Description
数多くの抗腫瘍化合物が合成されてきた。各種のインドリル化合物が抗腫瘍効果を有していると指摘されている(WO 2013063492)が、どの実験データもそれらの化合物が抗癌効果を有していることを示してはいない。
カテコールアミン作動性神経系またはセロトニン作動性神経系が、脳内における腫瘍発現の作用機序に関連しているであろうということは、これまでに示唆されたことはない。
デプレニルは選択的にCAE物質であり、セロトニン作動性ニューロンの非常に弱いエンハンサーであるが、例えばJP04953041B2に教示されているように、BPAPはカテコールアミン作動性およびセロトニン作動性エンハンサー効果(すなわちCAE/SAE効果)の両方を有する化合物である。
本発明者らの知る限りにおいて、ニューロン活性エンハンサー化合物は、癌または腫瘍の予防または治療に使用することは示唆されていない。
該化合物は、中枢神経系のモノアミン作動性ニューロンにおいてインパルス伝播を介したモノアミン神経伝達物質の遊離を増強するモノアミン作動性活性エンハンサー化合物であるニューロン活性エンハンサー化合物であり、その化合物の癌またはその転移を予防または治療するための使用に関するものであり、好ましくは、モノアミン作動性エンハンサー化合物の効果、すなわち、モノアミン作動性ニューロンからのインパルス伝播を介したモノアミン神経伝達物質の遊離の増強が、以下の方法によって直接的または間接的に測定可能または検出可能である。
- カテコールアミン量の測定、好ましくは、前記エンハンサー化合物の存在下で単離されたラットの脳幹からの電気刺激により遊離された [3H]-ノルエピネフリン、[3H] -ドパミンの量の測定;
- セロトニン量の測定、好ましくは、前記エンハンサー化合物の存在下で単離されたラットの脳幹から電気刺激により遊離された [3H]-セロトニンの量の測定;
- 条件回避反射(CAR)解析、例えばテトラベナジン処置によって学習障害を来したマウスなどを用いて行ったとき、合成CAE物質またはA型MAO阻害薬の投与により拮抗しうるが、B型MAOの選択的阻害またはカテコールアミンまたはセロトニンの再取り込みの阻害は、それぞれ無効である; そして
- 非常に広い用量範囲での候補エンハンサー化合物の投与、二峰性、ベル型の濃度効果曲線はエンハンサー効果に特徴的である(少なくともMAO阻害より低い範囲); 低用量範囲は「特異的エンハンサー効果」を示し、高用量範囲は「非特異的エンハンサー効果」を示す(Knoll J 2012)。

我々が、例えば3分間の回収期間中に単離したラット脳幹から電気的刺激により遊離した[3H]-ノルエピネフリン、[3H]-ドパミンまたは[3H]-セロトニン量を測定した時、また、特異的なエンハンサー効果を発揮するデプレニルやBPAPを至適濃度の存在下、測定を繰り返した時、標識された伝達物質の遊離量は有意に高い。これは、エンハンサー感受性ニューロン群が、全体として、合成エンハンサー物質の存在下で高い活性レベルですぐに働くことを示している。単回ウォッシュアウト後、ニューロンは再びそれらの正常な活性レベルですぐに働く。ニューロンは、「悉無律」の様式で刺激に反応するので、ニューロン群(最も興奮性のもの)の一部のみが電気刺激に反応することは明らかである。エンハンサー物質はニューロンの興奮性を増幅させるので、エンハンサー物質の存在下、ニューロン群のより高い割合が興奮し、電気的刺激により遊離された標識伝達物質量が、有意に増加する(Knoll,2005.3.1.3レビュー参照)。
材料および方法
材料
(-)-1-(ベンゾフラン-2-イル)-2-プロピル-アミノペンタンHCl [(-)−BPAP] 藤本製薬株式会社、大阪、日本;
(-)デプレニル(セレギニン)はサノフィ−キノイン、ブタペスト、ハンガリー;
テトラベナジン HCl(ハンガリー、ブダペストの技術科学大学、有機化学講座、C. Szantay教授によって合成された。)
