実施例1
キュウリモザイクウイルス(CMV)のコートタンパク質(CP)の単離およびクローニング
CMV感染ユリ葉由来の全RNAをTRI試薬(Sigma、Saint Louis、USA)を使用して、製造者の指示にしたがい単離した。cDNA合成では、OneStep RT-PCRキット(Qiagen、Venlo、オランダ)を使用した。CMV CP遺伝子の増幅では、プライマー配列をGenBankからのCMV配列の分析後に選択した:CMcpF(CACCATGGACAAATCTGAATCAACCAGTGCTGGT)(SEQ ID NO:8)およびCMcpR(CAAAGCTTATCAAACTGGGAGCACCCCAGATGTGGGA)(SEQ ID NO:9);NcoIおよびHindIII部位には下線が付けられている。対応するPCR産物をpTZ57R/Tベクター(Fermentas、Vilnius、リトアニア)にクローニングした。大腸菌XL1-Blue細胞をクローニングおよびプラスミド増幅のための宿主として使用した。PCRエラーを含むクローンの選択を回避するために、いくつかのCP遺伝子含有pTZ57プラスミドクローンを、BigDyeサイクル配列決定キットおよびABI Prism3100ジェネティックアナライザ(Applied Biosystems、Carlsbad、USA)を使用して、配列決定した。配列決定後、SEQ ID NO:1のCMVコートタンパク質をコードする、配列エラーのないCMV CP遺伝子のcDNA(SEQ ID NO:10)をその後、pET28a(+)発現ベクターのNcoI/HindIII部位にサブクローニングし(Novagen、San Diego、USA)、発現プラスミドpET-CMVwtを得た(図1)。
CMVのVLPにつながる、大腸菌におけるSEQ ID NO:1のCPの発現
CMV VLPを得るために、大腸菌C2566細胞(New England Biolabs、Ipswich、USA)を、CMV CP遺伝子含有プラスミドpET-CMVwtで形質転換させた。最高発現レベルの標的タンパク質を有するクローンの選択後、大腸菌培養物を、回転式振盪機(200rev/分;Infors、Bottmingen、スイス)上にて、カナマイシン(25mg/l)を含む2×TY培地中で、30℃にて、0.8-1.0のOD600まで増殖させた。その後、細胞を、0.2mM IPTGで誘導し、培地に5mM MgCl2を補充した。インキュベーションを回転式振盪機上で20℃にて18時間の間続けた。得られたバイオマスを低速遠心分離により収集し、-20℃で凍結させた。氷上での解凍後、細胞を、50mMクエン酸ナトリウム、5mMホウ酸ナトリウム、5mM EDTA、5mMメルカプトエタノールを含むバッファー(pH9.0、バッファーA)中に懸濁させ、超音波処理により破壊させた。不溶性タンパク質および細胞デブリを遠心分離により除去した(13,000rpm、5℃で30分)。清澄ライセート中の可溶性CMV CPタンパク質を、飽和硫酸アンモニウム(1:1、vol/vol)を用いて、一晩+4℃でペレット化した。沈殿したタンパク質を同じバッファーA(メルカプトエタノールなし)中で4時間の間+4℃で可溶化した。不溶性タンパク質を低速遠心分離により除去した(13,000rpm、4℃で15分)。可溶性CMV CP含有タンパク質溶液を細胞タンパク質から、超遠心分離法(SW28ローター、Beckman、Palo Alto、USA;25,000rpmで6時間、5℃)により、スクロース勾配中で分離した(バッファーA中20-60%スクロース、メルカプトエタノールなし、0.5%トリトンX-100補充)。勾配を勾配の底部で開始して、6つの画分に分割し、画分をSDS-PAGEにより分析した(データ示さず)。組換えCMV CPを含む画分No.2およびNo.3を合わせ、200体積のバッファー(5mMホウ酸ナトリウム、2mM EDTA、pH9.0)に対して透析し、スクロースおよびトリトンX-100を除去した。透析後、CMV CP溶液を0.2μフィルタに通す濾過により滅菌した。次に、CMV CPを、Type70ローター(Beckman、Palo Alto、USA)超遠心分離法を用いて、20%スクロース「クッション」により、無菌条件下で(50000rpm、4時間、+5℃)濃縮した。精製CMVwtの濃度を、QuBit蛍光光度計を用いて、製造者の推奨にしたがい推定した(Invitrogen、Eugene、USA)。濃VLP溶液(approx.3mg/ml)を、+4℃で、5mMホウ酸ナトリウム、2mM EDTA、バッファー(pH9.0)中で保存した。VLPの発現および精製に関与する全てのステップをSDS-PAGEにより12.5%ゲルを用いてモニタした。
