本発明の核となる用語の解釈
本明細書で使用されるように、用語「抗体」または「免疫グロブリン」は、2つの同一の軽鎖(L)と2つの同一の重鎖(H)からなる、同じ構造特性を持つ約150000ダルトンのヘテロ四量体糖タンパク質である。各軽鎖は1つの共有ジスルフィド結合によって重鎖に接続され、異なる免疫グロブリンアイソタイプの重鎖間のジスルフィド結合の数は異なる。各重鎖および軽鎖には、規則的に間隔を置いた鎖内ジスルフィド結合もある。

本発明は、診断製剤を調製するため、またはTRAILR2関連疾患を予防および/または治療するための薬物を調製するための、本発明の抗体の用途をさらに提供する。前記TRAILR2関連疾患は、腫瘍の発生、成長および/または転移血栓関連疾患、炎症、代謝関連疾患などを含む。
(i)腫瘍の発生、成長および/または転移、特にTRAILR2陽性発現する固形腫瘍の診断、予防および/または治療。
前記腫瘍は、非小細胞肺癌、肝臓癌、結腸癌、乳癌、卵巣癌、膵臓癌、甲状腺癌、頭頸部扁平上皮癌、食道扁平上皮癌、肺扁平上皮癌、肺腺癌、子宮頸部の扁平上皮癌、膵扁平上皮癌、結腸扁平上皮癌、胃扁平上皮癌、前立腺癌、骨肉腫または軟部組織肉腫、より好ましくは非小細胞肺癌、肝癌、卵巣癌および膵臓癌などを含む(しかし、これらに限定されない)。
(ii)血液系の腫瘍の診断、予防および/または治療。前記腫瘍は、包括(しかし、これらに限定されない):リンパ性白血病、顆粒球性白血病、非T非Bリンパ性白血病、濾胞性リンパ腫、マントル細胞リンパ腫、バーキットリンパ腫、びまん性大細胞型B細胞リンパ腫、非ホジキン病、末梢T細胞リンパ腫、血管免疫芽球性T細胞リンパ腫、未分化大細胞リンパ腫などを含む(しかし、これらに限定されない)。より好ましくは、急性Tリンパ性白血病、Bリンパ性白血病、非T非Bリンパ性白血病である。
本発明は、組成物をさらに提供する。好ましい例では、前記組成物は、上記の抗体またはその活性断片またはそのは融合タンパク質またはそのADC、および薬学的に許容される担体を含む薬学的組成物である。一般に、これらの物質は、非毒性で不活性で薬学的に許容される水性担体媒体に配合することができ、pHは通常約5~8、好ましくは約6~8であるが、pHは調製される物質の性質および治療される疾患によって変化される。調製された薬学的組成物は、腫瘍内、腹腔内、静脈内、または局所投与を含むがこれらに限定されない従来の経路によって投与することができる。
(1)本発明のADCは、新しい、広域スペクトル、高効率、特異的に抗腫瘍性の抗TRAILR2抗体-毒素-複合体(ADC)であり、TRAILR2陽性腫瘍の治療に使用され、腫瘍の治療、特にリンパ性白血病、肝臓癌、肺癌、膵臓癌、卵巣癌などの治療に使用される。
(2)本発明のADCの治療用量が低く、薬物の安全性を大幅に改善する。
以下、添付の図面を参照し、例を挙げて本発明の実施例を参照して本発明をより詳細に説明するが、本発明はこれらに限定されない。以下の実施例に記載されている実験方法は、特に指定のない限り従来の方法であり、試薬および生物学的材料は、特に指定のない限り、商業チャネルから入手できる。
本発明によって解決される技術的問題は、以前の臨床試験から教訓を引き出し、腫瘍治療のための抗TRAILR2モノクローナル抗体の治療効果の問題をどのように解決するかである。この目的のために、本発明者らは、ADC複合体を調製する戦略を採用し、以下の一連の研究工程を経て、本発明のADC薬物候補を得た。すなわち:
(1)Zaptuzumabを発現するCHOオリジナル細胞バンク、マスター細胞バンク、生産細胞バンクを構築し、小規模プロセス研究を完了し、5リットルのバイオリアクターで3.5g/Lの発現レベルに達した。
(2)マウスの尾静脈から注射した131I標識Zaptuzumabは、肺癌移植腫瘍などのTRAILR2陽性腫瘍を特異的に標的とすることができる。
(3)免疫細胞化学技術を使用して、蛍光標識されたZaptuzumabは、肺癌などの腫瘍細胞の表面のTRAILR2受容体に結合した後、迅速にエンドサイトーシスされ、リソソームに入り、Zaptuzumabが抗体-毒素-複合体を調製する2つの重要な機能を有することを証明した。すなわち、腫瘍特異的ターゲティングと、腫瘍細胞によってエンドサイトーシスによってリソソームに取り込まれ、小分子毒素を放出。
