Description
a)目的の前記抗原に対して特異的な抗体を産生することができる少なくとも1種のB細胞、又は目的の前記抗原に対して特異的な抗体を産生することができるB細胞に発達することができる少なくとも1種のB細胞を選択する工程と、
b)前記少なくとも1種のB細胞において、BCL6の発現を誘導し、増強させ及び/又は維持する工程、並びに抗アポトーシス核酸の発現を誘導し、増強させ及び/又は維持する工程と、
c)前記少なくとも1種のB細胞を増殖させて第1のB細胞培養物とする工程と、
d)前記第1のB細胞培養物におけるB細胞の、目的の前記抗原に対する平均結合能よりも高い目的の前記抗原に対する結合能を有する少なくとも1種のB細胞を前記第1のB細胞培養物から選択する工程と、
e)好ましくは、工程d)において選択された前記少なくとも1種のB細胞を増殖させて第2のB細胞培養物とする工程と、
f)工程d)において選択された前記少なくとも1種のB細胞によって又は前記第2のB細胞培養物によって産生された抗体の安定性を決定する工程と、
g)前記第1のB細胞培養物のB細胞によって産生された抗体の平均安定性と比較して高い安定性を有する抗体を産生することができる少なくとも1種のB細胞を選択する工程と
を含む方法を提供する。好ましくは、工程g)で選択された前記少なくとも1種のB細胞は、増殖させて更にB細胞培養物とする。
a)目的の抗原に対して特異的な抗体を産生することができるex vivo B細胞培養物を準備する工程と、
b)前記ex vivo B細胞培養物のB細胞の、目的の前記抗原に対する平均結合能よりも高い目的の前記抗原に対する結合能を有する少なくとも1種のB細胞を前記ex vivo B細胞培養物から選択する工程と、
c)工程b)において選択された前記少なくとも1種のB細胞によって産生された抗体の安定性を決定する工程と、
d)前記ex vivo B細胞培養物のB細胞によって産生された抗体の平均安定性と比較して高い安定性を有する抗体を産生することができる少なくとも1種のB細胞を選択する工程と
を含む方法を提供する。
のB細胞培養物を開始した11〜12日後に、少なくとも100ng/mlの抗体濃度がすでに得られる。当業者が認識するように、培養が1種超のB細胞から開始される場合、所望の抗体濃度をより早い時点で得ることができる。

b)前記少なくとも1種のB細胞において、BCL6の発現を誘導し、増強させ及び/又は維持する工程、並びに抗アポトーシス核酸の発現を誘導し、増強させ及び/又は維持する工程と、
c)前記少なくとも1種のB細胞を増殖させて第1のB細胞培養物とする工程と、
d)前記第1のB細胞培養物におけるB細胞の、目的の前記抗原に対する平均結合能よりも高い目的の前記抗原に対する結合能を有する少なくとも1種のB細胞を前記第1のB細胞培養物から選択する工程と、
e)好ましくは、工程d)において選択された前記少なくとも1種のB細胞を増殖させて第2のB細胞培養物とする工程と、
f)工程d)において選択された前記少なくとも1種のB細胞によって又は前記第2のB細胞培養物によって産生された抗体の安定性、及び目的の前記抗原に対する親和性を決定する工程と、
g)前記第1のB細胞培養物のB細胞によって産生された抗体の平均安定性と比較して高い安定性を有し、前記第1のB細胞培養物によって産生された抗体の、又は目的の前記抗原に対する平均親和性と同様である又はこれより低い目的の前記抗原に対する親和性を有する抗体を産生することができる少なくとも1種のB細胞を選択する工程と
を含む方法を提供する。
a)目的の抗原に対して特異的な抗体を産生することができるex vivo B細胞培養物を準備する工程と、
b)前記ex vivo B細胞培養物のB細胞の、目的の前記抗原に対する平均結合能よりも高い目的の前記抗原に対する結合能を有する少なくとも1種のB細胞を前記ex vivo B細胞培養物から選択する工程と、
c)工程b)において選択された前記少なくとも1種のB細胞によって産生された抗体の安定性、及び目的の抗原に対する親和性を決定する工程と、