破傷風トキソイドエピトープを含むCMVの改変コートタンパク質のクローニング(CMV-Ntt830)
SEQ ID NO:1のCMV CPのN末端の元のアミノ酸を破傷風トキソイドエピトープコード配列で置き換えるために、pET-CMVwtプラスミドをPCR増幅および変異誘発のために使用した。CMVwt遺伝子内に位置するSalI部位(図1)を対応するPCR産物をクローニングするために使用した。
CMVの改変VLPにいたる、大腸菌におけるCMV-Ntt830の発現
CMV-Ntt830 VLPを得るために、大腸菌C2566細胞(New England Biolabs、Ipswich、USA)を、CMV-Ntt830遺伝子含有プラスミドpET-CMV-Ntt830で形質転換させた。最高発現レベルの標的タンパク質を有するクローンの選択後、大腸菌培養物を、カナマイシン(25mg/l)を含む2×TY培地中で、回転式振盪機(200rev/分;Infors、Bottmingen、スイス)において30℃で、0.8-1.0のOD600まで増殖させた。細胞をその後、0.2mM IPTGで誘導し、培地に5mM MgCl2を補充した。インキュベーションを回転式振盪機上で20℃で18時間の間続けた。得られたバイオマスを低速遠心分離により収集し、-20℃で凍結させた。氷上での解凍後、細胞を、50mMクエン酸ナトリウム、5mMホウ酸ナトリウム、5mM EDTA、5mMメルカプトエタノールを含むバッファー中に懸濁させ(pH9.0、バッファーA)、超音波処理により破壊した。不溶性タンパク質および細胞デブリを遠心分離により除去した(13,000rpm、5℃で30分)。清澄ライセート中の可溶性CMV-Ntt830タンパク質を、飽和硫酸アンモニウム(1:1、vol/vol)を一晩+4℃で用いてペレット化した。沈殿したタンパク質を、バッファーA(メルカプトエタノールなし)中で4時間の間+4℃で可溶化した。不溶性タンパク質を低速遠心分離により除去した(13,000rpm、4℃で15分)。可溶性CMV-Ntt830含有タンパク質溶液を細胞タンパク質から超遠心分離法(SW28ローター、Beckman、Palo Alto、USA;25,000rpm、6時間、5℃)により、スクロース勾配中で(バッファーA中20-60%スクロース、メルカプトエタノールなし、0.5%トリトンX-100補充)分離した。勾配を、勾配の底部で開始して、6画分に分割した。組換えCMV-Ntt830を含む画分を合わせ、200体積の5mMホウ酸ナトリウム、2mM EDTA(pH9.0)に対して透析し、スクロースおよびトリトンX-100を除去した。透析後、CMV-Ntt830溶液を0.2μフィルタを通す濾過により滅菌した。次に、CMV-Ntt830を、Type70ローター(Beckman、Palo Alto、USA)超遠心分離法を用いて、20%スクロース「クッション」により、無菌条件下で(50000rpm、4時間、+5℃)濃縮した。精製CMV-Ntt830の濃度を、QuBit蛍光光度計を用いて、製造者の推奨にしたがい推定した(Invitrogen、Eugene、USA)。濃VLP溶液(approx.3mg/ml)を、+4℃で、5mMホウ酸ナトリウム、2mM EDTA、バッファー(pH9.0)中で保存した。VLPの発現および精製に関与する全てのステップをSDS-PAGEにより12.5%ゲルを用いてモニタした。CMV VLP中の破傷風トキソイドエピトープの存在を証明するために、精製CMV-Ntt830VLPの質量分析を使用した。図3Cに示されるように、得られた主ピークは、大腸菌細胞中のタンパク質合成中に生じる第1のメチオニンが除去される場合、タンパク質の理論分子質量に対応する。動的光散乱および電子顕微鏡法により、CMVwt VLPに類似する等軸粒子形態が確認された(図4Aおよび4B)。
PADREエピトープを含むCMVの改変コートタンパク質のクローニング
(CMV-Npadr)
PADREエピトープコード配列をCMVwt遺伝子に導入するために、PCR変異誘発を、増幅およびサブクローニングのための鋳型としてpET-CMVwtプラスミドを使用して実施した(実施例2および3も参照されたい)。増幅のために、下記プライマーを使用した:pET-220(SEQ ID NO:11)およびCMV-padrSal-R(GACGTCGACGCGCGGCCGCCTTGAGGGTCCACGCGGCCACAAATTTCGCCATGGT)(SEQ ID NO:15)。得られたPCR産物(SEQ ID NO:7のCMV-NpadrをコードするSEQ ID NO:16のcDNA)をpET-CMVwtのBglII/SalI部位で重ねてサブクローニングさせた。妥当なクローンを配列決定により同定し、pET-CMV-Npadrと指定した。
CMVの改変VLPに至る大腸菌におけるCMV-Npadrの発現
CMV-Npadrの発現および精製についての手順は、CMV-Ntt830に対するものと本質的に同じであり、実施例4で記載される。CMV VLPにおけるPADREエピトープの存在を証明するために、精製CMV-Npadr VLPの質量分析を使用した。図3Bに示されるように、得られた主ピークは、大腸菌細胞中のタンパク質合成中に生じる第1のメチオニンが除去される場合、タンパク質の理論分子質量に対応する。動的光散乱および電子顕微鏡分析により等軸粒子形態が確認された(図5Aおよび図5B)。
6×HisタグおよびC末端システインを有するイヌIL-31のクローニングおよび産生
大腸菌最適化コドン、N末端6×Hisタグ、TEVプロテアーゼ切断部位、C末端システインおよび隣接しているNcoIおよびPstI制限部位を有するイヌIL31cDNA配列(SEQ ID NO:17)をIDT社で製造した。さらに、NcoI/PstI断片をベクターpET42の対応する部位中に連結させた。コンストラクトを化学的コンピテント大腸菌DH5α細胞において形質転換させ、コロニーをアンピシリンを含むLB寒天プレート上に播種した。得られたクローンの配列を、Sanger配列決定により検証した。得られたコンストラクトpET42_6HcIL31Cを化学的コンピテント大腸菌BL21-DE3細胞においてさらに形質転換させた。
組換え天然イヌIL-31のクローニングおよび産生
イヌIL-31を、SEQ ID NO:17を含むプラスミドから、PCRを用いて、PstIおよびNcoI部位を含む、プライマーcIL31NATf(TACACCATGGCCTCCCACATGGCTCCAACG、SEQ ID NO:19)およびcIL31NATr(CATACTGCAGTTACTGCGGTCCACTGTTTAAG、SEQ ID NO:20)を使用して増幅させた。得られたcIL-31PCR断片を、PstIおよびNcoI制限酵素で消化させ、pET42ベクターの対応する部位において連結させた。コンストラクトを化学的コンピテント大腸菌DH5α細胞において形質転換させ、コロニーをアンピシリンを含むLB寒天プレート上に播種した。陽性クローンを、Sanger配列決定により同定した。cIL31-pET42コンストラクトをその後、化学的コンピテント大腸菌BL21-DE3細胞において形質転換させた。
組換えイヌIL-31コンストラクトのCMV-Npadr VLPおよびCMV-Ntt830 VLPへのカップリングならびにマウスおよびイヌの免疫化
A.cIL-31のVLPへのカップリング
