Description
更に、再生医療において用いられる多能性幹細胞(例えば、iPS細胞(induced Pluripotent Stem Cell))の作製において、p53遺伝子の発現を抑制する必要があることが知られている。
このため、種々の疾患の治療及び再生医療において、p53タンパク質の量(発現)を減少させる方法論が検討されている。
このため、癌治療において、変異型p53タンパク質の量(発現)を減少させる方法論が検討されている。
(1)ユビキチンリガーゼには多数の種類があり、標的特異性がある。このため、特定の標的タンパク質をユビキチン化するためには、例えば、標的タンパク質に合わせて分子を設計するなど、個々に対応する必要がある。
(2)ユビキチン化シグナルのコントロールが困難である。例えば、タンパク質のユビキチン化は、タンパク質の分解以外に、分化、癌化等のシグナルに関連することが知られている。また、タンパク質のユビキチン化には、組織特異性及び時間特異性があることが知られている。このため、標的タンパク質のユビキチン化が、標的タンパク質の分解ではなく、その他のシグナルとなるケースがあると推定される。
(3)ユビキチン又はユビキチン化酵素に不具合があるケースがある。例えば、変異等により、ユビキチンやユビキチン化酵素が正常に機能しない(マルファンクションとなっている)場合があり、それが疾患の原因である場合も多い。このため、標的タンパク質のユビキチン化が、標的タンパク質の分解を誘導しないケースがあると推定される。
<1> p53タンパク質又はp53複合体に対して親和性を有するp53親和性分子と、プロテアーゼに対して親和性を有し、且つ、プロテアーゼによるタンパク質の分解を阻害しないタンパク質分解誘導タグとのコンジュゲートであり、前記p53タンパク質又はp53複合体の分解を誘導可能なp53分解誘導分子。

本明細書においてアミノ酸は、本技術分野で周知の一文字表記(例えば、グリシンであれば「G」)又は三文字表記(例えば、グリシンであれば「Gly」)で表記する。
本開示のp53分解誘導分子は、p53タンパク質又はp53複合体に対して親和性を有するp53親和性分子と、プロテアーゼに対して親和性を有し、且つ、プロテアーゼによるタンパク質の分解を阻害しないタンパク質分解誘導タグとのコンジュゲートであり、p53タンパク質又はp53複合体の分解を誘導可能である。本開示のp53分解誘導分子によれば、p53タンパク質又はp53複合体のユビキチン化を介することなく(すなわち、ユビキチン非依存的に)、p53タンパク質又はp53複合体をプロテアーゼ(例えば、プロテアソーム)による分解(ノックダウン)へと導くことが可能となる。
本開示のp53分解誘導分子を構成するp53親和性分子は、p53タンパク質又はp53複合体に対して親和性を有する分子である。
また、心機能障害、糖尿病等の疾患において、心組織、膵臓組織等におけるp53タンパク質の発現量が増加していることが知られており、これらの疾患は、p53タンパク質に対する阻害剤の投与により改善することが知られている。
また、p53タンパク質は、老化因子として、生活習慣病に起因する動脈硬化、代謝異常等との関連も指摘されている。
更に、再生医療において用いられる多能性幹細胞(例えば、iPS細胞)の作製において、p53遺伝子の発現を抑制する必要があることが知られている。
本開示のp53分解誘導分子を構成するタンパク質分解誘導タグは、プロテアーゼに対して親和性を有し、且つ、プロテアーゼによるタンパク質の分解を阻害しない分子である。以下では、このタンパク質分解誘導タグをCiKD(Chemical interactions and KnockDown)タグ又はCANDDY(Chemical AffiNities and Degradation DYnamics)タグとも称する。
26Sプロテアソームは、20Sプロテアソームに19Sプロテアソームが2つ結合したものである。20Sプロテアソームは、α1~α7の7つのサブユニットから構成されるαリングと、β1~β7の7つのサブユニットから構成されるβリングとが、αββαの順に積み重なった筒状構造をしており、β1、β2、及びβ5がそれぞれカスパーゼ様活性、トリプシン様活性、及びキモトリプシン様活性という触媒活性を発揮する。
免疫プロテアソームは、触媒サブユニットβ1、β2、及びβ5がそれぞれβ1i、β2i、及びβ5iに置き換わったものである(Science, 1994, 265, 1234-1237)。
