Description
本出願は、2012年12月14日に出願された「VIRAL VECTOR NANOCAPSULE FOR TARGETING GENE THERAPY AND ITS PREPARATION」と題する、同時係属中の米国仮特許出願第61/737,233号の米国特許法第119条(e)の下での利益を主張し、その内容は本明細書に参照によって組み込まれる。

正確な特異性を有する導入遺伝子を所望の標的細胞または標的組織に導入する能力は、遺伝子治療のより容易な適用への鍵である。ここで、我々は、宿主細胞受容体特異性の認識が改変されたレンチウイルスベクターをナノテクノロジーを用いて生成する方法を記載する。簡潔には、VSV−G偽型レンチウイルスベクターの感染力を、ウイルスエンベロープ上にてその場で合成された薄いポリマーシェルによって遮蔽し、当該ポリマーシェル上に環状RGD(cRGD)ペプチドをコンジュゲートすることによって、当該遮蔽されたウイルスに対して新たな結合能力を付与した。我々は、結果として生じるウイルスを「ターゲティングナノウイルス」と称した。ターゲティングナノウイルスはVSV−G偽型とほぼ同等の力価を有し、高い形質導入効率でHela細胞に特異的に形質導入された。更に、ポリマーシェルによるVSV−G偽型レンチウイルスのカプセル化は、VSV−G偽型が侵入して細胞と融合する経路、ならびに逆転写および遺伝子発現などの後続の事象を変更しなかった。更に、ターゲティングナノウイルスは、ヒト血清の存在下で改善された安定性を有し、ポリマーシェルによるヒト血清補体不活性化からのウイルスの保護を示した。ナノテクノロジーのこの新規の使用は、実験的および臨床的目的のための、ウイルスベクターの再方向付けのためにより一般的に適用され得るアプローチを実証する。
レトロウイルスベクターおよびレンチウイルスベクターの安定的な組込みが、遺伝子送達のために一般的に利用されている(例えば、Aiuti et al.(2009)The New England Journal of Medicine 360,447−458、Aiuti et al.(2002)Science 296,2410−2413、Cartier et al.(2009)Science 326,818−23、およびCavazzana−Calvo et al.(2000)Science 288,669−672を参照されたい)。現在のベクターは、典型的にはVSV−Gエンベロープで偽型化されているために幅広い宿主範囲を有する(例えば、Marsh and Helenius(1989)Adv Virus Res 36,107−51を参照されたい)ため、当該ベクターの使用は、所望の標的細胞および標的組織が形質導入のために精製および/または物理的に単離され得る用途に限定されている。混合された集団内で特定の細胞をターゲティングし得るレトロウイルスベクターの作製は、遺伝子治療のより一般的な用途を可能にする。主要な障害は、ビリオン安定性および力価を同時に維持しながら、特異的にターゲティングするためのベクターエンベロープの修飾であった(Han et al.(1995)Proc Natl.Acad Sci U.S.A.92,9747−9751、Kasahara et al.(1994)Science 266,1373−1376、Marin et al.(1996)J.Virol.70,2957−2962、Nilson et al.(1996)Gene Therapy 3,280−286、Somia et al.(1995)Proc Natl.Acad Sci U.S.A.92,7570−7574、Valsesia−Wittmann et al.(1994)J.Virol.68,4609−4619、Yu and Schaffer(2005)Adv Biochem Eng Biotechnol 99,147−67)。更に、ウイルスエンベロープは、ヘパリン硫酸、ラミニン、インテグリン、炭水化物、脂質などに対する結合などの、非標的細胞特異的である様々な受容体結合部分をコードする(Haywood(1994)J Virol 68,1−5)。
ウイルス産生および力価
