Description
[1]高吸水性高分子化合物を基材とする微粒子を含む腫瘍封止剤。
[2]高吸水性高分子化合物が、下記:
PS1−X−PS2 (1)
(式中、PS1及びPS2は多糖類であり、Xはスペーサーである)
で表される構造を有する、上記[1]に記載の腫瘍封止剤。
[3]PS1及びPS2は、同一であるか又は異なっていてもよく、独立して、βグルカン、キチン、キトサン、及びそれらの誘導体、並びにこれらの混合物からなる群から選択される、上記[2]に記載の腫瘍封止剤。
[4]βグルカン又はその誘導体が、セルロース、カルボキシメチルセルロース、又はTEMPO酸化セルロースである、上記[3]に記載の腫瘍封止剤。
[5]Xが、1種以上のポリカルボン酸無水物である、上記[2]〜[4]のいずれかに記載の腫瘍封止剤。
[6]Xが、1,2,3,4−ブタンテトラカルボン酸二無水物(BTCA)、ポリエチレングリコールジグリシジルエーテル(PEGDE)、ビフェニルテトラカルボン酸二無水物(BPDA)、ジフェニルスルホンテトラカルボン酸二無水物(DSDA)、PEG(ポリエチングリコール)400、PEG2000、及びエピクロロヒドリンからなる群から1種以上選択される架橋剤に由来する、上記[2]〜[4]のいずれかに記載の腫瘍封止剤。

本発明に係る腫瘍封止剤は、高吸水性高分子化合物を基材とし、腫瘍血管内皮細胞の間隙を透過し得る粒子径を有する微粒子であって、この微粒子がその間隙を透過後、高い生体接着性により間隙近傍に存在する腫瘍組織に付着した後、生体内の水分と接触することによって膨潤し、ゲル状組成物によるバリアを形成して、腫瘍への酸素と栄養の供給並びに腫瘍からの誘導因子の伝達を遮断することによって、その腫瘍細胞を壊死に至らしめ、その後、付着した微粒子が生体内で分解及び/又は代謝されることを特徴とする。また、本発明に係る腫瘍封止剤は、上記特徴を有するが、腫瘍への栄養血管を圧迫して血流を阻止することにより、腫瘍部位での血中の酸素濃度及び栄養供給を減少させ、腫瘍細胞を壊死させる効果も有することから、本発明の腫瘍封止剤には、上記の付加的効果を有する血流阻止剤としても機能する。
本発明の腫瘍封止剤は、水分により膨潤し、ゲル状組成物を形成することができる高吸水性高分子化合物を含有し、生体分解性及び/又は生体代謝性であってもよい。本明細書で使用するとき、用語「高吸水性」とは、吸水量又は膨潤率が、乾燥時における基材の重量又は容積の少なくとも2〜10,000倍となる材料の特性を意味する。また、用語「高分子」化合物とは、一般的には、複数の単量体の繰り返し構造から構成される重合体を意味するが、本明細書においては、さらに架橋された重合体を含むものを指す。高分子化合物の分子量は、典型的には、約10kDa〜2,000kDaであるが、これに限定されず、使用の目的に応じて適宜変更可能である。なお、本発明において使用される高吸収性高分子化合物は、後述する式(1)で表される化合物の他に、ポリアクリルアミド、グリコサミノグリカン、グリコサミノグリカン架橋体、コラーゲン、コラーゲン架橋体、ポリアクリル酸、ポリメチルメタクリレート、ポリビニルアルコール、ポリ−L−乳酸、またはこれらの誘導体であってもよい。また、本発明において使用される高吸水性高分子化合物の膨潤度は、上記の通り、重量比又は体積比で2〜10,000倍であってもよく、例えば、2〜1,000倍、2〜500倍、2〜300倍、2〜200倍、又は2〜100倍(上限値として、90倍、80倍、70倍、60倍、50倍、40倍、30倍、20倍、15倍、10倍、9倍、8倍、7倍、6倍、5倍、4倍、3倍)であり得る。後述する式(1)で表される化合物では、X(スペーサー)は架橋剤に由来する分子であるが、架橋剤を減少させて化合物を合成すると膨潤度及び膨潤速度が高くなる傾向にあった(データ示さず)。

で表される構造を有するものであることが好ましい。より具体的には、上記「PS1」及び「PS2」は同一であるか又は異なっていてもよく、独立して、βグルカン、キチン、キトサン、及びそれらの誘導体、並びにこれらの混合物からなる群から選択される。当業者であれば、本発明には、例えば、「PS1」がβグリカンであり、「PS2」がキチンである高吸水性高分子化合物、又は「PS1」がβグリカンであり、「PS2」がキトサンである高吸水性高分子化合物が包含されることを理解し得る。

本発明に係る生体分解性及び/又は生体代謝性の腫瘍封止剤に用いる高吸水性高分子化合物は、水分と接触することによって膨潤し、ゲル状組成物を形成することができる。高吸水性高分子化合物は、上述したように、広範な環境条件下で高い安定性を示す。上記高吸水性高分子化合物を腫瘍封止剤として用いる場合、該高吸水性高分子化合物が、生体内の水分によりゲル状物質として元の容積の数十倍〜数百倍に膨潤することによって、腫瘍細胞表面へ物理的に密着する現象を利用する。
本発明に係る生体分解性及び/又は生体代謝性の腫瘍封止剤は、高吸水性高分子化合物を基材とし、腫瘍血管内皮細胞の間隙を透過し得る粒子径を有するナノ微粒子であって、この微粒子がその間隙を透過後、高い生体接着性により間隙近傍に存在する腫瘍組織に付着した後、生体内の水分と接触することによって膨潤し、ゲル状組成物を形成して、腫瘍への酸素と栄養の供給並びに腫瘍からの誘導因子の伝達を遮断することによって、その腫瘍細胞を壊死に至らしめ、その後、付着した微粒子が生体内で分解及び/又は代謝されることを特徴とする。
(1)シリンジ又はマイクロカテーテル通過性
本発明に係る腫瘍封止剤は、前述したように、上記高吸水性高分子化合物をナノ微粒子化して、油性媒体に分散したエマルションを含む。本エマルションによって、シリンジ又はマイクロカテーテルの通過性を確保している。
(2)腫瘍血管透過性
腫瘍組織に特異的に形成された腫瘍新生血管は、血管を覆うペリサイト(血管周皮細胞)が減少しているため、腫瘍血管における内皮細胞間の間隙は、正常細胞と比較して広がっている。その間隙を漏れ出る粒子が存在する。本発明の腫瘍封止剤は、その間隙を通過するのに十分小さな粒子径を持つ。間隙の大きさは個人差があり、腫瘍種別、部位にもよるが、一般に5nm以上700nm以下とされている。
(3)異なる粒子径で構成される分散性
上記腫瘍血管の間隙は大小様々である。本発明の腫瘍封止剤は、粒子径は単一ではなく数百nm幅の分布をもつエマルションのため、どの間隙からも透過することによって集積密度を増加させることができる。
(4)腫瘍組織集積性
前述のように腫瘍血管を透過した微粒子は、腫瘍周辺に放出されるが、正常組織と異なり腫瘍組織では、リンパ管網が未発達である。このため体液と共にリンパ管経由で、他の部位に運ばれることは無い。従って、放出された微粒子即ち本発明の腫瘍封止剤は、腫瘍部位に留まる集積性がある。前記(2)と(4)を合わせてEPR効果と呼ぶ。
(5)酸素と栄養の遮断性
本発明の腫瘍封止剤がゲル状態となって集合体化するため、稠密に腫瘍組織を封止することによって、腫瘍細胞は増殖に必要な栄養と酸素が遮断され、細胞死(壊死)に至る。
(6)伝達系転写誘導因子の伝達阻害性
腫瘍組織・細胞からは、各種誘導因子が発せられる。例えば、低酸素ストレス下のがん細胞から発せられるHIF(低酸素誘導因子)は、VEGF(血管内皮細胞増殖因子)に伝達して腫瘍血管新生を促し、腫瘍への栄養と酸素の供給量を増大させ増殖に寄与している。本発明の腫瘍封剤は、腫瘍組織を隙間なく覆い封じ込めることによって、これら腫瘍から発せられる伝達因子の拡散を阻止することで悪性化を食い止めることができる。
(7)速やかな代謝・排泄
本発明の腫瘍封剤は、腫瘍細胞が壊死するのに十分な時間・期間を経た後は、体内酵素による分解又は可溶化、一部はさらなる分解などを経て、体外へ排泄される。
(8)視認性及び可視化性
(5)までは本発明の腫瘍封剤の基本的な機能に限定した性質であるが、実用上の機能として、医師が使用する場面で有用な機能を付加している。
第一が、目視による視認性である。色素(蛍光色素を含む)を包含することによって、他の医薬品との取り違えリスクを軽減し、更にはマーカーとして手術部位・範囲を表示することで、間違った箇所への施術を回避できる。
