しかしながら、一般に100nm未満のサイズのリポソームを作製できるという報告は少ない。従って、ドラッグデリバリーの効率を向上するのを目的に、ナノ材料のサイズをより小さくすることが期待できる新たな生体材料を開発することが求められている。

(定義)
以下に、本明細書および特許請求の範囲中で使用する用語の定義を示す。
グリセロリン脂質としては、例えばホスファチジルグリセロール、ホスファチジルセリン、ホスファチジルコリン、及びホスファチジルエタノールアミンを挙げられ、ホスファチジルコリンが好ましい。アシル基におけるアルキル基の炭素数が約9〜約23(つまり、炭素数が約10〜24の脂肪酸基)と、好ましくは約11〜約17(つまり、炭素数が約12〜18の脂肪酸基)と、より好ましくは約13〜約17(つまり、炭素数が約14〜18の脂肪酸基)と長鎖のグリセロリン脂質が好ましい。ここで、該脂肪酸基は1個以上の炭素−炭素不飽和二重結合を含んでいてもよいが、該炭素−炭素不飽和二重結合を含まないことが好ましい。典型的な例としては、ジラウロイルホスファチジルコリン(DLPC)、ジミリストイルホスファチジルコリン(DMPC)、ジパルミトイルホスファチジルコリン(DPPC)、ジステアロイルホスファチジルコリン(DSPC)、エッグホスファチジルコリン(PC)、および1−パルミトイル−2−オレオイルホスファチジルコリン(POPC)を挙げられるが、これらに限定されるものではない。当該リン脂質は、単独でまたは2種以上を組み合わせて使用することができる。本発明におけるリン脂質とは、例えば脂質膜の硬さを調整し、また安定化した複合体が形成する、さらには薬物等の化合物を内包するのに寄与する役割を果たしているものと考えられる。
本発明における細胞親和性ペプチドとは、天然のタンパク質由来の天然ペプチドおよび人工的に製造する合成ペプチド(例えば、キメラペプチド)を含む。細胞親和性ペプチドは、そのアミノ酸配列による化学的特性により、塩基性細胞親和性ペプチド、両親媒性細胞親和性ペプチド、および疎水性細胞親和性ペプチドを含む。塩基性細胞親和性ペプチドが好ましく、塩基性細胞膜透過性ペプチドがより好ましい。例えば、塩基性細胞親和性ペプチド(例えば、HIV−1ウイルスのTATタンパク質(Trans-activator of transcription protein)由来のTATペプチド、ショウジョウバエ由来のペネトラチン(PEN)、ポリアルギニン(例えば、アルギニンの8量体(R8))、ポリヒスチジン((例えば、ヒスチジンの16量体(H16))、抗菌性ペプチド由来のLL−37ペプチド)、両親媒性細胞親和性ペプチド(例えば、キメラペプチドであるトランスポータンやPep−1ペプチド)、および疎水性細胞親和性ペプチド(例えば、ファージディスプレー法によって得られる合成ペプチドであるPep−1ペプチド、膜タンパク質のシグナルペプチド由来のペプチドであるミトコンドリアターゲティングシグナル(MTS))からなる群から選ばれる少なくとも1種を挙げられるが、これらに限定されるものではない。
アポリポタンパク質を例えば約0.01〜0.5μmoL/L、典型的には約0.05〜0.1μmoL/L、
リン脂質を例えば約0.5〜250μmoL/L、典型的には約2.5〜20μmoL/L、および
適宜、アポリポタンパク質と等量のモル比で融合した細胞親和性ペプチド
を含む。
また、適宜、蛍光標識物質、生体活性物質または薬物から選ばれる少なくとも1種類の化合物を、例えば約0.01〜25μmoL/L、典型的には約0.02〜約10μmoL/L、約0.05〜約2μmoL/L(例えば、0.03μmoL/L、0.05μmoL/L、0.1μmoL/L、0.2μmoL/L)の割合で含んでいてもよい。
本発明の目的の細胞親和性ペプチドを結合(例えば、融合)した高密度リポタンパク質(cHDL)の製造方法の1例としては、
i)アポリポタンパク質に、細胞親和性ペプチドを遺伝子工学的手法により、融合させて、融合タンパク質を得る;そして、
ii)得られた融合タンパク質に、リン脂質、更に必要に応じて蛍光標識物質、生体活性物質または薬物から選ばれる少なくとも1種類の化合物を含むリポソームを混合することにより、粗cHDLを製造する(Spontaneous interaction法);あるいは、リポソーム形成を経由しない、コール酸ミセルとの混和(例えば、リン脂質の相転移温度で混和する)により、粗cHDLを製造する(コール酸透析(cholate-dialysis)法);