従前に記載されたように、全てのレンチウイルスベクターを、293T細胞のリン酸カルシウム媒介一過性トランスフェクションによって産生した(Morizono et al.(2001)Journal of Virology 75,8016−8020、Morizono and Chen(2005)Cell Cycle 4,854−6)。簡潔には、293T細胞(1.8×107細胞)を、12.5μgのpCMVR8.2_VPRと、(cppt2eと称する)中央ポリプリントラクトを有する12.5μgのSIN18−RhMLV−Eと、5μgのVSV−G発現プラスミドとでトランスフェクトした。ベータラクタマーゼを有するVSV−G偽型ウイルスのパッケージングの生成のために、ベータラクタマーゼ−Vpr融合タンパク質をコードするプラスミドを6ug含めた。ウイルスベクターを、抗生物質を有するAIM V(登録商標)培地(Invitrogen,Carlsbad,CA,USA)中で採取した。レンチウイルスベクターを、SW32ローター(Beckman,Palo Alto,CA,USA)による90分間、4℃、28,000RPMの超遠心法によって濃縮した。ペレットを、100倍少ない量のPBSで再懸濁した。ウイルス力価を、抗p24Gag酵素免疫吸着アッセイ(ELISA)によって測定した。レポーター遺伝子発現を、フローサイトメトリーによってモニタリングした。データを、Cytomics FC500(Beckman Coulter,Fullerton,CA,USA)上で収集し、FCS発現(De Novo Software,Los Angeles,CA,USA)を用いて分析した。
cRGDとして標識化される、環状[Arg−Gly−Asp−d−Phe−Lys(PEG−PEG)]を、Peptides International,Inc.から発注した。アクリル−cRGDの調製を、cRGDをアクリル酸、ヒドロキシスクシンイミドエステル(NAS)と反応させることによって達成した。簡潔には、cRGD(5mg)を1mLのpH=8、50mM、HEPES緩衝液中に溶解し、NAS(1.2mg)を100uLのDMSO中に溶解した。その後、NAS溶液をcRGD溶液中に徐々に室温で添加した。一晩の反応の後、1×PBS緩衝液をストックとして混合物を0.02%に希釈した。
100×濃縮VSV−G偽型レンチウイルスを、1×PBS緩衝液中で4℃で一晩透析した。ウイルス力価を、抗p24Gag ELISAによって測定した。5mgのN−アクリロキシスクシンイミド(NAS)(Sigma#A8060)を、0.1mLのDMSO中に溶解し、氷冷の1×PBS緩衝液で0.002%に希釈した。NAS溶液を、異なるモル比(m/m)1×104、2×104、5×104、および1×105でウイルスに添加(p24=60ng)した。反応を1時間、4℃で実行した。1μLの100mM、pH=7、TRIS緩衝液をマイクロチューブに添加し、NASによる表面修飾を停止させた。2%(w/v)のアクリルアミド(Sigma#A3553)と、2%(w/v)のGMA(Sigma#436895)と、0.5%(w/v)のAPS(Sigma#A3678)と、0.1%(w/v)のN,N,N',N'−テトラメチルエチレンジアミン(Sigma#T9281)とを含有するカクテル溶液を、2×104のモル比のアクリルオキシ化cRGD(0.02%)と共にまたは無しで、125、250、500、750の異なる重量比(w/w)でNAS修飾ウイルスに添加して、表面でのラジカル重合を開始させた。反応を、4℃で更に60分間進行させた。ウイルスおよびナノウイルス(p24=60ng)のサイズ分布を、Malvern粒径分析計Nano−ZSを有する動的光散乱(DLS)によって1×PBS緩衝液中で測定した。
VSV−G偽型およびターゲティングナノウイルス(p24=10ng)の両方を使用して、1×105個のHela細胞を4時間形質導入し、その後、細胞を1×PBSで2回洗浄し、500μlのDMEM/10%FBS/1%GPS中で2日間培養した。レポーター遺伝子発現(EGFP)を、フローサイトメトリーによってモニタリングした。ブロッキングアッセイのために、cRGD(1mg/ml)か、cRAD(1mg/ml)か、抗インテグリン抗体の混合物(20μg/mlの抗インテグリンαVβ3およびαVβ5抗体の両方)(Chemicon)か、またはアイソタイプ対照抗体(40μg/ml)を、感染の30分前および感染中にウイルスと共にインキュベートした。融合アッセイのために、バフィロマイシン−A(Sigma,B1793)を、形質導入の30分前および形質導入中に125nMの最終濃縮のウイルスと共にインキュベートした。逆転写アッセイのために、AZT(Sigma,A2169)を、ウイルス感染中に25μMの最終濃縮のウイルスと共にインキュベートした。細胞を1×PBSで洗浄し、フローサイトメトリー分析の前に2日間、AZTの存在下で培養を続けた。