d)前記ex vivo B細胞培養物のB細胞によって産生された抗体の平均安定性と比較して高い安定性を有し、前記ex vivo B細胞培養物によって産生された抗体の、目的の抗原に対する平均親和性と同様である又はこれより低い目的の前記抗原に対する親和性を有する抗体を産生することができる少なくとも1種のB細胞を選択する工程と
を含む方法を提供する。
より、本発明による安定な抗体を産生することができる産生細胞株が生じる。
- Bcl-xL及び/若しくはMcl-1及び/若しくはBcl-2及び/若しくはA1及び/若しくはBcl-w及び/若しくはBcl2L 10の発現を直接的若しくは間接的に増強させることができる化合物を有する前記B細胞を準備する工程、並びに/又は
- Bcl-xL及び/若しくはMcl-1及び/若しくはBcl-2及び/若しくはA1及び/若しくはBcl-w及び/若しくはBcl2L 10の発現を直接的若しくは間接的に増強させることができる化合物の存在下で前記B細胞を培養する工程を含む本発明による方法が提供される。
- Blimp-1の発現を直接的若しくは間接的に増大させることができる化合物を有する前記B細胞を準備する工程、及び/又は
- Blimp-1の発現を直接的若しくは間接的に増大させることができる化合物の存在下で前記B細胞を培養する工程を更に含む本発明による方法が提供される。
a)目的の前記抗原に対して特異的な抗体を産生することができる少なくとも1種のB細胞、又は目的の前記抗原に対して特異的な抗体を産生することができるB細胞に発達することができる少なくとも1種のB細胞を選択する工程と、

BCL6及び/又はSTAT5、又はその機能的部分若しくは機能的誘導体をコードする核酸分子を有する前記ex vivo B細胞培養物を準備することによって、前記ex vivo B細胞培養物において、BCL6の発現を誘導し、増強させ及び/又は維持する工程、好ましくは、STAT3、又はその機能的部分若しくは機能的誘導体をコードする核酸分子を有する前記ex vivo B細胞培養物を準備することによって、又はIL21の存在下で前記ex vivo B細胞培養物を培養することによって、前記ex vivo B細胞培養物において、Blimp-1の発現を誘導し、増強させ及び/又は維持する工程、並びに前記ex vivo B細胞培養物において、BCL2ファミリー、好ましくはBcl-xL又はMcl-1の抗アポトーシス分子をコードする遺伝子の発現を誘導し、増強させ及び/又は維持する工程を更に含む方法を提供する。

- ex vivo B細胞培養物から、本発明による方法で、前記ex vivo B細胞培養物によって産生された抗体の平均安定性と比較して高い安定性を有する抗体を産生することができる少なくとも1種のB細胞を選択する工程と、
- 任意選択で、前記選択された少なくとも1種のB細胞を増殖させてB細胞培養物とする工程と、
- 高い安定性を有する前記抗体の重鎖及び/又は軽鎖のアミノ酸配列を決定する工程と、
- 第2の細胞において、前記抗体の重鎖及び/又は軽鎖をコードする核酸分子を発現させる工程と
を含む方法が提供される。好ましくは、前記第2の細胞は、いわゆる産生細胞、例えば、好ましくは市販の抗体産生に適合される、チャイニーズハムスターの卵巣(CHO)の細胞、NSO(マウスの骨髄腫)又は293(T)細胞株等である。このような産生細胞の増殖により、本発明による安定な抗体を産生することができる産生細胞株が生じる。好ましくは、前記産生細胞株は、ヒトにおいて使用するための抗体を産生するのに適している。したがって、前記産生細胞株は、好ましくは、病原性微生物等の病原体を含まない。

ヒト記憶B細胞を、Kwakkenbosら(Nature medicine、16(1)、123〜128. doi:10.1038/nm.2071;Methods 2013 doi:10.1016/j.ymeth.2013.07.002;特許出願WO2007/067046)によって記載されたBCL6/Bcl-xL技術を使用して不死化した。交差反応性抗インフルエンザヘマグルチニン特異的B細胞クローンAT10_004の同定及び特性評価が、特許出願WO2013/081463及びWO2012/072814に記載されている。AT10_004抗体の異なるヘマグルチニンタンパク質への結合を、固相ELISAを使用して、又はウイルス感染細胞を用いたFACSアッセイを使用して試験した(特許出願WO2013/081463に記載された)。AT10_004は、群2のヘマグルチニンタンパク質に最も強い結合を示すが、群1のヘマグルチニンタンパク質には中程度の結合しか検出されない[Table 1(表1)]。