ゼンメルワイス(Semmelweis)大学の飼育コロニーから提供を受けた体重250-350gのWistar系雄性ラット(チャールズ・リバー)で実験を行った。動物を12時間の明暗サイクルおよび温度(22±2℃)および相対湿度(55±5%)に制御された条件下で維持した。室温および相対湿度は毎日確認した。ラットを標準実験室用飼料および水道水を自由摂取および飲水で飼育した。すべての操作は、実験的および他の科学的目的のために使用される脊椎動物の保護のための欧州条約に準拠している。この研究は、ブダペストのゼンメルワイス大学の動物倫理委員会によって承認された(承認番号:1810/003/2004)。
シャトルボックスの改変版では、双方向条件回避反射(CAR)の獲得を5日間連続して解析した。ラットを、中央に小さなゲートを有する仕切りによって2つの部分に分割されたボックスに入れ、動物を条件刺激(CS、光刺激)の影響下で仕切りを横切るように訓練した。5秒以内に反応しなかった場合には、ラットは無条件刺激(US)であるフットショック(1mA)が与えられた。ラットがUSに5秒以内に応答しなかった場合、それは逃避失敗(EF)として分類された。1回の試行は、10秒の試行間隔と20秒のCSとで構成された。CSの最後の5秒はUSの5秒をオーバーラップさせた。各学習セッションでは、CAR、EF、および信号間反応(IR)の数が自動的に計数され、多元配置分散分析(ANOVA)によって評価された。
長寿研究は、ゼンメルワイス大学の繁殖コロニーから提供された200匹の雄性ラットを用いて実施された。ラットは2010年2月末に生まれた。2ヶ月の馴化期間の後、ラットを無作為に5つの群に分けた。長寿試験は2010年5月初旬に開始された。動物は自然死するまで観察された。この研究のデータは、生理食塩水処置群の最後の動物が2012年10月31日に死亡したため、2012年10月31日までに観察された変化を表す。後で示すように、エンハンサー処置群の数匹のラットは、 2012年11月1日時点でなお生存していた。
我々は、皮下腫瘍の出現を注意深く観察し、腫瘍の展開を死亡まで追跡した。組織学的解析は、例として各群から採取した数匹のラットについて死後に行った。
最終的な組織学的診断は、皮下組織における粘液線維肉腫であった。
デプレニルおよびBPAPが「特異的」および「非特異的」エンハンサー効果を発揮する低用量での延命効果
薬物のCAE効果の発見前に行ったデプレニルの2つの長寿試験では、脳のMAO-B活性を完全に阻害する0.25mg/kg用量のデプレニルを使用した。この研究では、シャトルボックス実験で発揮された化合物が「非特異的」および「特異的」エンハンサー効果を示す(図3参照)、デプレニルのピーク用量を使用した。
我々の長寿研究の過程で、我々はラットの脳内で以前は知られていなかったユニークなエンハンサー感受性TMS機構が作動していることを発見した。
肉眼的観察
ラットの皮下の局所において、結節が出現した。写真は、粘液線維肉腫の典型的な皮下局在を説明している。
腫瘍は限局性であったが、検出可能な被膜は見られなかった。写真は、特徴的な構造を持たない、灰白色の腫瘍の切断面を示している。時折、小さな出血領域と黄色の同質性の壊死領域が見られることがあった。
ヘマトキシリン-エオシン(HE)染色切片では、腫瘍細胞は、多数の小さな血管を有する、疎性の淡い好酸性マトリックスに局在していた。腫瘍細胞は、中央性または偏心性に局在した丸みを帯びた核および淡い好酸球性の細胞質を有する細長いまたは星状の形態を有していた。時折有糸分裂像が検出された。PAS反応では、マトリックスは淡陽性反応を示し、腫瘍細胞は主に陰性であった。写真は、皮下腫瘍の典型的な組織学を示している。
免疫組織化学反応により、腫瘍細胞はビメンチンに対して強く陽性に染色された。SMA(平滑筋抗原)の抗体は血管のみを染色し、腫瘍細胞は陰性であった。抗デスミン抗体は、筋肉構成成分のみと反応し、陰性染色された腫瘍細胞が浸潤していた。
このデータは、我々の研究で使用されたWistar系ラットでは、急速に増殖する皮下粘液線維肉腫の発現を示す割合が、時間につれて連続的に増加するという実験的証拠を提供し、この腫瘍の発現はこの系統の本来の特性であるということの証拠であると思われる。
