Description
本発明は、分子医薬と、治療剤又は診断剤の標的化送達の分野に関する。より具体的には、本発明は、生物の脳脊髄液(CSF)及び脳を標的化するペプチドに関する。
例えば低い脳透過性を有する大型バイオ治療薬又は小分子薬などの神経障害薬の脳透過及び/又はCSF透過は、神経血管ユニット(NVU)中の他の細胞成分と一緒に広範囲で不透過性の血液脳関門(BBB)又は血液−CSF関門BCSFB(BCSFB)によって厳密に制限されている。この障壁を克服するための多くの戦略が試験されており、一つは、脳毛細血管内皮上に発現された内因性レセプターによって仲介されるトランスサイトーシス経路を利用することである。脳へのバイオ治療薬及び診断薬のレセプター介在性送達を可能にするために、モノクローナル抗体又はペプチドなどの組換えタンパク質がこれらのレセプターに対して設計されている。しかし、脳内皮細胞(BEC)内への誤分類及びBEC中のある細胞小器官(特にバイオ治療薬の分解に導く細胞小器官)内の蓄積度を最小化する一方で脳への取り込みを最大化するための戦略は、まだ探求されていない。
第一の態様では、本発明は、脳エフェクター本体と、
からなる群より選択された3アミノ酸ペプチドモチーフを含む脳標的化ペプチドとを含む、血液脳関門シャトルを提供する。
からなる群より選択されたペプチドモチーフを模倣した配列を含む単離されたペプチド模倣体、又は、配列番号1〜配列番号36からなるペプチドを模倣した配列を含むペプチド模倣体を含み得、該ペプチドは、CSF及び脳において豊富である。
定義
「血液脳関門」又は「BBB」は、脳毛細血管内皮形質膜内のタイトジャンクションによって形成される、末梢循環と脳及び脊髄との間の生理学的関門をいい、尿素(60ダルトン)などの非常に小さな分子であっても脳内への分子の輸送を制限する密着関門を作る。脳内のBBB、脊髄内の血液−脊髄関門、及び網膜内の血液−網膜関門は、CNS内の近接毛細血管関門であり、本明細書において、まとめて血液脳関門又はBBBと呼ばれる。BBBは、関門が毛細血管内皮細胞よりもむしろ上衣細胞から構成される血液−CSF関門(脈絡叢)も包含する。
からなる群より選択された少なくとも1つの3アミノ酸ペプチドモチーフを含む。本発明の脳標的化ペプチド内の3アミノ酸ペプチドモチーフは、連続的であっても及び/又は重複していてもよいことが理解されなければならない。例えば、重複モチーフの場合、脳標的化ペプチドにおいて、最初の3アミノ酸モチーフの2番目のアミノ酸は、2番目の3アミノ酸モチーフの最初のアミノ酸であってもよく、及び/又は、最初のアミノ酸モチーフの3番目のアミノ酸は、3番目のアミノ酸モチーフの最初のアミノ酸であってもよい。
インビボでのファージディスプレイによる血管マッピングにより、様々な脈管構造内で選択的に発現された生化学的「アドレス」が明らかとなる。このタイプのアプローチは、薬剤を特定の組織へと送達するために使用することのできるリガンドシステム及びレセプターシステムを発見するために使用されている(Arap et al, 1998, Pasqualini et al, 1996, Arap et al, 2002, Kolonin et al, 2001, Pasqualini et al, 2000)。このスクリーニングアプローチは、コンビナトリアルライブラリー(M13をベースとしたファージベクターにディスプレイされる)に由来する短いリガンドペプチドが全身投与後に特定の器官に標的化する能力に基づく(Pasqualini et al, 2000)。組織及び疾患状態を標的化するペプチドが単離され、いくつかの場合では、対応する血管レセプターが同定された(Arap et al, 1998, Pasqualini et al, 1996, Arap et al, 2002, Kolonin et al, 2001, Rajotte and Ruoslahti, 1999, Kolonin, et al, 2002, Kolonin et al, 2004)。特に、研究者は、癌患者におけるファージディスプレイライブラリーの使用を報告した。