特許出願WO2012/072814では、モノクローナルB細胞クローンの不均一な亜集団内で、増加した抗原結合能力を有する細胞は、抗原染色(H3-Alexa-647)とBCR染色の組合せを使用して選択されうることが示されている。BCR染色は、BCRの重又は軽鎖に結合する抗体を用いて実施した。高いH3染色及び等しい又は低いBCR染色は、その特定のサブクローンのBCRの高い抗原結合能力を示し、一方低いH3染色及び等しい又は高いBCR染色は、BCR当たりの抗原結合が少ないことを示す。
これまでは、抗原結合性における差異は、全く同じ数の細胞をBCR抗体及び標識した抗原で染色することで、同様の細胞/染色溶液の比を有するようにすることによって検出した。ここでの実験においては、増加した抗原結合性を示すサブクローンを、抗原競合実験を使用して検出する。AT10_004細胞を採取し、氷上でPe-Cy7標識したCD19抗体で染色した。15分後、細胞を洗浄し、1ウェル当たり40.000個の細胞で播種し、100μlのサブクローン細胞を添加する。次に、細胞混合物を洗浄し、標識した抗原(H1 HA:A/New Caledonia/20/1999又はH3 HA:A/Wyoming/03/2003)と標識したBCR抗体で染色する。細胞を氷上で1〜3時間インキュベートし、洗浄し、FACSCanto(BD Biosciences社)で測定する。CD19陽性細胞(親AT10_004細胞)に結合する標識した抗原の量を、サブクローン(CD19陰性)に結合した標識した抗原の量と比較する。図2でプロットされているのは、H1又はH3抗原のいずれかに対し増加した抗原結合性を示す6つのサブクローンである。
親AT10_004クローンと比較して増強された抗原結合性に基づいて、サブクローンのパネルを選択した。これらのサブクローンのうち、本発明者らは、RNeasy(登録商標)ミニキット(Qiagen社)で全RNAを単離し、cDNAを生成し、PCRを実施し、配列解析を実施した。組換えヒトIgGを産生するために、本発明者らは、ヒトIgG1及びカッパ定常領域とフレームが合うように、重及び軽鎖可変領域をpcDNA3.1(Invitrogen社)ベースのベクターにクローニングし、293T細胞に一過性に形質導入した。本発明者らは、MAbSelect Sureカラム(GE Healthcare社)を使用して、培養物上清から組換えIgGを精製した。
本発明者らは、iCycler RealTime PCR機器(BIORAD社)を使用して、動的走査蛍光法(DSF)(Phillips及びHernandez de la Pena、2001、The Combined Use of the Thermoflour Assay and ThermoQ Analytical Software for the Determination of Protein Stability and Buffer Optimization as an Aid in Protein Crystallization. Hoboken, NJ, USA:John Wiley & Sons, Inc. doi:10.1002/0471142727.mb1028s94)を用いて組換え親及び変異AT10_004抗体の構造的安定性を試験した(図3)。DSFアッセイでは、抗体は、温度の上昇に供され、それらの構造をアンフォールドさせる。タンパク質のアンフォールディングを、蛍光リポーター色素(SyPro、Invitrogen社)で測定する。本来のAT10_004抗体のDSF溶解曲線は、異なる温度での抗体サブドメイン(Fab、CH2及びCH3)のアンフォールディングを表す、多峰性のパターンを示す(Garber及びDemarest、2007、Biochemical and Biophysical Research Communications、355、751〜757)。軽鎖変異S30N(図示せず)、D50G及びS92Yを含有する変異は、親抗体で観察された±60℃での第1の「アンフォールディングピーク」を欠いており(図3)、これらの選択された変異が増大された熱安定性を有することを示す。