胸腺プロテアソームは、β1i及びβ2iとともに、胸腺皮質上皮細胞(cTEC)特異的に発現するβ5tが組み込まれたものである(Science, 2007, 316, 1349-1353)。
ハロゲノ基としては、フルオロ基、クロロ基、ブロモ基等が挙げられる。
プロテアソーム阻害剤は、抗癌剤等として研究が進んでおり、医薬品として認可済みの化合物及び臨床試験中の化合物が数多く存在する。また、プロテアソーム阻害剤は、分子量が比較的小さく、疎水性が低いものが多く、細胞膜透過性、細胞毒性等の問題も生じ難い。このため、プロテアソーム阻害剤を基にタンパク質分解誘導タグを合成することは、非常に合理的且つ効率的である。
p53親和性分子とタンパク質分解誘導タグとのコンジュゲートの様式は、p53親和性分子のp53タンパク質又はp53複合体との親和性、及びタンパク質分解誘導タグのプロテアーゼとの親和性が維持される限り、特に制限されない。なお、p53親和性分子及びタンパク質分解誘導タグがいずれもタンパク質である場合、両者を融合した融合タンパク質を合成することが可能であるが、このような融合タンパク質は「コンジュゲート」には含まれない。
一方、タンパク質分解誘導タグにおけるp53親和性分子との連結位置は、プロテアーゼとの親和性が維持される限り、特に制限されない。例えば、タンパク質分解誘導タグが、前述のように、プロテアーゼ阻害剤(例えば、プロテアソーム阻害剤)の活性部位を他の構造部分に置き換えた構造である場合、この置き換えた他の構造部分においてp53親和性分子と連結することができる。具体的に、プロテアーゼ阻害剤の活性部位をカルボキシ基に置き換えた場合、カルボキシ基を介してp53親和性分子と連結することができる。
例えば、p53親和性分子としては、p53タンパク質又はp53複合体に親和性を有する既知の分子を使用することができるが、この既知の分子とタンパク質分解誘導タグとを直接連結することが困難な場合も想定される。このような場合には、タンパク質分解誘導タグと連結可能な構造を当該既知の分子に導入し、p53親和性分子として使用してもよい。
本開示の医薬組成物は、本開示のp53分解誘導分子を含むものである。前述したとおり、本開示のp53分解誘導分子によれば、p53タンパク質又はp53複合体のユビキチン化を介することなく(すなわち、ユビキチン非依存的に)、p53タンパク質又はp53複合体をプロテアーゼ(例えば、プロテアソーム)による分解(ノックダウン)へと導くことが可能となる。したがって、p53分解誘導分子を含む本開示の医薬組成物は、p53タンパク質が仲介する疾患又は症状の予防又は治療に用いることができる。本開示によれば、p53分解誘導分子を含む医薬組成物を投与することを含む、p53タンパク質が仲介する疾患又は症状の予防又は治療方法もまた提供される。
また、癌の放射線治療時に野生型p53タンパク質の機能を回復することにより、治療効果が改善することが知られている。放射線治療と本開示の医薬組成物を用いた治療とを併用することにより、変異型p53タンパク質を有する癌における放射線治療の効果が改善することも期待できる。

下記合成スキームに従ってCANDDY_MLNを合成した。
下記合成スキームに従ってTIBC-CANDDY_MLNを合成した。
TIBC-CANDDY_MLNの物性データを以下に示す。

実施例2では、TIBC-CANDDY_MLNを添加したHCT116細胞(ヒト大腸癌細胞)における内在の野生型p53タンパク質及びMDM2タンパク質の分解(ノックダウン)について、ウェスタンブロット解析により評価した。
HCT116細胞を8×104個/ウェルの細胞密度で24ウェルプレートに播種した後、37℃、5体積% CO2の条件下で16時間培養した。
細胞播種から16時間後に、以下のようにして、HCT116細胞にTIBC-CANDDY_MLN又はTIBCを添加した。培地としては、D-MEM(High D-Glucose, Phenol Red, Sodium Pyruvate)(和光純薬工業(株))に1質量% L-グルタミン溶液(Sigma-Aldrich)を添加した無血清培地(37℃)を用いた。なお、L-グルタミン溶液は使用直前に添加した。TIBC-CANDDY_MLN又はTIBCを含有するDMSO溶液をDMSO濃度が1体積%となるように培地と混合し、各ウェル当たり500μL添加して、37℃、5体積% CO2の条件下で培養した。コントロールとしてはDMSOを用いた。