細胞株
ヒト髄芽腫細胞株DaoyはATCCCから購入し、UW-228はSilber教授(ワシントン州シアトル、ワシントン大学)から好意により提供されたものである。
DaoyおよびUW-228-2細胞株は、(50ml FCS(Gibco)、40mgゲンタマイシン(Sandoz)、5mlのピルビン酸ナトリウム(S8636、Sigma, St Louis, USA)、5mlの非必須アミノ酸溶液(M7145, Sigma, St Louis, USA)、10 mlのL-グルタミン(Sigma, St Louis, USA)を含有した500mlのイーグル最小必須培地, α改変型(M8042, Sigma, St Louis, USA)で、37℃、5% CO2中で維持された。
96ウェルプレート(Sarstedt)の各ウェルに、3x10 3個のDaoyまたはUW-228-2細胞をに播種し、100μlの10%FCS含有培地に加えた。播種24時間後、さらに100μlの培地に溶解した薬物によって細胞を72時間処理した。最初に、両細胞株を単独の(-)-BPAPおよび(-)-デプレニルで処置し、10-6、10-7、10-8、10-9、10-10、10-11、10-12、10-13および10-14M濃度で用量−効果曲線を評価した。併用処置では、10-3、3.3×10-4、1.1×10-5および3.7×10-5Mのテモゾロミド(Schering Plough, USA)、または、0.04, 0.2, 1, 5μMのシスプラチン(Ebewe Pharma, Austria)、または0.04, 0.2, 1, 5μMのエトポシド(Ebewe Pharma, Austria)、または10-7、10-6、10-5および10-4μM(UW228-2)または0.001, 0.005, 0.025および0.125μM(Daoy)のビンクリスチン(Richter Gedeon, Hungary)の単独または、10-13または10-8Mの(-)-BPAPまたは(-)-デプレニルとの併用で行われた。
シャトルボックスにおける群間のパフォーマンスの有意性は、多因子分散分析(ANOVA)によって評価した。ラットにおける腫瘍発現から死亡までの期間およびエンハンサー物質で慢性的に処置されたラットの加齢に関連した体重変化に関する有意性は、一元配置 ANOVAによって解析した。生存率および腫瘍発現の統計解析は、Kaplan-Meier法によって行った。
我々は、2種のヒト培養髄芽腫細胞株、すなわち、線維形成性小脳髄芽細胞腫由来のDaoy HTB-186細胞株(Jacobsenら、1985)および二倍体DNA量をもつ後頭蓋窩髄芽腫由来のUW-228-2細胞株(Kelesら、1985)におけるエンハンサー物質の効果を試験し、10-14及び10-6Mの9つの濃度範囲でデプレニルとBPAPの効果を調べた。適用したいずれの濃度においてもデプレニルまたはBPAPは培養髄芽細胞腫細胞上の増殖に影響を与えなかった。さらに、BPAPとデプレニルは、調べられた既知の腫瘍細胞増殖阻害薬の有効性を変化させなかった(方法における調べられた組み合わせを参照)。
(-)-BPAP処置が肺腺癌細胞の成長に影響を及ぼするか否かを調べるために、in vivo異種移植モデルが計画された。
皮下に維持されたマウスの原発性肺腺癌腫瘍が、本研究で利用された。直径3mmの大きさの腫瘍が、体重25gのFVB/N雌性マウス計18匹の皮下に移植された。腫瘍接種翌日に、それぞれ6匹ずつの動物に、0.0001mg/kg(低用量)または0.05mg/kg(高用量)のいずれかの用量で、(-)-BPAPを毎日皮下注射された。腫瘍サイズはデジタルノギスを使用して週2回測定した。腫瘍体積は以下の式で計算した。
対照のFVB/N系動物における腫瘍の成長は、(-)-BPAP処置されたマウスより速かった(図14)。腫瘍移植後12日目から、対照マウスと低用量の(-)-BPAP処置動物との間で腫瘍体積に有意差が認められた(P<0.05)。高用量の(-)-BPAP処置マウスは、低用量の(-)-BPAP処置マウスと同様の腫瘍体積を示し、これらの変化には、統計的有意性がなかった(図14)。
