Description
本出願は、2011年1月18日に出願された米国仮出願61/433,890号および2011年8月24日に出願された米国仮出願61/527,049号に基づく優先権を主張するものであり、その内容全体が参照により本明細書に組み込まれる。
最近の研究では、γcサイトカインの発現調節異常および機能異常によって、様々なヒトの免疫疾患および造血器疾患が引き起こされている可能性が示されている。
IL−2は、T細胞増殖因子のプロトタイプであると歴史的に考えられてきたが、IL−2の発現を欠くノックアウトマウスの作製によって、IL−2はインビボにおける通常のT細胞の増殖および発生には重要ではないことが明らかとなっている。しかしながら、IL−2の過剰発現は、T細胞のサブセットである制御性T細胞(Treg)の選択的増加につながる(Antonyら, 2006, J. Immunol. 176:5255-66参照)。Tregは、他の細胞の免疫応答を抑制することにより末梢性寛容を維持する作用を発揮している(Sakaguchiら, 2008, Cell 133:775-87における検討)。末梢性寛容の破綻はヒトにおいて自己免疫疾患を引き起こすと考えられている。したがって、Tregの免疫抑制能は自己免疫疾患の発症を防いでいると考えられている(Sakaguchiら, 2008, Cell 133:775-87参照)。また、Tregはがんにも関与しており、充実性腫瘍および血液悪性腫瘍はTreg数の増加を伴う(De Rezendeら, 2010, Arch. Immunol. Ther. Exp. 58:179-190参照)。
IL−4は、ヘルパーT細胞からTh2(ヘルパーT2型)サブセットへの分化に関与する非重複サイトカインであり、Th2は未熟B細胞からIgE産生形質細胞への分化を促進する。IgEレベルはアレルギー性喘息において上昇する。したがって、IL−4はアレルギー性喘息の発症に関連する。IL−4を標的とする抗体はアレルギー性喘息の治療、さらには発症の予防に使用することができる(Le Buanecら, 2007, Vaccine 25:7206-16参照)。
IL−7は、B細胞の発生および胸腺におけるT細胞の初期発生に必須である。マウスにおいては、IL−7の異常発現によってT細胞性白血病が引き起こされる(Fisherら, 1993, Leukemia 2:S66-68参照)。しかしながら、ヒトでは、IL−7の制御異常はT細胞性白血病を引き起こさないように思われる。ヒトにおいて、IL−7のアップレギュレーションは、単独でも別のγcサイトカインファミリーメンバーであるIL−15のアップレギュレーションとの組み合わせにおいても、大型顆粒リンパ球性(LGL)白血病と関連付けられている。
IL−9の役割は、γcサイトカインファミリーの他のメンバーと比べて未だ明らかになっていない点が多い。IL−9遺伝子欠失マウスは一見正常であり、リンパ球コンパートメントおよび造血コンパートメントにおける細胞サブセットの欠失も見られない。しかしながら、最近の研究では、IL−9がインビボにおいてTh17(インターロイキン17によって誘導されるヘルパーT)細胞の発生に関与していることが明らかとなっている(Littmanら, 2010, Cell 140(6):845-58;およびNowakら, 2009, J. Exp. Med. 206: 1653-60参照)。
IL−15は、NK細胞、NKT細胞、上皮内リンパ球(IEL)のサブセットのいくつか、γδ T細胞、およびメモリー型CD8T細胞の発生において極めて重要な役割を果たしている(Waldmann, 2007, J. Clin. Immunol. 27:1-18;およびTagayaら, 1996, EMBO J. 15:4928-39参照)。マウスにおけるIL−15の過剰発現は、NKT細胞型T細胞白血病およびCD8細胞型T細胞白血病を引き起こす(Fehnigerら, 2001, J. Exp. Med. 193:219-31;およびSatoら, 2011 Blood(近日中に刊行予定)参照)。いずれの文献においても白血病細胞がCD8抗原を発現していることから、これらの文献において実験的に誘導された白血病はヒトのLGL(大型顆粒リンパ球)白血病に類似していると考えられる。
IL−21はγcファミリーのメンバーのうち最も新しく発見されたサイトカインである。γcファミリーの他のメンバーとは異なり、IL−21は強力な成長促進効果を有するとは考えられていない。IL−21は細胞増殖制御因子としてよりも、むしろ分化因子として機能すると考えられている(Tagaya, 2010, J. Leuk. Biol. 87:13-15参照)。
γcサイトカインは多くのヒト疾患に関連すると考えられており、このためγcサイトカインファミリーの活性を阻害してγcサイトカイン関連疾患を治療する方法が提案されている。このような方法として、インビボにおいてサイトカイン特異的モノクローナル抗体を使用して標的とするサイトカイン活性を中和する方法;特定のサイトカインに固有のサイトカイン特異的受容体サブユニット(共有γcサブユニット以外のサブユニット)を標的とするモノクローナル抗体を使用して選択的にサイトカイン活性を阻害する方法;および細胞内のサイトカイン信号伝達経路の下流をブロックする化学阻害剤を使用する方法が挙げられる。治療剤を設計する際には、サイトカイン特異的抗体が第一の選択肢となる場合が多い。しかしながら、受容体の構成成分を共有するサイトカインは重複した機能を示し(Paul, W.E., 1989, Cell 57:521-24参照)、2種以上のサイトカインが同時に作用して疾患を引き起こしていることもある(後述の例を参照のこと)。このことから、1種のサイトカインのみの中和を対象とする手法は、サイトカイン関連ヒト疾患の治療に効果的ではない可能性がある。
γcサブユニットとγcサイトカインとの相互作用によって、ヤヌスキナーゼ3(Jak3)と称される細胞内タンパク質チロシンキナーゼが活性化される。次いで、Jak3は、STAT5やPI3キナーゼなどの複数種のシグナル伝達分子をリン酸化する。γcサブユニットとJak3との相互作用は非常に特異的である。実際に、シグナル伝達のためにJak3を必要とする受容体分子は他に存在しない(O'Shea, 2004, Ann. Rheum. Dis. 63:(suppl. II):ii67-7参照)。したがって、γcサブユニットを介したサイトカインシグナル伝達の阻害は、Jak3キナーゼの活性をブロックすることにより達成することができる。これに応じて、Jak3キナーゼ活性を標的とする複数種の化学阻害剤が市場に導入されている(Pesuら, 2008, Immunol. Rev. 223:132-142参照)。その一例としてCP690,550が挙げられる。
γcサイトカインは、免疫系を構成するリンパ系細胞(特にT細胞、B細胞およびNK細胞)の発生において重要な役割を果たしている。さらに、γcサイトカインは様々なヒト疾患に関連している。したがって、γcサイトカイン活性を阻害する因子は、リンパ球サブセットの発生機序を解明し、かつ免疫障害およびγcサイトカイン媒介性疾患を治療するための有用なツールになると考えられる。
γcサイトカインのC末端(Dヘリックス)には、マルチユニットサイトカイン受容体の共通γcサブユニットと相互作用するとされている部位が含まれている(Bernardら, 2004 J. Biol. Chem. 279:24313-21)。マウスおよびヒトにおいて同定されたすべてのγcサイトカインのアミノ酸配列について生化学的特性を比較することによって、これらのアミノ酸配列の疎水性、親水性、塩基性/酸性などの化学的性質がγcサイトカインファミリーのメンバー間で保存されており、これらの化学的性質が全く同一でない場合でもDヘリックスの多くの位置において保存されていることが明らかとなっている。これとは対照的に、γcサイトカインであるIL−4と関連性を有しているがγcサブユニットとは結合しないIL−13の配列は、Dヘリックス領域においてγcサイトカインと有意な相同性を示さない。このことは、Dヘリックス領域における配列相同性がγcサブユニットに対する結合性と相関することを示唆している。図1に示すように、ヒトγcサイトカインファミリーメンバーのDヘリックス領域のアミノ酸配列をアライメントすると、これらのサイトカインにおいて中程度の配列相同性を有するモチーフ(本明細書において「γcボックス」と呼ぶ)が存在することが分かる。