TIBC-CANDDY_MLN又はTIBCの添加48時間後、培地を除去し、PBSを添加して細胞を洗浄した。PBSの除去後、細胞溶解バッファー(CelLytic M, Sigma)とプロテアーゼ阻害剤(cOmplete Mini, EDTA-free, Roche)との混合溶液を各ウェル当たり27μL添加した。4℃にて15分間静置した後、ピペットチップを用いて氷上で細胞を剥がした。細胞溶液を1.5mLチューブに回収し、液体窒素で瞬間凍結した後、氷上で融解させた。融解後、遠心分離(13800rpm×20分間、4℃)し、上清(細胞抽出物)を回収した。
実施例3では、TIBC-CANDDY_MLNを添加したHeLa細胞(ヒト子宮頸癌細胞)における内在の野生型p53タンパク質の分解(ノックダウン)について、ウェスタンブロット解析により評価した。同時に、プロテアソーム阻害剤(MLN2238)によるp53タンパク質分解のレスキューを評価した。
HeLa細胞を4×104個/ウェルの細胞密度で24ウェルプレートに播種した後、37℃、5体積% CO2の条件下で16時間培養した。
細胞播種から16時間後に、以下のようにして、HeLa細胞にTIBC-CANDDY_MLNを添加した。培地としては、D-MEM(High D-Glucose, Phenol Red, Sodium Pyruvate)(和光純薬工業(株))に1質量% L-グルタミン溶液(Sigma-Aldrich)を添加した無血清培地(37℃)を用いた。なお、L-グルタミン溶液は使用直前に添加した。TIBC-CANDDY_MLNを含有するDMSO溶液をDMSO濃度が1体積%となるように培地と混合し、各ウェル当たり500μL添加して、37℃、5体積% CO2の条件下で培養した。コントロールとしてはDMSOを用いた。また、TIBC-CANDDY_MLNを含有するDMSO溶液を添加する実験群のほかに、TIBC-CANDDY_MLN及びMLN2238の両方、又はMLN2238を含有するDMSO溶液を添加する実験群も準備した。
TIBC-CANDDY_MLN又はMLN2238の添加24時間後、培地を除去し、PBSを添加して細胞を洗浄した。PBSの除去後、細胞溶解バッファー(CelLytic M, Sigma)とプロテアーゼ阻害剤(cOmplete Mini, EDTA-free, Roche)との混合溶液を各ウェル当たり27μL添加した。4℃にて15分間静置した後、ピペットチップを用いて氷上で細胞を剥がした。細胞溶液を1.5mLチューブに回収し、液体窒素で瞬間凍結した後、氷上で融解させた。融解後、遠心分離(13800rpm×20分間、4℃)し、上清(細胞抽出物)を回収した。
実施例4では、TIBC-CANDDY_MLNを投与したマウス個体における内在の野生型p53タンパク質及びMDM2タンパク質の分解(ノックダウン)について、ウェスタンブロット解析により評価した。
投与の直前に、TIBC-CANDDY_MLNをDMSOに溶解した後、DMSOの濃度が10体積%となるようにトウモロコシ油(Code No. 25606-55, Nacalai Tesque)に溶解し、50mg/kg体重又は100mg/kg体重の投与量で、C57BL/6J野生型マウス(7~8週齢、雄)(日本クレア(株))に腹腔内投与した(n=3)。コントロールとしては、注射液の担体(10体積% DMSOを含有するトウモロコシ油)を用いた。マウスは、餌及び水が自由摂取できる環境下で飼育した。投与48時間後に、ソムノペンチル(共立製薬(株))による深麻酔下でマウスを解剖した。開腹してから肝臓を摘出し、液体窒素で瞬間凍結させた。液体窒素で凍結後の組織は、-80℃のディープフリーザーにて保管した。
凍結した組織(0.04g)を粉砕した後、980μLの1×TKM組織溶解バッファー(50 mM トリエタノールアミン(pH 7.8), 50 mM KCl, 5 mM MgCl2, 0.25 M sucrose, 1 mM PMSF, protein inhibitors cocktail-EDTA free(Code No.03969-21, Nacalai Tesque), 1 mM DTT,Recombinant RNase inhibitor 5ul/ml (40 U/μl, Cat No. 2313A, Lot No. K8402DA, TAKARA Bio))を加え、15分間回転(1rpm、25℃)して溶解させた。その後、遠心分離(3000rpm×15分間、4℃)し、上清(組織抽出物)を回収した。組織抽出物をDEPC処理水により20倍希釈したものを用いて、分光光度計で組織抽出物におけるタンパク質濃度を定量した。
