Description
オリーブの植物水/搾りかすからの濃縮されたポリフェノールの製造
全体の製造工程は、膜ベースの接線流濾過及び分離技法の使用を中心とする。
ポリフェノール濃縮物の組成が、下記表Iに示される:
サンプルの希釈度は、以下の表IIに存在するmg/ml及びμMの値に対応する:
ヒト内皮細胞(HUVEC)の増殖に対するサンプルA009及びA012の効果を、MTT生存性アッセイ(テトラゾイル塩、[3−(4,5−ジメチルチアゾール−2−イル)]−2,5−ジフェニルテトラゾリウム臭化物)により評価した。アッセイは、ミトコンドリア酵素琥珀酸デヒドロゲナーゼにより代謝されるMTT化合物の能力を基礎とする。塩の還元は、水に不溶性である青色生成物ホルマザンの結晶の形成を導く。生存細胞は、非生存性細胞とは異なって、塩を還元し、そして生成されるホルマザンの量は、存在する細胞の数に比例する。形成される結晶は、溶解され、そして570nmの波長での吸光度値を、分光光度計により読み取る。
濃縮物により処理された内皮細胞のアポトーシス(プログラムされた細胞死)の誘発を、アネキシンV及び7−アミノ−アクチノマイシンD(7−ADD)によりマーキングすることにより評価した。それらのマーカーの細胞蛍光分析は、同じ細胞集団内で、異なる段階(早期、後期、高度)で細胞死を誘発する処理の能力の区別を可能にする。アネキシンVは、ホスファチジルセリンを、結合できる分子、すなわち細胞損傷の結果として細胞外側上に露出される、内部細胞質層に通常関連する、細胞膜のグリセロリン脂質である。7−AADは、損傷された細胞又はアポトーシスにおいてのみ細胞膜を通過できる化合物であり;従って、このマーカーに対する陽性は、細胞死及び製品の毒性を示す。
それらが細胞外マトリックスにおいてインビトロで培養され、そして適切な活性化刺激に暴露される場合、内皮細胞は血管の内腔の構造を模倣する管状構造に組織化することができる。
内皮細胞を、走化性グラジエントに続いて、特定部位の方に移動する能力により特徴づける。HUVECの移動能力を評価するために、移動アッセイを、Boydenチャンバー、及びコラーゲン(50μg/ml)によりソーキングされた、12μmのカットオフを有する多孔性フィルターを用いて設定した。
現場リクルートされると、内皮細胞は、走化性部位へのそれらの実際の移動に対する物理的バリヤーを構成する、細胞外マトリックスを分解できる因子を生成する、周囲組織に浸潤する能力を有する。
反応性酸素種(ROS)は、細胞損傷の腫瘍機構の1つを表し、そして炎症工程において、及び結果として、炎症に関する血管新生において基本的役割を演じる。従って、DCFH−DA(2′、7′−ジクロロフルオレセイン−ジアセテート)によるマーキングにより、H2O2による処理の後、植物水濃縮物による処理にゆだねられたHUVE細胞に関する抗酸化力による評価が行われた。
150,000個のHUVE細胞を、1%ゼラチンにより被覆された6−ウェルプレートの各ウェルにおける完全培地に播種した。続いて、細胞をトリプシン処理し、そして完全M199培地、及び植物水濃縮物(A009及びA012)、ヒドロキシチロソール、又はエタノール含有培地(標準希釈素で)に再懸濁した。
インビボでの血管形成を阻害する植物水濃縮物の能力を、Albini et al. in “Angiogenic potential in vivo by Kaposi's sarcoma cell-free supernatants and HIV-1 tat product: inhibition of KS-like lesions by tissue inhibitor of metalloproteinase-2 (AIDS, 1994)”により記載されるように、マトリゲルスポンジアッセイにより評価した。

Claims (10)
- 炎症の治療及び/又は予防への使用のための、1.5〜5g/Lの範囲の濃度のヒドロキシチロソール及び1〜6g/Lの範囲の濃度の3,4−DHPEA−EDAを含む、オリーブ植物水及び/又はオリーブ搾りかすの濃縮物であって、当該「g/L」が、1リットルの濃縮物あたりのグラムを表し、当該濃縮物が、以下の工程:
(i)マイクロ濾過の濃縮物及び透過物を得るために、オリーブ植物水及び/又はオリーブ搾りかすのサンプルをマイクロ濾過する工程、ここで当該マイクロ濾過工程が、1)酸化アルミニウム及びジルコニア製、及び2)管状形状、によって特徴付けられる、少なくとも1つのセラミック膜の使用を包含する;及び
(ii)工程(i)のマイクロ濾過透過物を逆浸透により濃縮する工程、ここで当該逆浸透はポリアミド製のポリマー膜を用いて実施される;
を含む方法を用いて取得される、当該濃縮物。 - 前記ヒドロキシチロソールの含有量が2〜3g/Lの範囲であり、前記3,4−DHPEA−EDAの含有量が1.5〜2.5g/Lの範囲である、請求項1に記載の濃縮物。
- 好ましくは、チロソール、クロロゲン酸、β‐ヒドロキシベルバスコシド、ルチン、ベルバスコシド、及びルテオリンから選択された少なくとも1つのフェノール化合物;及び/又は好ましくは、ナトリウム、カルシウム、マグネシウム及びカリウムから選択された少なくとも1つの金属;及び/又は好ましくは、塩化物、硫酸塩、リン酸塩及び硝酸塩から選択された少なくとも1つのアニオン;及び又はグルコース、フルクトース、マンニトール及びスクロースから選択された少なくとも1つの炭水化物;及び/又は窒素をさらに含む、請求項1又は2のいずれか1項に記載の濃縮物。

- 炎症の治療及び/又は予防への使用のための、請求項1〜5のいずれか1項に記載の濃縮物を含有する経口製剤。
- 前記炎症が腫瘍性又は非腫瘍性炎症である、請求項1〜5のいずれか1項に記載の濃縮物、又は請求項6若しくは7のいずれか1項に記載の飲料、又は請求項8に記載の経口製剤。
- 前記非腫瘍性炎症が、リウマチ性疾患、好ましくはリウマチ性関節炎及び痛風; 結腸直腸の炎症性疾患、好ましくは、クローン病、過敏性及び潰瘍性腸症候群;気管支の病変、好ましくは気管支慢性閉塞性肺疾患及び喘息; 肝疾患、好ましくは、線維症及び肝硬変; 前立腺の疾患、好ましくは、良性前立腺肥大症及び急性/慢性前立腺炎; 粘膜炎;皮膚炎又は前腫瘍性病変、好ましくは、乳房、子宮、肺又は口の病変に関連付けられる、請求項9に記載の濃縮物又は飲料又は経口製剤。