インターロイキン−11様ペプチドとして同定されたリガンドモチーフの1つ、及び、インターロイキン−11レセプターへのその標的化は、ヒト前立腺癌における治療薬の標的化送達のための可能性ある戦略として活用されている(Zurita et al, 2004)。インビボでのファージディスプレイランダムペプチドライブラリーの選択の重要な工程は試料採取戦略であり、したがってどのようにどこでファージを回収するかである。好ましくは、インビボでの選択戦略は、組織又はレセプターへの結合に基づくだけではなく、例えば細胞障壁を超えての輸送のような機能的な工程も含むべきである。これは、目的が、血液脳関門(BBB)又は血液CSF関門(BCSFB)の関与する新規な輸送システムを同定することである場合には特に重要である。
本発明は、CSF、脳、又は関連した脈管構造への組成物の局在化のために使用することができる、1つ以上の脳標的化ペプチド又は分子標的を同定するための方法を含む。被験体の液体及び器官を、N個の残基(Nは5、6、7、8、9、10、11、12、13、14、15個以上の残基であり得る)を有するペプチドについて、いくつかのペプチドモチーフを生じるランダムファージライブラリーからスクリーニングする。一例では、トリペプチドモチーフ
を含む様々なクローンが、CSF及び脳において、高い頻度、選択的な標的化又は結合、及び存在を示した。オンラインタンパク質データベースにおいて入手可能な配列と、選択されたペプチドモチーフとの比較により、多くの候補タンパク質が、これらのペプチドモチーフと相同な又は類似した配列を共有することが示唆される。これらのモチーフを取り囲む機序の研究が、CSF液及び脳、及び関連した脈管構造、並びにそれらの組合せを標的化するための、ペプチド及び標的の同定のための新規プラットフォームを提供するために探究されている。所見はまた、新規に同定されたモチーフがリードペプチド模倣薬として働き得、様々な治療部分の直接送達のために最適化させることができるという、重要な臨床的意義も有する。1つの方法は、ファージライブラリーを静脈内注射することを含み、数分後に試料を回収する。
マウスで行なわれた以前のインビボでの選択試験は、fUSE5ベクターにおいて遺伝子III被包タンパク質との融合タンパク質として発現されるランダムペプチドライブラリーを優先的に使用した(Pasqualini and Ruoslahti, 1996)。輸送体ペプチドを同定するために使用された好ましいファージ系は、T7ファージ系である。T7ファージ粒子は非常に小さく(約50nmの直径)、細胞を通って天然の細胞小器官の細胞内選別を使用して輸送されることを可能とする。M13ファージ系は、サイズが有意により大きく、細胞への取り込み及び細胞内選別を妨げ得る。したがって、血液脳関門(BBB)及び血液CSF関門(BCSFB)などの細胞膜によって保護された液体及び器官における標的ペプチドの濃縮は妨げられる。所与のライブラリーに存在する個々のクローンの数及び多様性は、インビボでの選択の成功のための重要な因子である。欠陥のあるファージクローンを過剰提示する可能性がより低い、一次ライブラリーを使用することが好ましい。全てのファージクローンの比率が等しく高い多様性を有する一次ライブラリーを使用することが好ましい。ライブラリーの調製は、108〜109個のプラーク形成単位(pfu)/mlとなるように最適化されるべきである。ある実施態様では、容積増幅戦略を、各回の選択操作の間に適用する。直鎖状、環状、又は二環式ペプチドを示すファージライブラリーを、本発明の範囲内で使用し得る。しかし、5、6、7、8、9、10、11、12、13、14、15個以上の残基を示すファージライブラリーが好ましい。環式ライブラリーも好ましい。しかし、同族合成ペプチドの産生も可能であるが、ジスルフィド橋の配置が異なる複数の配座異性体に因り複雑であり得る。
脈管構造に標的化するペプチドを、細胞傷害性薬物又はアポトーシス誘発性ペプチドと結合させることにより、親化合物よりも効果的でより毒性の少ない化合物が得られる。
