Abstract
Description
本発明で使用される用語「薬学的に許容可能な塩」とは、化合物が投与される有機体に深刻な刺激を誘発せず、化合物の生物学的な活性と物性とを損傷させない化合物の剤形を意味する。前記薬学的塩は、薬学的に許容される陰イオンを含む無毒性酸付加塩を形成する酸、例えば、塩酸、硫酸、硝酸、リン酸、臭化水素酸、ヨウ化水素酸のような無機酸;酒石酸、ギ酸、クエン酸、酢酸、卜リクロロ酢酸、トリフルオロ酢酸、グルコン酸、ベンゾ酸、乳酸、フマル酸、マレイン酸、サリチル酸のような有機カーボン酸;メタンスルホン酸、エタンスルホン酸、ベンゼンスルホン酸、p−トルエンスルホン酸のようなスルホン酸などによって形成された酸付加塩が含まれる。例えば、薬学的に許容されるカルボン酸塩には、リチウム、ナトリウム、カリウム、カルシウム、マグネシウムなどによって形成された金属塩またはアルカリアルカリ土類金属塩;リシン、アルジニン、グアニジンなどのアミノ酸塩;ジシクロヘキシルアミン、N−メチル−D−グルカミン、トリス(ヒドロキシメチル)メチルアミン、ジエタノールアミン、コリン及びトリエチルアミンのような有機塩などが含まれる。

2)基質金属蛋白質分解酵素3(Mmp3)または基質金属蛋白質分解酵素13(Mmp13)の発現低減
3)Sox−9活性上昇
4)p−ERKの不活性化増大
軟骨細胞(chondrocyte)は、生後5日の正常マウスの大腿骨頭(femoral heads)、大腿骨関節(femoral condyles)及び頚骨高原部(tibial plateaus)に由来する軟骨組織から得た。得られた軟骨細胞は、10%(v/v)ウシ胎児血清(fetal bovine serum,Gibco、米国)、50μg/mlのストレプトマイシン(Sigma-Aldrich、米国)及び50unit/mlペニシリン(Sigma-Aldrich、米国)が含有されたDMEM培地(Gibco、米国)から培養した。
2−1:タイプIIコラーゲン(Col2a1)の発現増大
3’−または6’−シアリルラクトースによる軟骨形成及び再生効果を確認するために、実施例1で得られた軟骨細胞を36時間培養した後、3’−または6’−シアリルラクトースを、0,10,50,100,250μMの濃度別にそれぞれ処理し、36時間追加培養した。

IL−1βは、軟骨細胞において、Col2a1の発現を抑制させる代表的な炎症性サイトカインであり、軟骨細胞を36時間培養した後、5ng/mlのIL−1β(GeneScript、アメリカ)を24時間処理し、IL−1βにより、Col2a1発現が低減されることを確認した。
軟骨の形成及び再生に重要なCol2a1の発現は、Sox−9という転写因子によって調節されるので、3’−シアリルラクトースにより、Sox−9転写因子が調節されるか否かということを調べた。
IL−1βは、軟骨細胞において、軟骨形成に重要なCol2a1を低減させながら、同時に関節炎症及び軟骨組織破壊を促進する代表的な炎症性サイトカインである。軟骨細胞に、5ng/mlのIL−1βを時間別に処理した後、実施例2−1の方法で、下記表1の条件及びプライマーを使用し、qRT−PCRを行い、Mmp3、Mmp13の発現抑制を確認した。
IL−1βによって増加する軟骨破壊因子であるMmp3、Mmp13は、軟骨細胞内において、多様な信号伝逹経路を介して活性化される。それゆえに、3’−シアリルラクトースが、IL−1βによって調節される多様な信号伝逹経路(signal transduction pathway)を遮断させるか否かということを確認した。
本発明の全ての実施例の結果は、マンキンスコア(Mankin score)のような序数等級システムに基づいて定量されたデータであり、非母数(non-parametric)統計学的方法を利用して分析した。相対的な倍数変化(fold changes)によって表示されたqRT−PCRデータは、まず、シャピロ・ウィルク(Shapiro-wilk)検定を利用し、正規分布(normal distribution)を確認した後、双対比較(pair-wise comparisons)及び多重比較(multi-comparisons)のために、それぞれStudent’s t検定及び事後検定(post hoc test)を含むANOVA(analysis of variance)を利用した。統計的な有意性は、0.05レベルの確率で受容された(P<0.05)。

- シアリルラクトース、またはその薬学的に許容可能な塩を有効成分として含む退行性関節炎の予防用または治療用の薬学組成物であって、前記シアリルラクトースは、3’−シアリルラクトースまたは6’−シアリルラクトースであることを特徴とする薬学組成物。
- 前記3’−シアリルラクトースの塩は、下記化学式1の構造を有することを特徴とする請求項1に記載の薬学組成物:

- 下記特性のうち1以上を有することを特徴とする請求項1に記載の薬学組成物:
1)タイプIIコラーゲン(Col2a1)の発現増大
2)基質金属蛋白質分解酵素3(Mmp3)または基質金属蛋白質分解酵素13(Mmp13)の発現減少
3)Sox−9活性上昇
4)p−ERKの不活性化増大。 - 薬学的に許容される担体、賦形剤または希釈剤を追加して含むことを特徴とする請求項1に記載の薬学組成物。
- シアリルラクトース、またはその薬学的に許容可能な塩を有効成分として含む退行性関節炎の予防用または改善用の食品であって、前記シアリルラクトースは、3’−シアリルラクトースまたは6’−シアリルラクトースであることを特徴とする食品。
- 前記3’−シアリルラクトースの塩は、下記化学式1の構造を有することを特徴とする請求項6に記載の食品:
