Description
架橋を行う方法は当該技術分野で周知であり、特定の架橋方法を選択することは、当業者にとって十分に理解の範囲内である。
例えばシステインの化学的性質に関連したジスルフィド結合、例えばトランスグルタミナーゼ等の酵素処理、により誘導される共有結合、水素結合、疎水性結合、又はイオン結合等の非共有結合、あるいはトリポリホスフェートのような塩の付加、又はグルタルアルデヒド、ゲニピンのような化学物質、により誘導される結合が挙げられる。
・脂質及び/又は親油性有効成分(2)と、任意に重量剤及び/又は乳化剤とを、溶液に添加し混合することと、
・上記形成された混合物を均質化することと、
・ポリマーマトリックス(3)のポリマーを上記混合物に添加することと、
・上記混合物を、ポリマーマトリックスのポリマーが融解し、かつ混合物が均質になるまで加熱することと、
・任意に、ポリマーマトリックス(3)のポリマーを、例えば加熱し、酸及び/又は塩基(HCl及び/又はNaOH)並びにNaCl又はCaCl2等の塩の添加によって、上記形成された混合物のpHを例えば6.5〜8に調整し、かつイオン強度を所望の値に調整することにより架橋することと、
・上記混合物から、例えばモールディング、テンプレーティング若しくは射出法を使用して形状を形成するか、又は混合物から、エマルションテンプレーティング、3Dプリント、マイクロ若しくはミリフルイディクス、若しくはジェット押出成形を使用して小液滴を形成することと、
・上記形成された形状又は小液滴を、可消化ポリマーシェル(4)の可消化ポリマーの溶液中に、任意に増粘剤と共に、配置することと、
・上記混合物から形状を形成し、可消化ポリマーシェル(4)の可消化ポリマーを架橋すること、例えば加熱し、酸及び/又は塩基(例えばHCl及び/又はNaOH)並びに一価の塩(例えばNaCl)、二価の塩(例えばCaCl2)又は多価の塩(例えばFe(III)Cl3)の添加によって、上記形成された混合物のpHを例えば6.5〜8に、かつ/又は、イオン強度を所望の値に調整することにより架橋することと、
を含む方法によって調製することができる。
本発明の送達媒体を、下記の2工程プロセスによって製造した。第1の工程は、コアの形成を伴った。6.67重量% type Bゼラチン、100mM NaCl、pH8の溶液に、50〜60℃で、Silversonロータースターターホモジナイザー(rotor stator homogeniser)を使用し(6000rpm、2分間)、40重量%コリアンダー油を分散して、予備エマルションを調製した。次に、2段バルブホモジナイザー(Niro Panda 2000、第1段1000バール、第2段200バール)を使用して、予備エマルションを高せん断/圧均質化に供することにより、微細エマルションを調製した。次に、このエマルションを型に注ぎ込み、続いて4℃に冷却し、ゲル化剤にゲル化させて所望の形状を得ることにより、コアを形成した。

本発明の送達媒体を、下記の2工程プロセスによって製造した。第1の工程は、ゲル内に取り込まれたエマルションからなる、コアの形成を伴う。2重量%ポリソルベート80溶液に、Silversonロータースターターホモジナイザーを使用し(6000rpm、2分間)、60重量%高DHA/EPAマグロ油を分散して、予備エマルションを調製する。次に、2段バルブホモジナイザー(Niro Panda 2000、第1段1000バール、第2段200バール)を使用して、予備エマルションを高せん断/圧均質化に供することにより、微細エマルションを調製する。次に、このエマルションをK−カラギーナン(Genugel WR−78)及び塩化カルシウム溶液で希釈し、最終組成物:40重量%乳化油脂、0.6重量% K−カラギーナン、及び30mM CaCl2を形成した。次に、ゲル化剤を含有するエマルションを型に注ぎ込み、続いて4℃に冷却し、ゲル化剤にゲル化させて所望の形状を得ることにより、コアを形成する。