Vpr−β−ラクタマーゼ融合タンパク質を組み込んだ、VSV−G偽型およびターゲティングナノウイルスの両方(p24=130ng)を使用し、1×105個のHela細胞を5、15、30、45、60、90、および120分間形質導入した。細胞を1×PBSで2回洗浄した。ベータラクタマーゼ基質CCF2−AM(Invitrogen,K1039)を、企業プロトコルに従って、暗所で室温で2時間、細胞と共にインキュベートした。蛍光を、フローサイトメトリーによってモニタリングした。
ターゲティングナノウイルスの合成
ウイルスエンベロープは、タンパク質、脂質、および炭水化物からなる。タンパク質がエンベロープの主成分であるため、我々は、合成タンパク質ナノカプセルについての我々の従来の方法が、ウイルスナノカプセルを合成するために適用され得ると考えた。我々は、ビリオンの周囲で合成される架橋されたポリマーシェルは、ウイルスの本来の感染力を除去し、ポリマーシェルの表面にコンジュゲートされたリガンドを介して新たな標的結合能力が付与され得ると仮定する。ターゲティングウイルスナノカプセルを合成するために、我々は、3段階の手順を使用して、EGFPを発現するVSV−G偽型レンチウイルスベクターを修飾した(図5)。I)重合可能な分子アンカーであるN−アクリロキシスクシンイミド(NAS)を使用し、VSV−Gエンベロープタンパク質のリジンと反応させて、重合可能な基を生成した。II)その後、これらの重合可能な基は、モノマー(アクリルアミド)のビニル基と反応して、ウイルス表面にポリマーを形成する。架橋剤(GMA)を反応に含めて、ポリマー構造を安定化した。架橋剤はpH6未満で分解可能であり、それにより、エンドソームなどの酸性環境においてポリマーシェルからのビリオンの放出が可能となる。III)その後、ポリマー上の過剰ビニル基との反応(マイケル付加反応)を介してポリマーシェル上に化学的にコンジュゲートされたターゲティング分子によって、新たな結合活性が付与される。Hela細胞をターゲティングするために、我々は、αVβ3インテグリンに対する強い親和性および選択性を示しかつ腫瘍内皮および腫瘍細胞上に十分に発現される環状アルギニン−グリシン−アスパラギン酸(cRGD)とコンジュゲートされているターゲティングナノウイルスを生成した。
ターゲティングナノウイルスのポリマーシェルの組成、サイズ、および分解性は、それらの送達効率および効力に影響を与える最も重要なパラメータである。NASの量は、後続の重合、ひいてはポリマー間の間隙距離のための表面アンカーの量を決定する。モノマーの量は、ポリマーの長さ、ひいてはポリマーシェルのサイズを制御する。NASおよびモノマーの濃度の上昇は本来のウイルス感染力のより良好な遮蔽につながったが、ウイルスベクターの過剰遮蔽はターゲティングナノウイルスの形質導入効率の減少にもつながり得る。したがって、ターゲティングナノウイルスの設計において、遮蔽およびターゲティング形質導入効率のバランスが必要となる。まず、我々は、NAS:ビリオン比(m:m=1×104)とモノマー:ビリオン比(w:w=250)を試験し、VSV−G偽型の58%と比較して、cRGDとのコンジュゲート有りで26%、cRGDとのコンジュゲート無しで11%の形質導入効率を達成し、これはポリマーシェルによるウイルス感染力の部分的な除去を示した(図6)。最適なターゲティング形質導入効率および最小のバックグラウンド感染力を達成するために、我々は、異なる量のNASおよびモノマー(cRGD有り、無し)の一連の組み合わせを更に試験した(図7)。我々は、HeLa細胞におけるターゲティングナノウイルスの形質導入効率を検査した。図7に示すように、cRGD無しでのナノウイルスの形質導入は、cRGD有りでのナノウイルスの形質導入よりも常に低く、これはポリマーシェルがVSV−G偽型レンチウイルスの本来の感染力を遮蔽し、cRGDがウイルスに対して新たな結合能力を付与したことを示している。我々の試験において、我々は、NAS:ウイルス(m/m=2×104)およびモノマー:ウイルス(w/w=125)の組み合わせが本来のウイルス感染力を完全に遮蔽することができ、cRGD有りの場合の結果は形質導入効率35%であり、これは同量のp24を有するVSV−G偽型(42%)とほぼ同等である(図7)ことを発見し、これにより、ターゲティングナノウイルスの高い感染力はポリマーシェルの間隙距離およびサイズの調整によって達成され得ることが示された。cRGD無しでのナノウイルスの形質導入効率は、この組み合わせ比で0%であった。我々はまた、動的光散乱(DLS)によってナノウイルスのサイズを決定した。ナノウイルスのサイズは約100nm〜約150nmの範囲であった(図7)。予想通り、ナノウイルスのサイズは、モノマーの量の増加と共に増加した。cRGD有りまたは無しでのナノウイルスのサイズは、ほぼ同等であった(図7)。別のRGD発現細胞、すなわちヒト臍静脈内皮細胞(HUVEC)において、NAS対ウイルス比およびモノマー対ウイルス比が最適な場合のターゲティングナノウイルスの形質導入効率を試験すると、8.1%の形質導入効率が、VSVG偽型による11.7%の形質導入効率、およびcRGD無しのナノウイルスによる0.2%の形質導入効率と共に観察された。この結果は、様々なRGD発現細胞においてターゲティングナノウイルスを使用する可能性を示している。