実施例5では、TIBC-CANDDY_MLNを添加した老化関連酸性β-ガラクトシダーゼ(SA-β-gal)誘導TIG3細胞(ヒト胎児繊維芽細胞)における抗老化作用について、FACS解析により評価した。
正常細胞であるTIG3細胞を8×104個/ウェルの細胞密度で24ウェルプレートに播種した後、37℃、5体積% CO2の条件下で16時間培養した。
細胞播種から16時間後に、ドキソルビシンを各ウェル当たり150nMとなるように添加し、細胞老化を誘導した。培地としては、D-MEM(High D-Glucose, Phenol Red, Sodium Pyruvate)(和光純薬工業(株))に1質量% L-グルタミン溶液(Sigma-Aldrich)を添加した無血清培地(37℃)を用いた。なお、L-グルタミン溶液は使用直前に添加した。ドキソルビシンの添加後、37℃、5体積% CO2の条件下で24時間培養した。
老化誘導から24時間後に、以下のようにして、TIG3細胞にTIBC-CANDDY_MLNを添加した。培地としては、D-MEM(High D-Glucose, Phenol Red, Sodium Pyruvate)(和光純薬工業(株))に1質量% L-グルタミン溶液(Sigma-Aldrich)を添加した無血清培地(37℃)を用いた。なお、L-グルタミン溶液は使用直前に添加した。TIBC-CANDDY_MLNを含有するDMSO溶液をDMSO濃度が1体積%となるように培地と混合し、各ウェル当たり500μL添加して、37℃、5体積% CO2の条件下で48時間培養した。コントロールとしてはDMSOを用いた。
老化マーカーであるSA-β-galの定量には、市販のキット(Cellular Senescence Detection Kit-SPiDER-βGal((株)同仁化学研究所))を用いた。
参考例1では、タンパク質親和性分子とタンパク質分解誘導タグとを連結することにより、タンパク質分解誘導分子であるTMP-CANDDY_DMTを合成した。
また、タンパク質分解誘導タグとしては、前述した式(I)においてR1及びR2をいずれもメトキシ基とした化合物(DMT)を用いた。DMTは、プロテアソーム阻害剤に由来しないものの、プロテアソームに対して親和性を有する化合物である。
参考例2では、TMP-CANDDY_DMTのプロテアソーム阻害活性及びプロテアソームとの親和性を評価した。ポジティブコントロールとしては、プロテアソーム阻害剤であるMG-132を用いた。
図5A~図5Cに示すとおり、TMP-CANDDY_DMTは、MG-132と比較して、プロテアソーム阻害活性が著しく低いことが確認できた。また、β1、β2、及びβ5のいずれに対しても、TMP-CANDDY_DMTの濃度依存的に阻害活性が高まることから、TMP-CANDDY_DMTがプロテアソームと穏やかな親和性を有していることが示唆された。すなわち、DMTは、プロテアソームと親和性を有するものの、分解を阻害しないと評価された。
参考例3では、HeLa細胞において強制発現させたecDHFRタンパク質のTMP-CANDDY_DMTを介した分解(ノックダウン)について、FACS解析により評価した。
ecDHFRタンパク質を発現するプラスミド(pMIR-DsRed-IRES-ecDHFR-HA-GFP)を大腸菌で増幅した後、Miniprep Kit(QIAGEN)で精製した。
HeLa細胞にプラスミドを導入することにより、細胞内に標的タンパク質であるecDHFRタンパク質(詳細には、HAタグを介したecDHFRタンパク質とGFPとの融合タンパク質)又は比較用のDsRedタンパク質を一過的に過剰発現させた。
プラスミドを導入してから40時間培養した後、以下のようにして、HeLa細胞にTMP-CANDDY_DMTを添加した。培地としては、D-MEM(High D-Glucose, Phenol Red, Sodium Pyruvate)(和光純薬工業(株))に1質量% L-グルタミン溶液(Sigma-Aldrich)を添加した無血清培地(37℃)を用い、各ウェル当たり297μL添加した。なお、L-グルタミン溶液は使用直前に添加した。TMP-CANDDY_DMTを含有するDMSO溶液を各ウェル当たり3μL添加して、37℃、5体積% CO2の条件下で培養した。コントロールとしては、TMPを含有するDMSO溶液又はDMSOを用いた。
