Description
したがって、人工多能性幹細胞由来の分化細胞の再生医療への応用は、大変注目されている。
例えば、人工多能性幹細胞の製造過程で、樹立された人工多能性幹細胞を培養する工程において、ゲノム構造に変化が起き、細胞増殖にとって有利な変化を起こした細胞集団が出現する可能性が報告されている。細胞増殖に有利に働くゲノム構造の変化は癌化と密接な関連があるため、当該変化をおこした細胞集団は、人工多能性幹細胞の再生医療応用におけるリスク要因となる。
ゲノム構造の変化と、ゲノム構造の変化のしやすさ、即ちゲノム不安定性とは密接な関係をもつと考えられるため、当該リスク要因を排除するために、臨床応用に使用する人工多能性幹細胞は、長期培養を実施した後に、最終的に、細胞や核の形態の観察や、分裂中期の染色体の数や形態を観察する核型解析、遺伝子の変異を検査する全ゲノム配列解析、一塩基多型解析であるSNPs(Single Nucleotide Polymorphism)解析、コピー数多型解析であるCNV(Copy Number Variation)解析などの試験を実施することで、ゲノム構造の安定性の高い高品質な人工多能性幹細胞を選抜する必要があった(非特許文献1)。

以下、本明細書で共通して用いられる用語は、特記しない限り、以下の意味である。
核型解析は、染色体の構造の形態学的特徴あるいは染色体数の増減を直接的に観察することで異常の種類を同定することができる点に特徴があるが、一方で個々の染色体の観察には熟練が必要である。また、観察に多大な労力を要することから検査できる細胞数がごく少数に限られ、低頻度に発生する異常の検出や、異常が発生する頻度の定量的な見積もりや細胞間での定量的な比較が困難であるという課題がある。加えて、核型解析はある一時点における染色体の特徴を解析するため、培養における細胞分裂に伴う経時的変化の定量的な比較が困難である。
全ゲノム配列解析は非常に解像度の高い解析手法であり、後述するSNPsやCNVに関する情報が得られる他、低頻度に発生したDNAの変異を1塩基レベルで検出可能である。一方で、当該解析には多大なるコストと期間を要するという課題があり、樹立された多数の人工多能性幹細胞の全てを検査して品質のよい株を選抜するには非実用的である。
(1)培養された人工多能性幹細胞を提供する工程、
(2)提供された人工多能性幹細胞中の異常核酸構造を有する細胞小器官の自然出現頻度を測定する工程、及び
(3)自然出現頻度が基準値以下である人工多能性幹細胞と、自然出現頻度が基準値を超える人工多能性幹細胞とを識別する工程を含むことを特徴とする、人工多能性幹細胞の評価方法(以下「本発明の評価方法」と略記する。)を提供する。
(1)樹立され、培養された人工多能性幹細胞を提供する工程、
(2)提供された人工多能性幹細胞中の異常核酸構造を有する細胞小器官の自然出現頻度を測定する工程、
(3)前記自然出現頻度が基準値以下である人工多能性幹細胞と、前記自然出現頻度が基準値を超える人工多能性幹細胞とを識別する工程、
(4)前記自然出現頻度が基準値以下である人工多能性幹細胞を選抜する工程、及び
(5)(4)で選抜された人工多能性幹細胞を分化誘導し、分化細胞を得る工程
を含むことを特徴とする、人工多能性幹細胞由来分化細胞の製造方法を提供する。
前記の人工多能性幹細胞由来分化細胞の製造方法は、前記(1)工程の前に、さらに下記工程を含むこともできる:
(A)人工多能性幹細胞を樹立する工程、及び
(B)樹立された人工多能性幹細胞を培養する工程。
以下、各工程について具体的に説明する。
人工多能性幹細胞を樹立する方法は、体細胞に特定の初期化因子を導入する工程を含む限り特に限定されない。例えば、体細胞を採取し、Oct3/4、Sox2、Klf4、Myc等の初期化因子を導入して人工的に発現させた後、多能性を獲得した細胞を選択して拡大培養する工程、レトロウイルスベクター又はセンダイウイルスベクターを用いて初期化因子(Oct3/4、Sox2、Klf4、及びc-Myc)をヒト末梢血リンパ球に導入し培養する工程(Nishimura, T. et al. Cell Stem Cell 2013, 12, 114-126)等を挙げることができる。