VSVG偽型レンチウイルスは、4℃での短期間の保管中、または−70℃で冷凍され解凍される際に安定である。ターゲティングナノウイルスがVSVG偽型とほぼ同等の安定性を有するかどうかを検査するために、我々は、4℃で4時間、8時間、および24時間保管後、ならびに凍結融解サイクル下での、ターゲティングナノウイルスによる形質導入を試験した。実験データは、種々の時点で、VSVG偽型およびターゲティングナノウイルスの両方について感染力の損失が存在しないことを示しており、これはターゲティングナノウイルスが、VSVG偽型のように、4℃で少なくとも24時間安定であることを示していた。凍結融解サイクル下にある場合、VSVG偽型およびターゲティングナノウイルスの感染力は、2サイクルの凍結融解後は安定であったが、3回目の凍結融解サイクルで著しく低減し、これは、ターゲティングナノウイルスがVSVG偽型のように、感染力の著しい損失なく、少なくとも2回冷凍および融解され得ることを示していた。
ターゲティング形質導入の特異性を確認するために、我々は、Hela細胞において非特異的ペプチド、すなわち環状RAD(cRAD)とコンジュゲートしたターゲティングナノウイルスの形質導入を試験し、形質導入がないことを観察し、これはHela細胞におけるナノウイルスの形質導入が、cRADではなくcRGDによって付与されたことを示していた(図8)。我々はまた、可溶性cRGDまたはcRADによってHeLa細胞をブロッキングし、その後、VSV−G偽型レンチウイルスまたはcRGDとコンジュゲートしたターゲティングナノウイルス(cRGD−nVSVG)のいずれかで細胞を形質導入した。cRGDまたはcRADのいずれかでのブロッキングは、VSV−G偽型レンチウイルスによる形質導入効率に対して影響を有さなかった(図9)。cRADではなく、cRGDによるブロッキングは、cRGD−nVSVGによる形質導入を、ブロッキング分子が不在の場合の形質導入の40%に阻害した(図9)。これらの結果は、Hela細胞におけるcRGD−nVSVGによる形質導入が、cRGD特異的であることを更に支持する。
我々は、古典的な標準VSV−G偽型と比較した、ターゲティングナノウイルスのウイルス学的特性を検査した。まず、我々は、これまでウイルスの細胞内への侵入の測定として使用されているβ−ラクタマーゼ(BlaM)アッセイによってウイルスの侵入キネティクスにアクセスした。BlaM−Vpr融合タンパク質を、VSV−G偽型およびターゲティングナノウイルスの両方の中に組み込んだ。続いて、内因性細胞質エステラーゼによってBlaMの基質となるように変換されてサイトゾル内に保持されるCCF2−AMを細胞に加えることによって、サイトゾルBlaM活性を検出した。CCF2−AMは、BlaMによって切断されると、緑色から青色の蛍光への推移を呈する。BlaM活性を、ウイルスの細胞との37℃でのインキュベートの5、15、30、45、60、90、および120分後に検査した。図10に示すように、5、15、30分で、BlaM活性は、ターゲティングナノウイルス形質導入細胞と比較して、VSV−G偽型形質導入細胞においてわずかに高かった。30分間のインキュベーション後、どちらのウイルス形質導入細胞においてもBlaM活性について著しい差は観察されなかった(図10)。これらの結果は、最初の30分以内に、VSV−G偽型がターゲティングナノウイルスよりもわずかに速く細胞に侵入し、BlaMを放出することを示していた。ターゲティングナノウイルスからのBlaMの放出の遅延は、ウイルス膜の融合が生じる前の、架橋剤の分解、ひいてはポリマーシェルの分解が原因であり得る。