得られた変異抗体が、親AT10_004抗体と比較して異なる中和能力を有するかどうかを決定するために、in vitro中和アッセイを実施した。アッセイは、MDCK-SIAT細胞で実施した(Matrosovich M.ら、2010、Journal of Virology、77(15)、8418〜8425)。MDCK-SIAT細胞は、96ウェルプレート(CellCarrier Plate、PerkinElmer社)において、80〜100%のコンフルエンシーまで、DMEM/8%FCS/PS/G418で増殖させた。中和アッセイを、FCS又はBSAなしでOptimem/PS/G418/Trypsin培地で実施する。50μlの組換えmAbをH3N2(A/Ned/177/2008)又はH1N1(A/Hawaii/31/2007)インフルエンザのウイルス懸濁液(100TCID50/50μl又は1000TCID50/50μl)50μlと混合し、37℃で1時間インキュベートした。次いで、懸濁液を、多連で、100μlのOptimem/PS/G418/Trypsin中にMDCK-SIAT細胞を含有する95ウェルプレートに移した。使用する前に、MDCK-SIAT細胞を150μlのPBSで2回洗浄した。次いで、プレートを2500rpmで室温で15分間遠心分離し、37℃/5%CO2に置いた。24時間後、細胞をPBSで2回洗浄し、室温で10分間ホルマリン(水中37%のホルムアルデヒド)で固定し、150μlのPBSで2回洗浄し、室温で、DAPI及び抗体でインフルエンザウイルス(NP-FITC、Abcam社)の核タンパク質に対して染色した。30分後、細胞を150μlのPBSで2回洗浄し、100μlのPBS+50%グリセロールをウェルに添加した。MDCK-SIAT細胞のウイルス感染は、10xの対物レンズを使用するOperetta(PerkinElmer社)で測定し、分析した。mAbの中和能力を定量化するために、感染した細胞の数を計数した(DAPI及びNP-FITCに対して陽性)。IC50値は、Prism(GraphPad software)を用いて計算した。LC:D50G又はLC:S92Y変異を含有する抗体は、最低TCID50でH3N2中和能力を維持し、中和アッセイが高いウイルス投与量(>3000TCID50)で実施された場合、低いIC50によって示されるように増大した中和能力を有しさえした[図5及びTable 3(表3)]。これらの軽鎖変異では、H1中和能力の増大は検出されなかった(150μg/mlまで試験した)。標識したH1抗原での選択によって同定されたAT10_004サブクローン(AT10_004.8、AT10_004.9及びAT10_004.10)は、低減したH3N2中和能力を示した[図5及びTable 3(表3)]が、AT10_004.8及びAT10_004.9は増大したH1N1中和能力を示した。本来のAT10_004抗体又は軽鎖変異がH1N1感染の阻害を示さないのに対して、抗体AT10_004.8は、H1N1感染に顕著な効果を示す。