10重量%水中油型エマルションは、予め調製された40重量%水中油型エマルションを、3.2重量%乳清タンパク質分離物で希釈することによって調製した。3.2重量%乳清タンパク質分離物は、ヒマワリ油240gを5重量%タンパク質水溶液360gと、穏やかに混合して組み合わせることによって調製した。予備エマルションは、水相と油相の混合物を、Silverson L5M−Aローター−スターターミキサー(rotor-stator mixer)(Silverson machines,UK)を使用し、7000rpmで2分間均質化することによって調製した。次に、2段バルブホモジナイザー(NIRO Soavi Panda,GEA,France)を使用し、2回の通過のうち第1段1000バール及び第2段160バールの圧力で、予備エマルションを更に均質化することによって、微細エマルションを調製した。
Claims (15)
- 動物又はヒトの体温よりも高い融点及び/又は軟化点を有する可消化ポリマーシェル(4)によって囲まれたコア(1)を含み、
前記コアは、連続ポリマーマトリックス(3)中に分散された、脂質及び/又は親油性有効成分(2)を含み、
前記可消化ポリマーシェル(4)内に含まれる可消化ポリマーは、架橋された可消化バイオポリマーであり、
前記可消化ポリマーシェル(4)又は任意で可消化ポリマーシェル上に施されたコーティングが、前記脂質及び/又は親油性有効成分(2)の放出に対して拡散の障壁となる成分を含有せず、前記拡散の障壁となる成分が、脂質、ワックスコーティング、カルナウバワックス、又は親水コロイドであり、
前記軟化点は、材料がその固体特性を失い、相対的に液体化する温度であり、
前記可消化ポリマーシェル(4)が増粘剤を含み、該増粘剤が、キサンタン、カラギーナン、グアーガム、及びこれらの組合せからなる群から選択され、
前記可消化ポリマーシェル(4)の厚さが0.005mm〜3mmの範囲である、送達媒体(ただし、ゼラチン及びアラビアゴムを含むものを除く。)。 - 前記ポリマーマトリックス(3)が、動物又はヒトの体温未満では固体であり、かつ動物又はヒトの体温以上では固体又は液体である、請求項1に記載の送達媒体。
- 前記ポリマーマトリックス(3)がバイオポリマーマトリックスであり、前記バイオポリマーが、好ましくは、ゼラチン、カラギーナン、ペクチン、アルギン酸塩、寒天、ジェラン、コンニャクマンナン、グアーガム、ジェランガム、ローカストビーンガム、及びこれらの組み合わせからなる群から選択される、請求項1又は2に記載の送達媒体。
- 前記脂質及び/又は親油性有効成分(2)が、医薬品、機能性食品、栄養補助食品、動物用医薬品、天然/伝統薬、食品原料成分、及びこれらの組み合わせからなる群から選択される、請求項1〜3のいずれか一項に記載の送達媒体。
- 前記脂質及び/又は親油性有効成分(2)が、トリグリセリド、リン脂質、ジグリセリド、モノグリセリド、セラミド、糖脂質及びスフィンゴ脂質、魚油、ペトロセリン酸、エイコサペンタエン酸、カロテノイド、フィトステロール、フィトステロールエステル、ポリフェノール、コレステロール、コレステロールエステル、レチノール、レチノールエステル、及びこれらの組み合わせからなる群から選択される、請求項1〜4のいずれか一項に記載の送達媒体。
- 前記コアが、乳化剤、好ましくは、タンパク質、レシチン、界面活性多糖類、合成界面活性剤、天然界面活性剤、イオン性界面活性剤、非イオン性界面活性剤、及びこれらの組み合わせからなる群から選択される乳化剤を含む、請求項1〜5のいずれか一項に記載の送達媒体。