天然VSV−Gウイルスは、エンドサイトーシスを介して細胞に侵入し、エンドソームで融合する。空胞型H+−ATPaseの阻害剤であるバフィロマイシン−Aは、エンドソーム内のpHを中和し、ひいてはpHに依存したウイルス融合、および続く形質導入を阻害するために使用されている。ターゲティングナノウイルスのポリマーシェルの架橋剤は低いpHで分解され、それによりビリオンの放出が可能となる。したがって、我々は、ターゲティングナノウイルスもまた、エンドサイトーシスを介して細胞に侵入し得、架橋剤がエンドソームで分解されて、融合のためのVSV−Gタンパク質を暴露すると仮定した。エンドソームがターゲティングナノウイルスの融合部位であるかどうかを確認するために、VSV−G偽型およびターゲティングナノウイルスを使用して、バフィロマイシン−A有りまたは無しでHeLa細胞を形質導入した。バフィロマイシンAの存在下で、VSV−G偽型およびターゲティングナノウイルスの両方の形質導入は阻害され(図11A)、これは両方のウイルスがエンドソームを介して侵入および融合し、ポリマーシェルによる遮蔽がウイルスの結合のみを除去し、融合は除去しなかったことを示していた。
ターゲティングナノウイルスの形質導入中に逆転写が生じることを確認するために、逆転写阻害剤であるAZTを使用して形質導入をブロッキングした。VSV−G偽型およびターゲティングナノウイルスを使用して、AZT有りまたは無しでHeLa細胞を形質導入した。AZT無しの場合、両ウイルスが、それぞれ43%および39%の形質導入効率でHeLa細胞を形質導入した(図11B)。AZT有りの場合、両方の形質導入がブロッキングされ(図11B)、それにより、ターゲティングナノウイルスによる感染力には逆転写が必要であることが実証された。
ヒト細胞内で産生されるVSV−G偽型HIVベクターは、ヒト血清補体によって不活性化され得ることが報告されており、これは臨床的用途における治療ベクターのためにはエンベロープのより高い安定性が必要とされることを示している。我々は、ターゲティングナノウイルスのポリマーシェルが、ヒト血清補体による不活性化からの保護をウイルスに提供し得るかどうかを更に検査した。VSV−G偽型またはターゲティングナノウイルスを、1:1の比で、PBS、熱不活性化ヒト血清、または非不活性化ヒト血清と共に、感染前および感染を通して30分間インキュベートした。図12に示すように、ヒト血清処理の後、VSV−G偽型の形質導入効率は低減し、これは公開されたデータと一貫性がある。ヒト血清の存在下において、ターゲティングナノウイルスによる形質導入は、VSV−G偽型と比較して5倍高かった(図12)。PBSまたは熱不活性化ヒト血清による処理の場合は、VSV−G偽型およびターゲティングナノウイルスの形質導入効率は、ほぼ同等であった。これらのデータによって、VSV−G偽型がヒト血清補体によって不活性化され得、ポリマーシェルがターゲティングナノウイルスをヒト血清による不活性化から保護し得ることが実証された。
ヒト疾患の治療のための遺伝子治療の成功した適用は、発現の望ましい部位への、治療遺伝子の効率的かつ安全な送達を必要とする。最も有効なアプローチは、特定の細胞および組織が存在し、それを形質導入するベクターを開発することであろう。特定の細胞および組織をターゲティングし得るレトロウイルスベクターを開発するための、数多くの努力がこれまで行われてきた。典型的には、これは天然エンベロープの修飾、および/または他のウイルスエンベロープでの偽型化を伴った。天然エンベロープにおける修飾はウイルス力価の大きな低減につながり、他のウイルスエンベロープでの偽型化は多くの異なる型の細胞および組織のターゲティングに対して一般的に適用可能でないため、これらのアプローチは一般的に適用可能ではなかった。
これは、本発明の好適な実施形態の記載を結論付ける。本発明の1つまたは複数の実施形態についての前述の記載は、例示および説明の目的のために提示されている。それは、包括的であること、または本発明を開示される正確な形態に制限することが意図されない。上記の教示に照らして、多くの修正および変更が可能である。
Claims (7)
- ウイルスベクターの細胞特異性を調節する方法であって、該方法が、
ウイルスエンベロープと標的組織または標的細胞系列に対する第1の特異性とを有するウイルスベクターを選択する段階、および
該ウイルスベクターをポリマーシェルにカプセル化する段階であって、
該ウイルスベクターを薄いポリマーシェル内にカプセル化するように、該ウイルスエンベロープ上で直接重合されたモノマーから該ポリマーシェルが形成され、かつ
該ポリマーシェルが、
インビボで分解されるポリマーシェルを形成するように分解性架橋剤によって架橋された複数のポリマーと
該架橋された分解性ポリマーシェルに結合したターゲティング剤であって、該架橋された分解性ポリマーシェルが、N−アクリロキシスクシンイミド(NAS)、アクリルアミド、およびグリシジルメタクリレート(GMA)を含み、かつ該ターゲティング剤が標的組織または標的細胞系列に対する第2の特異性を有するように選択された、ターゲティング剤と、を含む、段階