第二に、造影剤を包含させることにより、術中の画像装置によるモニタリングで的確な箇所へ十分な施術が行われている又は行われたかを確認することができる。さらには、造影剤のみを使用した時のように流出せずに腫瘍部位に定着するため、再度造影剤を注入する手術をすることなく、術後の腫瘍の状況を画像装置でモニタリングできる可視化性を有する。
(9)塞栓性
本発明の腫瘍封止剤は、動脈塞栓用塞栓材としても使用できる。塞栓材としては、永久塞栓材と同等の塞栓効果を発揮する一時塞栓材として、永久留置リスクを回避でき、更に腫瘍封止効果と塞栓効果との相乗効果を期待することもできる。
上記の通りに調製した腫瘍封止剤は、例えば、固形悪性腫瘍を診断及び/又は治療するために使用することができる。本明細書で使用するとき、「固形悪性腫瘍」には、有棘細胞がん、乳がん、皮膚リンパ腫、血管肉腫、肝胆道系のがん、頭頸部がん、肺がん、中皮腫、縦隔がん、食道がん、胃がん、膵臓がん、小腸がん、結腸がん、結直腸がん、大腸がん、肛門がん、腎臓がん、尿道がん、膀胱、前立腺、尿道がん、陰茎がん、睾丸がん、婦人科臓器のがん、卵巣がん、内分泌系のがん、皮膚がん、脳を含む中枢神経系のがん;軟組織及び骨肉腫;並びに皮膚及び眼内起源の黒色腫を含む。使用形態としては、限定されないが、1種類又は2種類以上の高吸水性高分子を基材とする微粒子を油性媒体中に分散させたエマルションの形態で使用してもよく、又は該微粒子を含む医薬組成物として使用してもよい。このように、本発明の腫瘍封止剤は、固形悪性腫瘍などのがんの診断に適用可能であるだけでなく、がんの治療に応用することもできることから、「セラノスティクス・デバイス」として称することができる。
1gのセルロース(株式会社スギノマシン)を塩化リチウム(LiCl)/N−メチルピロリドン(NMP)溶液(5g LiCl/95g NMP)に添加し、室温で2日間十分に撹拌し溶解させた。触媒としてN,N−ジメチル−4−アミノピリジン(DMAP)を1.14g添加し、架橋剤として1,2,3,4−ブタンテトラカルボン酸二無水物(BTCA)を3.07g加えて、撹拌しながら24時間室温下で反応させた。このエステル架橋反応によって、セルロース分子鎖間に架橋を形成された。その後、反応生成物を1Lのメタノール溶液中に析出させた後、10%水酸化ナトリウム水溶液を用いてpH7.0に調整した。続いて、沈殿物をホモジナイザーで粉砕し、ガラスフィルターを用いてろ過した。ろ過後、メタノールと水を用いて洗浄及びろ過を数回繰り返し、最終的に70℃の条件で減圧乾燥してセルロース高吸水性高分子化合物を得た。なお、使用した試薬は、特に明示されていない場合、いずれも東京化成工業株式会社から購入したものである(以下、同様)。
1.5M水酸化ナトリウム水溶液100mlをビーカー内で撹拌しながら、5gのCMC(三晶株式会社)をダマにならないよう少量ずつ加えて完全に溶解させた。架橋剤としてポリエチレングリコールジグリシジルエーテル(PEGDE)を0.5ml添加し、室温で10分間よく撹拌し、その後、60℃で3時間架橋反応させた。反応生成物のハイドロゲルにメタノール200mlを加え、ホモジナイザーで湿式粉砕した。粉砕後、粒状になったハイドロゲルの粒子を、ガラスフィルターを用いてろ過した。ろ過後、洗浄液のpHが中性付近になるまでメタノールと水を用いて洗浄及びろ過を数回繰り返し、最終的に70℃の条件で減圧乾燥してCMC高吸水性高分子化合物を得た。
(1)乳癌細胞移植マウスの作製
ルシフェラーゼを安定に発現する4T1乳癌細胞(4T1−Luc)を入手し、ヌードマウスBalb/c nu/nu(日本チャールス・リバー株式会社)の皮下に2.5×107細胞/mlのPBS/Geltrex(登録商標)=1/1(vol/vol)の細胞懸濁液を0.04ml移植した。