γcファミリーサイトカインの活性は、γcサイトカインとγcサブユニットとの相互作用を破壊することによりブロックしてもよい。たとえば、マルチサブユニットサイトカイン受容体のシグナル伝達を刺激することなく、γcサブユニットと相互作用することが可能な競合阻害剤を導入することにより上記のブロッキングを行ってもよい。特定の理論に拘束されることを望むものではないが、γcファミリーサイトカインのγcサブユニットへの結合に関与する保存γcボックスモチーフは、γcサイトカインシグナル伝達のペプチド阻害剤の設計に利用することが可能な、コアとなる基本的なアミノ酸配列を提示するものである。
で示されるアミノ酸配列からなり、太字で表記したアミノ酸はIL−2とIL−15との間で保存されており、下線を引いたアミノ酸は当該アミノ酸の生理化学的特性が保存されている位置を示している。
実施形態のいくつかは、γcサイトカイン媒介性疾患の治療におけるγcアンタゴニストペプチドの使用に関する。本発明の実施形態によるカスタムペプチド誘導体を使用することにより、治療剤の設計(ペプチドのカスタム設計)を柔軟に行うことができ、抗サイトカイン抗体または抗サイトカイン受容体抗体を用いた従来の戦略では達成が不可能なより包括的な成果を上げることが可能となる。
本発明の実施形態はさらに、疾患治療剤を製造するためのγcアンタゴニストペプチドの使用を包含する。本発明の実施形態はさらに、γcアンタゴニストペプチドと薬学的に許容される担体とを組み合わせて含む医薬組成物を包含する。該医薬組成物は、薬学的に許容される担体、および無毒性かつ治療に有効な量のγcアンタゴニストペプチド、または本発明の実施形態の別の組成物を含むことができる。
本明細書において「患者」は治療処置のレシピエントを指し、動物界の生物すべてを含む。好ましい実施形態では、上記の動物は、ヒト、ウシ、ヒツジ、ブタ、ネコ、スイギュウ、イヌ、ヤギ、ウマ、ロバ、シカ、霊長類などの哺乳動物科の動物である。動物としてはヒトが最も好ましい。
(i)標的とする生物学的病態に関連する症状をまだ示していないが、その恐れのある患者における、該生物学的病態に関連する症状の発生の予防もしくは遅延;
(ii)標的とする生物学的病態を有すると診断された患者における、該生物学的病態に関連する症状の改善;
(iii)標的とする生物学的病態を有すると診断された患者またはその恐れのある患者における、該生物学的病態に関連する合併症、病態もしくは疾患に関連した症状の発生の予防、遅延もしくは改善;
(iv)標的とする生物学的病態の進行速度の遅延、進行の遅延、もしくは進行の阻止;または
(v)細胞における炎症の予防、炎症の遅延、炎症の拡大の遅延、炎症の阻止、もしくは炎症の改善
を包含する。
(1)プロモーターおよびエンハンサーなどの、遺伝子発現を制御する単一または複数の遺伝学的要素、
(2)本発明の実施形態のポリペプチドをコードする構造またはコード配列、ならびに
(3)適切な転写開始配列および翻訳開始配列、および所望に応じて終止配列
からなるアセンブリを含む転写単位を含むプラスミドを指す。酵母系および哺乳動物系における使用を意図した構成要素として、酵母宿主細胞または哺乳動物宿主細胞において翻訳されたポリペプチドを細胞外分泌させることが可能なシグナル配列が含まれていることが好ましい。
γcアンタゴニストペプチドの阻害活性の評価方法
哺乳動物細胞アッセイを使用してγcサイトカインファミリーメンバーに対する哺乳動物細胞の増殖応答を測定することにより、本発明の実施形態に従って調製した、γcサイトカインファミリーの1メンバーの作用を阻害するカスタムペプチド誘導体の能力を測定する。