(4)ジスルフィド結合架橋カップリング技術を使用して、Zaptuzumabは異なる化学リンカーを介して毒素にカップリングされ、異なる化学構造を持つさまざまなADCを得た。インビボoおよび/またはインビトロでの抗腫瘍活性のスクリーニングを繰り返した後、優れた薬効性を持つ2つのADC候補(それぞれZapadcine-1およびZapadcine-3と命名)を得、TRAILR2陽性のさまざまな腫瘍の治療に使用される。
実験動物
BALB/c雌ラットは上海Xipuer-Bikai実験動物有限会社(中国、上海)から購入し、SPF実験動物室で飼育し、NOD-SCID雌ラットは上海Lingchang生物化学有限会社(中国、上海)から購入し、SPF実験動物室で飼育し、12時間明暗サイクルを維持し、十分な食物と清潔な飲料水を提供し、8週齢で実験を開始した。すべての動物実験は承認され、上海市動物管理および使用委員会のガイダンスに従って実施した。
ヒトリンパ性白血病、肺癌細胞、肝臓癌細胞、卵巣癌細胞、膵臓癌細胞などの腫瘍細胞、正常細胞は、ATCCまたは中国医学科学院基礎医学研究所のセルセンター、または中国科学院上海生命科学研究院の細胞資源センター、または Saiqi(Shanghai)Bioengineering Co.、Ltd.またはCyberkon(Shanghai)Biotechnology Co.、Ltd.またはMiaotong(Shanghai)Biotechnology Co.、Ltd.から購入した。パクリタキセル、ビンクリスチンおよびエピルビシンはすべてSelleckから購入した。CellTiter-Glo発光細胞生存率検出キットは、Promegaから購入した。
さまざまな種類TRAILR2を高発現するリンパ性白血病細胞株(Jurkat E6-1)、肺癌細胞株(NCI-H460およびNCI-H1975)、肝癌細胞株(SMMC-7221)、卵巣癌細胞株(A2780)、および膵臓癌Mia PaCa-2およびヒト正常細胞株または末梢血細胞、例えばヒト正常末梢血単核細胞(PBMC)、ヒト正常結腸上皮細胞(NCM-460)、ヒト正常結腸組織細胞(CCD-18Co)およびヒト正常肺上皮細胞(BEAS-2B)を使用して、さまざまな腫瘍細胞および正常細胞に対するZapadcine-1の細胞毒性効果を評価した。具体的な研究プロセスは次のとおりである:トリプシン(0.25%、V/V)を使用して、付着培養細胞(NCI-H460やSMMC-7221など)を消化し、細胞を剥がし、および/または浮遊培養で細胞を直接収集し(Jurkat E6-1)、100μLの完全培地に再懸濁した。5000個の付着細胞または16000個の浮遊細胞を96ウェルプレートに接種し、37℃で一晩培養した。次に、異なる濃度の抗TRAILR2裸抗体またはZapadcine-1を含む100μLの培地を追加し、72時間インキュベーターに入れてから、CellTiter-Gloルシフェラーゼ活性検出キット(Promega、ロット番号:0000217738)を使用して、インビトロで培養したさまざまな腫瘍細胞に対する試験薬物の細胞毒性を測定した。
ELISA法を使用して、Zapadcine-1aとヒト化組換えタンパク質TRAILR2の結合状況を評価し、具体的なプロセスは次のとおりである:1×PBSバッファー(pH 7.4)で100μl/ウェルの容量で2μg/mlのヒト化組換えタンパク質TRAILR2を96ウェルプレートにコーティングし、4℃で一晩放置した。上清を捨て、プレートをPBST(0.05%Tweeen20を含むpH 7.4 PBS)バッファーで3回、各回5分間洗浄し、240μl/ウェルごとに5%脱脂粉乳を含むPBSを加え、ブロッキングのために37℃で3時間インキュベートした。ブロッキング溶液を捨て、プレートを300μl /ウェルのPBSTで3回、各回5分間洗浄し、50μl/ウェルごとに1%スキムミルク粉末またはBSAを含むPBSで希釈したテストする抗体(一次抗体)またはADCを添加し、濃度は 4μg/mlで2倍勾配希釈を開始し、合計12の濃度、3つのウェルを複製し、室温で1時間インキュベートした。上清を捨て、プレートをPBSTで3回、各5分間洗浄し、50μl/ウェルごとに1%スキムミルク粉末またはBSAを含むPBSで希釈した二次抗体を加え、濃度は1μg/ml であり、37℃で40分間インキュベートした。上清を捨て、プレートをPBSTで3回、各回5分間洗浄し、50μl/ウェルごとにTMBカラー現像液を添加し、室温で5~10分間インキュベートした。発色効果に従い、50μl/ウェルの1M H2SO4を添加して反応を停止した。SPARK 10M多機能マイクロプレート検出器で450nmでOD値を読み取った。GraphPad Prism 5.0ソフトウェアを使用してデータ分析した。具体的な結果を図4に示したとおりである。