臨床適用を考える場合、医薬組成物発現ベクター、ウイルスストック、タンパク質、抗体、及び目的とする適用に適した剤形の薬物を調製することが必要であり得る。一般的に、これは、ヒト又は動物に対して有害である可能性がある不純物を実質的に含まない組成物を調製することを包含する。一般的には、送達ベクターを安定化し、標的細胞による取り込みを可能とする適切な塩及び緩衝液を使用することを所望するだろう。組換え細胞を患者に導入する場合には緩衝液も使用される。本発明の水性組成物は、薬学的に許容される担体又は水性媒体に溶解又は分散させた、有効量のタンパク質、ペプチド、抗体、融合タンパク質、組換えファージ、及び/又は発現ベクターを含み得る。「薬学的に又は薬理学的に許容される」という語句は、動物又はヒトに投与された場合に有害反応、アレルギー反応、又は他の望ましくない反応を生じない分子本体及び組成物をいう。本明細書において使用される「薬学的に許容される担体」は、任意の及び全ての溶媒、分散媒体、コーティング剤、抗細菌剤及び抗真菌剤、等張化剤、及び吸収遅延剤などを含む。薬学的に活性な物質のためのこのような媒体及び薬剤の使用は、当技術分野において周知である。あらゆる従来の媒体又は薬剤が本発明のタンパク質又はペプチドと非適合性でない限り、治療組成物へのその使用が考えられる。補助活性成分もまた該組成物に取り込まれ得る。本発明の活性組成物は、古典的な医薬調製物を含み得る。本発明に記載のこれらの組成物の投与は、標的の液体、組織、又は器官がその経路を介して到達可能である限り、任意の一般的な経路を介する。これは、経口、鼻腔、頬側、直腸、膣内、又は局所を含む。あるいは、投与は、同所性、皮内、皮下、筋肉内、腹腔内、動脈内、又は静脈内投与により得る。このような組成物は通常、上記された薬学的に許容される組成物として投与されるだろう。注射用途に適した剤形は、無菌水溶液又は分散液、及び、無菌注射溶液又は分散液の即時調製のための無菌粉末が挙げられる。全ての場合において、剤形は無菌でなければならず、シリンジが容易に扱える程度まで流動性でなければならない。それは、製造及び保存の条件下で安定でなければならず、細菌及び真菌などの微生物の汚染作用から防御されていなければならない。担体はまた、例えば、水、エタノール、(例えば、グリセロール、プロピレングリコール、及び液体ポリエチレングリコールなど)、その適切な混合物、及び植物油を含有する、溶媒又は分散媒体であり得る。適切な流動性は、例えば、レシチンなどのコーティング剤の使用によって、分散液の場合には必要とされる粒径の維持によって、及び界面活性剤の使用によって維持され得る。微生物の作用の防御は、様々な抗細菌剤及び抗真菌剤、例えばパラベン、クロロブタノール、フェノール、ソルビン酸、チメロサールなどによってもたらされ得る。多くの場合、等張化剤、例えば糖又は塩化ナトリウムを含めることが好ましい。注射用組成物の吸収延長は、組成物中に、吸収を遅延させる物質、例えばモノステアリン酸アルミニウム及びゼラチンなどを使用することによってもたらされ得る。無菌注射溶液は、必要量の活性化合物を、適切な溶媒中に、必要であれば上記に列挙された他の様々な成分と共に取り込み、その後、滅菌濾過することによって調製される。一般的には、分散液は、様々な滅菌された活性成分を、基本的な分散媒体及び上記に列挙された成分の中からの必要とされるその他の成分とを含有する無菌ビヒクルに取り込むことによって調製される。無菌注射溶液の調製のための無菌粉末の場合、好ましい調製法は真空乾燥及び凍結乾燥技術であり、これにより、以前に滅菌濾過されたその溶液から活性成分と任意の追加の所望の成分の粉末が得られる。
以下の実施例は、本発明の好ましい実施態様を実証するために含められる。以下の実施例に開示された技術は、本発明者らによって本発明の実施においてよく機能することが発見された技術を示し、したがって、その実施のための好ましい形態を構成すると考えられ得ることが当業者によって理解されるべきである。しかし、当業者は、本開示に鑑みて、多くの変更を、開示された具体的な実施態様に行なうことができ、依然として本発明の精神及び範囲から逸脱することなく似たような又は類似した結果を得ることができることを理解すべきである。
