利用可能であるマウスモデルを次に採用した。まず、B細胞応答を、2つのワクチン接種のそれぞれの後に20日間、40μgのそれぞれの一価DNAワクチンの注射の間に3週間あけて、H-2dマウス(n=5/群)において評価した。これらの実験からのデータが、図3A~3Cに示される。図3Aおよび図3Bに示される通り、出血前対照標本においてGP特異性IgGはほとんど観察されなかったが、ワクチン接種後には全動物において著しい増加が検出された。精製されたSGPは利用可能ではなかったため、精製されたZGPを代わりに使用した。SUDVワクチン接種を受けたマウスにおけるIgGはZGPに結合し、ワクチン誘導Ab生成能力、および異種種間認識能力を実証している。加えて、セロコンバージョンが、僅か1回の免疫化後にワクチン接種を受けた動物の100%に生じ、その後、応答は同様の強化によって著しく増加された。AVE逆数エンドポイント希釈力価は、pMARV免疫付与されたマウスにおいて22.1倍、またpEBOSおよびpEBOZワクチン接種を受けた動物においてそれぞれ3.4倍および8.6倍強化された。次に、標本は、BSL-4の施設内でZEBOV、SUDV-Boniface、およびMARV-Angolaの中和に関して分析された。中和分析の結果が図3Cに示される。NAb価における著しい増加が、全動物においてワクチン接種後に検出された。
次に、ZEBOV攻撃に対するワクチン有効性が、前臨床マウスにおいて評価された。マウスは、強いNAb誘導のため、1回のみワクチン接種を受け、防御データが観察された。マウス(H-2k、n=10/群)は、40μgのpEBOZ DNAで免疫付与され、防御が、1,000LD50のマウス適合したZEBOV(mZEBOV)による攻撃によって、28日後にBSL4の施設の施設内で評定された。全ての対照動物は攻撃後7日目までに感染により死亡したが、図5Aは、DNAワクチン接種を受けたマウスは、完全に防御されたことを示す(P=0.0002)。それに加えて、図5Bに示される通り、対照マウスは、死亡までに進行性の体重損失を見せた(P<0.0001)。

Claims (11)
- 組成物であって、
a)コンセンサスザイールエボラウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列を含む核酸分子であって、前記コンセンサスザイールエボラウイルスエンベロープ糖タンパク質免疫原のアミノ酸配列が、配列番号1(ZEBOV CON)、およびIgEシグナルペプチドに連結される配列番号1(ZEBOV CON)からなる群から選択される、核酸配列を含む核酸分子、および/または、
b)コンセンサススーダンエボラウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列を含む核酸分子であって、前記コンセンサススーダンエボラウイルスエンベロープ糖タンパク質のアミノ酸配列が、配列番号2(SUDV CON)、およびIgEシグナルペプチドに連結される配列番号2(SUDV CON)からなる群から選択される、核酸配列を含む核酸分子
を含み、
前記核酸分子が、核酸が投与される個体または哺乳動物の細胞において発現を指示することができるプロモーターおよびポリアデニル化シグナルを含む調節エレメントに作動可能に結合している開始および終結シグナルを更に含む、組成物。 - i)前記コンセンサスザイールエボラウイルスエンベロープ糖タンパク質免疫原をコードするヌクレオチド配列は、配列番号64(ZEBOV CON)およびIgEシグナルペプチドをコードするヌクレオチド配列に連結される配列番号64(ZEBOV CON)からなる群から選択され、
ii)前記コンセンサススーダンエボラウイルスエンベロープ糖タンパク質をコードするヌクレオチド配列は、配列番号65(SUDV CON)およびIgEシグナルペプチドをコードするヌクレオチド配列に連結される配列番号65(SUDV CON)からなる群から選択される、請求項1に記載の組成物。 - 前記コンセンサスザイールエボラウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列を含むプラスミド、または前記コンセンサススーダンエボラウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列を含むプラスミドを含む、請求項1に記載の組成物。
- 電気穿孔を用いた個体への送達のために製剤化される、請求項1~3のいずれか一項に記載の組成物。
- IL-12、IL-15、およびIL-28からなる群から選択される1個以上のタンパク質をコードする核酸配列を含む核酸分子を更に含む、請求項1~3のいずれか一項に記載の組成物。
- フィロウイルスに対する免疫応答を誘導するための請求項1~3のいずれか一項に記載の組成物であって、個体に免疫応答を誘導するのに有効な量で前記個体に投与される、組成物。
- エボラウイルス・スーダン、およびエボラウイルス・ザイールからなる群から選択されるフィロウイルスに対する免疫応答を誘導するための請求項1~3のいずれか一項に記載の組成物。
- フィロウイルスに感染していると診断された個体を治療するための請求項1~3のいずれか一項に記載の組成物であって、その治療的に有効な量が個体に投与される、組成物。
- エボラウイルス・スーダン、およびエボラウイルス・ザイールからなる群から選択されるフィロウイルスと診断された個体を治療するための請求項1~3のいずれか一項に記載の組成物。
- 個体のフィロウイルス感染を予防するための請求項1~3のいずれか一項に記載の組成物であって、その予防的に有効な量が個体に投与される、組成物。
- 個体のフィロウイルス感染を予防するための請求項1~3のいずれか一項に記載の組成物であって、前記フィロウイルスは、エボラウイルス・スーダン、およびエボラウイルス・ザイールからなる群から選択される、組成物。