BNZ−γペプチドによるIL−9およびIL−15の増殖促進活性の特異的阻害
上記のPT−18β細胞を使用して、選択したγcサイトカインの増殖促進活性に対するBNZ−γペプチドの特異的阻害能を測定した(図3A)。PT−18β細胞の増殖を補助する非γcサイトカインであるIL−3をネガティブコントロールとして使用した。簡潔に述べると、HEK293T細胞により産生されたBNZ−γペプチドを異なる希釈倍率で希釈し(BNZ−γ発現構築物でトランスフェクトしたHEK293T細胞の上清の1:20または1:50希釈液)、これらの希釈液のいずれかとともにまたはBNZ−γペプチドを使用せずにIL−3、IL−9、IL−15またはIL−4(培養物中の各サイトカインの濃度:1nM)の存在下でPT−18β細胞をインキュベートした。BNZ−γペプチドおよびサイトカインを導入した2日後に、WST−1アッセイを用いて細胞の増殖応答を測定した。IL−3(非γcサイトカイン)の増殖促進活性はBNZ−γによって阻害されなかった。これに対して、IL−15およびIL−9の活性は、BNZ−γペプチドによって顕著に低下した(p<0.01、スチューデントt検定)。別のγcサイトカインであるIL−4によって刺激された細胞増殖は、BNZ−γペプチドの添加による影響を受けなかった。IL−3、IL−9、IL−15およびIL−4についての結果を図3Aに示す。
細胞増殖マーカーとしての 3 Hチミジンの取り込みを分析することによる、γcサイトカイン活性の阻害の測定方法
3Hチミジン取り込みアッセイを用いて、γcサイトカインにより誘導された指標細胞集団の増殖に対するアンタゴニストカスタムペプチド誘導体による阻害を測定する。簡潔に述べると、サイトカイン存在下において増殖中の20〜50,000個の細胞に、放射性同位体で標識したチミジン(1μCi)を添加する。慣用のハーベスター装置(たとえば、パーキンエルマー社製Filtermateユニバーサルハーベスター)を使用して、細胞に結合した放射能をガラス繊維フィルターで捕捉し、次いで、βカウンター(たとえば、1450Triluxマイクロプレートシンチレーションカウンター)を使用して放射能を測定することにより、細胞に取り込まれた放射能を測定する。
細胞増殖マーカーとしての細胞追跡色素の取り込みを分析することによる、γcサイトカイン活性の阻害の測定方法
選択したγcサイトカインの存在下、または選択したγcサイトカインおよび選択したカスタムペプチド誘導体の存在下で指標細胞をインキュベートする。次いで、インビトロにおいて、たとえばCMFDA(インビトロジェン社製C2925)などの細胞追跡色素を使用して細胞集団を標識し、細胞分裂ごとの細胞内緑色蛍光の減衰をフローサイトメーター(たとえばベクトン・ディッキンソン社製FACScalibur)を使用してモニターする。通常、γcサイトカインによる刺激に応答して、細胞分裂回数に対応する7〜10個の異なるピークが緑色蛍光チャネルに現われる。選択したγcサイトカインおよびアンタゴニストカスタムペプチド誘導体とともにインキュベートした細胞では、ピークの数は1〜3個にまで低減され、その数は阻害の程度によって決まる。
アンタゴニスト(BNZ−γおよびその誘導体)による細胞内シグナル伝達の阻害
γcサイトカインとその受容体との結合は、細胞増殖を刺激するだけでなく、多様な細胞内事象を引き起こす(Rochmanら, 2009 Nat. Rev. Immunol. 9:480-90およびPesuら, 2005 Immunol. Rev. 203:127-142)。γcサイトカインがその受容体に結合すると、即座にJak3(ヤヌスキナーゼ3)と称されるチロシンキナーゼが形質膜上の受容体に動員される。このキナーゼは、γcサブユニット、STAT5(シグナル伝達兼転写活性化因子5)、PI3(ホスファチジルイノシトール3)キナーゼのサブユニットなどの多種類のタンパク質のチロシン残基をリン酸化する。これらのタンパク質のリン酸化のうち、STAT5のリン酸化は、γcサイトカインによって引き起こされる細胞増殖と関連することが多くの研究において示唆されている(HennighausenおよびRobinson, 2008 Genes Dev. 22:711-21における検討)。公表されているこれらの資料に従って、BNZ−γペプチドが、IL−15によって刺激されたPT−18β細胞におけるSTAT5分子のチロシンリン酸化を阻害するかどうかを試験した(図3Cに結果を示す)。