大槽(CM)に、Sarna et al.(1983)(Waring et al. 2010, 192 249-253)(Sarna et al. 1983, 383-388)によって以前に記載された方法の改変形を使用してカニューレを挿入した。麻酔したウィスターラット(200〜350g)を定位脳手術装置上に載せ、剃った頭頂部から肩甲骨間部まで正中切開を行なった。頭頂部に2つの穴を開け、取り付けねじを穴にしっかり固定した。外後頭陵に追加の穴を開け、これを使用してカニューレを大槽に定位的に誘導した。歯科用セメントをカニューレ及びねじの周辺に塗布して、適所に固定した。セメントが乾燥した後、皮膚創傷を4/0のsupramid糸を用いて縫合した。カニューレが上手に配置されると、脳脊髄液(CSF)が自然発生的に流れる。ラットを定位脳手術装置から取り出し、適切な術後の鎮痛処置を受け、CSF中に血液の兆候が観察されなくなるまで少なくとも1週間は回復させた。全ての動物手順は、施設動物実験当局(institutional animal care authorities)(認可番号2474)によって全て承認された。
大槽にカニューレを挿入された起きているラットを手で優しく握った。マンダリンをカニューレから取り出し、自然発生的に流れているCSF 10μlを収集した。血液による汚染又は変色の兆候が全くない透明なCSF試料のみを本試験に含めた。なぜなら、カニューレの開存性が最終的に損なわれるからである。平行して尾の先端の小さな切開から血液10〜20μLを、ヘパリン(シグマアルドリッチ社)を含有しているチューブに収集した。CSF及び血液を、バクテリオファージの静脈内注射後の様々な時点で収集した。全てのCSF試料を収集する前に、カテーテルのデッド容量を示す10〜15μLの液体を廃棄した。
ライブラリーを、T7Select System Manual(Novagen)(Rosenberg et al. InNovations 6, 1-6 1996)に概略が示されているようにT7Select 10-3bベクターを使用して構築した。簡潔に言えば、ランダムな12−mer挿入断片DNAを、以下の形式で合成した:
ブロスで又はプレートで増殖させたファージの可変領域は、社内で設計された454/Roche−アンプリコン融合プライマーを使用したPCR増幅の直接的な対象であった。正方向の融合プライマーは、可変領域(NNK)12(鋳型特異的)、GS FLXチタニウムアダプターA、及びライブラリーに重要な4塩基の配列(TCAG)にフランキングする配列を含有する:
ライブラリーからのファージ、wtファージ、CFS及び血液から回収したファージ、又は個々のクローンを、ブロス培地(TB培地)(Sigma Aldrich)又は500cm2のプレート(Thermo Scientific)上のいずれかの大腸菌(Escherichia coli)BL5615中で37℃で4時間かけて増殖させた。プレートをトリス−EDTA緩衝溶液(Fluka Analytical)で濯ぐか又は滅菌済みピペットチップを用いてプラークを拾い上げるかのいずれかによって、ファージをプレートから抽出した。ファージを、1回の操作のポリエチレングリコールによる沈降(PEG8000)(Promega)によって培養上清又は抽出緩衝液のいずれかから回収し、トリス−EDTA緩衝溶液に再度懸濁した。
Roche-454生データを、ベンダーソフトウェア(http://454.com/my454/documentation/gs-flx system/emanuals/Part_C/wwhelp/wwhimpl/js/html/wwhelp.htm#href=PartC.1.087.html#9003035)を使用することによって、バイナリ標準フローグラム形式(sff, http://www.ncbi.nlm.nih.gov/Traces/trace.cgi?cmd=show&f=formats&m=doc&s=format#sff)からヒトで解読可能なピアソン形式(fasta, http://emboss.sourceforge.net/docs/themes/SequenceFormats.html)へと変換する。さらなるヌクレオチド配列処理を、以下に記載されているように、社内で開発したC−プログラム及びスクリプト(公表されていないソフトウェアパッケージ)を用いて行なう。