Zapadcine-1aのインビボ抗リンパ性白血病効果は、NOD-SCIDマウスでのTRAILR2の高発現のヒトJurkat E6-1白血病細胞に対するその成長阻害効果によって評価した。具体的な研究プロセスは次のとおりである:ヒトTリンパ性白血病Jurkat E6-1の雌NOD/SCIDマウス(4週齢)皮下異種移植腫瘍モデルを構築した。Jurkat E6-1細胞をインビトロで培養し、対数増殖期まで拡大し、細胞数を数えた後、PBSバッファーで1×108/mlの細胞懸濁液に希釈して、超クリーンベンチで1 mlのシリンジを使用して、0.1 ml(1×107細胞)を吸引し、マウスの右背中の皮膚の下に接種した。接種部位の皮膚を70%アルコールで消毒し、接種後、定期的に動物の生存状態を観察し、移植された腫瘍の成長を測定して記録した。平均腫瘍サイズが80-100 mm3に達したとき、担腫瘍マウスをランダムに5つのグループに分け、それぞれ8匹のマウス、それぞれZapadcine-1a投与群(低用量群1 mg/kg、中用量群3 mg/kg、高用量群9 mg/kg、Q3D×3)、陰性対照群(PBS、Q3D×3)およびビンクリスチン陽性対照群(0.5 mg/kg、Q7D×3)である。Zapadcine-1aの投与時間は群分け後0、4、7日であり、ビンクリスチンの投与時間は群分け後0、7、14日であり、上記投与方法は全て尾静脈注射である。投与後3、7、10、14、17、21、24、28日目に腫瘍サイズ(最長径、最短径)を測定した。実験は最初の投与から28日後に終了し、動物は過剰摂取麻酔によって直ちに安楽死させ、マウスの体重を量った。腫瘍を取り出して重量を測定し、写真を撮った。血液サンプルを採取して肝臓と腎臓の機能指数(ALT、BUN、CERA)を検出した。腫瘍と主要組織および臓器(心臓、肝臓、腎臓、脾臓、肺)を分離し、液体窒素で凍結保存するか、4%パラホルムアルデヒドで固定し、後で使用するためにパラフィン切片を準備した。

具体的な結果は図7に示したとおりである。その結果、Zapadcine-1aの3回投与後7日目に、高、中、低用量群の肺癌移植腫瘍の増殖を抑制し始めたことを示す。
Zapadcine-1aのインビボ抗肺癌特性は、ヌードマウスインビボでのTRAILR2の陽性発現のヒトSMMC-7721肝癌細胞の増殖阻害効果によって評価した。具体的な研究プロセスは次のとおりである:ヒトSMMC-7721肝癌細胞の雌BALB/cヌードマウス(4週齢)皮下異種移植腫瘍モデルを構築し、SMMC-7721細胞をインビトロで培養し、対数増殖期まで拡大し、細胞数を数えた後、PBSバッファーで1×107/mlの細胞懸濁液に希釈して、超クリーンベンチで1 mlのシリンジを使用して、0.15ml(1.5×106細胞)を吸引し、マウスの右背中の皮膚の下に接種した。接種部位の皮膚を70%アルコールで消毒し、接種後、定期的に動物の生存状態を観察し、移植された腫瘍の成長を測定して記録した。腫瘍サイズが80-100 mm3に達したとき、担腫マウスをランダムに5つのグループに分け、それぞれ8匹のマウス、それぞれZapadcine-1a投与群(低用量群1 mg/kg、中用量群3 mg/kg、高用量群9 mg/kg、Q3D×3)、陰性対照群(PBS、Q3D×3)およびエピルビシン陽性対照群(5 mg/kg、Q3D×9)である。Zapadcine-1aおよび陰性対照群の投与時間はグループ化後0、4、7日であるが、エピルビシンの投与時間はグループ化後0、4、7、10、14、17、21日であり、上記投与方法は全て尾静脈注射である。投与後4、7、10、14、17、21、24、28日目に腫瘍サイズ(最長径、最短径)を測定した。実験は最初の投与から28日後に終了し、動物は過剰摂取麻酔によって直ちに安楽死させ、マウスの体重を量った。腫瘍を取り出して重量を測定し、写真を撮った。血液サンプルを採取して肝臓と腎臓の機能指数(ALT、BUN、CERA)を検出した。腫瘍と主要組織および臓器(心臓、肝臓、腎臓、脾臓、肺)を分離し、液体窒素で凍結保存するか、4%パラホルムアルデヒドで固定し、後で使用するためにパラフィン切片を準備した。