実施例8で得られた、100μlの、cIL-31タンパク質(SEQ ID NO:21)の溶液(PBSバッファー中4mg/ml)を、4.8μlの55mM N-スクシンイミジルS-アセチルチオアセテート(SATA、Thermo Fisher Scientific)を含むDMSOと混合し、30分間室温(RT)でインキュベートした。未反応SATAをアミコンウルトラ-0.5 3K濾過ユニット(Merck-Millipore)を用いたPBS(0.1Mリン酸ナトリウム緩衝液、pH7.2および0.15M NaCl)への4×バッファー交換により除去し、最終体積を100μlに調整した。スルフヒドリル基を10μLの脱アセチル溶液(0.5Mヒドロキシルアミンおよび25mM EDTAを含むPBS)の添加および2時間室温でのインキュベーションにより脱保護した。脱アセチル溶液を、アミコンウルトラ-0.5 3K濾過ユニット(Merck-Millipore)を用いたPBSへの4×バッファー交換により除去し、最終体積を100μlに調整した。
5匹の雌Balb/cマウスの群を第0日および第14日に、PBS中で製剤化された、50μgの、SEQ ID NO:21のcIL-31に、SATAを介してカップリングさせたCMV-Npadr VLPおよびCMV-Ntt830のいずれかを用いてs.c.で免疫化する。マウスを第0日(免疫前)、第14日、および第28日に採血し、血清をcIL-31特異的ELISAを用いて分析する。
5匹のイヌの群を、第0日および第14日、第28日および第42日に、PBS中で製剤化された、50μgの、SATAを介してSEQ ID NO:21のcIL-31にカップリングさせたCMV-Npadr VLPおよびCMV-Ntt830 VLPのいずれかを用いてs.c.で免疫化する。イヌを第0日(免疫前)、第14日、第28日、第42日および第56日に採血し、血清をcIL-31特異的ELISAを用いて分析する。
イヌアトピー性皮膚炎の治療のためのCMV-Ntt830 VLPにカップリングされた組換えイヌIL-31コンストラクトによるイヌの治療的免疫化
15匹のアトピー性皮膚炎を有するイヌの群を第0日および第14日、第28日、第56日よびおよび第80日に、Alum中で製剤化された、50μgの、SATAを介してSEQ ID NO:21のcIL-31にカップリングさせたCMV-Ntt830 VLPまたはCMV-Ntt830単独を用いてs.c.で免疫化する。疾患重症度を免疫化前ならびに第28日、第56日、第80日および第120日に目視検査により測定する。
CADスコアリング
CAD症状スコアリングでは、アトピー性皮膚炎病変指数(Atopic Dermatitis Lesion Index)(ADLI)およびそう痒ビジュアルアナログスケール(PVAS)を実験イヌにおいて使用する。ADLIスコアは、5つの特定の身体領域において評価されるイヌアトピー性皮膚炎と関連する、6つの臨床症状の検証された複合指数である。各特定の身体領域で、各パラメータがスコアラーにより0から5までスコア化され、0のスコアは病変なしとして規定され、5のスコアは重篤/広範な病変として規定される。PVASはスコアラーにより0から10のアナログスケールによりスコア化され、0のスコアはそう痒/咀嚼なしを表し、10のスコアは、絶え間ない、かつ激烈なそう痒/咀嚼に等しい。処置したイヌのCADスコアリングをベースライン、ならびにプラセボで処置したイヌのスコアと比較する。
屋内チリダニにアレルギー性のイヌのcIL-31-CMV-Ntt830 VLPを用いたワクチン接種およびその後の誘発
6匹のアレルギーイヌをAlum中で製剤化された、100μgのcIL-31-CMV-Ntt830 VLPを用いて免疫化し、6匹のアレルギーイヌをプラセボ(alum)で処置した。3週間後、イヌを300μgのワクチンまたはプラセボで追加免疫した。血液を第28日に採取した。d-28日に、毛を腹部皮膚からテープストリッピングにより除去し、屋内チリダニ誘発を1日1回10分間、連続5日間実施した。皮膚病変およびそう痒を12匹のイヌについて、6時間/日の間のイヌの臨床検査録画によりスコア化した。