本発明において小核自然頻度等の異常核酸構造を有する細胞小器官の自然出現頻度の測定方法は限定されるものではなく、種々の公知の方法が利用できる。
本工程では、本自然出現頻度を測定した人工多能性幹細胞を、本自然出現頻度が基準値以下であるか又は本自然出現頻度が基準値を超えるかによって識別する。
ここで、「基準値」は、安全性が高いことが既知であるES細胞の本自然出現頻度を基に設定することができる。また、複数のES細胞の細胞株における本自然出現頻度の統計値を基に設定することもできる。例えば、各ES細胞の細胞株について、複数回の実験によりそれぞれ本自然出現頻度の平均値を算定し、ES細胞の細胞株間での本自然出現頻度の平均値を比較し、平均値が最大であるES細胞の細胞株の該平均値を基準値として採用することができる。この場合において、好ましい基準値は例えば2%である。あるいは、各ES細胞の細胞株について、複数回の実験によりそれぞれ本自然出現頻度の最大値を算定し、ES細胞の細胞株間での本自然出現頻度の最大値を比較し、最大値が最大であるES細胞の細胞株の該最大値を基準値として採用することができる。
あるいは、「基準値」は、基準とするiPS細胞における本自然出現頻度の統計値を基に設定することもできる。例えば、既存の品質評価指標により安全性が高いことが確認されている特定のiPS細胞株群について、複数回の実験によりそれぞれ本自然出現頻度の平均値を算定し、iPS細胞株間での本自然出現頻度の平均値を比較し、平均値が最大であるiPS細胞株の該平均値を基準値として採用することができる。あるいは、各iPS細胞株について、複数回の実験によりそれぞれ本自然出現頻度の最大値を算定し、iPS細胞株間での本自然出現頻度の最大値を比較し、最大値が最大であるiPS細胞株の該最大値を基準値として採用することができる。また、iPS細胞株の本自然出現頻度の50パーセンタイル値(中央値)を基準値としてもよく、30パーセンタイル値を基準値としてもよく、平均値を基準値としてもよい。具体的には、複数の細胞株からなる特定のiPS細胞株群について、複数回の実験によりそれぞれ本自然出現頻度の平均値を算定し、該iPS細胞株群における本自然出現頻度の平均値の中央値、30パーセンタイル値または平均値等を基準値として採用することができる。
次いで、本自然出現頻度が基準値以下である人工多能性幹細胞をゲノム構造の安定性が高く、腫瘍化のリスクが低い人工多能性幹細胞として識別することができる。
また、後述の実施例に示す通り、本自然出現頻度が特定の基準値以下である人工多能性幹細胞は、本自然出現頻度が該基準値を超える人工多能性幹細胞と比べて、分化効率が高いことが示された。従って、本発明の別の態様において、前記識別する工程は、本自然出現頻度が基準値以下である人工多能性幹細胞を、本自然出現頻度が基準値を超える人工多能性幹細胞よりも分化効率が高い人工多能性幹細胞であると評価する工程に置き換えた、人工多能性幹細胞の評価方法が提供される。
本工程では、本自然出現頻度が基準値以下である人工多能性幹細胞を、ゲノム構造の安定性が高く、腫瘍化のリスクが低い人工多能性幹細胞として選抜する。既存の方法によりゲノム構造を評価する、あるいは分化効率を測定することにより、選抜された人工多能性幹細胞がゲノム構造の安定性が高く、腫瘍化のリスクが低い人工多能性幹細胞であることを確認することができる。また、小核自然頻度の基準値を2%以下、1.9%以下、1.8%以下、1.7%以下、1.6%以下、1.5%以下、1.4%以下または1.3%として人工多能性幹細胞を選抜することで、ゲノム構造の安定性が高く、腫瘍化のリスクが低い人工多能性幹細胞を得ることができる。