Zapadcine-1の急性毒性効果は、投与後の正常マウスの体調と生化学的指標の変化を調査することによって評価した。具体的な研究プロセスは次のとおりである:20匹の健康なBALB/c雌マウス(上海Xipuer-Bikai実験動物有限会社から購入)を取り、マウスは6~7週齢で、体重は18~22 gである。5つのグループに分け、各グループ4匹である。定期的な給餌の1週間後、投与し始めた。Zapadcine-1aの用量は、20 mg/kg、30 mg/kg、40 mg/kgおよび50 mg/kgで、正常対照としてブランク溶媒を使用した。尾静脈からゆっくりと単回投与し、死亡、食事、運動を含めて毎日マウスを観察し、3日ごとにマウスの体重を記録し、5日目と15日目に血液生化学検査のために血液サンプルを採取し、検査項目はALTとUREAを含む。投与15日目に、過量麻酔により安楽死させた。
具体的な結果は図11に示したとおりである。その結果、対照群と比較して、Zapadcine-1a 20 mg/kg、30 mg/kg、40 mg/kgの3つの用量群のマウスは正常に成長し、よく運動し、正常な体重変化を示し、正常な肝臓と腎臓の機能を示し、死ななかった。Zapadcine-1a 50 mg/kg用量群では、マウスの死亡率は50%に達し、マウスの体重は初期段階で減少し、マウスの成長は後期段階で正常に戻り、マウスの体重変化は正常であり、マウスの肝臓と腎臓の機能は正常であり、これは、Zapadcine-1aの単回投与に対するマウスの最大耐量が40~50 mg/kgであり、安全であることを示す。
さまざまな種類のTRAILR2高発見のリンパ性白血病細胞株(Jurkat E6-1、Reh)、肺癌細胞株(MSTO-211H)、神経膠腫細胞株(A172)、膵臓癌Mia PaCa-2、およびヒト正常細胞株または末梢血細胞、例えばヒト正常末梢血単核細胞(PBMC)、ヒト正常結腸上皮細胞(NCM-460)、ヒト正常結腸組織細胞(CCD-18Co)、ヒト正常肺上皮細胞(BEAS-2B)を使用して、さまざまな腫瘍細胞および正常細胞に対するZapadcine-3の細胞毒性効果を評価した。具体的な研究プロセスは次のとおりである:トリプシン(0.25%、V/V)を使用して、付着培養細胞(MSTO-211Hなど)を消化し、細胞を剥がし、および/または浮遊培養で細胞を直接収集し(Jurkat E6-1、Reh)、100μLの完全培地に再懸濁した。5000個の付着細胞または16000個の浮遊細胞を96ウェルプレートに接種し、37℃で一晩培養した。次に、異なる濃度の抗TRAILR2裸抗体またはZapadcine-3を含む100μLの培地を追加し、72時間インキュベーターに入れてから、CellTiter-Gloルシフェラーゼ活性検出キット(Promega、ロット番号:0000217738)を使用して、インビトロで培養したさまざまな腫瘍細胞に対する試験薬物の細胞毒性を測定した。
ELISA法を使用して、Zapadcine-3aとヒト化組換えタンパク質TRAILR2の結合状況を評価し、具体的なプロセスは次のとおりである:1×PBSバッファー(pH 7.4)で100μl/ウェルの容量で2μg/mlのヒト化組換えタンパク質TRAILR2を96ウェルプレートにコーティングし、4℃で一晩放置した。上清を捨て、プレートをPBST(0.05%Tweeen20を含むpH 7.4 PBS)バッファーで3回、各回5分間洗浄し、240μl/ウェルごとに5%脱脂粉乳を含むPBSを加え、ブロッキングのために37℃で3時間インキュベートした。ブロッキング溶液を捨て、プレートを300μl/ウェルのPBSTで3回、各回5分間洗浄し、50μl/ウェルごとに1%スキムミルク粉末またはBSAを含むPBSで希釈したテストする抗体(一次抗体)またはADCを添加し、濃度は4μg/mlで2倍勾配希釈を開始し、合計12の濃度、3つのウェルを複製し、室温で1時間インキュベートした。上清を捨て、プレートをPBSTで3回、各5分間洗浄し、50μl/ウェルごとに1%スキムミルク粉末またはBSAを含むPBSで希釈した二次抗体を加え、濃度は1μg/mlであり、37℃で40分間インキュベートした。上清を捨て、プレートをPBSTで3回、各回5分間洗浄し、50μl/ウェルごとにTMBカラー現像液を添加し、室温で5~10分間インキュベートした。発色効果に従い、50μl/ウェルの1M H2SO4を添加して反応を停止した。SPARK 10M多機能マイクロプレート検出器で450nmでOD値を読み取った。GraphPad Prism 5.0ソフトウェアを使用してデータ分析した。具体的な結果を図12に示したとおりである。