iii)超遠心法により、未反応のリポソーム、リン脂質ミセル、アポリポタンパク質を除去することにより、粗cHDLを精製する、
ことを含む。

融合前の高密度リポタンパク質(HDL)に対して等モル比の該細胞親和性ペプチドを仕込むことにより、融合タンパク質を作製する。
次に、仕込み量として、融合前の高密度リポタンパク質(HDL)の1モルに対して、数倍〜数千倍モル、数十倍〜数百倍モル、例えば約5倍〜約2000倍モル、約20倍〜約500倍モル、約30倍〜約200倍モルのリン脂質を仕込むことにより、cHDLを調製する。
以下の方法により、高密度リポタンパク質(HDL)を作製した。
細胞親和性HDL(cHDL)の作製
(方法)
遺伝子改変型apoA−IのC末端に、細胞親和作用を有するTAT(Trans-activator of transcription protein)ペプチドをつなげた融合タンパク質を作製した。この融合型apoA−Iに、リン脂質としてジミストリルホスファチジルコリン(DMPC)と蛍光標識物質としてのクマリン−6からなる粒径100nm程度の小サイズのリポソームを混和することにより、cHDLを作製した(Spontaneous interaction法)。遺伝子改変型apoA−Iとリポソームの混和物から反応しなかったリポソームやリン脂質ミセル、またリン脂質や蛍光物質と結合していないアポリポタンパク質を除去するために、超遠心法によりcHDLを精製した。
同様な方法で、比較対照として、TATペプチドを含まないリコンビナントHDL(rHDL)をも作製した。また、比較対照として、リポソームのssLip(submicron sized small uni-lamella vesicle)のジステアロイル ホスファチジルコリン(DSPC)を国際公開第2009/107753号パンフレットに記載の方法に従って調製した。上記の得られたHDLおよび調製したDSPCのそれぞれの組成を、タンパク質、リン脂質およびクマリン−6の各含量、並びに粒径、表面電位について調べた。タンパク質はローリー(Lowry)法で、リン脂質はCテスト(C test Wako(登録商標))で、クマリン−6は蛍光分光光度計FluoroMaxを用いる蛍光分光法で、粒径および表面電荷状態は動的光散乱法(DLS)を用いて、それぞれ体積平均径(MV)およびゼータ電位として求めた。
Spontaneous interaction法と超遠心法により、粒径が直径10〜20nmの小サイズのHDLを得ることができた。各HDLのサイズおよびそれを構成するタンパク質、リン脂質およびクマリン−6の各濃度、並びにゼータ電位は下記表の通りである。
次に、製造した各HDLを用いた膜毒性試験を行った。
cHDLの毒性評価
(方法)
得られた各HDLの角膜毒性を調べるために、ヒト角膜培養細胞を使って細胞毒性試験を行った。該試験は、cHDL、rHDL、および比較対照の塩化ベンザルコニウム、酵素活性を測定する比色定量法で行い、検出試薬はCCK−8(Cell Counting Kit-8)を用いる、CCK−8試験を使用した。
cHDL、rHDL共に、ネガティブコントロールの代表例であるHBSS/HEPES溶液と同等以上の細胞生存率を示し、ポジティブコントロールの代表例である塩化ベンザルコニウム(市販点眼剤に含まれている防腐剤)と比べて有意に細胞毒性が低かった(図3)。
次に、製造したcHDLの網膜への到達効率を調べた。
cHDLの網膜への到達効率
(方法)
実験動物マウスであるC57/B6に、クマリン−6濃度が0.1μmoL/mLの、cHDL、rHDL、粒径100nmのDSPCリポソームをそれぞれ3μLずつマウスの眼表面に点眼し、30分後に眼球摘出を行い、凍結切片を作製し、凍結標本で網膜を観察し、コンフォーカル顕微鏡で網膜内層の蛍光強度を観察し、結果を比較した。
(結果)
共焦点顕微鏡観察により、cHDL点眼群では、クマリン−6を有しないHBSS/HEPESバッファーの点眼群には見られない蛍光が網膜内層に観察された(図4)。
以下の方法により、後眼部薬物デリバリーシステムのモデル試験として血管新生モデルの試験を行った。