BNZ−γから誘導されるアンタゴニストペプチドの論理的設計
ペプチド誘導体は、D/E−F−L−E/Q/N−S/R−X−I/K−X−L/I−X−Q(配列番号2)(式中、Xは任意のアミノ酸を示す)で示されるコア配列の特定のアミノ酸を、図2に示したような同一の生理化学的特性を有するアミノ酸で置換することにより調製する。
アンタゴニストカスタムペプチド誘導体の阻害特異性を同定する方法
6種のγcサイトカインに応答したサイトカイン応答性細胞株の増殖に対するカスタムペプチド誘導体の阻害能を分析することによって、アンタゴニストカスタムペプチド誘導体のγcサイトカイン阻害特異性を決定する。たとえば、マウス細胞株CTLL−2は、候補ペプチドがIL−2およびIL−15の機能を阻害するかどうかを判断するために使用される。PT−18(β)細胞は、候補ペプチドがIL−4およびIL−9の機能を阻害するかどうかを判断するために使用される。PT−18(7α)細胞は、候補ペプチドがIL−7の機能を阻害するかどうかを判断するために使用される。PT−18(21α)細胞は、候補ペプチドがIL−21の機能を阻害するかどうかを判断するために使用される。PT−18(β)は、遺伝子導入によりヒトIL−2Rβを外因的に発現するPT−18細胞のサブクローンである(Tagayaら, 1996参照)。PT−18(7α)は、遺伝子導入によりヒトIL−7Rαを発現するサブクローンである。PT−18(21Rα)細胞はヒトIL−21Rαを発現する。
γcアンタゴニストペプチドの調製
カスタムγcアンタゴニストペプチド誘導体は、手動の工程および自動の工程によって化学的に合成される。
1つのアミノ酸のカルボキシル基またはC末端を、別のアミノ酸のアミノ基またはN末端とカップリングすることを含む古典的な液相合成を使用する。別の方法においては、固相ペプチド合成(SPPS)が使用される。
多数の民間企業が有償で自動ペプチド合成を提供している。このような会社では、アプライドバイオシステムズ(ABI)より提供されている合成機などの様々な市販のペプチド合成機が使用されている。カスタムγcアンタゴニストペプチド誘導体は自動ペプチド合成機で合成される。
組換え技術を使用したカスタムγcアンタゴニストペプチド誘導体の生物学的製造
カスタムγcアンタゴニストペプチド誘導体は、BNZ−γペプチドの構造を強化もしくは安定化させ、その生物学的活性を向上させる公知のヒトタンパク質由来ペプチド、適切なタグペプチド、およびシグナルペプチドを含むペプチド前駆体として生物学的に合成される。所望に応じて、ペプチドのN末端の前に適切な酵素切断配列を設計し、最終的に得られるタンパク質からタグペプチドまたは任意のペプチド部分を取り除いてもよい。
免疫化を目的としたBNZ−γおよびその誘導体の担体タンパク質との共役ならびに抗BNZ−γ抗体の作製
BNZ−γおよび他のカスタムペプチド誘導体は、動物を免疫化してポリクローナル抗体およびモノクローナル抗体を得るために用いられる。グルタルアルデヒドまたはm−マレイミドベンゾイル−N−ヒドロキシスクシンイミドエステルを使用した慣用の方法によって、適切な担体タンパク質(たとえば、ウシ血清アルブミン、キーホールリンペットヘモシアニン(KLH)など)のN末端またはC末端にペプチドを共役させる。次いで、適切なアジュバントとともに共役ペプチドを用いて、ウサギ、げっ歯類、ロバなどの動物を免疫化する。慣用の方法を用いて、得られた抗体の特異性を試験する。得られた抗体が免疫原性ペプチドと反応する場合、実施例1〜3に記載の細胞増殖アッセイを用いて各γcサイトカイン活性に対する阻害能を試験する。ペプチド誘導体が複合体であることから、2種の異なるサイトカインを同時に認識する単一の抗体を作製することが可能である。