TMP-CANDDY_DMTの添加24時間後、培地を除去し、PBSを添加して細胞を洗浄した。PBSの除去後、37℃のトリプシン(0.25 w/v% Trypsin-1 mmol/L EDTA・4Na Solution with Phenol Red)(和光純薬工業(株))を各ウェル当たり300μL添加し、37℃、5体積% CO2の条件下で1分間培養した。培養後、D-MEM(Low D-Glucose, L-Glutamine, Phenol Red)(和光純薬工業(株))に10質量% FBS及び1質量% PenStrep(100 U/mL Sodium Penicillin G and 100 μg/mL Streptomycin Sulfate)(和光純薬工業(株))を添加した培地を各ウェル当たり500μL添加して懸濁し、15mLチューブに細胞溶液を回収した。
参考例4では、HeLa細胞において強制発現させたecDHFRタンパク質のTMP-CANDDY_DMTを介した分解(ノックダウン)について、ウェスタンブロット解析により評価した。
参考例3と同様にして、ecDHFRタンパク質を発現するプラスミドを作製した。
参考例3と同様にして、HeLa細胞にプラスミドを導入することにより、細胞内にecDHFRタンパク質又は比較用のDsRedタンパク質を一過的に過剰発現させた。プラスミド導入後のHeLa細胞は、4×104個/ウェルの細胞密度で24ウェルプレートに播種した後、37℃、5体積% CO2の条件下で40時間培養した。
プラスミドを導入してから40時間培養した後、以下のようにして、HeLa細胞にTMP-CANDDY_DMTを添加した。培地としては、D-MEM(High D-Glucose, Phenol Red, Sodium Pyruvate)(和光純薬工業(株))に1質量% L-グルタミン溶液(Sigma-Aldrich)を添加した無血清培地(37℃)を用いた。なお、L-グルタミン溶液は使用直前に添加した。TMP-CANDDY_DMTを含有するDMSO溶液をDMSO濃度が1体積%となるように培地と混合し、各ウェル当たり300μL添加して、37℃、5体積% CO2の条件下で培養した。また、TMP-CANDDY_DMTを含有するDMSO溶液を添加する実験群のほかに、TMP-CANDDY_DMT及びボルテゾミブ(Bortezomib)を含有するDMSO溶液を添加する実験群も準備した。TMP-CANDDY_DMTの添加12時間後に、タンパク質合成阻害剤であるシクロへキシミドを50μg/mLの濃度となるように培地中に添加した。なお、コントロールとしては、TMPを含有するDMSO溶液又はDMSOを用いた。
TMP-CANDDY_DMTの添加24時間後、培地を除去し、PBSを添加して細胞を洗浄した。PBSの除去後、細胞溶解バッファー(CelLytic M, Sigma)とプロテアーゼ阻害剤(cOmplete Mini, EDTA-free, Roche)との混合溶液を各ウェル当たり55μL添加した。4℃にて15分間静置した後、ピペットチップを用いて氷上で細胞を剥がした。細胞溶液を1.5mLチューブに回収し、液体窒素で瞬間凍結した後、氷上で融解させた。凍結融解を3回繰り返した後、遠心分離(13000rpm×20分間、4℃)し、上清(細胞抽出物)を回収した。
参考例5では、タンパク質親和性分子とタンパク質分解誘導タグとを連結することにより、タンパク質分解誘導分子であるTMP-CANDDY_ALLNを合成した。
また、タンパク質分解誘導タグとしては、プロテアソーム阻害剤であるALLNの活性部位(ホルミル基)をカルボキシ基に置き換えた化合物(CANDDY_ALLN)を用いた。
ナスフラスコにALLN(87.2 mg, 0.23 mmol, 1 eq, Code No. 07036-24, ナカライテスク(株))を仕込み、脱水DMFを2mL加えた。室温にて5分間撹拌した後、Oxone(212.1 mg, 0.69 mmol, 3 eq, Code No. 228036, Sigma-Aldrich)を反応溶液に直接加え、室温にて5時間撹拌した。反応溶液を水で希釈した後、クロロホルムで3回抽出した。無水硫酸ナトリウムで乾燥させた後、溶媒を減圧留去した。シリカゲルクロマトグラフィー(Code No. 30511-35, ナカライテスク(株))(クロロホルム/メタノール=20/1~10/1, gradient)を用いた分離精製処理により、CANDDY_ALLN(27.0 mg, 0.068 mmol, 30%)を得た。