上記の乳癌細胞を移植されたマウス腫瘍部位に、実施例1で作製した腫瘍封止剤(エマルション)を局所投与した。投与後、ルシフェラーゼの基質であるD−ルシフェリンを含むリン酸緩衝生理食塩水を腹腔内投与により投与して血流循環させ、上記乳癌細胞のルシフェラーゼ(Luc)活性をIn vivoイメージング装置(IVIS)(Spectrum)で経時的に観察した。腫瘍部位では、血中のルシフェリンの存在によりLuc活性に基づいて蛍光が確認される。図3Aに示されるように、エマルションの非投与マウスでは、移植した乳癌細胞のLuc発現により継続的に蛍光を観察できるのに対して、エマルションを投与したマウスでは、投与直後から蛍光が減少したことから、エマルションの投与により、血中のD−ルシフェリンの腫瘍への送達、エマルション投与により腫瘍部位への血流が阻止されることが示唆された。また、この観察結果について、非投与マウスに対する投与マウスにおけるLuc活性の比を算出し、経時的な変化を図3Bとして示す。
エマルションを複数回投与した場合の腫瘍体積の変化について検討した。上記(1)に記載したように、4T1−Lucを移植されたヌードマウスを用いて経時的に観察した。エマルションの投与は、初回(1日目)、2日目、3日目、及び4日目に行った。腫瘍体積の経時的な変化を図4に示す。図4から明らかなように、生理食塩水を投与した対照と比較して、エマルションを投与したマウスの腫瘍体積は減少した。これは、およそ4回投与後から、腫瘍体積の減少は顕著となった。
本実施例では、実施例3で採用された乳癌細胞を皮下移植したマウスを担癌動物モデルとして用い、封止剤として市販のポリアクリルアミド系の高吸水性高分子化合物(粒径:500〜600nm)(三洋化成工業株式会社)(以下、「MD2」と称する場合がある)、及びカルボキシメチルセルロース(CMC)を原料とした高吸水性高分子化合物(粒径:600〜1500nm)(以下、「MD3」と称する場合がある)を用いて、それらによるバリア形成に基づく抗腫瘍効果を検討した。
高吸収性高分子化合物を含む腫瘍封止剤(生理食塩水に含有する)及び生理食塩水のみ(対照)をマウスに局所投与し、腫瘍成長の変化を観察した。
図5に示されるように、生理食塩水(100μl)のみを投与した対照では、腫瘍成長が経時的に観察された。これとは対照的に、生理食塩水に高吸水性高分子化合物を含有させた溶液(10mg/ml)を投与した場合には、腫瘍成長を顕著に抑制することができた。6日目で比較してみると、両者に有意な差が認められた。
(b)CMC系の高吸水性高分子化合物(MD3)の場合
高吸収性高分子の投与前後による腫瘍体積の変化を測定した結果を図6にまとめた。図6Aは、ビヒクルとして生理食塩水のみを用いた場合の結果を示す。投与直前の腫瘍体積を基準として、投与8日後に腫瘍を摘出し、腫瘍体積を比較検討した。生理食塩水のみを投与した対照では、腫瘍は342%に増大したが、高吸収性高分子を投与した対象では、69%まで腫瘍体積が減少した。さらに、ビヒクルとして、生理食塩水+リピオドールを用いた場合の結果を図6Bに示す。図6Aの結果と同様に、腫瘍体積の減少を観察することができたが、リピオドールの含有によって、腫瘍抑制効果は増大した。
本発明の高吸収性高分子化合物(MD3)による腫瘍組織への酸素供給の遮断効果を検証した。実験動物の腫瘍部位における、高吸収性高分子化合物による低酸素状態を可視化するために、インビボ光音響3次元断層イメージングMSOT(MultiSpectral Optoacoustic Tomography)(機種名:MSOT inVision 256−TG;住友ファーマインターナショナル株式会社)を用いた。このシステムでは、ヘモグロビンの酸素飽和度を可視化することにより、画像として、酸素が豊富な酸化ヘモグロビンを赤色、及び脱酸素の還元ヘモグロビンを青色に区別するこができる。該システムによる低酸素状態を可視化した画像を図7に示す。左右の腫瘍に対して、高吸収性高分子化合物を投与していない対照の腫瘍(左側)では、酸素が豊富な状態(赤色)として確認できた。これに対して、高吸収性高分子化合物を投与した腫瘍(右側)では、脱酸素の状態(青色)として視認され、腫瘍部位は低酸素状態にあることが確認できた。これにより、本発明の高吸収性高分子化合物が封止剤として抗腫瘍効果を有することが示された。