本発明において選抜された人工多能性幹細胞を分化誘導する方法は限定されるものではなく、種々の公知の分化手法が利用できる。公知の分化手法としては、例えば、T細胞への分化手法、間葉系幹細胞への分化手法、造血幹細胞への分化手法、赤血球への分化手法、血小板への分化手法、血管内皮細胞への分化手法、心筋細胞への分化手法、骨格筋への分化手法、神経幹細胞への分化手法、大脳皮質神経細胞への分化手法、大脳辺縁系神経細胞への分化手法、ドパミン神経細胞への分化手法、肝細胞への分化手法、骨への分化手法、軟骨への分化手法、始原生殖細胞への分化手法、膵臓細胞への分化手法、網膜細胞への分化手法、角膜細胞への分化手法、腎臓細胞への分化手法、肺胞上皮細胞への分化手法、気管支上皮細胞への分化手法、腸管への分化手法等を挙げることができる。T細胞への分化手法としては、種々の公知の分化誘導方法を適宜選択して用いることができる。公知の分化誘導方法としては、例えば、「Timmermans, F. et al., J. Immunol., 2009, 182, 68879-6888」に記載のヒトES細胞からTリンパ球を誘導する方法が挙げられる。また、例えば、「Nishimura, T. et al. Cell Stem Cell, 2013, 12, 114-126」に記載の方法により、ヒト末梢血Tリンパ球より樹立したヒトiPS細胞をTリンパ球に分化誘導することができる。
(1)ヒトES細胞
京都大学再生医科学研究所より提供を受けた5種類のヒトES細胞株KhES-1、KhES-2、KhES-3、KhES-4、及びKhES-5について、小核自然頻度の測定を行った。ヒトES細胞は、「Ueno, M. et al. PNAS 2006, 103(25), 9554-9559」、「Watanabe, K. et al. Nat Biotech 2007, 25, 681-686」に記載の方法に倣い、マイトマイシンC処理したマウス線維芽細胞(リプロセル)上に播種し、37℃、2% CO2条件下で維持培養した。その際の培地には、DMEM/F12培地(Sigma-Aldrich)に、20% KnockOutTM Serum Replacement(KSR、Invitrogen)、0.1mM 非必須アミノ酸(NEAA;nonessential amino acids、Invitrogen)、2mM L-glutamine(Sigma-Aldrich)、及び0.1mM 2-メルカプトエタノール(Wako)が添加された培地(以下、hES培地と記す。)に、bFGF(Wako)(KhES-1では10ng/ml、KhES-2では30ng/ml、KhES-3では15ng/ml、KhES-4及びKhES-5では20ng/ml)を添加して用いた。KhES-2、KhES-3、KhES-4、及びKhES-5では、Y-27632(Wako)を10μMさらに上記培地に添加して用いた。維持培養開始1日後から毎日、Y-27632を含まないこと以外はそれぞれ同じ組成の培地への培地交換を行った。
(1)で得た細胞懸濁液を遠心分離(1000rpm、5分間)し、上清をリン酸緩衝生理食塩液(PBS、日水製薬)で置換した。得られた細胞懸濁液を再度遠心分離(1000rpm、5分間)して上清を除去し、37℃に加温した75mM KCl(Wako)を添加して5分間の低張処理を行った。低張処理後の細胞懸濁液に1/5容の固定液(メタノール(nacalai tesque):氷酢酸(nacalai tesque)=3:1)を添加し、遠心分離(4℃、1000rpm、5分間)しながら細胞を半固定した。遠心分離後、上清を新鮮な固定液に置換し、遠心分離(4℃、1000rpm、5分間)しながら細胞を固定した。上清を除去した後、懸濁液がわずかに白濁する程度の細胞密度となるようにメタノール中に再懸濁してスライドグラス上に滴下、風乾して小核標本とした。小核標本は100μg/mL アクリジンオレンジ(Wako)溶液で染色し、蛍光顕微鏡下で広帯域Blue励起のフィルターを用いて観察した。細胞株ごとに2枚のスライドグラスを観察に供し、スライドグラス1枚あたり1000個、すなわち細胞株あたり2000個のヒトES細胞中に認められる小核を有する細胞数を測定し、小核自然頻度を算出した。結果を表1に示した。表1より、小核自然頻度が最も高いものは2%であった。ES細胞は、腫瘍化リスクが低く安全であると考えられているため、この2%を基準値として採用することができる。
