Description
ロスバスタチンカルシウム20mgと同量の塩基性添加剤を乳鉢を用いて混合し、開放の状態で60℃75%RHの環境下に保管した。保管10日間後における分解物の量を測定した。
ロスバスタチンカルシウム2.6g、D−マンニトール67.15g、クロスポビドン3.75gを3分間混合し、さらにステアリン酸マグネシウム1.5gを1分間混合して混合末を得た。得られた混合末をロータリー打錠機にて圧縮成形し、素錠を得た。
ロスバスタチンカルシウム2.6g、乳糖水和物52.21g、結晶セルロース15.5g、クロスポビドン3.75gを3分間混合し、さらにステアリン酸マグネシウム0.94gを1分間混合して混合末を得た。得られた混合末をロータリー打錠機にて圧縮成形し、素錠を得た。
ロスバスタチンカルシウム2.6g、D−マンニトール64.55g、クロスポビドン3.75g、第三リン酸カルシウム2.6gを3分間混合し、さらにステアリン酸マグネシウム1.5gを1分間混合して混合末を得た。得られた混合末をロータリー打錠機にて圧縮成形し、素錠を得た。
ロスバスタチンカルシウム2.6g、D−マンニトール64.55g、クロスポビドン3.75g、含水二酸化ケイ素(アドソリダー101、フロイント産業(株))2.6gを3分間混合し、さらにステアリン酸マグネシウム1.5gを1分間混合して混合末を得た。得られた混合末をロータリー打錠機にて圧縮成形し、素錠を得た。
ロスバスタチンカルシウム2.6g、D−マンニトール64.55g、クロスポビドン3.75g、含水二酸化ケイ素(カープレックス#65、エボニック(株))2.6gを3分間混合し、さらにステアリン酸マグネシウム1.5gを1分間混合して混合末を得た。得られた混合末をロータリー打錠機にて圧縮成形し、素錠を得た。
実施例1〜8、比較例1〜2、参考例1〜3を、開放の状態(無包装)で60℃75%RHの環境下に保管した。保管10又は21日間後における分解物の量を測定した。
本品を粉末とし、表示量に従いロスバスタチン20mgに対応する量をとり、水/アセトニトリル混液(7:3)20mLを加え、10分間振り混ぜた後、遠心分離した。上澄液を孔径0.45μm以下のメンブランフィルターでろ過した。初めのろ液5mLを除き、次のろ液を試料溶液とした。この液1mLを正確に量り、水/アセトニトリル混液(7:3)を加えて正確に100mLとし、標準溶液とした。試料溶液及び標準溶液10μLずつを正確にとり、次の条件で液体クロマトグラフィーにより試験を行う。それぞれの液の各々のピーク面積を自動積分法により測定した。
検出器:紫外吸光光度計(測定波長:242nm)
カラム:内径3mm、長さ15cmのステンレス管に3μmの液体クロマトグラフィー 用オクタデシルシリル化シリカゲルを充填する。
カラム温度:40℃付近の一定温度
移動相A:水/アセトニトリル/トリフルオロ酢酸混液(700:300:1)
移動相B:アセトニトリル/水/トリフルオロ酢酸混液(750:250:1)
流量:毎分0.75mL
移動相の送液:移動相A及び移動相Bの混合比を次のように変えて濃度勾配制御する。
表8、9に示すとおり、D−マンニトール及び安定化剤を含有する実施例1〜4は、D−マンニトールを含有するが安定化剤を含有しない比較例1ならびにD−マンニトール及び安定化剤を含有しない比較例2と比べて類縁物質の増加を大幅に抑えることが確認された。
Claims (1)
- ロスバスタチンカルシウム、D−マンニトール、安定化剤、及び崩壊剤を含有する錠剤であって、D−マンニトールを1錠剤あたり50重量%以上含み、安定化剤として含水二酸化ケイ素を1錠剤あたり10.4/3〜20/3重量%を含むことを特徴とする錠剤。