Roche-454に特異的なシークエンスのエラー(例えばホモポリマーストレッチをシークエンスする問題)を克服し、あまり些細ではない重複性(例えばシークエンスエラーに起因する)を除去するために、以前にフィルターにかけたN−mer挿入断片DNA配列(挿入断片)を、以下のように定義された反復アルゴリズムによって類似の挿入断片(2つまでのミスマッチが許容される)に分類する:挿入断片は主に、その頻度(最も高いものから最も低いものまで)、第二に、頻度が等しい場合には、その長さ(最長から最短まで)によって分類される。結果として、最も頻度が高く最も長い挿入断片が第一の「グループ」を規定し、「グループの頻度」は、挿入断片の頻度と等しくなるように設定された。続いて、分類したリストにおける全ての残りの挿入断片を、ペアワイズNeedleman-Wunschアラインメントによってグループに加えることを試みる。アラインメントにおけるミスマッチ、挿入、又は欠失の数が2の閾値を超えない場合、挿入断片をグループに加え、したがって、挿入断片の頻度をグループの頻度の合計に加える。グループに加えられた挿入断片は使用したとしてフラグが付され、さらなる処理から排除される。挿入断片の配列を既存のグループに加えることができない場合、挿入断片を使用して、対応する挿入断片の頻度を有する新しいグループを作り、したがって、そのフラグを付けたものを「使用された」と設定する。全ての挿入断片配列を新たなグループを形成するために使用したか又は既存のグループに含めることができた場合に、反復を終了する。結局、ヌクレオチドから構成されるグループ分けされた挿入断片は、ペプチド配列(ペプチドライブラリー)へと翻訳される。この分析の結果は、「グループ分けされた挿入断片」、及び、1つの挿入断片あたりにシークエンスされた数に相当するその対応する頻度のリストである。
独特なペプチドのリストに基づいて、全ての可能なアミノ酸パターンを含有しているライブラリーを以下のように作成する。長さ3の全ての可能なアミノ酸パターン(例えば「ABC」)をペプチドから抽出し、その逆のパターン(例えば「CBA」)と一緒に、全ての可能なパターンを含有している普遍的なモチーフライブラリーに加える。この高度に重複したモチーフライブラリーを分類し、重複性を除去する。次いで、ライブラリー中の各モチーフを、それがペプチドライブラリーに存在するかどうかを反復試験する。これが事実である場合、モチーフが見られるペプチドの頻度を加え、モチーフライブラリーのモチーフに割り当てる。これらの頻度は「モチーフ数」と呼ばれる。モチーフ作成の結果は、存在している全ての3アミノ酸パターン(モチーフ)及びその対応するモチーフ数を含有している2次元アレイであり、これはシークエンス解読値の数の指標であり、これにより、上記に詳述されているような解読値のフィルター、グループ分け、及び翻訳時に対応するモチーフがもたらされる。

本発明は、CSF、脳、又は関連した脈管構造への組成物の局在化のために使用することができる、1つ以上の標的化ペプチド又は分子標的の同定法を構成する。被験体の液体及び器官をX(N)(Nは7、8、9、10、11、12又はそれ以上の残基であり得る)について、いくつかのペプチドモチーフを生じるランダムファージライブラリーからスクリーニングする。一例では、様々なクローン(トリペプチドモチーフ
を含む)が、CSF及び脳において高い頻度、選択的な結合、及び存在を示した。
合計して924回のシークエンス(短い解読値、1回しか見られなかった解読値、及び配列を全く与えなかったシークエンスを含む)。各ペプチドの頻度は、合計で924回のシークエンス操作に関連している。
CNSにおけるペプチドにより媒介される薬理学的作用を調べるために、本発明者らは、配列番号8を有する脳シャトルペプチドを、ストレプトアビジン(SA)を有するBACE1ペプチド阻害剤(どちらもビオチニル化されている)と、2つの異なる比で混合した実験を行なった。1つの試料に対しては、本発明者らはBACE1ペプチド阻害剤のみを使用し、他の試料に対しては本発明者らはBACE1ペプチド阻害剤と、配列番号8を有する脳シャトルペプチドを1:3の比で使用し、脳送達を促進する、配列番号8を有する大半の脳シャトルペプチドと単一のBACE1ペプチド阻害剤との混合物を有するコンジュゲートしたSA複合体の一部を生成する。2つの試料を静脈内注射し、血液及びCSF中のアミロイド−βペプチド40(Aβ40)のレベルを決定した。予想された通り、両方の試料において、血液中のAβ40レベルは実質的に減少していた。しかし、配列番号8を有する脳シャトルペプチドと、SAにコンジュゲートしたBACE1ペプチド阻害剤との混合物を含む試料のみが、CSFにおける明確なAβ40の減少を誘導した(図7Aの灰色の線を参照)。このデータは、配列番号8を有する脳シャトルペプチドが、大型タンパク質(SA)をCNSに輸送し、SAにコンジュゲートしたBACE1ペプチド阻害剤による薬理作用を誘導することができることを示す。