(1)ヒトiPS細胞
京都大学iPS細胞研究所金子研究室にて、「Nishimura, T. et al. Cell Stem Cell 2013, 12, 114-126」に記載の方法に従ってヒト末梢血リンパ球より樹立したヒトiPS細胞株「2」、「3」、「5」、「9」、「10」、「11」、及び「12」の、計7株のヒトiPS細胞について小核自然頻度および分化効率の測定を行った。
(1)に記載のヒトiPS細胞を、「Nishimura, T. et al. Cell Stem Cell 2013, 12, 114-126」に記載の方法に倣い、マイトマイシンC処理したマウス線維芽細胞(リプロセル)上に播種し、37℃、2% CO2条件下で維持培養した。その際の培地には、DMEM/F12培地(Sigma-Aldrich)に、20% KSR、2mM L-glutamine、1% NEAA、及び10μM 2-メルカプトエタノールが添加された培地に、bFGFを5ng/ml添加して用いた。
(1)に記載のヒトiPS細胞について、「Nishimura, T. et al. Cell Stem Cell 2013, 12, 114-126」に記載の方法に従ってT細胞系列への分化誘導を行った。
ヒトiPS細胞株「2」、「12」について、「Nishimura, T. et al. Cell Stem Cell 2013, 12, 114-126」に記載の方法に従ってCD8SP(Single Positive)となるT細胞への分化誘導を行った。得られたCD8SP T細胞(当該細胞を、以下、再分化T細胞と記す)を拡大培養した後、以下の方法に従って小核自然頻度を測定した。すなわち、再分化T細胞を1.0μg/mL 抗CD3抗体OKT3(eBioscience)でコートした12ウェルプレート(BD Falcon)上に1.0×106細胞/mLの細胞密度で播種し、増殖刺激を与えた。播種時の培地には、L-glutamine含有RPMI-1640培地(Wako)に、10% human serum AB(Nova Biologics)、1% Penicillin-Streptomycin(nacalai tesque)、100U/mL IL-2(Wako)、10ng/mL IL-7(Wako)、及び10ng/mL IL-15(R&D SYSTEMS)が添加された培地(以下、Rh培地と記す)を用いた。17時間の増殖刺激の後、再分化T細胞を全量回収し、細胞懸濁液90μLにRh培地60μLを加えて、OKT3でコートしていない96ウェルプレート(nunc)上に再播種した。再播種から約24時間後に、上清15μLを除去し、60μg/mL サイトカラシンB(Wako)を添加したRh培地15μLを加えた(サイトカラシンBの最終濃度:6μg/mL)。サイトカラシンB添加から33時間後、上清を除去し、PBSを添加して撹拌した。遠心分離(1500rpm、3分間)の後、再度上清をPBSで置換して撹拌した。再度遠心分離(1500rpm、3分間)して上清を除去し、37℃に加温した75mM KClを添加して5分間の低張処理を行った。低張後の細胞懸濁液に1/5容の固定液(メタノール:氷酢酸=3:1)を添加し、遠心分離(4℃、1500rpm、3分間)しながら細胞を半固定した。遠心分離後、上清を新鮮な固定液に置換し、遠心分離(4℃、1500rpm、3分間)しながら細胞を固定した。再度上清を新鮮な固定液に置換し、遠心分離(4℃、1500rpm、3分間)しながら細胞を固定した。更に上清をメタノールに置換し、遠心分離(4℃、1500rpm、3分間)しながら細胞を固定した。上清を除去した後、懸濁液がわずかに白濁する程度の細胞密度となるようにメタノール中に再懸濁してスライドグラス上に滴下、風乾して小核標本とした。小核標本は40μg/mL アクリジンオレンジ溶液で染色し、蛍光顕微鏡下で広帯域Blue励起のフィルターを用いて観察した。株「2」では1000個、株「12」では1500個の2核細胞を観察して小核を有する2核細胞数を測定し、総2核細胞に占める小核を有する2核細胞の割合を算出した。結果を表3に示した。表3には、実施例1で測定した小核自然頻度を付記した。
