Description
本発明は、式(I)の化合物またはその薬学的に許容される塩、およびJAKの調節におけるそれらの使用に関する。それゆえに、本発明の化合物は、JAK調節に感受性のある疾患および/または障害の治療において有用である可能性がある。そのような疾患および/または障害は、典型的には、とりわけ、アトピー性皮膚炎、乾癬、および例えば以下に記載されるような他の疾患および/または障害を含む。本発明は、さらに、例えば式(I)の新規ジアミノピリジン誘導体を含む医薬組成物、様々な疾患および障害の治療において前記化合物を使用する方法、および前記新規化合物を調製するためのプロセスに関する。

本発明は、式(I)の新規化合物および/またはその薬学的に許容される塩、ならびにJAKの調節におけるそれらの使用に関し、とりわけ、アレルギー性疾患、喘息および慢性閉塞性肺疾患(COPD)のような気道疾患、関節リウマチ、全身性発症若年性特発性関節炎(SOJIA)、痛風、尋常性天疱瘡、特発性血小板減少性紫斑病、全身性紅斑性狼瘡、多発性硬化症、重症筋無力症、シェーグレン症候群、血栓性血小板減少性紫斑病、慢性自己免疫性蕁麻疹、アレルギー(アトピー性皮膚炎、接触性皮膚炎、アレルギー性鼻炎)、アテローム性動脈硬化、1型糖尿病、2型糖尿病、炎症性腸疾患、潰瘍性結腸炎、クローン病、膵炎、糸球体腎炎、グッドパスチャー症候群、橋本甲状腺炎、グレーブス病、抗体関連型移植拒絶反応(AMR)、移植片対宿主病、B細胞関連性の超急性または急性および慢性の移植拒絶反応のような疾患および/または障害の治療をさらに含んでいてもよい。

本発明は、そのもっとも広い実施形態(実施形態1)において、発明の概要の節に上述されたように、式(I)の化合物および/またはその薬学的に許容される塩に関する。

本明細書において使用される際に、用語「C1〜C6アルキル」は、6個までの炭素原子を有する完全飽和された分岐または非分岐の炭化水素部分をいう。別段の定めがない限り、それは、1個から6個の炭素原子、1個から4個の炭素原子、または1個から2個の炭素原子を有する炭化水素部分をいう。アルキルの代表例は、メチル、エチル、n−プロピル、イソプロピル、n−ブチル、sec−ブチル、イソブチル、tert−ブチル、n−ペンチル、イソペンチル、ネオペンチル、n−ヘキシルなどを含むがそれらに限定されない。

本発明の化合物は、典型的には、JAKが役割を果たす障害または疾患の予防または治療において、例えば、アレルギー性疾患、喘息および慢性閉塞性肺疾患(COPD)のような気道疾患、関節リウマチ、全身性発症若年性特発性関節炎(SOJIA)、痛風、尋常性天疱瘡、特発性血小板減少性紫斑病、全身性紅斑性狼瘡、多発性硬化症、重症筋無力症、シェーグレン症候群、血栓性血小板減少性紫斑病、慢性自己免疫性蕁麻疹、アレルギー(アトピー性皮膚炎、接触性皮膚炎、アレルギー性鼻炎)、アテローム性動脈硬化、1型糖尿病、2型糖尿病、炎症性腸疾患、潰瘍性結腸炎、クローン病、膵炎、糸球体腎炎、グッドパスチャー症候群、橋本甲状腺炎、グレーブス病、抗体関連型移植拒絶反応(AMR)、移植片対宿主病、B細胞関連性の超急性および急性または慢性の移植拒絶反応から選択される疾患または障害において、有用である。
上述の使用のために、必要とされる用量は、もちろん、投与のモード、治療されるべき特定の状態、および所望とされる効果によって変わることとなる。概して、満足のいく結果は、体重あたり約0.02〜25mg/kgの1日の用量においてシステム上得られることが示されている。より大きな哺乳類、例えばヒトにおいて、示される1日の用量は、典型的には約0.2mg〜約2gの範囲であってもよく、例えば分割された1回分において1日最高4回または遅らせる形で、便利に投与されてもよい。経口投与のための適当な単位投薬形態は、典型的には、約0.1〜500mgの活性成分を含んでいてもよい。
本発明の化合物は、任意の慣用の経路によって、特には、非経口で、例えば、注射可能な溶液または懸濁液の形態で、経腸的に、例えば、経口的に、例えば、錠剤またはカプセル剤の形態で、局所的に、例えばローション剤、ゲル剤、軟膏剤、またはクリーム剤の形態で、または鼻腔のまたは坐剤の形態で、投与されてもよい。局所投与は、例えば、皮膚に対するものであってもよい。局所投与のさらなる形態は、目に対するものであってもよい。少なくとも1つの薬学的に許容される担体または希釈剤を伴って本発明の化合物を含む医薬組成物は、慣用の方法で、薬学的に許容される担体または希釈剤と混合することによって、製造されてもよい。

(1)医薬品としての使用のため、または医薬としての使用のための、本発明の化合物またはその薬学的に許容される塩、

式(I)の化合物は、単独の活性成分として投与されてもよく、または、例えば、他の薬物に対する補助剤として、例えば免疫抑制剤または免疫修飾剤、または、例えば、同種移植または異種移植急性または慢性拒絶反応もしくは炎症性または自己免疫性障害の治療または予防のための他の抗炎症剤、または化学療法薬、例えば、悪性細胞抗増殖活性剤と併せて投与されてもよい。例えば、式(I)の化合物は、カルシニューリン阻害剤、例えば、シクロスポリンAまたはFK506、もしくはmTOR阻害剤、例えば、ラパマイシンとの組み合わせにおいて使用されてもよい。