Balb/c系ヌードマウスで行った実験では、以前のFVB/N系の動物での結果と類似した結果が得られた(図15)。低用量の(-)-BPAP処置は、腫瘍体積の測定によって示される腫瘍増殖を有意に阻害した(図15)。我々の以前の観察と同様に、高用量の(-)-BPAPは、対照サンプルに比べて、より低い腫瘍体積であったものの、腫瘍サイズの有意な変化を引き起さなかった(図15)。これらの結果は、BPAPの腫瘍抑制作用は、免疫系のTまたはB細胞を必要としないことを示唆している。
この研究において、15日齢のFVB/N系マウスにジエチルニトロソアミンの単回腹腔内注射(15μg/g体重)によって、原発性肺腺癌が誘発された。
3群の動物が定義された:
1.対照群:8週齢から毎日生理食塩水が皮下注射された。
2.BPAP低用量群:8週齢から毎日0.0001mg/kgが皮下投与された。
3.BPAP高用量群:8週齢から毎日0.05mg/kgが皮下投与された。
1年後、動物を屠殺し、体重および肺重量を記録した。さらに、肉眼的に観察可能な腫瘍を計数した。
これらの予備的な結果は、低用量のBPAPが原発性肺腺癌の形成または発現をよく予防しうることを示している。
この研究では、速やかに摘出されたラットの青斑核のオーガンバスに(-)-BPAPを添加した。8個の組織が、各濃度の解析のために使用された。異なる濃度の(-)-BPAP存在下で、組織から20分以内に遊離されたノルエピネフリンの量を、KnollおよびMiklya(1995)の方法に従って測定した。(対応のあるスチューデントのt検定。*P<0.01、**P<0.001)。
ノルエピネフリン量(組織湿重量1g当たりのmmolとしてプロット)は、BPAPモル濃度に対してプロットされている(図18参照)。二峰性、ベル型の濃度効果曲線は、ラットの単離青斑核に対する(-)-BPAPのエンハンサー効果に特徴的である。
この研究では、30,000個のマウス結腸癌細胞(C38)をC57B1/6系マウスの脾臓内に接種され、接種後23日目には肝転移が肉眼的に観察された。その実験において、1つのマウスの群は、30,000個の細胞/動物の接種に先立って、0.0001mg/kgBPAPを一週間毎日皮下に前処置し、さらに実験終了(第23日目)まで処置したが、対照動物にはBPAP処置をしなかった。
結果
BPAP処置群では、2個の腫瘍/肝臓が現れた(P <0.05)が、対照においては、接種後23日目の肉眼的に観察可能な肝転移の平均数は14であった。
マウスの寿命における(-)-デプレニルと(-)-BPAPの効果を調べるために、「特異的」(低い、すなわち特異的エンハンサー用量)および「非特異的」(より高い、すなわち、非特異的エンハンサー用量)エンハンサー効果を発揮する投与量により処置されたマウスをランダムに選択し剖検した:6ヶ月、1年、1.5年と2年の時点で屠殺し、ランダムに選んだマウスにおける肺癌の発現を検出した。
5群の性的に成熟したマウスは、異なる2用量の(-)-BPAPおよび(-)-デプレニルでそれぞれ毎日処置された。
1群:対照:生理食塩水
2群:BPAP低用量:0.0001mg/kgの(-)-BPAP
3群:BPAP高用量:0.05mg/kgの(-)-BPAP
4群:デプレニル低用量:0.001mg/kgの(-)-デプレニル
5群:デプレニル高用量:0.1mg/kgの(-)-デプレニル
合計172匹のマウスを実験に供した。体重は隔月に測定した。モニタリングのために、6ヶ月、1年、1.5年と2年の時点で、各群からマウスを屠殺する計画がなされた。残りの動物は自発死まで飼育された。
長寿研究がまだ実行されている;現時点(2015年11月)まで、6ヶ月、1年、1.5年間処置したマウスで剖検が行われた。肺癌は、6ヶ月または1年間の生理食塩水処置したマウスでは検出されなかった。これに対し、1.5年間生理食塩水処置したマウスの75%に腫瘍の発生が観察されたが、1.5年齢のエンハンサー処置マウスにおいては、腫瘍の発生は有意に低いかまたは全く発生しなかった。