カスタムγcアンタゴニストペプチド誘導体の大量生産方法

Claims (17)
- I/P−K/V−E−F−L−E/Q/N−S/R−F/W−V/I/K−H/T/S−L/I/F−V/C/L−Q−M/S/K−F/I/M−I−N/S/H−T/Q−S/L/Hで示されるアミノ酸配列を含み、かつIL−2、IL−15およびIL−21からなる群から選択される2種以上のγcサイトカインの活性を阻害することができる単離または精製されたペプチドであって、該ペプチドが19〜50アミノ酸の長さを含む単離または精製されたペプチド。
- IL−15の活性を阻害することを特徴とする請求項1に記載の単離または精製されたペプチド。
- 細胞内でγcサイトカインファミリーの2以上のメンバーによるシグナル伝達をブロックするための薬剤を製造するための請求項1又は2に記載の単離または精製されたペプチドの使用。
- γcサイトカインのγcサブユニットへの結合を阻害するための薬剤を製造するための請求項1又は2に記載の単離または精製されたペプチドの使用。
- 治療有効量の請求項1又は2に記載の単離または精製されたペプチドを含む医薬組成物。
- γcサイトカイン媒介性疾患を改善または治療するための、請求項5に記載の医薬組成物。
- 前記γcサイトカイン媒介性疾患が、CD4白血病、CD8白血病、LGL白血病、全身性エリテマトーデス、シェーグレン症候群、ヴェーゲナー肉芽腫症、セリアック病、橋本甲状腺炎、関節リウマチ、炎症性腸疾患、糖尿病、乾癬、多発性硬化症、ブドウ膜炎、眼炎症、および移植片対宿主病(GvHD)からなる群から選択されることを特徴とする請求項6に記載の医薬組成物。
- HTLV−1関連脊髄症(HAM)/熱帯性痙性不全対麻痺(TSP)に関連する疾患を改善または治療するための、請求項5に記載の医薬組成物。
- 前記HAM/TSPに関連する疾患が、成人T細胞性白血病(ATL)、HTLV関連脊髄症/熱帯性痙性不全対麻痺(HAM/TSP)、ならびにHTLVに関連する他の非新生物性炎症性疾患(ブドウ膜炎(HU)、関節症、肺疾患、皮膚炎、外分泌腺症および筋炎)からなる群から選択されることを特徴とする請求項8に記載の医薬組成物。
- 炎症性呼吸器疾患を改善または治療するための、請求項5に記載の医薬組成物。
- 前記炎症性呼吸器疾患が、喘息、副鼻腔炎、花粉症、気管支炎、慢性閉塞性肺疾患(COPD)、アレルギー性鼻炎、急性耳炎、慢性耳炎、および肺線維症からなる群から選択されることを特徴とする請求項10に記載の医薬組成物。
- ざ瘡の治療、脱毛の治療、日焼けの治療、爪のケア、および外見の老化の軽減からなる群から選択される美容目的用の軟膏であって、治療有効量の請求項1又は2に記載の単離または精製されたペプチドを含む軟膏。
- γcサイトカインの活性を調節する請求項1又は2に記載の単離または精製されたペプチドを設計する方法であって、
少なくとも2種のILタンパク質のそれぞれのγcボックスDヘリックス領域の部分配列を含む配列を得ること;および、
少なくとも2種のILタンパク質のγcボックスDヘリックス領域の部分配列を含む配列を含む単離または精製されたペプチドを構築することを含み、
該配列が19〜50アミノ酸を含み、
該単離または精製されたペプチドが1以上のγcサイトカイン活性を調節するものである方法。 - 前記ペプチドがN末端での共役、C末端での共役または側鎖残基での共役の1つ以上をさらに含む、請求項1又は2に記載の単離または精製されたペプチド。
- 前記ペプチドがシグナルペプチドをさらに含む、請求項1、2又は14に記載の単離または精製されたペプチド。
- ペプチドの構造を安定化させ、その生物学的活性を向上させるタンパク質をさらに含む、請求項5に記載の医薬組成物。
- 前記タンパク質が、BSA、アルブミン、IgGのFc領域、スキャフォールドとして機能する生体タンパク質、ポリエチレングリコール(PEG)またはそれらの誘導体からなる群から選択される、請求項16に記載の医薬組成物。