- 前記可消化ポリマーシェル(4)が、ウシ血清アルブミン、β−ラクトグロブリン、ラクトフェリン、乳清タンパク質分離物、パタチン、大豆タンパク質、カノーラタンパク質、エンドウ豆タンパク質、卵タンパク質、リゾチーム、エンドウ豆タンパク質、ルピナス、米タンパク質、グルテン、リゾチーム、ゼイン、及びこれらの組み合わせからなる群から選択される1つ以上のバイオポリマーから形成される、請求項1〜6のいずれか一項に記載の送達媒体。
- 前記可消化ポリマーシェル(4)の平均メッシュ/細孔サイズが50nm未満である、請求項1〜7のいずれか一項に記載の送達媒体。
- 前記脂質及び/又は親油性有効成分(2)の50%以上が、対象による前記送達媒体の摂取後の規定のラグタイムの終了後45分以内に放出され、かつ/又は、前記脂質及び/又は親油性有効成分(2)の80%以上が、対象による前記送達媒体の摂取後の規定のラグタイムの終了後75分以内に放出される、請求項1〜8のいずれか一項に記載の送達媒体。
- 請求項1〜9のいずれか一項に記載の送達媒体の調製方法であって、
・脂質及び/又は親油性有効成分(2)と、任意に重量剤及び/又は乳化剤とを、溶液に添加し混合することと、
・前記形成された混合物を均質化することと、
・前記ポリマーマトリックス(3)のポリマーを前記混合物に添加することと、
・前記混合物を、前記ポリマーマトリックス(3)のポリマーが融解し、かつ前記混合物が均質になるまで加熱することと、
・任意に、ポリマーマトリックス(3)のポリマーを、例えば加熱し、かつ酸及び/又は塩基(HCl及び/又はNaOH)並びにNaCl又はCaCl2等の塩の添加によって、前記形成された混合物のpHを例えば6.5〜8に、かつ/又はイオン強度を所望の値に調整することにより架橋することと、
・前記混合物から、例えばモールディング、テンプレーティング若しくは射出法を使用して形状を形成するか、又は前記混合物から、エマルションテンプレーティング、3Dプリント、マイクロ若しくはミリフルイディクス、若しくはジェット押出成形を使用して小液滴を形成することと、
・前記形成された形状又は小液滴を、前記可消化ポリマーシェル(4)の1種又は複数種の前記可消化ポリマーの溶液中に、任意に増粘剤と共に、配置することと、
・前記混合物から形状を形成し、前記可消化ポリマーシェル(4)の前記可消化ポリマーを、好ましくは加熱し、酸及び/又は塩基(例えばHCl及び/又はNaOH)並びに一価の塩(例えばNaCl)、二価の塩(例えばCaCl2)又は多価の塩(例えばFe(III)Cl3)の添加によって、前記形成された混合物のpHを例えば6.5〜8に、かつ/又はイオン強度を所望の値に調整することにより架橋することと、を含む方法。 - 請求項1〜9のいずれか一項に記載の送達媒体を含む消費可能な製品であって、好ましくは、前記組成物及び/又は消費可能な製品が、医薬製品、機能性食品、医療食、ペットフード及びペットケア製品を含む獣医製品、並びに機能性食品及び栄養補助食品を含むあらゆる種類の食品材料からなる群から選択される、消費可能な製品。
- 脂質及び/又は親油性有効成分(2)を、既定のラグタイムの終了後に胃腸管内に放出するための、請求項1〜9のいずれか一項に記載の送達媒体の使用。
- 睡眠障害、気分障害、又は、肥満、糖尿病、若しくは、潰瘍性大腸炎、クローン病、胃腸管の感染、大腸癌、及びこれらの組み合わせ等の胃腸管に影響を及ぼす他の状態等の病的状態の治療又は予防に使用するための、請求項1〜9のいずれか一項に記載の送達媒体。
- 過体重及び老化等の美容的な問題を処置及び/又は予防するための、請求項1〜9のいずれか一項に記載の送達媒体の非治療的使用。
- 送達媒体中に含まれる脂質又は親油性有効成分(2)の、代謝による修飾又は代謝を防止又は緩和し、それによって前記脂質又は親油性有効成分(2)の最小有効濃度を低減するための、請求項1〜9のいずれか一項に記載の送達媒体。