後眼部薬物デリバリーシステムのモデル試験(例、血管新生モデル)
6〜8週齢のC57BL6マウスの網膜にアルゴンレーザーを照射し、脈絡膜新生血管を作製した。レーザー照射直後から1週間、点眼液を一日三回、一回3μL点眼し、1週間後に眼球摘出した。免疫染色法にて脈絡膜新生血管を染色し、フラットマウント法にてコンフォーカル顕微鏡下で脈絡膜新生血管の面積を測定することで治療効果を判定した。1)1mg/mLのアポリポタンパク質濃度を有するcHDL溶液1mLおよびcHDLを含まない緩衝液1mLに、それぞれ1mgのパゾパニブ(Pazopanib)を混和したもの(図6)、2)アポリポタンパク質と等モル比のPazopanibを内包するcHDL溶液およびPazopanibを内包していないcHDL溶液(いずれもアポリポタンパク質濃度:1.5mg/mL)(図7)、をそれぞれ二者間で比較した。ここで、Pazopanibを内包したcHDL溶液は、実施例1におけるクマリン−6の代わりにPazopanibを用いて調製した。
結果、いずれの場合も基剤を内包したcHDL溶液の点眼で脈絡膜新生血管の面積が縮小しており、血管新生阻害作用を有する薬剤がcHDLへの内包を通じて疾患部位まで到達していることが示唆された(図6、図7)。このことは、薬剤としてのパゾパニブが後眼部へ到達し、血管新生を抑制する効果を示唆する。
網膜への薬剤到達に対するリン脂質の種類の役割を検討するために、種々のリン脂質を用いて細胞親和性HDL(cHDL)を作製し、網膜への到達効率を調べた。
1)細胞親和性HDL(cHDL)の作製
(方法)
異なるリン脂質(具体的には、DMPC、DPPC、DSPCの三種類)を用いて、各種高密度リポタンパク質(cHDL)を、以下の方法により作製した。
具体的には、クマリン−6・リン脂質・コール酸からなるミセルを事前に作製し、そのミセルとapoA−Iをリン脂質の相転移温度で混和することにより作製した(コール酸透析法)。ここで、各リン脂質の相転移温度は以下の通りである。DMPCは24℃、DPPCは40−41℃、及びDSPCは55℃。
クマリン−6の濃度は0.05μmol/mlに統一して使用した。得られたcHDLは、実施例1の方法に準じて超遠心法により精製した。
上記の得られたcHDLのそれぞれの組成を、実施例1の方法に準じて、タンパク質、リン脂質およびクマリン−6の各含量、並びに粒径について調べた。
Spontaneous interaction法と超遠心法により、粒径が直径10〜20nmの小サイズのcHDLを得ることができた。各cHDLの粒径サイズおよびそれを構成するタンパク質、リン脂質およびクマリン−6の各濃度は下記表2の通りである。
(方法)
次に、上記製造した各種cHDLの網膜への到達効率を調べた。
具体的には、実施例3と同様の方法でコンフォーカル顕微鏡で網膜内層の蛍光強度を観察し、結果を比較した。
共焦点顕微鏡観察により、網膜内層の蛍光強度を比較した。その結果を、図8に示す。DSPC>DPPC>DMPCの順に蛍光強度が高かった。
また、これら蛍光強度の結果を実施例3の方法に準じて数値化した(図9)。
この結果から、上記本発明のHDLは有意な大きさでクマリン−6を網膜内層へ到達させ得ることが示唆されており、特に細胞親和性ペプチドを結合したcHDLは極めて高い程度でクマリン−6を網膜に到達し得ることが示唆される。また、スフィンゴリン脂質のアシル基におけるアルキル基の炭素数が多い、長鎖のリン脂質ほど網膜への到達効率を向上させることが示唆された。
種々の細胞膜透過性ペプチド(CPP)を用いる細胞親和性HDL(cHDL)
網膜への薬剤到達に対する網膜への薬剤到達に対する細胞膜透過性ペプチド(CPP)の種類の役割を検討するために、種々の細胞膜透過性ペプチドを用いて細胞親和性HDL(cHDL)を作製し、網膜への到達効率を調べた。
1)細胞親和性HDL(cHDL)の作製
(方法)
異なるCPP(具体的には、TATペプチド、ペネトラチン(PEN)ペプチド、ポリアルギニン(R8))の三種類)を用いて、前記[0042]に記載する遺伝子工学的手法と同様の方法に従って、N末端43アミノ酸欠損apoA−Iに融合したタンパクを作製した。次に、実施例5と同様の方法でDSPC・クマリン−6からなるcHDLを作製した。クマリン−6の濃度は0.03μmol/mlに統一して使用した。得られたcHDLは、実施例1の方法に準じて超遠心法により精製した。
