Claims (18)
- 未分化多能性細胞を低減または除去する方法であって、有効量のNAMPT阻害剤を、異種細胞集団または、分化した細胞種及び未分化多能性幹細胞を含むか含むことが疑われるサンプルに接触することを含むことにより、前記NAMPT阻害剤が、前記細胞集団またはサンプルからの未分化幹細胞を、選択的に低減または除去する方法。
- 前記NAMPT阻害剤が、STF−31、FK866、GMX−1778、GNE−617、及びGNE−618からなる群から選択される、請求項1に記載の方法。
- 前記有効量が1ナノモル(nM)から100マイクロモル(μΜ)の間である、請求項1に記載の方法。
- 前記有効量が5μΜのSTF−31である、請求項3に記載の方法。
- 前記有効量が25nMのFK866である、請求項3に記載の方法。
- 以下の(a)〜(c)を含む、実質的に未分化多能性幹細胞を含まない幹細胞由来の細胞集団を得るための方法。
(a)未分化多能性幹細胞の1つ以上の幹細胞由来細胞種への分化または、部分的に分化誘導すること、
(b)有効量のNAMPT阻害剤を前記誘導された細胞集団に接触することにより、前記誘導された細胞種から、選択的に未分化多能性幹細胞を低減または除去すること、及び
(c)1つ以上の細胞種及び幹細胞由来細胞種の集団を得るために接触した細胞を分離すること、ここで前記集団は実質的に未分化多能性幹細胞を含まない。 - 前記NAMPT阻害剤がSTF−31、FK866、GMX−1778、GNE−617、及びGNE−618からなる群から選択される、請求項6に記載の方法。
- 前記有効量が1nMから100μΜの間である、請求項6に記載の方法。
- 前記有効量が5μΜのSTF−31である、請求項8に記載の方法。
- 前記有効量が25nMのFK866である、請求項8に記載の方法。
- 前記1つまたは複数の幹細胞由来細胞種が、心筋細胞神経前駆細胞、神経細胞、網膜色素上皮細胞、肝細胞、及び間葉系幹細胞からなる群から選択される、請求項6に記載の方法。
- 単細胞クローンとして前記単離された幹細胞由来細胞種を拡大することを含む、請求項6に記載の方法。
- 以下の(a)、及び(b)を含む、奇形腫形成の可能性が低減された細胞組成物を得る方法。
(a)多能性幹細胞由来の細胞集団を提供すること、及び
(b)未分化多能性幹細胞または部分的に分化した多能性幹細胞由来の子孫を、選択的に低減するかまたは除去する有効量のNAMPT阻害剤と、前記集団を接触することにより、前記接触した集団は、前記NAMPT阻害剤に接触していない多能性幹細胞由来細胞集団と比較して低減された細胞の奇形腫形成の可能性を有し、かつ前記接触した集団を使用すること。 - 前記NAMPT阻害剤はSTF−31、FK866、GMX−1778、GNE−617、及びGNE−618からなる群から選択される、請求項13に記載の方法。
- 前記有効量が1nMから100μΜの間である、請求項13に記載の方法。
- 前記有効量が5μΜのSTF−31である、請求項15に記載の方法。
- 前記有効量が25nMのFK866である、請求項15に記載の方法。
- 前記細胞がヒトにとって同種異系または自源性である、請求項13に記載の方法。