イヌ血清における抗体応答を第28日に分析した。cIL-31に特異的な抗体を、ELISAプレートを、3μg/mlの濃度の組換えcIL-31を含む100μlの体積のPBS(pH7.2)で4℃にて一晩コートすることにより分析した。ELISAプレートを5×、0.05%Tween20を含む200μlのPBS(pH7.2、PBST)で洗浄した。非特異的結合を回避するために、ELISAプレートを200μlの2%BSAを含むPBSTでブロックし、2時間の間室温でインキュベートした。血清試料を2%BSA/PBST中で希釈する。予備希釈された血清(10倍)をコートプレート上に移し、さらに段階希釈し(3倍ステップ)、OD50計算に基づく抗体力価を取得した。室温での2時間のインキュベーション後、ELISAプレートを5×、200μlのPBSTで洗浄した。血清抗体の結合をセイヨウワサビペルオキシダーゼコンジュゲートヤギ抗イヌIgG(JacksonImmunoResearch)により検出した。検出抗体を2%BSA/PBST中で1:1000希釈し、100μlの体積/試料を移した。プレートを1時間の間室温でインキュベートした。ELISAプレートを前に記載されるように洗浄する。洗浄前に、5つの基質溶液を調製した。この目的を達成するために、1錠剤(10mg)のOPD(1,2-フェニレンジアミン二塩酸塩)および9μlの30%H2O2を25mlクエン酸バッファー(0.066MNa2HPO4、0.035Mクエン酸、pH5.0)に溶解した。100μlの体積の基質溶液をプレート上にピペッティングし、7分間室温で正確にインキュベートした。反応を停止させるために、50μlの停止液(5%H2SO4のH2O溶液)をプレート上に直接ピペッティングする。1,2-フェニレンジアミン二塩酸塩呈色反応の450nmでの吸光度測定値を分析し、10倍予備希釈血清のlog3として表した(図13B)。図13BはIL-31特異抗体力価とひっかき強度の間の相関を示す。依然としてひっかいていた1匹のイヌは最低抗体応答を開始し、<1/300の指示力価が保護的であることが見出された。

- 前記CMVポリペプチドは、CMVのコートタンパク質のアミノ酸配列を含み、前記アミノ酸配列は、SEQ ID NO:1またはSEQ ID NO:1の少なくとも95%の配列同一性を有するアミノ酸配列を含み;ならびに、前記アミノ配列はSEQ ID NO:23を含み、前記ヘルパーT細胞エピトープは前記CMVポリペプチドのN末端領域にとって代わり、前記CMVポリペプチドの前記置き換えられたN末端領域は11~13の連続アミノ酸から構成される、請求項5~8のいずれか一項に記載の組成物。
- 前記改変CMVポリペプチドは、SEQ ID NO:6またはSEQ ID NO:7のアミノ酸配列を含む、請求項5~9のいずれか一項に記載の組成物。
- 前記少なくとも1つのcIL-31抗原は、下記から選択されるアミノ配列を有するタンパク質を含む、請求項1~10のいずれか一項に記載の組成物:
(a)SEQ ID NO:18;
(b)SEQ ID NO:21;
(c)SEQ ID NO:22;または
(d)SEQ ID NO:26。 - 前記組成物の前記投与は少なくとも1つのCADパラメータまたは症状を、前記投与前の前記少なくとも1つのCADパラメータまたは症状と比べて低減させる、請求項1~11のいずれか一項に記載の組成物。
- (a)少なくとも1つの第1の付着部位を有するウイルス様粒子(VLP);
(b)少なくとも1つの第2の付着部位を有する少なくとも1つのイヌインターロイキン-31抗原(cIL-31抗原)
を含む組成物であって、
前記cIL-31抗原は、SEQ ID NO:22から選択されるアミノ配列を有するタンパク質、または、SEQ ID NO:22と少なくとも90%アミノ酸配列同一性のアミノ酸配列を有するタンパク質を含み;
(a)および(b)は前記少なくとも1つの第1の付着部位および前記少なくとも1つの第2の付着部位により、少なくとも1つの非ペプチド共有結合を介して連結される、組成物。 - 前記VLPはキュウリモザイクウイルス(CMV)の改変VLPであり、前記改変CMVポリペプチドは、SEQ ID NO:6またはSEQ ID NO:7のアミノ酸配列を含む、請求項12または13に記載の組成物。
- 前記第2の付着部位はスルフヒドリル基である、請求項13または14に記載の組成物。