(1)ヒトiPS細胞
京都大学iPS細胞研究所金子研究室にて、「CytoTune(登録商標)-iPS 2.0」(iDファーマ)を用いてヒト末梢血リンパ球より樹立したヒトiPS細胞株「D-1R(-)#1」、「D-1R(-)#2」、「F-1R(-)#2」、「F-1R(+)#1」、及び「F-3R(+)#1」の、計5株のヒトiPS細胞について小核自然頻度の測定を行った。
また、上記5株に実施例1に記載のヒトiPS細胞7株を加えた、計12株のヒトiPS細胞について核形態の観察を、該12株から「F-1R(+)#1」を除いた、計11株のヒトiPS細胞について核型解析を、それぞれ行った。
(1)に記載の小核自然頻度測定に供した5株のヒトiPS細胞を、「フィーダーフリーでのヒトiPS細胞の樹立および維持培養(CiRA HPより)」に記載の方法に倣い、iMatrix(ニッピ)上に播種し、37℃、2% CO2条件下で維持培養した。その際の培地には、Stemfit AK03N(味の素)を用いた。
(2)で作製した5株のヒトiPS細胞の小核標本スライドグラス、および実施例1(2)で作製した7株のヒトiPS細胞の小核標本スライドグラスを、小核自然頻度の測定と同様にアクリジンオレンジ溶液で染色し、蛍光顕微鏡下で広帯域Blue励起のフィルターを用いて核形態を観察した。
[1]核膜の外縁が明瞭であり、1細胞につき1個の核を有する。
[2]核膜に損傷あるいは突起等の異常構造が認められず、滑らかな円形あるいは楕円形である。
(1)に記載の計11株のヒトiPS細胞について、(2)あるいは実施例1(2)と同様に維持培養を行った上で細胞を回収して細胞懸濁液とし、核型解析検体とした。核型解析は株式会社LSIメディエンスに委託して実施した。細胞懸濁液から分裂期の染色体標本を作製し、Gバンド法による分染を行って染色体を染色体番号ごとに分類した上でヒトiPS細胞株あたり20細胞について解析を行った。
(1)ヒトiPS細胞
「Nishimura, T. et al. Cell Stem Cell 2013, 12, 114-126」に記載の方法に従ってヒト末梢血リンパ球よりヒトiPS細胞株を樹立する。
(1)に記載のヒトiPS細胞を、「Nishimura, T. et al. Cell Stem Cell 2013, 12, 114-126」に記載の方法に倣い、マイトマイシンC処理したマウス線維芽細胞(リプロセル)上に播種し、37℃、2% CO2条件下で維持培養する。その際の培地には、DMEM/F12培地(Sigma-Aldrich)に、20% KSR、2mM L-glutamine、1% NEAA、及び10μM 2-メルカプトエタノールが添加された培地に、bFGFを5ng/ml添加して用いる。
小核自然頻度が1.3%以下である人工多能性幹細胞と、小核自然頻度が1.3%を超える人工多能性幹細胞とを識別する。そして、小核自然頻度が1.3%以下である人工多能性幹細胞を選抜することで、該ヒトiPS細胞株を、小核自然頻度が1.3%以下であるヒトiPS細胞として得る。
Claims (25)
- 下記(1)~(3)の工程を含むことを特徴とする人工多能性幹細胞の評価方法。
(1)培養された人工多能性幹細胞を提供する工程、
(2)提供された人工多能性幹細胞中の異常核酸構造を有する細胞小器官の自然出現頻度を測定する工程、及び
(3)前記自然出現頻度が基準値以下である人工多能性幹細胞と、前記自然出現頻度が基準値を超える人工多能性幹細胞とを識別する工程 - 前記異常核酸構造を有する細胞小器官が小核である、請求項1記載の評価方法。
- 前記基準値が胚性幹細胞における自然出現頻度の統計値である、請求項1又は2に記載の評価方法。
- 前記胚性幹細胞がヒト胚性幹細胞である、請求項3記載の評価方法。
- 前記自然出現頻度の基準値が、基準とする人工多能性幹細胞における自然出現頻度の統計値である、請求項1又は2に記載の評価方法。