ストレプトアビジン−ペプチド−BACE1阻害剤複合体の機能性を、製造業者のプロトコール(和光、294−64701)に従ってAβ(1−40)のELISAによって評価した。簡潔に言えば、CSF試料を、標準的な希釈剤(1:23)で希釈し、捕獲抗体BNT77でコーティングされた96穴プレート上で4℃で一晩インキュベートした。5回の洗浄工程の後、HRPにコンジュゲートしたBA27抗体を加え、4℃で2時間インキュベートし、続いて5回の洗浄工程を行なった。Aβ(1−40)を室温で30分かけてのTMB溶液中でのインキュベーションによって検出した。停止溶液を用いて発色を停止した後に450nmでの吸光度を解読した。血漿試料は、Aβ(1−40)ELISAの前に固相抽出を受けた。血漿を96穴プレート中の0.2%DEA(Sigma)に加え、室温で30分間インキュベートした。SPEプレート(Oasis, 186000679)を100%メタノールで、次いで水で洗浄した後、血漿試料をSPEプレートに加え、全ての液体を除去した。試料を洗浄し(5%メタノール、次いで30%メタノール)、2%NH4OH/90%メタノールで溶出した。一定のN2気流下で55℃で99分間溶出液を乾燥させた後、試料を標準的な希釈剤で復元し、Aβ(1−40)を上記に示されているように測定した。
Claims (14)
- 脳エフェクター本体と、
3アミノ酸ペプチドモチーフである、Arg−Leu−Ser(RLS)を含む脳標的化ペプチドとを含み、脳標的化ペプチドは、配列番号8の配列を含む、血液脳関門シャトル。 - 脳エフェクター本体と、リンカーと、3アミノ酸ペプチドモチーフを含む脳標的化ペプチドとを含み、ここで、該リンカーが、エフェクター本体を、請求項1に記載される3アミノ酸ペプチドモチーフを含む脳標的化ペプチドに結合させている、請求項1の血液脳関門シャトル。
- 脳エフェクター本体が、神経障害薬、神経栄養因子、成長因子、酵素、細胞傷害性薬剤、脳標的に対する抗体、脳標的に対するモノクローナル抗体、脳標的に対するペプチドからなる群より選択される、請求項1〜2のいずれか一項記載の血液脳関門シャトル。
- 脳標的が、β−セクレターゼ1、Aβ、上皮成長因子、上皮成長因子レセプター2、タウ、リン酸化タウ、アポリポタンパク質E4、αシヌクレイン、αシヌクレインのオリゴマー性フラグメント、CD20、ハンチンチン、プリオンタンパク質、ロイシンリッチリピートキナーゼ2、パーキン、プレセニリン2、γセクレターゼ、デスレセプター6、アミロイド前駆タンパク質、p75ニューロトロフィンレセプター、及びカスパーゼ6から成る群より選択される、請求項3の血液脳関門シャトル。
- 脳エフェクター本体が、タンパク質、ポリペプチド、及びペプチドからなる群より選択される、請求項1〜4のいずれか一項記載の血液脳関門シャトル。
- 脳エフェクター本体が、脳標的に対する完全長抗体を含む、請求項1〜5のいずれか一項記載の血液脳関門シャトル。
- 脳エフェクター本体が、Aβに対する完全長抗体である、請求項6の血液脳関門シャトル。
- 脳エフェクター本体が、リン酸化タウに対する完全長抗体である、請求項6の血液脳関門シャトル。
- 脳エフェクター本体が、αシヌクレインに対する完全長抗体である、請求項6の血液脳関門シャトル。
- 請求項1〜9のいずれか一項記載の血液脳関門シャトルと薬学的な担体とを含む、医薬製剤。
- 医薬品としての使用のための請求項1〜9のいずれか一項記載の血液脳関門シャトル。
- 医薬品の製造における、請求項1〜9のいずれか一項記載の血液脳関門シャトルの使用。
- 前記医薬品が神経変性疾患の処置用である、請求項12の使用。
- 血液脳関門を横断して脳エフェクター本体を輸送するための、請求項1〜9のいずれか一項記載の血液脳関門シャトル。