ナスフラスコにCANDDY_ALLN(26.8 mg, 0.067 mmol, 1 eq)及び別途合成したTMP-NH2(Long, M.J. et al., Chem. Biol., 2012, 19(5), 629-637)(26.0 mg, 0.060 mmol, 0.9 eq)を仕込み、脱水DMFを2mL加えた。室温にて5分間撹拌した後、DIPEAを0.1mL加え、溶液を中性にした。室温にて5分間撹拌した後、DMT-MM(30.0 mg, 0.11 mmol, 1.6 eq, Code No. 329-53751, 和光純薬工業(株))を反応溶液に直接加え、室温にて2時間撹拌した。冷却下で10mLの10質量%食塩水/0.1N 塩酸水溶液を加え、酢酸エチルで3回抽出した。0.5N 塩酸水溶液、次いで食塩水で洗浄した後、無水硫酸ナトリウムで乾燥させた。溶媒を減圧留去した後、シリカゲルクロマトグラフィー(Code No. 30511-35, ナカライテスク(株))(クロロホルム/メタノール=10/1)を用いた分離精製処理により、TMP-CANDDY_ALLN(8.2 mg, 0.010 mmol, 15%, isolated yield)を得た。
参考例6では、参考例2と同様にして、TMP-CANDDY_ALLNのプロテアソーム阻害活性及びプロテアソームとの親和性を評価した。
図8A~図8Cに示すとおり、β2及びβ5の活性に対して、TMP-CANDDY_ALLNでは、ALLN単独と比較して阻害活性が弱まり、ALLNの阻害活性が失活していることが確認できた。β1については、ALLNではあまり阻害されないことが報告されており(Kaiser, M. et al., Chem. Bio. Chem., 2004, 5, 1256-1266)、この報告と矛盾しない結果であった。
また、β1、β2、及びβ5のいずれに対しても、TMP-CANDDY_ALLNの濃度依存的に阻害活性が高まることから、TMP-CANDDY_ALLNとプロテアソームとの親和性が確認された。
参考例7では、HeLa細胞において強制発現させたecDHFRタンパク質のTMP-CANDDY_ALLNを介した分解(ノックダウン)について、FACS解析により評価した。
参考例3と同様にして、ecDHFRタンパク質を発現するプラスミド(pMIR-DsRed-IRES-ecDHFR-HA-GFP)を準備した。

プラスミドを導入してから40時間培養した後、以下のようにして、HeLa細胞にTMP-CANDDY_ALLNを添加した。培地としては、D-MEM(High D-Glucose, Phenol Red, Sodium Pyruvate)(和光純薬工業(株))に1質量% L-グルタミン溶液(Sigma-Aldrich)を添加した無血清培地(37℃)を用い、各ウェル当たり300μL添加した。なお、L-グルタミン溶液は使用直前に添加した。TMP-CANDDY_ALLNを含有するDMSO溶液を各ウェル当たり3μL添加して、37℃、5体積% CO2の条件下で培養した。コントロールとしては、TMPを含有するDMSO溶液又はDMSOを用いた。
TMP-CANDDY_ALLNの添加24時間後、培地を除去し、PBSを添加して細胞を洗浄した。PBSの除去後、37℃のトリプシン(0.25 w/v% Trypsin-1 mmol/L EDTA・4Na Solution with Phenol Red)(和光純薬工業(株))を各ウェル当たり200μL添加し、37℃、5体積% CO2の条件下で1分間培養した。培養後、D-MEM(Low D-Glucose, L-Glutamine, Phenol Red)(和光純薬工業(株))に10質量% FBS及び1質量% PenStrep(100 U/mL Sodium Penicillin G and 100 μg/mL Streptomycin Sulfate)(和光純薬工業(株))を添加した培地を各ウェル当たり300μL添加して懸濁し、15mLチューブに細胞溶液を回収した。

Claims (11)
- 前記プロテアソーム阻害活性が、カスパーゼ様活性、トリプシン様活性、及びキモトリプシン様活性から選ばれる少なくとも1種に対する阻害活性である請求項1に記載のp53分解誘導分子。
- 請求項1~請求項3のいずれか1項に記載のp53分解誘導分子を含む医薬組成物。
- p53タンパク質が仲介する疾患又は症状の予防又は治療に用いられる請求項8に記載の医薬組成物。
- 前記p53タンパク質が仲介する疾患又は症状が、癌、細胞老化、神経疾患、神経細胞死、糖尿病、又は心機能障害である請求項9に記載の医薬組成物。
- 前記p53タンパク質が仲介する疾患又は症状が細胞老化である請求項10に記載の医薬組成物。