結果を表4と図19および図20に示した。有意水準は、両面のカイ二乗検定(Chi-square test)に基づいて算出した。
図19は、表4に示すように、4匹の生理食塩水処置マウスと、18ヶ月間エンハンサー物質を毎日処置した合計14匹のマウスとの間の有意差の計算結果を示している。
図20は、特異的エンハンサー効果を発揮する非常に低用量の(-)-BPAPが、FVB/N系マウスにおける肺癌の発現を完全に抑制することを示している。
この測定は、EP1052259B1(WO2000026204A1に相当)の実施例2に記載された方法と同様にして、必要な変更を加えて、行うことができる。この方法は、[Knoll J、 Knoll BおよびMiklya I Life Sci, 58、2101-2114(1996)]にも記載されている。
電気的に刺激された脳(例えば、脳幹)からのモノアミン神経伝達物質の遊離を引き起こす、本発明の化合物の低用量または最小用量を明確にする場合、エンハンサー化合物の最小用量がMAO阻害薬のそれより低く、特に有意に低く、典型的には、0.04または0.02μg/ml、またはさらにより好ましくは0.015μgまたは0.01μg/mlのより高くなく、またはより低い。
この測定は、EP1052259B1(WO2000026204A1に相当)の実施例4に記載の測定と同様に、必要な変更を加えて、行うことができる。この方法は、[Knoll J. Knoll BおよびMiklya I、Life Sei, 58, 2101−2114(1996)]に記載されている。

Claims (9)
- 中枢神経系のモノアミン作動性ニューロンにおけるインパルス伝播を介したモノアミン神経伝達物質の遊離を増強する、モノアミン作動性エンハンサー化合物であり、一般式IIIで示される化合物またはその薬学的に許容される塩を、哺乳動物に、1日投与量として、0.05mg/kg体重より少ない量で投与されるように用いられる、一般式IIIで示される化合物またはその薬学的に許容される塩を含有する癌またはその転移の予防または治療剤。
(式IIIにおいて、
R 1 はC 2-4アルキル;
R 2 は水素、C 1-3アルキル、C 2-3アルキルカルボニルまたはC 6-10アリール;
R 3 は水素、メチルまたはエチルである。) - モノアミン作動性エンハンサー化合物がカテコールアミン作動性活性エンハンサー(CAE)物質および/またはセロトニン作動性活性エンハンサー(SAE)物質であり、モノアミン作動性ニューロンがカテコールアミン作動性および/またはセロトニン作動性ニューロンである請求項1に記載の癌またはその転移の予防または治療剤。
- 一般式IIIで示される化合物が、(2R)-1-(1-ベンゾフラン-2-イル)-N-プロピルペンタン-2-アミン[(-)-BPAP]である請求項1または2に記載の癌またはその転移の予防または治療剤。
- 癌が悪性腫瘍、または癌腫、肉腫、白血病、リンパ腫および胚細胞腫からなる群から選択される癌である請求項1から3のいずれかに記載の癌またはその転移の予防または治療剤。
- 悪性腫瘍が粘液線維肉腫、腺癌、結腸癌および肝転移癌からなる群から選択されるものである請求項4に記載の癌またはその転移の予防または治療剤。
- 哺乳動物がヒトである請求項1から5のいずれかに記載の癌またはその転移の予防または治療剤。
- モノアミン作動性エンハンサー化合物の1日投与量が、0.0001mg/kg体重未満の特異的エンハンサー用量である請求項1から6のいずれかに記載の癌またはその転移の予防または治療剤。
- モノアミン作動性エンハンサー化合物が、ヒトに少なくとも1ヶ月間投与されるように用いられる請求項1から7のいずれかに記載の癌またはその転移の予防または治療剤。
- モノアミン作動性エンハンサー化合物の光学純度が80%以上である請求項1から8のいずれかに記載の癌またはその転移の予防または治療剤。