上記の得られたcHDLのそれぞれの組成を、実施例1の方法に準じて、タンパク質、リン脂質およびクマリン−6の各含量、並びに粒径、表面電位について調べた。
Spontaneous interaction法と超遠心法により、粒径が直径10〜20nmの小サイズのcHDLを得ることができた。各cHDLの粒径サイズおよびそれを構成するタンパク質、リン脂質およびクマリン−6の各濃度、並びにゼータ電位は下記表3の通りである。
(方法)
次に、上記製造した各種cHDLの網膜への到達効率を調べた。
具体的には、実施例3と同様の方法で共焦点顕微鏡で網膜内層の蛍光強度を観察し、結果を比較した。
共焦点顕微鏡観察により、網膜内層の蛍光強度を比較した。その結果を、図10に示す。PEN>R8≧TAT>CPPなしの順に蛍光強度が高かった。
また、これら蛍光強度の結果を実施例3の方法に準じて数値化した(図11)。
この結果から、上記本発明のHDLは有意な大きさでクマリン−6を網膜内層へ到達させ得ることが示唆されており、特に細胞親和性ペプチドを結合したcHDLは極めて高い程度でクマリン−6を網膜に到達し得ることが示唆される。また、PENが最も網膜への到達効率を向上させることが示唆された。
網膜への薬剤到達における濃度依存性
PENペプチド・DSPC・クマリン−6を含むcHDLを用いて、網膜への薬剤到達における濃度依存性を検証した。
クマリン−6の濃度が0.03μmol/ml、0.05μmol/ml、0.1μmol/ml、0.2μmol/mlでそれぞれ調整したcHDL溶液を、上記実施例5または6と同様な方法を用いて製造した。次いで、各溶液を実施例3と同様の方法で網膜内層の蛍光強度を比較した。その結果を、図12に示す。クマリンー6の濃度が高いほど蛍光強度が高い傾向が認められた。
また、これら蛍光強度の結果を実施例3の方法に準じて数値化した(図13)。
この結果から、cHDL中に含有されている薬剤の濃度が高ければ、網膜へ高濃度の薬剤を到達させることができることが示唆され、よって網膜への薬剤到達における濃度は、適用するcHDL中に含有されている薬剤の濃度に依存することが分かった。
本願発明のcHDLと従来のキャリアとの薬物デリバリー用キャリアとしての比較実験
後眼部への薬物デリバリー用キャリアとしての有用性について、本願発明のcHDLを従来のキャリアと比較検討した。
薬剤としてパゾパニブを選択し、パゾパニブのポジティブコントロールキャリアとして治験などで一般的に使われているカプチゾール、および実施例3において比較対象とした小サイズのリポソーム(ssLip)を選択した。そして、パゾパニブを有する本願発明のパゾパニブ内包cHDLを、パゾパニブ内包したこれらキャリアの複合体と、キャリアとしての有用性を比較検討した。
具体的には、本願発明のcHDLとしては、実施例4の2)と同様の方法にて調製したcHDLを使用した。一方で、上記比較対象キャリアは、文献1(Yafai et al. Eur J Pharmacol. 2011; 666: 12-8)および文献2(特表第2013-525501号公報)に記載の方法と同様に、7%カプチゾール溶液にパゾパニブを溶解することで比較カプチゾール溶液を作製した。また、ssLipへのパゾパニブの内包は、実施例1と同様の方法で作製したssLipに、DMSOに溶解したパゾパニブを混和し、次いでゲル濾過カラムを用いて内包されていないパゾパニブを除去することで精製し、比較ssLip溶液を作製した。
本実験ではパゾパニブ濃度を統一するために、文献3(国際公開第2011−069053号パンフレット)及び文献4(Escudero-Ortiz et al. Ther Drug Monit. 2015; 37: 172-9)に記載する方法に準ずる方法で超高速液体クロマトグラフィー(UPLC)を用いて各サンプルの濃度測定を行った。濃度が既知のパゾパニブ標準物質溶液を用いてクロマトグラフィーの保持時間を確認し、保持時間に位置するピークの面積を計量することで検量線を作成した。各サンプルでも同一の保持時間に位置するピーク面積を計量し、検量線からパゾパニブ濃度を定量し、各サンプルが同一濃度になるよう適宜希釈した。