SPR分析を、前述のように、IBIS MX96 SPRイメージングシステム(IBIS Thchnoligies BV社、Enschede、The Netherlands)で実施した(Lokateら、2007、J Am Chem Soc. 129(45):14013〜8)。この実験において、抗体(カップリング緩衝液中1.0μg/ml:10mM MES-NaOH、pH 4.5+0.05% Tween20)を、連続流マイクロスポッター装置(continuous flow microspotter device)(Wasatch Microfluidics社、Salt Lake City、UT、USA)を使用して、Easy2Spot gold-film gel-type SPR-sensor(Ssens社、Enschede、The Netherland)で直接的に固定化した。スポッティング後に、センサーを50mMのエタノールアミン、pH8.5で不活性化した。不活性化後、それぞれ、エンプティアッセイ緩衝液(PBS+0.05%アジ化ナトリウム、0.05% Tween20及び0.01% BSA)を用いる45分の注入に続き、再生緩衝液(50mMのグリシン-HCl、pH2.0)を用いる2分のインキュベーションからなる、数回のブランク注入サイクルがなされた。
Claims (9)
- 目的の抗原に対する抗体を産生することができるB細胞を生成するための方法であって、
a)目的の前記抗原に対して特異的な抗体を産生することができる少なくとも1種のB細胞、又は目的の前記抗原に対して特異的な抗体を産生することができるB細胞に発達することができる少なくとも1種のB細胞を選択する工程と、
b)前記少なくとも1種のB細胞において、BCL6の発現を誘導し、増強させ及び/又は維持する工程、並びに抗アポトーシス核酸の発現を誘導し、増強させ及び/又は維持する工程と、
c)前記少なくとも1種のB細胞を増殖させて第1のB細胞培養物とする工程と、
d)前記第1のB細胞培養物におけるB細胞の、目的の前記抗原に対する平均結合能よりも高い目的の前記抗原に対する結合能を有する少なくとも1種のB細胞を前記第1のB細胞培養物から選択する工程と、
e)好ましくは、工程d)において選択された前記少なくとも1種のB細胞を増殖させて第2のB細胞培養物とする工程と、
f)工程d)において選択された前記少なくとも1種のB細胞によって又は前記第2のB細胞培養物によって産生された抗体の、安定性及び目的の前記抗原に対する親和性を決定する工程と、
g)前記第1のB細胞培養物のB細胞によって産生された抗体の平均安定性と比較して高い安定性を有し、前記第1のB細胞培養物によって産生された抗体の目的の前記抗原に対する平均親和性と同じである又はこれより低い、目的の前記抗原に対する親和性を有する抗体を産生することができる少なくとも1種のB細胞を選択する工程と
を含む方法。 - より高い安定性を有する抗体を産生することができる前記少なくとも1種のB細胞の重鎖及び/又は軽鎖のアミノ酸配列の少なくとも一部を決定する工程を更に含む、請求項1に記載の方法。
- 前記第1のB細胞培養物におけるB細胞の平均結合能より高い結合能を有するとして選択された前記少なくとも1種のB細胞によって産生された抗体の安定性、又は前記第2のB細胞培養物によって産生された抗体の安定性は、前記第1のB細胞培養物におけるB細胞の平均結合能より高い結合能を有する前記少なくとも1種のB細胞を選択してから4カ月以内に決定される、請求項1または2に記載の方法。
- 前記第1のB細胞培養物におけるB細胞の平均結合能より高い結合能を有するとして選択された前記少なくとも1種のB細胞によって産生された抗体の安定性、又は前記第2のB細胞培養物によって産生された抗体の安定性は、前記第1のB細胞培養物におけるB細胞の平均結合能より高い結合能を有する前記少なくとも1種のB細胞を選択してから1カ月以内に決定される、請求項3に記載の方法。
- 前記抗アポトーシス核酸が、抗アポトーシス分子をコードする遺伝子を含む、請求項1〜4のいずれか一項に記載の方法。
- 前記抗アポトーシス核酸が、BCL2ファミリーの抗アポトーシス分子をコードする遺伝子を含む、請求項5に記載の方法。
- 前記抗アポトーシス核酸が、Bcl-xL又はMcl 1の抗アポトーシス分子をコードする遺伝子を含む、請求項6に記載の方法。
- 前記少なくとも1種のB細胞におけるBlimp-1発現生成物の量を、直接的又は間接的に誘導し、増強させ及び/又は維持する工程を更に含む、請求項1〜7のいずれか一項に記載の方法。
- 前記少なくとも1種のB細胞が、目的の前記抗原に以前にさらされた個体に由来する、請求項1〜8のいずれか一項に記載の方法。