- 前記自然出現頻度の基準値が2%である、請求項2記載の評価方法。
- 前記人工多能性幹細胞がヒト人工多能性幹細胞である、請求項1~6のいずれか一項記載の評価方法。
- 下記(1)~(4)の工程を含むことを特徴とする人工多能性幹細胞の選抜方法。
(1)培養された人工多能性幹細胞を提供する工程、
(2)提供された人工多能性幹細胞中の異常核酸構造を有する細胞小器官の自然出現頻度を測定する工程、
(3)前記自然出現頻度が基準値以下である人工多能性幹細胞と、前記自然出現頻度が基準値を超える人工多能性幹細胞とを識別する工程、及び
(4)前記自然出現頻度が基準値以下である人工多能性幹細胞を選抜する工程 - 前記異常核酸構造を有する細胞小器官が小核である、請求項8記載の選抜方法。
- 前記自然出現頻度の基準値が胚性幹細胞における自然出現頻度の統計値である、請求項8又は9に記載の選抜方法。
- 前記胚性幹細胞がヒト胚性幹細胞である、請求項10記載の選抜方法。
- 前記自然出現頻度の基準値が、基準とする人工多能性幹細胞における自然出現頻度の統計値である、請求項8又は9に記載の選抜方法。
- 前記自然出現頻度の基準値が2%である、請求項9記載の選抜方法。
- 前記人工多能性幹細胞がヒト人工多能性幹細胞である、請求項8~13のいずれか一項記載の選抜方法。
- 下記(1)~(4)の工程を含むことを特徴とする人工多能性幹細胞の製造方法。
(1)樹立され、培養された人工多能性幹細胞を提供する工程、
(2)提供された人工多能性幹細胞中の異常核酸構造を有する細胞小器官の自然出現頻度を測定する工程、
(3)前記自然出現頻度が基準値以下である人工多能性幹細胞と、前記自然出現頻度が基準値を超える人工多能性幹細胞とを識別する工程、及び
(4)前記自然出現頻度が基準値以下である人工多能性幹細胞を選抜する工程 - 前記(1)工程の前に、さらに下記工程を含むことを特徴とする請求項15記載の製造方法。
(A)人工多能性幹細胞を樹立する工程、及び
(B)樹立された人工多能性幹細胞を培養する工程 - 下記(1)~(5)の工程を含むことを特徴とする、人工多能性幹細胞由来分化細胞の製造方法。
(1)樹立され、培養された人工多能性幹細胞を提供する工程、
(2)提供された人工多能性幹細胞中の異常核酸構造を有する細胞小器官の自然出現頻度を測定する工程、
(3)前記自然出現頻度が基準値以下である人工多能性幹細胞と、前記自然出現頻度が基準値を超える人工多能性幹細胞とを識別する工程、
(4)前記自然出現頻度が基準値以下である人工多能性幹細胞を選抜する工程、及び
(5)(4)で選抜された人工多能性幹細胞を分化誘導し、分化細胞を得る工程 - 前記(1)工程の前に、さらに下記工程を含むことを特徴とする請求項17記載の製造方法。
(A)人工多能性幹細胞を樹立する工程、及び
(B)樹立された人工多能性幹細胞を培養する工程 - 前記異常核酸構造を有する細胞小器官が小核である、請求項15~18のいずれか一項に記載の製造方法。
- 前記自然出現頻度の基準値が胚性幹細胞における自然出現頻度の統計値である、請求項15~19のいずれか一項記載の製造方法。
- 前記胚性幹細胞がヒト胚性幹細胞である、請求項20記載の製造方法。
- 前記自然出現頻度の基準値が人工多能性幹細胞における自然出現頻度の統計値である、請求項15~19のいずれか一項記載の製造方法。
- 前記自然出現頻度の基準値が2%である、請求項19記載の製造方法。
- 前記人工多能性幹細胞がヒト人工多能性幹細胞である、請求項15~23のいずれか一項記載の製造方法。
- 下記(1)~(3)の工程を含むことを特徴とする、人工多能性幹細胞の評価方法。
(1)培養された人工多能性幹細胞を提供する工程、
(2)提供された人工多能性幹細胞中の異常核酸構造を有する細胞小器官の自然出現頻度を測定する工程、及び
(3)前記自然出現頻度が基準値以下である人工多能性幹細胞を、前記自然出現頻度が基準値を超える人工多能性幹細胞よりも分化効率が高い人工多能性幹細胞であると評価する工程