これら調製した各溶液を、マウスへの点眼の回数を1日2回に変更した点を除いて実施例4に記載する方法に準じて血管新生モデルのマウスに点眼適用して、脈絡膜新生血管(CNV)の面積の縮小の効果を調べた。
その結果を図14に示す。本発明のcHDLをキャリアとして用いた場合は、比較対象のカプチゾール及びssLipのいずれかを用いた場合と比較して、脈絡膜新生血管の面積が有意に縮小していた(図14)。このことから、血管新生作用を有する薬剤がcHDLへの内包を通じて疾患部位まで有効に到達していることが示唆された。

- 該高密度リポタンパク質1分子あたり、1分子以上の蛍光標識物質、生体活性物質または薬物から選ばれる少なくとも1種類の化合物を更に含む、請求項1乃至6のいずれか1項に記載の細胞親和性ペプチド結合高密度リポタンパク質を含有する複合体。
- 請求項1乃至6のいずれか1項に記載の細胞親和性ペプチド結合高密度リポタンパク質、該高密度リポタンパク質中に含まれる後眼部疾患の診断、予防または治療に有効な薬物、および医薬的に許容され得る添加剤を含む、後眼部疾患の診断、予防または治療のための医薬組成物。
- 後眼部疾患が、加齢黄斑変性症、糖尿病網膜症、糖尿病黄斑浮腫、緑内障、網膜動静脈閉塞症、網膜変性疾患、変性近視、黄斑円孔、黄斑上膜、網膜剥離、白内障、硝子体混濁、およびぶどう膜炎からなる群から選ばれる少なくとも1種の疾患である、請求項8に記載の後眼部疾患の診断、予防または治療のための医薬組成物。
- i)アポリポタンパク質に細胞親和性ペプチドを結合させて、結合タンパク質を得る(ここで、該細胞親和性ペプチドは、ペネトラチンまたはポリアルギニン(R8)である);
ii)a)上記i)で得られた結合タンパク質に、リン脂質、適宜、蛍光標識物質、生体活性物質または薬物から選ばれる少なくとも1種類の化合物を含むリポソームを混合することにより、細胞親和性ペプチドを結合した粗高密度リポタンパク質を製造する;あるいは、
b)得られたコール酸ミセル、および生体活性物質または薬物から選ばれる少なくとも1種類の化合物との混和により、細胞親和性ペプチドを結合した粗高密度リポタンパク質を製造する;
iii)超遠心法により、未反応のリポソーム、リン脂質ミセル、アポリポタンパク質を除去することによって、該細胞親和性ペプチドを結合した粗高密度リポタンパク質を精製する、
ことを含む、請求項1乃至6のいずれか1項に記載の該細胞親和性ペプチド結合高密度リポタンパク質または請求項7に記載の複合体の製造方法。 - 点眼用である、請求項8に記載の医薬組成物。
- 該薬物が、眼内の脈絡膜新生血管の抑制のための薬剤、あるいは加齢黄斑変性、糖尿病網膜症、糖尿病黄斑浮腫、および網膜動静脈閉塞症からなる群から選ばれる疾患の診断、予防又は治療剤の少なくとも1種類の化合物である、加齢黄斑変性症、糖尿病網膜症、糖尿病黄斑浮腫、および網膜動静脈閉塞症からなる群から選ばれる少なくとも1種の後眼部疾患の診断、予防または治療のための請求項8または9のいずれかに記載の医薬組成物。
- 該薬物がパゾパニブである、請求項8、9または12のいずれか1項に記載の医薬組成物。
- アポリポタンパク質A−Iおよびその遺伝子改変型アポリポタンパク質A−Iからなる群から選ばれるアポリポタンパク質、およびジステアロイルホスファチジルコリン(DSPC)からなる高密度リポタンパク質、およびペネトラチンからなる細胞親和性ペプチド結合高密度リポタンパク質、
パゾパニブ、および
医薬的に許容され得る添加剤を含む、
後眼部疾患の診断、予防または治療のための点眼用医薬組成物。 - 後眼部疾患が、加齢黄斑変性症、糖尿病網膜症、糖尿病黄斑浮腫、または網膜動静脈閉塞症からなる群から選ばれる少なくとも1種の疾患である、請求項14に記載の後眼部疾患の診断、予防または治療のための点眼用医薬組成物。
- 請求項1乃至6のいずれか1項に記載の細胞親和性ペプチド結合高密度リポタンパク質を含む、後眼部薬物デリバリー用キャリア。
- アポリポタンパク質A−Iおよびその遺伝子改変型アポリポタンパク質A−Iからなる群から選ばれるアポリポタンパク質、およびジステアロイルホスファチジルコリン(DSPC)からなる高密度リポタンパク質、およびペネトラチンからなる細胞親和性ペプチド結合高密度リポタンパク質を含む、請求項16に記載の点眼用の後眼部薬物デリバリー用キャリア。