本発明の高吸収性高分子化合物による腫瘍組織への血中の各種物質の供給遮断は、生体内の水分と接触による高吸収性高分子化合物の膨潤によるものと考えられる。これを検証するために、本実施例では、インビトロにおいて高吸収性高分子化合物をゲル化し、ゲルに対するタンパク質及び栄養素の通過(透過性)を指標として遮断効果を検討した。まず、高吸収性高分子化合物(MD3)のゲルをPBS(−)を用いて調製した(最終濃度:0.01g/mL)。次に、底を綿栓した2.5mlのシリンジに、上記で調製したゲルを所定の容積(0.2、0.5、及び1.0ml)で注入した。続いて、ウシ胎児血清(FBS)(100%又は10%)をシリンジ内のゲル上に充填した。その後、ゲルを通過したFBSを回収し、該FBS中に残存する各成分(タンパク質/栄養素、ヘモグロビン、及びヘモグロビン)の量を、対照の各成分量と比較することによって、ゲルによる封止効果を検証した。なお、対照は、ゲルを充填せずに、底に綿栓をしたシリンジを通過させたFBSである。
ゲルを通過して、回収されたFBS中に残存するタンパク質の量を紫外分光光度計により測定し、標準曲線から求めた。ゲルによって遮断されたタンパク質の割合は、対照のFBS中に存在するタンパク質の量とゲルを通過させた後のFBSに残存する量とを比較することにより算出した。より具体的には、以下の式:
遮断されたタンパク質(%)=(対照中のタンパク質の量−ゲルを通過させた後に残存するタンパク質の量)×100/対照中のタンパク質の量
により求めた。結果を図8に示す。希釈されたFBS(10%)を用いた場合、ゲルが飽和に至っていないため、タンパク質の遮断効果が顕著に見られた。希釈していないFBS(100%)を用いた場合には、ゲルが飽和した状態であるため、希釈したFBSと比較して、タンパク質の遮断効果は小さかった。容積が0.5ml及び1.0mlのMD3を用いた場合には、有意差が認められた。
上記(1)のタンパク質/栄養素の遮断実験と同様にして、ゲルによるヘモグロビンの遮断効果について検討した。結果を図9に示す。容積が0.5mlと1.0mlのMD3を用いた場合には、ヘモグロビンの遮断効果に有意差が認められた。
上記(1)のタンパク質/栄養素の遮断実験と同様にして、ゲルによるグルコースの遮断効果について検討した。結果を図10に示す。1.0mLのMD3を用いた場合、約80%のグルコースがゲルに捕捉されたが、使用したゲルの量により、その効果は顕著に異なった。また、装填したFBSの濃度の違いによって、遮断効果に差は生じなかった。
4T1−Luc細胞を移植したマウスを用いて、さらに高吸収性高分子化合物の投与による腫瘍血管の阻血効果(酸素濃度減少)を検証した。該高吸収性高分子化合物(MD3)の投与から1日後に、光音響3次元断層イメージングで腫瘍内の酸素状態を測定すると、MD3を投与した側の腫瘍組織(左)では、還元型ヘモグロビン(青色)が増加していた(図11)。これにより、腫瘍組織内で血中酸素濃度が減少していることが観察され、血流が遮断されたことが推定された。
Claims (5)
- 高吸水性高分子化合物を基材とするナノ微粒子を含む、腫瘍封止剤であって、
当該高吸水性高分子化合物を基材とするナノ微粒子は、水分により膨潤し、その膨潤度は、乾燥時における基材の体積の少なくとも2〜1000倍であり、
当該高分子化合物は、分子間で架橋され、
ナノ微粒子は、20nm以上700nm以下であり、
腫瘍封止剤は、固形悪性腫瘍組織内に投与されるか、固形悪性腫瘍近傍に投与されるか、又は固形悪性腫瘍への動脈に投与されるものであり、当該投与は、ナノ微粒子が、腫瘍部位に集積し、かつ生体内の水分と接触することによって膨潤して、当該腫瘍への血流または栄養供給を減少させ、腫瘍を壊死させるように行われる、
腫瘍封止剤。 - 高吸水性高分子化合物を基材とするナノ微粒子を含む、腫瘍封止剤であって、
当該高吸水性高分子化合物を基材とするナノ微粒子は、水分により膨潤するものであり、その膨潤度は、乾燥時における基材の体積の少なくとも2〜1000倍であり、

腫瘍封止剤。 - フリーズドライされた、請求項1または2に記載の腫瘍封止剤。
- 前記ナノ微粒子が油性媒体に分散させたエマルション形態である、請求項1または2に記載の腫瘍封止剤。
- 薬剤、造影剤、および色素からなる群から選択される機能性物質をさらに包含する、請求項1〜4のいずれか一項に記載の腫瘍封止剤。
